Changes of RuBisCO Content and Protease Activity during the Life Span of Tobacco Leaf (담배잎의 일생에 있어서 RuBisCO 함량과 Protease활성의 변동)

  • Changes in the amount of ribulose 1, 5-bisphosphate carboxylase/oygenase(=RuBisCO) protein, namely fraction I protein, and the protease activity were determined in the 10th leaf of tobacco(Nicotiana tabacum, var. Ky-57) from 10 days after emergence through senescence at 5 days interval. The amount of RuBisCO per deveined leaf rapidly increased during the early growing season, reached a maximal quantity at the around 20 days after leaf emergence, when the leaf has gone through its most rapid expansion, and began gradually to decrease till 30 days after leaf emergence, thereafter significantly declined to 45 days that the leaf has been dried up partly. The pattern of the ratio of RuBisCO protein to soluble protein in quantity changed similar to that of RuBisCO contents in a leaf, that was 43%, 60%, and 21% at the around 10 days, 20 days, and 45 days, respectively. And RuBisCO contents was linearly correlated with the concentration of chlorophyll(r=0.98) throughout the life span of the leaf. So, it was assumed that the leaf color can be a useful indicator for judging whether RuBisCO contents higher or not in tobacco leaves without homogenization. On the other hand, the protease activities for degradation of casein were assayed at pH 5.5. 7.0. and 8.5 with crude extracts desalted on Sephadex G-25. The highest caseolytic activity was found at pH 5.5 throughout the life sawn of the leaf. Also, the activity at 5.5 became gradually to increase from 30 days after leaf emergence, when RuBisCO protein had became to disappear and remarkably increased in the last stage of senescence, although nitrogen contents of the leaf had reached low levels. The caseolytic activity at pH 5.5 was in negative correlation with RuBisCO contents throughout the life span of the leaf, but not in lineality between them. In other words, the caseolytic activity increased in a rapid exponential manner when RuBisCO contents had reached some low levels. These results showed that the leaf age, namely harvesting time, is a very important factor for the production of the tobacco leaf containing higher RuBisCO protein. It was concluded that the practical harvesting time is between 20 days and 30 days after the leaf emergence from the present results.

  • Telomeres consist of TTAGGG tandem repeated DNA sequences with specific proteins and locate at chromosome ends. Telomeres are essential for chromosome stability and are related with cell senescence, apoptosis and cancer. Telomerase is a ribonucleoprotein which has a template for the synthesis of telomeric DNA. This study was carried out to analyze the amount of telomeric DNA and telomerase activity in cattle cells. Analysis of the quantity of telomere in lymphocytes was done at different ages, sex and among Korean cattle and Holstein breeds. The telomerase activity was also analyzed in liver, brain, heart, kidney, and testis tissues of fetal calf and of 18 month old cattle. The amount of telomeres in lymphocytes and other tissue cells was analyzed by Quantitative-Fluorescence in situ Hybridization (Q-FISH) technique using a telomeric DNA probe. Telomerase activity was analyzed by Telomeric Repeat Amplification Protocol assay (TRAP). The amount of telomeric DNA on the lymphocytes during the whole life span was decreased along with age. Quantity of telomeres in Korean cattle was significantly higher than that in Holstein breed. The amount of telomeric DNA in males was significantly higher than that in females. Telomerase activity was up-regulated in most bovine tissues during fetal stage, but was down-regulated in most tissues at mature 18 month age except the testis cells. This study indicates that the amount of telomeres and telomerase activity of cells can be used as an age marker or/and a physiological marker of cattle.

Changes in enzyme activity and expression of DHFR of Toxoplumc gondii by antifolates (Antifolate 약제에 대한 톡소포자충의 DHFR 효소활성 및 유전자 발현의 변화)

  • The responses to antifolales of ToxopLasmc Bondii were investigated by measuring the dihydrorolate redLlctase (DHFR) activity. quantity of DHFR mRNA, and single-strand conformational polymorphism (SSCP) pattern. Pyrimethamine (PYM) and methotrexate (MTX) were tested ds anlifolates. When T. gondii was treated wish PYM, the viability was decreased by the increasing concentration of PYM. DHFR activity tended to increase as the passage proceeded. and the quantity of mRNA expressed was also increased according to passages. The viability of 7. gonnii was decreased by the increasing concentration of MTX, but it was maintained over 40% up to $100{\;}{\mu\textrm{m}}$ MTX. DHFR activity was 77.4% in the 1st passage ($1{\;}{\mu\textrm{m}}$). 82.2% in the 4th passage ($10{\;}{\mu\textrm{m}}$), and 141.3% in the 7th passage ($100{\;}{\mu\textrm{m}}$) But no changes were detected in SSCP pattern of T gondii rxposvd to FYM and MTX. both. These results suggested that the response of T gondii to FYN was rofulalcd by transrriptional level and that, in MTX. the viability of T. gonnii was derived from increasing DHFK activity.

