치자(梔子) 약침(藥鍼)이 백서(白鼠) 모델 족과 염좌(捻挫) 통증(痛症)에 미치는 영향(影響) (Effect of Frutus gardeniae herbal acupuncture on the rat model of ankle sprain pain)

  • Objective : Frutus gardeniae, seed of Gardenia jasminoides Ellis is one of the crude drugs used for the treatment of inflammatory condition in oriental medicine. Methodes : The present study aimed to examine the analgesic effect and anti-inflammatory effect of Frutus gardeniae extract (FGE) on a rat model of ankle sprain pain, and the relations between FGE-induced effect and endogenous nitric oxide (NO) and inducible NO synthase (iNOS), cyclooxygenase-2 (COX-2), and c-Fos protein expression in the spinal cord. As a chronic pain model, ankle sprain pain model was used to test the effect of FCE injection applied to acupuncture point. After the induction of ankle sprain, rats subsequently showed a reduced stepping force of the affected limb for at least the next 4 days. The reduced stepping force of the limb was presumably due to a painful knee. FGE dissolved in normal saline was injected several acupoints. Results : After the treatment, behavioral tests measuring stepping force were periodically conducted during the next 8 hours. FGE produced significant improvement of stepping force of the hindlimb affected by the ankle sprain lasting at least 4 hours. FGE produced the improvement of stepping force of the affected hindlimb in a dose-dependent manner. In addition, FGE injection showed inhibitory effect on the paw edema induced by ankle sprain. Both NO production and iNOS, COX-2 protein expression increased by ankle sprain were suppressed by FGE. FGE on combination with electroacupuncture (EA) produced more powerful and longer lasting improvement of stepping force of the hindlimb affected by the ankle sprain than either FGE or EA did. The present study suggest that FGE produces a potent analgesic effect on the ankle sprain pain model of the rat and that FGE-induced analgesia modulate endogenous NO through the suppression of iNOS/COX-2 protein expression.

Effect of the Electroacupuncture at ST36 in TMT-induced Memory Deficit Rats

  • In order to the neuroprotective effect of electroacupuncture (EA), the present study examined the effects of electroacupuncture inacupoint ST36 (Stomach 36) on trimethyltin chloride (TMT)-induced cognitive impairments rat using the Morris water maze (MWM) task and immunohistochemistry staining. The rats were randomly divided into the following groups: naive rat (Normal), TMT injection rat (Control), TMT injection + EA treated rat inacupoint ST36 (ST36) and TMT injection + EA treated rat in non-acupoint, base of tail (Non-AC). Electroacupuncture (2Hz, 2mA, and 10 minutes)was applied either to the acupuncture point ST36 or the nonacupuncture point in the tail for the last 14 days. In the water maze test, the animals were trained to find a platform in a fixed position during 4d and then received 60s probe trial on the $5^{th}$ day following removal of platform from the pool. Rats with TMT injection showed impaired learning and memory of the tasks and treatment with EA in acupoint ST36 (P<0.05) produced a significant improvement in escape latency to find the platform after $2^{nd}$ day and retention trial in the Morris water maze. Consistent with behavioral data, treatment with EA in acupoint ST36 also significantly increased expression of Choline acetyltransferase (ChAT) and Acetylcholinesterase (AChE) immunoreactive neurons in the hippocampus compared to the Control group. These results demonstrated that EA in acupoint ST36 has a protective effect against TMT-induced neuronal and cognitive impairments. The present study suggests that EA in acupoint ST36 might be useful in the treatment of TMT-induced learning and memory deficit.

Phamacopuncture and Dermal Application of Sebalgukhwa-san: Effects on Hair Growth in a Mouse Model of Alopecia

  • Background: This study was conducted to evaluate the effects of pharmacopuncture and dermal application of Sebalgukhwa-san extracts on hair growth in an alopecia mouse model. Methods: Twenty-one C57BL/6 mice were divided into 3 groups; control group-normal saline injection or vehicle solution application, positive control group-minoxidil (MNXD), experimental group-pharmacopuncture and applied Sebalgukhwa-san (SGS) extract. The effects of the treatment on hair growth, were determined through photographs, and phototrichogram analysis by folliscope. Hair follicle morphometry by hematoxylin-eosin staining was performed, and hair growth-related protein expression of vascular endothelial growth factor, insulin like growth factor-1, and transforming growth factor-beta 1 were monitored by Western blotting. Serum levels of aspartate aminotransferase and alanine aminotransferase were measured for liver function test. Results: Body weight increased consistently in all groups. Hair growth was improved in the MNXD and SGS groups compared with the control. Hair density and thickness improved statistically significantly in the MNXD and SGS groups compared with the control p < 0.05. The number of hair follicles improved in the MNXD and SGS groups compared with the control but the size did not. The expression of vascular endothelial growth factor and insulin like growth factor-1 increased, and there was a decrease in the expression of transforming growth factor-beta 1 in the MNXD and SGS groups compared with the control, however, there was no significant difference. Sebalgukhwa-san treatment had no toxicity in liver function tests. Conclusion: Pharmacopuncture and dermal application of Sebalgukhwa-san extract may be therapeutically beneficial for the treatment of alopecia.

