Quantification of an active ingredient in tablets by NIR transmission measurements

  • For the quality control of tablets several parameters have to be checked. The most important one is the content of an active ingredient which has to match a narrow range around the designated content. The only useful measurement mode is transmission which provides information of the complete tablet. A measurement in diffuse reflectance would register only the surface which is useless especially in case of a coated tablet. In this work tablets for a clinical study (placebo/verum studies) with very low concentrations of the active ingredient were measured. The concentration range was 0 to 6 mg with a total weight of the tablets of 105 mg, leading to a highest concentration of the active component of 5.7% by weight. Especially the spectroscopic distinction between the placebo and the low dosage forms with 0.25 and 0.5 mg active agent requires an extraordinarily accurate sampling technique. Using the VECTOR 22/N-T in transmission mode allows the collection of the information from the complete tablets. A quantitative PLS-model with transmission spectra from the tablets described above shows that the active substance can be predicted with a RMSECV (root mean square error of cross validation) of 0.04% absolute for this special application. The results are compared with those of measurements in diffuse reflectance using different accessories.

The Effect of Surface Environment on the Mechanism in Oblique Cutting (3차원 절삭에서 표면환경이 절삭기구에 미치는 영향)

  • The object of the study is to discuss the effect of magic ink as a surface active substance on the mechanism of chip formation in oblique cutting. The Rehbinder effect has been known as a phenomenon that the mechanical strength reduces when the metal is coated with some surface active substances. In order to interpret these surface effects defined by Rehbinder, the influence on the shear strength of shear plane by coating surface active substances, cutting force by the depth of cut, surface roughness and hardness ratio were observed. The results are as follows: 1. By coating the magic ink on free surface of the forming chip, the effective shear angle increases, and the cuttinbg force and the deformed chip thickness decreases. 2. With the large inclination angle the effective shear angle increases, and the specific cutting force and the friction angle decrease. 3. Cutting of the coated surface improves the surface roughness and the hardness ratio drops, which means another Rehbinder effect.

  • This study was conducted to further purify and characterize a polysaccharide fraction from whole parts of Duchesnea indica (Andr.) Focke, which was previously reported to show an antitumor activity by us. Mild acid-treatment of the fraction by 5% acetic acid yielded three subtractions (Fr. A,B and C) on a Sephadex G-25. Fr. A was composed of uronic acid, hexose, pentose and phenolic substance, Fr. C mainly phenolic substance, and Fr. B contained hexose. All three fractions exhibited colony stimulating factor activity.

  • High-performance liquid chromatography (HPLC) screening method revealed that a propolis product marketed as a functional food contained an undeclared substance similar to tadalafil, the active ingredient of the prescription drug Cialis$^{(R)}$ approved for the treatment of male erectile dysfunction. In order to identify the illegal additive, the propolis product was extracted with methylene chloride, and the extract was purified further using semipreparative HPLC. The chemical structure of the isolated substance was elucidated based on IR, LC/MS-ESI, and $^1H$- and $^{13}C$-NMR spectroscopy, which showed the characteristics similar to tadalafil. The only difference was the substitution of the methyl group at the piperazinedione ring of tadalafil to the amino group of the identified illegal additive.

  • Acidic polysaccharide from Korean red ginseng was found to inhibit pancreatic lipase activity and cause reduction of plasma triglyceride level after oral administration of corn oil emulsion to rats. Thus acidic polysaccharide may reduce plasma triglyceride through its inhibitory action on pancreatic lipase and successive inhibition of intestinal absorption of fat due to reduction of lipolysis. In the course of this experiment, we found an unknown ninhydrin positive substance in Korean red ginseng. The unknown substance was identified to be arginyl-fructosyl glucose(Arg - Fru - Glc). Coment of this new compound was $5.37\%$ in Korean red ginseng powder. Sucrase and maltase activities in mucous layer of rat jejunum were found to be inhibited by Arg-Fru-Glc. Physiological significance of the new compound was discussed based on these experimental results.

  • An antitumor antibiotic named B-1123 substance was isolated from the culture filtrate of a new isolate fungus identified as Aspergillus terreus. The fermentation yield reached about 23 mg per liter of the broth. The B-1123 substance, chlorine containing antibiotic, has the molecular formular of $C_{17}H_{12}0_7C_{12}$. Its structure was determined to be geodin by spectroscopic data. It is active against some Gram-positive bacteria and it prolongs the life span of mice inoculated with Ehrlich carcinoma.