  • The results of the investigation on the effects of plant nutrio-physiological status and reemergence of sudden wilting in rice on the occurrence of sudden wilting were summarized as follows: 1. The occurrence of sudden wilting, degree of damage and occurring time in the investigated variety showed to concur with the results of the previous experiment. 2. Nitrogen and Potassium contents in leaves related an occurrence of sudden wilting at heading stage have a very high negative co-relation (the value is -0.9224 at Yushin and -0.8243 at Milyang 23). 3. Regarding the relation between occurrence of sudden wilting and root activity capacity at the late growth stage, the root activity capacity at productive growth stage showed higher than at reproductive growth stage, which is the same in the two varieties but Milyang 23 is higher than Yushin. It is confirmed that root activity capacity in the two varieties has become very low at reproductive growth stage. 4. The toot activity capacity in the field with potassium showed a small occurrence of sudden wilting and the root activity capacity in the field with no potassium showed a very high co-relation value (the value is 0.8947 at Yushin and 0.7301 at Milyang 23). 5. Regarding the relation between the occurrence of sudden wilting and water evaporation quantity, water evaporation quantity after the heading date has a more direct relation to sudden wilting than that which occurrs prior to the heading date. Water evaporation quantity is higher in the field with no potassium, low leaves and Yushin, than in the field with potassium high leaves and Milyang 23. 6. The starch content in culm of Yushin and Milyang 23 at ripening stage showed high value in proportion to potassium quantity, and the starch content is lower as internode is lower. However, in fourth internode, the starch content is greatly increased in the field with potassium contrasted to the field with no potassium.

  • Bioconversion, the enzymatic process by microorganism on organic precursor to desired products. The natural extract is converted into a form that can be easily absorbed into the skin, while scaling up of to higher quantity is possible. Selection of naturally processed raw material rather than chemically processed is preferred. Therefore, fermentation was carried out by mixing Rubus coreanus Miquel, soybean, Zanthoxylum schinifolium as bioconverting materials, the possibility of inflammation, whitening material were checked. In this study, useful microorganisms were isolated from various salted fish, and 16S rDNA sequence was analyzed to confirm their genetic characteristics. Combining the three natural materials using bioconversion technology to study their activity before and after fermentation. To evaluate the antioxidant activity and the active ingredient content the DPPH radical scavenging activity and the total polyphenol content were examined. Raw 264.7 cells were used to evaluate MTT assay, NO and $TNF-{\alpha}$ production inhibitory activity. Also, to evaluate the whitening activity, tyrosinase inhibitory activity and melanin formation inhibitory activity were measured using B16F10 cells. In total 34 strains were obtained from various salted fish. The effective strains useful for the bioconversion process, showed that DPPH radical scavenging ability and polyphenol content were increased in the two kinds of microbial treatment groups compared to the untreated group. 16S rDNA sequencing analysis of the strains showed excellent in Pediococcus pentosaceus B1 in comparison. An increase of up-to 156% in anti-oxidative activity and 141% in polyphenol content was observed after bioconversion. In addition, after mixed fermentation the toxidty of Raw 264.7 and B16F10 cells tended to decrease and a significant increase was observed in anti-inflammatory activity as well. Also, tyrosinase activity and melanin significantly. synthesis decreased significantly.

Metal Ions' Effect on Activity of Pine Needle Tyrosinase (금속이온이 솔잎 tyrosinase의 활성에 미치는 영향)

