Effects of Supplemental Vegetable Oils on Ruminal Characteristics, Digestibility and Formation of Fatty Acid Soaps in Sheep (식물성유 첨가가 면양의 반추의 발효특성과 소화율 및 지방산염 형성에 미치는 영향)

  • This experiment were conducted to determine the formation of fatty acid soaps, digestibility and ruminal characteristics when vegetable oils were added. Three Corriedale${\times}$ 3 Latin square design. Sheep were fed one of three diets consisting of alfalfa hay and concentrates in a ratio of 70:30. Dietary treatments were 1) no oil, 2) 7% soybean oil and 3) 7% corn oil. At the end of each experimental period, rumen contents from each sheep were collected before feeding and at 1, 3, 6 and 9h after feeding. pH decreased up to the 3 hour after feeding and increased until the end of the collection(p<0.0001). Whereas the concentration of $NH_3$-N showed inverse changes to pH(p<0.0001). Type of oil did not affect the molar concentration of total VFA, acetate, propionate and butyrate. Acetic/propionic acid (A/P) ratio decreased with increasing time(P<0.0001), but the ratio at the end of the experiment became similar to the initial value. No effects were found in the digestibility of dry matter, total-N, crude fiber, ash, NFE, NDF and ADF. The formation of fatty acid soaps(FAS) increased after feeding than 1 hour before feeding(P<0.05) and increased in 7% oils than no oil addition(P<0.05). Long chain fatty acids formed mainly FAS. ince the formation of FAS might decrease negative effects on ruminal haracteristics and total tract digestibility.

Spatio-temporal Variability and Size Fractionation of Chlorophyll a in the Jeju Marine Ranching Area(JMRA) with Special Reference to the Signification of Nanoplankton (제주 바다목장 해역 크기별 엽록소 a의 시·공간적 분포 특성과 미소플랑크톤의 중요성)

  • To understand size fractioned chlorophyll a and material cycle characteristics in Jeju marine ranching area (JMRA), 4 times of survey were conducted from April to November 2008. Picoplankton on the surface in JMRA was on average, $0.30{\mu}g/L$(annual mean(M):17.3%) in the $0.03{\sim}0.84{\mu}g/L$ range, accounting for 17.3%. Nannoplankton and picoplankton was on average, $1.35{\mu}g/L$(M:78.0%) in the $0.22{\sim}3.93{\mu}g/L$ range, and $1.73{\mu}g/L$(M:4.7%) in the nd ~ 0.24 range, respectively. The 10m layer was similar to the surface. The measured values changed according to the measurement times but the nanoplankton composition ratio was higher throughout the year. In addition, the size fractioned chlorophyll a distribution in JMRA was similar to that of tropical sea area affected by the Monsoon rather than South Korean offshore coast geographically adjacent to the East China Sea and Japan coastal waters affected by the Kuroshio/Tsushima warm currents. That is, the material cycle of JMRA consists of a microbial food web rather than traditional food chain at a lower trophic levels. Primary production is deemed to have a higher possibility of being adjusted by top-down dynamics, such as micro-zooplankton grazing pressure rather than nutrients supply.

  • Poly(amino acid) derivatives have been widely investigated as a drug carrier in drug delivery system. Particularly,polysuccinimide (PSI) is one of the most promising drug carriers since it possesses suitable physicochemical characteristics for development of macromolecular prodrugs, due to biocompatibility and biodegradability. In this study, we deal with the synthesis of polyaspartamide having various functional groups such as methoxy-poly(ethylene glycol) (MPEG) via ring closing of PSI. PSI was synthesized by polyonensation polymerization of spartic acid. The variety of average molecular weight was confirmed with reacion time and catalyst content to observe the optimum condition of synthesis. MPEG, hydrophilic chain, was bonded to fabricate polymeric micell composed of hydrophilic and hydrophobic polymer. All materials were characterized by 1H-NMR, FT-IR and GPC. In addition, the formation of nanoparticle micelle as drug carrier were also examined. Micelle size was measured by ELS and AFM. The functionalized polysparamide formed nanoparticle micelle whose size ranged from 90 to 130 nm. In conclusion, we prepared polyaspartamide functionalized with PEG examined the possibility as drug carriers.

