A-factor I a microbial hormone that can positively control cell differentiation leading to spore formation and secondary metabolite formation in Streptomyces griseus. to identify a protease that is deeply involved in the morphological and physiological differentiation of Streptomyces, the proteases produced by Streptomyces griseus IFO 13350 and its A-factor deficient mutant strain, Streptomyces griseus HH1, as well as Streptomyces griseus HH1 transformed with the afsA gene were sturdied. In general Streptomyces griseus showed a higher degree of cell growth and protease activity in proportion to its ability to produce a higher amount of A-factor. In particular, the specific activity of the trypsin of Streptomyces griseus IFO 13350 was greatly enhanced more than twice compared with that of Streptomyces griseus HH1 in the later stage of growth. The specific activity of the metalloprotease of Streptomyces griseus HH1 was greatly enhanced more than twice compared with that of Streptomyces griseus IFO 13350, and this observation was reversed in the presence of thiostreptione, However, Streptomyces griseus HH1 transformed with the afsA gene showed a significantly decreased level of trypsin and metalloprotease activity compared with that of the HH1 strain. There was no significant difference between Streptomyces griseus IFO 13350 and HH1 strain in their chymotrypsin and thiol protease activity, yet the level of leu-amionpeptidase activity was 2 times higher in Streptomyces griseus HH1 than in strain IFO 13350 . Streptomyces griseus HH1 harboring afsA showed a similar level of enzyme activity , however, all the three protease activities sharply increased and the thiol protease activity was critically increased at the end of the fermentation. When a serine protease inhibitor, pefabloc SC, and metalloprotease inhibitor, EDTA, were applied to strain IFO 13350 to examine the in vivo effects of the protease inhibitors on the morpholofical differentiation, the formation of aerial meycelium and spores was delayed by two or three days.
Cadmium is a strong toxic heavy metal which presents in paints and liquid wastes and causes oxidative stress in fish. On the other hand, lead is widely used for different purposes, e.g. lead pipes, it targets vital organs such as liver and kidney causing biochemical alterations. The present study evaluates the effects of 60 days exposure to Cd and Pb either single or combined together in African catfish. Sixty-four fishes were divided into 3 groups and exposed to $CdCl_2$ (7.02 mg/L) or $PbCl_2$ (69.3 mg/L) or a combination of them along with control group. Activities of acid phosphatase (ACP), lactate dehydrogenase (LDH) and glucose-6-phosphate dehydrogenase (G-6-PDH) were estimated. Moreover, gill, liver and kidney were assayed for activities of superoxide dismutase (SOD), catalase (CAT) and levels of glutathione (GSH) and malondialdehyde (MDA). Individual exposure showed that both Cd and Pb significantly decreased LDH activity and SOD activity in the kidney. Pb significantly increased G-6-PDH activity and decreased GSH level in the gill. CAT activity in liver and kidney elevated significantly on Cd exposure while lead caused a significant depletion in the liver and significant elevation in the kidney. Both Cd and Pb significantly increased MDA levels in liver and kidney while Pb increased its level in gills. The combined exposure resulted in normalization of LDH, G-6-PDH activity, and CAT activity in liver and kidney as well as GSH level in both tissues and MDA in gill and kidney. The combination increased SOD activity and MDA level in liver and decreased SOD activity in kidney and GSH level in gills. In conclusion, the antioxidant system of African catfish was adversely affected by prolonged exposure to Cd and Pb. The combined exposure caused less damage than individual exposure and returned most parameters to those of controls.