금연침에 대한 반응군과 비반응군의 유전자 다형성 차이 연구 (The association of genetic polymorphism between responder and nonresponder to acupuncture in smoking cessation)

  • Objectives : This study was designed to investigate the differences of effects in smoking cessation after acupuncture treatment and we hypothesized that the discrepancies might be caused by individual genetic differences. Methods : Acupuncture treatment was given to the subjects three times a week for the 231 healthy male Korean smokers without personal or familial history of psychiatric or neurological illness. We evaluated for differentiate responder and non-responder who showed more than 50% decrease in the cigarette consumption or the desire for smoking were regarded as responder, and less than 25% decrease in the cigarette consumption or the desire for smoking were regarded as non-responder, respectively. Allele and genotype frequencies of the Taq1 A polymorphism of dopamine D2 receptor (DRD2) gene were compared in 231male smokers. Chai-square analyses were performed to test for an interactive effect between the DRD2 Taq1 A allele. Results : The allele frequencies and genotype distributions of DRD2 gene among the smokers (n = 231) showed significant the differences in their genotype distributions. The responder and non-responder showed the difference in genotype distribution with a prevalence of A1 allele. A slightly positive association of DRD2 Taq1 A1 genotypes with smoking was observed. Conclusions : This experiment results indicate that the present of DRD2 allele genotype showing significant difference in the genotype distributions between responders and non-responders could be explained by the difference in the genetic effect of DRD2 A1 allele.

Results : Moxi-tar produced significant improvement of stepping force of the hindlimb affected by the arthritis lasting at least 9 hours. The magnitude of this improvement was equivalent to that obtained after an oral injection of 3 mg/kg of indomethacin and this improvement of stepping force was interpreted as an analgesic effect. Moxi-tar produced the improvement of stepping force of the affected hindlimb in a dose-dependent manner. Both NO production and iNOS, COX-2 protein expression increased by arthritis were suppressed by moxi-tar. moxi-tar on combination with electroacupuncture (EA) produced more powerful and longer lasting improvement of stepping force of the hindlimb affected by the arthritis than either moxi-tar or EA did. Conclusion : The present study suggest that moxi-tar produces a potent analgesic effect on the chronic knee arthritis pain model in the rat and that moxi-tar-induced analgesia modulate endogenous NO through the suppression of iNOS/COX-2 protein expression.

  • Objectives: This study was produced to examine the effects of moxibustion that had been played important role to traditional oriental medical treatment on disease. Recently, it was reported that moxi-tar which is generated in the process of moxibustion as burning combustibles decreased nitric oxide(NO) and inducible NO synthase (iNOS) generation in cellular experiments. Methods: Carrageenan-induced arthritis rat model was used to test the effect of moxi-tar as a chronic pain model. Diluted moxi-tar was single injected in several acupoints or combined with electroacupuncture (l ms, 2 Hz, and 2 mA) into contralateral ST36 acupoint for 30 min to assess the synergic effects. After the treatment, behavioral tests measuring stepping force were periodically conducted during the next 12 hours. Endogenous NO and iNOS, cyclooxygenase-2 (COX-2), and c-Fos protein expression in the spinal cord were examined on a rat model of carrageenan-induced arthritis. 경혈 및 경락 안마자극 프로그램이 적용된 안마의자 사용이 수면의 질에 미치는 영향 (The Effect of Using Massage Chair with Acupoint and Meridian Massage Program on Sleep Disorder)

  • Objectives: The purpose of this study was to investigate the effect of the massage program stimulating the acupuncture points and meridians on the improvement of sleep quality. Methods: 16 subjects suffering from sleep disorders were recruited and the massage program was conducted 3 times a week for a month. VAS (Visual Analog Scale) and KPSQI (Korean Pittsburgh Sleep Quality Index) were collected through questionnaires, and salivary cortisol concentration test and HRV (Heart Rate Variability) were also used to evaluate the effect. The collected data were analyzed by independent statisticians. Results: VAS and KPSQI values decreased significantly after 4 weeks of massage chair application. There was no significant difference in salivary cortisol concentration and HRV test results. In the group with normal HRV test results and salivary cortisol concentration, the change of VAS and KPSQI values was not significant, while the group with abnormal range showed a statistically significant decrease. Conclusions: The massage chair program stimulating meridians according to Korean medicine theory was effective in improving sleep quality. Massage of acupoints and meridians through a massage chair is expected to be useful as an adjunctive therapy in addition to standard Korean medicine therapy.