Synthesis and Characterization of Polyamide Having Photoreactive Group in the Backbone and its Liquid Crystal Aligning Property (광반응성 작용기를 주사슬에 갖는 폴리아미드의 합성과 특성 분석 및 액정분자의 배향 특성)

  • A soluble polyamide containing photosensitive l,4-phenylenediacrylic acid (PDA) in main chain with biphenyl moiety was synthesized. The chemical structure of synthesized polyamide was investigated by means of $^1H$ NMR spectroscopy. The polymer was stable up to $280^{\circ}C$ and soluble in organic solvents, giving a good quality of thin films. The photoreaction of unpolarized LTV irradiated films was investigated by means of UV-vis absorption spectroscopy and FTlR spectroscopy, and liquid crystals (LCs) alignment property was examined by exposing to linearly polarized UV light (LPUVL) of 260~380 nm. The polyamide in film has excellent photoreactivity to unpolarized UV light. Direction selective photoreaction of PDA moiety in Lhe film was found to further induce nematic liquid -crystals to align along a perpendicular direction with respect to the electric vector of LPUVL, regardless of exposure energy of LPUVL. In addition, pretilt angle was measured by means of crystal rotation method. LPUVL-exposed polymer film induced the alignment of liquid-crystals (LC) with a pretilt angle of 0.2~$0.5^{\circ}$.

Effects of Fucoidan on Nitric Oxide Production and Activator Protein-1 Activation in Lipopolysaccharide-Stimulated Porcine Peripheral Blood Mononuclear Cells (LPS로 자극한 돼지 말초혈액 단핵구세포의 Nitric Oxide (NO) 생산 및 Activator Protein-1 (AP-1) 활성화에 있어 Fucoidan의 효과)

  • Fucoidan which is sulfated polysaccharide extracted from brown seaweed has a wide variety of internal biological activities. The objectives of this study were to examine the effect of fucoidan on nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated porcine peripheral blood mononuclear cells (PBMCs) and to investigate whether this effect is involved in the expression of inducible nitric oxide synthase (iNOS) and the activation of activator portein-1 (AP-1). The levels of NO production and AP-1 activity in the culture supernatants from porcine PBMCs were measured by the enzyme-linked immunosorbent assay and the levels of iNOS and AP-1 mRNA were determined by real time polymerase chain reaction. Fucoidan in LPS-naïve PBMCs has no effects on the production of NO and activity of AP-1. Expressions of iNOS and AP-1 mRNA in LPS-naïve PBMCs were also not affected by treatment of fucoidan. However, NO production, AP-1 activity and expressions of iNOS and AP-1 mRNA were dramatically increased in PBMCs stimulated with LPS. Enhancing effects of NO production and AP-1 activity in PBMCs induced by LPS were reduced by addition of fucoidan. Fucoidan also inhibited an increase in expressions of iNOS and AP-1 mRNA in LPS-stimulated PBMCs. These results suggested that fucoidan exerts anti-inflammatory effect by down-regulating production of NO via suppressing expression of iNOS and activity of AP-1 in LPS-stimulated porcine PBMCs.

Improvement Effect of Non-alcoholic Fatty Liver Disease by Curcuma longa L. Extract (강황 추출물의 비알코올성 지방간 질환 개선 효과)

  • Background: Non-alcoholic fatty liver disease (NAFLD) is a chronic liver disease associated with multiple metabolic disorders. The medicinal plant Curcuma longa L. is widely distributed in Asia and has been used to treat a spectrum diseases in clinical practice. To date, there are inadequate reports of the effects of C. longa 50% EtOH extract (CE) on NAFLD. Therefore, in this study, we evaluate the CE on an NAFLD animal and elucidate the mechanism of action. Methods and Results: C57BL/6J mice fed a methionine-choline deficient diet (MCD) were treated with CE or milk thistle, and changes in inflammation and stetosis were assessed. Experimental animals were divided into six group (n = 10); Normal, MCD, MCD + CE 50 mg/kg/day (CE 50), MCD + CE 100 mg/kg/day (CE 100), MCD + CE 150 mg/kg/day (CE 150), and the Control, MCD + Milk thistle 150 mg/kg/day (MT 150). Body weight, liver weight, liver function, and histological changes were assessed in experimental animals. Quantitative real-time polymerase chain reaction and western blot analyses were performed on samples collected after 4 weeks of treatment. We observed that CE administration improved MCD-diet-induced lipid accumulation, and triglyceride (TG) and total cholesterol (TC) levels in serum. Treatment with CE also decreased hepatic lipogenesis through modulation of the sterol regulatory element binding protein-1 (SREBP-1), CCAAT-enhancer binding protein α (C/EBPα), fatty acid synthase (FAS), and peroxisome proliferator-activated receptor γ (PPARγ) expresion. In addition, the use of CE increased adenosine monophosphate-activated protein kinase (AMPK) phosphorylation and inhibited the up-regulation of toll-like receptor (TLR)-2 and TLR-4 signaling and the production of inflammatory mediators. Conclusions: In this report, we observed that CE regulated lipid accumulation in an MCD dietinduced NAFLD model by decreasing lipogenesis. These data suggeste that CE could effectively protect mice against MCD-induced NAFLD, by inhibiting the TLR-2 and TLR-4 signaling cascades.