  • The purpose of this study is to explain the relations between pine needle tyrosinase's activity and quantity of minerals in the Waters' mineral water. Pine needle tyrosinase's activity was measured by metal ions' concentration like Ca, Mg, Na, K, and Fe in different kinds of drinking water. 1. Pine needle tyrosinase has the highest activity when Ca's concentration is 14.40mg/L while the activity decreases by 92% when it is 108.10 mg/L. Therefore, the resonable range of Ca concentration for drinking water is 10-100.0 mg/L. 2. Mg has higher Pine needle tyrosinase's activity than Ca by three times. The reasonable range of Mg concentration for drinking water is 3.0-10.0 mg/L. 3. Pine needle tyrosinase has the highest activity when Na's concentration is 15.70 mg/L. The reasonable range of Na concentration for drinking water is less than 15 mg/L. 4. The activity increases as K concentration rises. In normal kinds of drinking water, K concentration is less than 10 mg/L. Since K has impacts on the activity only when its concentration is more than 10 mg/L, no problem in expected. 5. Fe has some impacts on the activity when its concentration is more than 10mg/L. As most kinds of drinking water contain less than 0.3 mg/L, no problem is expected. With above-mentioned observations, it is concluded that Water's mineral water contains reasonable levels of minerals like Ca, Mg, K and Na.

SERCA2a: a prime target for modulation of cardiac contractility during heart failure

  • Heart failure is one of the leading causes of sudden death in developed countries. While current therapies are mostly aimed at mitigating associated symptoms, novel therapies targeting the subcellular mechanisms underlying heart failure are emerging. Failing hearts are characterized by reduced contractile properties caused by impaired $Ca^{2+}$ cycling between the sarcoplasm and sarcoplasmic reticulum (SR). Sarcoplasmic/endoplasmic reticulum $Ca^{2+}$ ATPase 2a (SERCA2a) mediates $Ca^{2+}$ reuptake into the SR in cardiomyocytes. Of note, the expression level and/or activity of SERCA2a, translating to the quantity of SR $Ca^{2+}$ uptake, are significantly reduced in failing hearts. Normalization of the SERCA2a expression level by gene delivery has been shown to restore hampered cardiac functions and ameliorate associated symptoms in pre-clinical as well as clinical studies. SERCA2a activity can be regulated at multiple levels of a signaling cascade comprised of phospholamban, protein phosphatase 1, inhibitor-1, and $PKC{\alpha}$. SERCA2 activity is also regulated by post-translational modifications including SUMOylation and acetylation. In this review, we will highlight the molecular mechanisms underlying the regulation of SERCA2a activity and the potential therapeutic modalities for the treatment of heart failure.

Effect of Fed Chlorella on the Change of Lipid Components and Enzyme Activity in Serum of Rat by Lead Exposure (납에 노출된 흰쥐에서 Chlorella 섭취가 혈청내 지방성분 및 효소활성에 미치는 영향)

  • This study was performed to investigate the influence of Chlorella which effected the change of Pb contents, enzyme activity and lipid compounds on the rats fed the beverage involved Pb and the different contented Chlorella added-diets for 18weeks. The rat's weight of 200ppm Pb group was decreased 6.04% and the cause of that was Pb intake. But the rat's weight of Chlorella added-diets +200ppm Pb group was increased 4.02% (p<0.01). When feeding the different(0%, 2%, 5%, 10% ) Chlorella added-diets with the Pb contented beverage to the rats, we could know that the Pb contents accumulated on tissue were decreased to 20.70(0%), 12.88(2%), 14.83 (5%) and 19.56(10%), compared with the quantity of Pb taken in. Total-cholesterol, triglyceride, glucose content and AST, ALT, ALP, LDH enzyme activity in serum were the highest on 200ppm Pb group and those were decreased by the order of different(2%∼5% >10%) Chlorella contents + 200ppmPb group. A significance was recognized on the level of 1%. Therefore, when rats were exposed to Pb, it was thought that the amount of Chlorella intake was adequate on content 2∼5% for reducing the lead toxicity.

Influence of DBD Plasma Exposure on Normal and Cancer Cells Activity

  • Non-thermal plasma has attracted medical researchers, since they showed higher apoptosis rate in cancer cells than normal cells. However, it is hard to conclude general cancer cell specific effect because comparison between normal and cancer cell activities after plasma treatment have not been reported yet. This research proposes a comparison of Dielectric Barrier Discharge (DBD) plasma effect on three normal cells lines and three cancer cells lines. We measured cell number, mitochondria activity (MTS assay) and amount of hydrogen peroxide (H2O2) for three days. The results show that the number of cancer cells decreased more than normal cells following of exposure time. On the other hand, mitochondria activities and amounts of H2O2 increased following of exposure time. In addition, we found that DBD plasma exposure on cell suspension in media and media only illustrated no difference in mitochondria activity, H2O2 quantity, and cell number. Thus, we can confirm higher apoptosis rate in cancer cells which is related to the reactive oxygen species (ROS) generated by DBD plasma. The related molecular mechanisms were investigated further.