Investigation of Brassinosteroid Substances in Undmia pinnatifida (미역의 Brassinosteroid 활성물질검색)

  • In order to examine the presence of brassinosteroid substance in sea mustard(Undaria pinnatifida), leaves of sea mustard were extracted with MeOH. The extract was purified by slovent fractionation, counter current distribution, silica gel adsorption chromatography, charcoal adsorption chromatography, Bondesil chromatography, and reverse phase HPLC, successively. The activity was monitored by the rice inclination test and its prescence could be confirmed in each purification step. Although sea mustard contained a less amount of the active substance than the vegetative tissue of higher plants, brassinosteroid was clearly present endogenously in sea mustard. We acknowledge that our work is probably the first publication reporting the presence of brassinosteroid in marine algae plants.

Antibiotics from Mushrooms (버섯의 항생물질(抗生物質))

  • Antibiotics which produced by mushrooms discovered for last 40 years were described. Any antibiotic has not been used as infectious disease remudy but will be used as physiological active substance in near future. The antibiotic of mushrooms have not been published much in papers and do not have various finds of structures, compared to those of Streptomyces. Triple bond having compounds, terpenoid compounds aromatic compounds and some other compound have been known. These compounds are not dissolved well in water and mainly fat-soluble, except for cordycepin. Also, they are generally neutral, and some of them are acidic and almost none of them are basic compounds. However, acetylene and terpenoid compounds are the characteristic compounds of mushroom, and are not found in other microorganisms and plants. Especially, there are various terpenoid compounds in mushrooms. These metabolites of mushrooms were not used as antibiotic, but are interested as physiological active substance, such as enzyme inhibitor and immunomodulator. To promote studying on the antibiotics of mushroom, new screening methods must be developed, because strain belonged to the different genus produces different antibiotics, even though mushrooms belonged to the same genus and species. It is also known that mushrooms collected in different areas produce different antibiotics. Now, it is difficult to separate each pure compound from mushroom. It is important to find mushrooms which is impossible to cultivate artificially, or grow in the back land where is difficult to collect. Thousands of mushrooms grow on earth now, so that which species will be screened if not known. The biochemical and mycological study for usability of the metabolites of mushrooms is thought, as one of the important research areas, must be performed.

Isolation of Antimicrobial Active Substance from Usnea longissima against Sclerotial Rot (Sclerotinia sclerotiorum) (송라(Usnea longissima)추출물로부터 균핵병 병원균(Sclerotinia sclerotiorum)에 대한 항균 활성물질 탐색)

  • To develop environment-friendly agricultural products with anti-microbial activity against Sclerotinia sclerotiorum as a pathogen of sclerotium disease, Usnea longissima was extracted by methanol and its extract was fractionated into several solvent fractions. The chloroform fraction, which showed the highest antimicrobial activity, was separated by silica gel-column chromatography and obtained into nine group subfractions. The nine group fractions were searched the antifungal activities by bioassay. The most active No. 3 subfraction was analyzed by GC-MS. Each mass spectra, corresponding to each peak of chromatogram, was compared to database of Wiley library. As a result, Usnic acid was identified as main compounds. In conclusion, Usnic acid isolated from Usnea longissima was antimicrobial chemical against Sclerotinia sclerotiorum as a pathogen of sclerotium disease.

Antibacterial Activities against Plant Pathogens and Identification of Agrimol B from Agrimonia pilosa LEDEB (식물병원균에 대한 짚신나물 (선학초) 추출물의 항균활성과 Agrimol B의 동정)

  • Eighty-five percent methanol extract of Agrimonia pilosa has antibacterial activity against Pseudomonas syringae pv. lacrymans (bacterial leaf spot pathogen), Ralstonia solanacearum (tomato bacterial wilt pathogen) and Pseudomonas syringae pv. tabaci (Tobacco wild fire pathogen.). The active substance was purified by silica gel column chromatography and HPLC. The molecular weight of the active compound was determined by LC-Mass as 687.2. With NMR analysis, the active substance was identified as Agrimol B.