Clinical and Laboratory Finding of the 2009 Pandemic influenza A (H1N1) in Children (소아에서 2009 신종 인플루엔자 A (H1N1) 바이러스 감염의 임상적 특징)

  • Purpose : 2009 Pandemic influenza A (H1N1) virus was identified in March 2009 and subsequently caused worldwide outbreaks. We described the clinical and epidemiological characteristics of H1N1 influenza infection. Methods : We used retrospective medical chart reviews to collect data on the visiting patients from a single institute. H1N1 infection was confirmed in specimens with the use of a RT-PCR (real time reverse transcriptase polymerase chain reaction assay). Result : 6,836 patients had H1N1 RT-PCR test, and 2,781 were confirmed with H1N1 virus infection. 158 patients (5.7%) had hospital treatment and inpatients were significantly younger (5.4${\pm}$3.3 years) than outpatients (7.5${\pm}$3.9 years) among H1N1 virus confirmed patients. Oxygen, steroid, immunoglobulin, ventilator treatment was provided in a substantial proportion among pneumonia patients accompanying wheezy respiration. In addition more intensive care was needed in patients accompanying segmental, lobar, interstitial, mixed pneumonia and lung effusion (27.2%) than patients with bronchopneumonia (7.3%) among H1N1 virus infection confirmed patients. Seventy-one infants had oseltamivir treatment out of 83 infants under 1 year, and no significant side effects and complications were identified. Conclusion : In 2009 pandemic influenza A (H1N1), hospital treatment was needed in younger patients. Early intensive care was needed in pneumonia patients accompanying wheezy respiration, and patients accompanying segmental, lobar, interstitial, mixed pneumonia and lung effusion.

Effect of Dietary Supplementation of DHA and AA on the Incorporation of Long Chain Fatty Acid and DNA Content in the Developing Brain of Rats (쥐에서 뇌발달 과정중에 식이에 첨가한 DHA와 AA가 뇌조직의 지방산조성과 DNA 함량에 미치는 영향)

  • The aim of this study was to observe whether the dietary supplementation of docosahexaenoic acid(DHA). In growing rats requires extra supplementation of arachidonic acid(AA) for brain development. Sprague-Dawley rats were divided into three groups, each fed a different diet. In the FO group, dams were fed a DHA-rich FO diet during pregnancy and lactation and pups were fed the same diet until 10 weeks old. In the AO group dams and pups were similarly fed a FO diet after weaning. DHA and AA were most effetively deposited in the developing brain during pregnancy and lactation in rats. However, FO-W pups showed significantly lower level of DHA at 0-3 weeks compared with the FO and AO groups and than slowly increased DHA levels to about 87% of other groups at 10 weeks with the introduction of the FO diet after weaning. The total amount of DNA in whole brain rapidly reached a maximum level at 3 weeks and then was sustained at a constant level after 5 weeks of age. The DNA content was positively correlated with DHA level but not with AA level in the developing brain. DNA content was significantly lower in the FO-W group compared to the FO and AO group at 3 weeks of age. However, the DNA content of brain in FO-W pups increased to 80% of the FO group level at 10 weeks after feeding the FO diet after weaning. The relative percentage of AA in brain lipids was significantly reduced in the early stage of brain development when only DHA was supplemented. However, DHA supplementation had no significant effect on the incorporation of AA when the approximately 35% of LA in the FO diet was substituted by preformed AA. These results suggest that large quantities of DHA could interfere with the normal conversion of LA to AA if LA is not supplemented enough together with DHA. Therefore, high DHA supplementation may require preformed AA in the diet even though AA has no significant correlation with the DNA content in brain. DHA supplementation after weaning also improved the incorporation of DHA into brain and content of DNA even though brain development was almost completed, suggesting that a low level of DHA supplementation without AA addition might be necessary to improve brain development during infancy as well as during pregnancy and lactation.