This study was undertaken to determine the effect of dietary fat to carbohydrate ratio on plasma glucose. free fatty acid level and hepatic glucokinase activity in normal or insulin treated diabetic rats. Sprague-Dawley male rats were fed with 3 different but isocaloric diets for 5 weeks. Diet 1 made to have low fat(4% corn oil and 65.8% corn starch wt/wt) : diet 2 medium fat (12% : 47.8%) : diet 3 high fat (20% : 29.8%) In the normal rats an apparent increase of GK activity was observed from the animal fed low fat diet when compared with other groups. GK activities were decreased in all the alloxan-diabetic rats than the normal rats. When insulin was injected the GK activities in all the livers of alloxan-diabetic rats restored to normal level and GK activity was highest in low fat group. In the entire group significant relationships were seen between the plasma glucose and GK activities(r=-0.6, p<0.001) FFA levels and GK activities(r=-0.63 p<0.001) Both in normal and insulin treated diabetic rats significantly depressed level of hepatic GK activity was observed in the livers of animals fed high fat diet for 5 weeks and depressed level of GK activity may be related to insulin resistance.
Rosae laevigatae Fructus extract (RLF) was tested for the effects on the urethral nitric oxide synthase (NOS) activity and Antioxidation in streptozotocin (STZ) induced diabetic rats. RLF was treated firstly into samples, and then STZ induced diabetic rats were set with them. In vitro, the urethral NOS activity was not noted but the type O activity and type conversion ratio of xanthine oxidase and the level of urethral lipid peroxide were decreased in the level of Dose of extract prepared from RLF. In vivo, after the extract was administered to the animal model for fifteen days, the urethral NOS activity increased in STZ induced diabetic rats to the level of normal rats. The content of urethral nitrite and glutathione followed by RLF pre-medicating administration, increased as highly as normal group in compare with the group treated with STZ. The type O activity and type conversion ratio of xanthine oxidase and the level of urethral lipid peroxide followed by RLF pre-medicating administration, decreased as lowly as normal group in compare with the group treated with STZ. In conclusion, the extract of RLF will be able to restore erectile dysfunction of STZ induced diabetic rats.
The regulatory mechanism of methionine biosynthesis in Corynebacterium glutamicum was analyzed at the protein arid gene expression level. O-Acetylhomoserine sulfhydraylase (encoded by metY) was inhibited by 10 mM methionine to a residual activity of 10% level, whereas no such inhibition was found with cystathionine $\gamma$-synthase (encoded by metB) and cystathionine $\beta$-lyase (encoded by metC). The enzymatic activity of homoserine acetyltransferase (encoded by metX) was repressed to a residual activity of 25% level by 10 mM methionine which was added to the growth medium. Cystathionine $\gamma$-synthase and cystathionine $\beta$-lyase were also repressed by 10 mM methionine, but only to a residual activity of 50-70% level. O-Acetylhomoserine sulfhydrylase was very sensitive to repression by 10 mM methionine, showing residual activity of 13%. In addition, homoserine acetyltransferase was also repressed by 10 mM cysteine to 50% of its original activity. No repression of the enzymes by S-adenosyl methionine was observed. The pattern of repression by methionine indicated that the metB and aecD genes might be regulated by a common mechanism, while the metA and metY genes are differently regulated.
This study examined the characteristics of children, parents, family, and the extra context related to the parenting stress of employed and unemployed mothers from a broader perspective. The subjects were 323 employed mothers 3nd 300 unemployed mothers of pre-school age children. Parenting stress due to the role of being a parent for both employed and unemployed mothers was correlated with the chid's activity level, husband support, quality of life, available social support, and satisfaction of early childhood program's location. Parenting stress due to child-rearing of both employed and unemployed mothers was correlated with child's birth order, activity level and rhythmicity of child, husband support, quality of life, available social support, and satisfaction of early childhood program. Number of children was the strongest predictor of parenting stress due to the role of being a parent for employed mothers and the child's activity level for unemployed mothers. Besides, the child's activity level was the strongest predictor of parenting stress due to child-rearing for both employed and unemployed mothers.
In order to prove the anticonvulsive effect of the fragrance of Magnoliae Flos in convulsion-induced mice, experiments were performed on anticonvulsive effect, GABA level, glutamic acid level, GABA-T activity and GAD activity. The results were obtained as follows: 1. As far as anticonvulsive effect was concerned, on the convulsion induced by such as maximal electric seizure, strychnine, bicuculline, or picrotoxin it was not significant, but the convulsion induced by pentylenetetrazole it was significant comparing to the control group. 2. GABA level was increased significantly in mice. 3. Glutamic acid level was decreased significantly in mice. 4. GABA-T activity was decreased by the fragrance of Magnoliae Flos. 5. The fragrance of Magnoliae Flos was not effective in GAD activity. From above result, the fragrance of Magnoliae Flos had significant effects on convulsion induced by pentylenetetrazole, so it is expected to clinical application on convulsive diseases such as epilepsy.