레이저침에 활용되는 830 nm, 905 nm 레이저의 안전성 연구 (A Study on Safety of 830 nm, 905 nm Laser Used for Laser Acupuncture Therapy)

  • Objectives : As the attempts to make good uses of laser acupuncture in korean medicine have been steadily carried on, to ensure the safety is needed to increase the use of laser acupuncture. The aim of this study was to examine the safety of using laser acupuncture therapy irradiated with 830 nm laser and 905 nm laser. Methods : Fifteen rats were randomly divided into three experimental groups - Intact ; no therapy group. LAT830 ; laser acupuncture therapy(830 nm) group at GB34 GB39. LAT905 ; laser acupuncture therapy(905 nm) group at GB34 GB39. During eight weeks of treatment, LAT830, LAT905 were treated 2 times weekly, frequency 20 Hz, intensity 20 mW. Test items were mortality, body weight, food consumptions, hematological values, blood chemical values, urinalysis values, absolute organ weights and relative organ weights. Results : Statistically significant differences were not found among 3 groups. The values measured in LAT830, LAT905 and Intact(no therapy) group have shown similar results and it indicated that LAT830, LAT905 had no abnormal influence on the reaction of living bodies. There was little difference on the liver weight of relative organ weight test between laser-treated groups(LAT830, LAT905) and Intact group. Conclusions : In conclusion, laser acupuncture therapies irradiated with 830 nm laser and 905 nm laser caused no abnormal reaction of living bodies. More studies are needed to further establish the safety of many other laser acupuncture therapies.

고의서에 나타난 경혈과 병증의 연관성 측정 및 시각화 - 침구자생경 분석 예를 중심으로 - (Measure of the Associations of Accupoints and Pathologies Documented in the Classical Acupuncture Literature)

  • Objectives : This study aims to analyze the co-occurrence of pathological symptoms and corresponding acupoints as documented by the comprehensive acupuncture and moxibustion records in the classical texts of Far East traditional medicine as an aid to a more efficient understanding of the tacit treatment principles of ancient physicians. Methods : The Classic of Nourishing Life with Acupuncture and Moxibustion(Zhenjiu Zisheng Jing; hereinafter ZZJ) was selected as the primary reference book for the analysis. The pathology-acupoint co-occurrence analysis was performed by applying 4 values of vector space measures(weighted Euclidean distance, Euclidean distance, $Cram\acute{e}r^{\prime}s$ V and Canberra distance), which measure the distance between the observed and expected co-occurrence counts, and 3 values of probabilistic measures(association strength, Fisher's exact test and Jaccard similarity), which measure the probability of observed co-occurrences. Results : The treatment records contained in ZZJ were preprocessed, which yielded 4162 pathology-acupoint sets. Co-occurrence was performed applying 7 different analysis variables, followed by a prediction simulation. The prediction simulation results revealed the Weighted Euclidean distance had the highest prediction rate with 24.32%, followed by Canberra distance(23.14%) and association strength(21.29%). Conclusions : The weighted Euclidean distance among the vector space measures and the association strength among the probabilistic measures were verified to be the most efficient analysis methods in analyzing the correlation between acupoints and pathologies found in the classical medical texts.

지각 약침액의 항산화 및 항염증 효과에 관한 연구 (Antioxidative and Anti-inflammatory Effects of Aurantii Fructus Immaturus Pharmacopuncture)

  • Objectives : Ulcerative colitis is a chronic relapsing inflammatory disease in the gastrointestinal tract. We investigated whether Aurantii fructus immaturus (AFI) pharmacopuncture has antioxidative and anti-inflammatory effects. Methods : in vitro experiments, 1,1-diphenyl-2-picryl hydrazyl (DPPH) free radical scavenging activity, superoxide dismutase (SOD) activity, prevention on $H_2O_2$-induced cell death in RAW264.7 cell line, DNA fragmentation, and cyclooxygenase-2 mRNA expression induced by lipopolysaccharide (LPS), were analyzed to investigate antioxidative and anti-inflammatory effect of AFI pharmacopuncture. in vivo experiment, a murine model of dextran sulfate sodium (DSS)-induced colitis was used to examine the effect of AFI pharmacopuncture on CV12 at different doses of 5 ${\mu}l$, 0.5 ${\mu}l$, 0.05 ${\mu}l$ for 10 days. Body weight, colon length and macroscopic features were investigated. Results : AFI pharmacopuncture showed DPPH free radical scavenging and SOD active effects in a dose-dependent manner. AFI pharmacopuncture showed a protective effect against $H_2O_2$-induced cell injury and also attenuated LPS-induced COX-2 mRNA expression. In a DSS- induced colitis murine model, however, AFI pharmacopuncture at CV12 had no anti-inflammatory effects. Conclusions : The present results suggest that AFI pharmacopuncture extract may have anti- inflammatory and antioxidative effects in vivo test, but further research on the underlying mechanism is required.