Effects of 17β-Estradiol on Colonic Permeability and Inflammation in an Azoxymethane/Dextran Sulfate Sodium-Induced Colitis Mouse Model

  • Background/Aims: Intestinal barrier dysfunction is a hallmark of inflammatory bowel diseases (IBDs) such as ulcerative colitis. This dysfunction is caused by increased permeability and the loss of tight junctions in intestinal epithelial cells. The aim of this study was to investigate whether estradiol treatment reduces colonic permeability, tight junction disruption, and inflammation in an azoxymethane (AOM)/dextran sodium sulfate (DSS) colon cancer mouse model. Methods: The effects of $17{\beta}$-estradiol (E2) were evaluated in ICR male mice 4 weeks after AOM/DSS treatment. Histological damage was scored by hematoxylin and eosin staining and the levels of the colonic mucosal cytokine myeloperoxidase (MPO) were assessed by enzyme-linked immunosorbent assay (ELISA). To evaluate the effects of E2 on intestinal permeability, tight junctions, and inflammation, we performed quantitative real-time polymerase chain reaction and Western blot analysis. Furthermore, the expression levels of mucin 2 (MUC2) and mucin 4 (MUC4) were measured as target genes for intestinal permeability, whereas zonula occludens 1 (ZO-1), occludin (OCLN), and claudin 4 (CLDN4) served as target genes for the tight junctions. Results: The colitis-mediated induced damage score and MPO activity were reduced by E2 treatment (p<0.05). In addition, the mRNA expression levels of intestinal barrier-related molecules (i.e., MUC2, ZO-1, OCLN, and CLDN4) were decreased by AOM/DSS-treatment; furthermore, this inhibition was rescued by E2 supplementation. The mRNA and protein expression of inflammation-related genes (i.e., KLF4, NF-${\kappa}B$, iNOS, and COX-2) was increased by AOM/DSS-treatment and ameliorated by E2. Conclusions: E2 acts through the estrogen receptor ${\beta}$ signaling pathway to elicit anti-inflammatory effects on intestinal barrier by inducing the expression of MUC2 and tight junction molecules and inhibiting pro-inflammatory cytokines.

CDC6 mRNA Expression Is Associated with the Aggressiveness of Prostate Cancer

  • Background: Cell division cycle 6 (CDC6) is an essential regulator of DNA replication and plays important roles in the activation and maintenance of the checkpoint mechanisms in the cell cycle. CDC6 has been associated with oncogenic activities in human cancers; however, the clinical significance of CDC6 in prostate cancer (PCa) remains unclear. Therefore, we investigated whether the CDC6 mRNA expression level is a diagnostic and prognostic marker in PCa. Methods: The study subjects included 121 PCa patients and 66 age-matched benign prostatic hyperplasia (BPH) patients. CDC6 expression was evaluated using real-time polymerase chain reaction and immunohistochemical (IH) staining, and then compared according to the clinicopathological characteristics of PCa. Results: CDC6 mRNA expression was significantly higher in PCa tissues than in BPH control tissues (P = 0.005). In addition, CDC6 expression was significantly higher in patients with elevated prostate-specific antigen (PSA) levels (> 20 ng/mL), a high Gleason score, and advanced stage than in those with low PSA levels, a low Gleason score, and earlier stage, respectively. Multivariate logistic regression analysis showed that high expression of CDC6 was significantly associated with advanced stage (${\geq}T3b$) (odds ratio [OR], 3.005; confidence interval [CI], 1.212-7.450; P = 0.018) and metastasis (OR, 4.192; CI, 1.079-16.286; P = 0.038). Intense IH staining for CDC6 was significantly associated with a high Gleason score and advanced tumor stage including lymph node metastasis stage (linear-by-linear association, P = 0.044 and P = 0.003, respectively). Conclusion: CDC6 expression is associated with aggressive clinicopathological characteristics in PCa. CDC6 may be a potential diagnostic and prognostic marker in PCa patients.