Objectives: The researchers investigated the relationship among fun seeking, self-efficacy, interest in physical activity and actual physical activities of male college students, in orders to provide valuable information to professionals in health promotion. Methods: The participants were 316 male college students whose ages ranged from 18 to 30(M=22.53, SD=2.09). The questionnaires and psychological tests used in this research included the following: sub-scale of Korean Version of BAS/BIS Scale, The General Self-Efficacy Scale, International Physical Activity Questionnaires, & sub-scale of The Intrinsic Motivation Inventory for physical activity. Results: Results indicated that male students who had moderate level of fun seeking were more likely to engage in physical activities than those who showed low or high level of fun seeking. And students with the strong self-efficacy were more likely to do physical activity than with the weak self-efficacy, especially high level of physical activity. Male students who had greater interest in physical activity were more likely to engage in physical activities than those who were not interested in physical activity. A regression analysis revealed that interest in physical activity accounted for the most variance; it accounted for around 14.5% variance of actual physical activity with self-efficacy. The interaction effect of self-efficacy and interest in physical activity was significant for actual physical activity. Conclusion: This study reiterates the roles of self-efficacy and the interest in physical activity on physical activity, and found that non-linear relation of fun seeking to physical activity. These results may provide useful information for health practitioners.
Purpose: The purpose of this study was to identify the relationship among the attitudes on exercise, physical activity and quality of life (QOL) in hemodialysis patients. Methods: A total of 42 patients in a hemodialysis unit participated in this study. Physical activity level was measured directly by 6 minute walking test and grip strength test. Structured questionnaires were also used for measuring their attitudes on exercise, physical activity and QOL. Participants' medical records were reviewed for obtaining their biochemical and clinical information. Statistical analysis was performed using Pearson correlation, and multiple liner regression. Results: A significant positive correlation between participants' attitudes and physical activity level measured by International Physical Activity Questionnaire (IPAQ) was found. And the physical activity level measured by Korea Activity Scale/Index (KASI) was significantly related to QOL. Conclusion: This study shows that QOL of the hemodialysis patients was significantly associated with their physical activity level.
The purposes of this study were: 1) to show the item difficulty hierarchy of walking/moving construct of the International Classification of Functioning, Disability and Health-Activity Measure (ICF-AM), 2) to evaluate the item-level psychometrics for model fit, 3) to describe the relevant physical activity defined by level of activity intensity expressed as Metabolic Equivalent of Tasks (MET), and 4) to explore what extent the empirical activity hierarchy of the ICF-AM is linked to the conceptual model based on the level of energy expenditure described as MET. One hundred and eight participants with lower extremity impairments were examined for the present study. A newly created activity measure, the ICF-AM using an item response theory (IRT) model and computer adaptive testing (CAT) method, has a construct on walking/moving construct. Based on the ICF category of walking and moving, the instrument comprised items corresponding to: walking short distances, walking long distances, walking on different surfaces, walking around objects, climbing, and running. The item difficulty hierarchy was created using Winstep software for 20 items. The Rasch analyses (1-parameter IRT model) were performed on participants with lower extremity injuries who completed the paper and pencil version of walking/moving construct of the ICF-AM. The classification of physical activity can also be performed by the use of METs that is often preferred to determine the level of physical activity. The empirical item hierarchy of walking, climbing, running activities of the ICF-AM instrument was similar to the conceptual activity hierarchy based on the METs. The empirically derived item difficulty hierarchy of the ICF-AM may be useful in developing MET-based activity measure questionnaires. In addition to convenience of applying items to questionnaires, implications of the finding could lead to the use of CAT method without sacrificing the objectivity of physiologic measures.
