로즈마리(Rosmarinus offcinalis L.) 분획물의 암세포에 대한 성장억제 및 세포사멸 유도 효과 (Effect of Rosmarinus officinalis L. on Growth Inhibition and Apoptosis Induction in Cancer Cells)

  • 최근 다방면에서 효능이 알려지고 있는 로즈마리를 이용하여 apoptosis 유도기전을 해석하기 위하여 인간유래 자궁암 Hela 세포주를 대상으로 조사한 결과는 다음과 같다. 로즈마리 분획물 중 hexane층과 chloroform층의 처리 농도 의존적으로 Hela 자궁암세포의 저해활성은 현저하게 감소되었으며 이는 apoptosis 유도와 연관성이 있었다. 로즈마리 분획물 중 hexane층과 chloroform층의 처리에 의하여 apoptosis 억제에 관여하는 Bcl-2 단백질의 발현은 감소되었으며, apoptosis 유도에 관여하는 Bax 유전자의 발현은 농도 의존적으로 증가되었다. 로즈마리 분획물 중 hexane층과 chloroform층의 처리에 의한 apoptosis 유도는 caspase-3, 7, 9 및 PARP의 활성화와 연관성이 있었다.

Age-Related Changes of Adult Neural Stem Cells in the MouseHippocampal Dentate Gyrus

  • This study was designed to investigate the changes in the properties of the neuronal setm cells or progenitor cells associated with age-related decline in neurogenesis of the hippocampal dentate gyrus (DG). Active whole cells cycle marker Ki67 (a marker of whole cell cycle)-positive and S phase marker bromodeoxyuridine (BrdU)-positive. Neural stem cells gradually were reduced in the hippocampal subgranular zone (SGZ) in an age-dependant manner after birth (from P1 month to P1 year). The ratio of BrdUpositivecells/Ki67-positive cells was gradually enhanced in an age-dependent manner. The ratio of Ki67-positive cells/accu-mulating BrdU-positive cells at 3 hrs after BrdU injection was injected once a day for consecutive 5 days gradually decreased during ageing. TUNEL- and caspase 3 (apoptotic terminal caspase)-positive cells gradually decreased in the dentate SGZ during ageing and immunohistochemical findings of glial fibrillary acid protein (GFAP) were not changed during ageing. NeuN, a marker of mature neural cells, and BrdU-double positive cells gradually decreased in an age-dependent manner but differentiating ratio and survival rate of cells were not changed at 4 wks after BrdU injection once a day for consecutive 5 days. The number of BrdU-positive cells migrated from the hippocampal SGZ into granular layer and its migration speed was gradually declined during ageing. These results suggest that the adult neurogenesis in the mouse hippocampal DG gradually decrease through reducing proliferation of neural stem cells accompanying with cells cycle change and reduced cells migration rather than changes of differentiation.

The Inhibitory Effects of Bee Venom and Melittin on the Proliferation of Vascular Smooth Muscle Cells

  • In the present study, I have investigated the bee venom (BV) and melittin (a major component of BV) -mediated anti-proliferative effects, and defined its mechanisms of action in cultured rat aortic vascular smooth muscle cells (VSMCs). BV and melittin $(0.4{\sim}0.8\;{\mu}g/ml)$ effectively inhibited 50 ng/ml platelet derived growth factor BB (PDGF-BB)-induced VSMCs proliferations. The regulation of apoptosis has attracted much attention as a possible means of eliminating excessively proliferating VSMCs. In the present study, the treatment of BV and melittin strongly induced apoptosis of VSMCs. I examined the effects on $NF-{\kappa}B$ activation to investigate a possible mechanism for anti-proliferative effects of BV and melittin, the PDGF-BB-induced $I{\kappa}B{\alpha}$ phosphorylation and its degradation were potently inhibited by melittin, and DNA binding activity and nuclear translocation of $NF-{\kappa}B$ p50 subunit in response to the action of PDGF-BB were potently attenuated by melittin. In further investigations, melittin markedly inhibited the PDGF-BB-induced phosphorylation of Akt but not ERK1/2, upstream signals of $NF-{\kappa}B$. Treatment of melittin also potently induced pro-apoptotic protein p53, Bax, and caspase-3 expression, but decreased anti-apoptotic protein Bcl-2 expression. These results suggest that the anti-proliferative effects of BV and melittin in VSMCs through induction of apoptosis via suppressions of $NF-{\kappa}B$ and Akt activation, and enhancement of apoptotic signal pathway. Based on these results, BV acupuncture can be a candidate as a therapeutic method for restenosis and atherosclerosis.

인체 전립선 암세포에서 Methyl Gallate의 항암효과 (Anticancer Activity of Methyl Gallate in RC-58T/h/SA#4 Primary Human Prostate Cancer Cells)

  • 본 연구에서는 MG의 RC-58T/h/SA#4 인체 전립선 암세포에 대한 증식 억제 및 apoptosis 유도효과에 대하여 확인하였다. MG는 RC-58T/h/SA#4 세포의 증식을 농도 의존적으로 억제하였다. 이러한 MG에 의한 RC-58T/h/SA#4 세포의 사멸이 apoptosis에 의해 일어나는지를 sub-G1 함량 측정 및 Hoechst 33258 염색을 이용하여 확인하였다. 그 결과 MG를 처리한 군의 sub-G1의 함량이 대조군에 비하여 증가하였으며, methyl gallate를 처리한 군에서 핵의 응축과 apoptotic body 형성을 Hoechst 33258 염색을 통하여 관찰할 수 있었다. 또한 RC-58T/h/SA#4 세포에서 MG가 유도하는 apoptosis의 기전을 확인하기 위하여 다양한 실험을 실시하였다. MG는 DNA의 분절량 증가 및 caspase 활성을 유도하였으며, 활성화된 caspase-8, -9 및 -3에 의해 PARP와 Bid 단백질의 분절을 발현시켰으며, Bcl-2 family 단백질의 발현에 영향을 미쳐 apoptosis를 유도하였음을 확인하였다. 한편 환경호르몬인 dioxin과 bisphenol A를 다양한 농도로 처리한 결과 각각 1 nM, $0.1{\mu}M$ 농도에서 가장 높은 전립선 암세포 과다증식을 유도하였으며, 이를 바탕으로 MG를 농도별로 처리한 결과 환경호르몬에 의해 유도된 인체 전립선 암세포의 증식을 농도 의존적으로 억제시켰다. 본 연구 결과 MG는 RC-58T/h/SA#4 전립선 암세포에서 apoptosis 유도를 통한 암세포 성장 억제효과를 가지고 있음을 확인하였으며, 환경호르몬에 의해 유발될 수 있는 암에 대해서도 유사한 보호효과를 가지고 있음을 증명하였다.

골관절염 랫드 모델에서 계피의 연골보호 효과 (Chondroprotective Effects of Cinnamomum cassia Blume in a Rat Model of Osteoarthritis)

  • 본 연구는 내측반월판 절제와 전십자인대 단열로 유발된 랫드 골관절염 모델을 이용하여 계피 (Cinnamomum cassia Blume, 육계(肉桂)) 추출물의 연골손상 방지에 대한 효과를 평가하였다. 골관절염유발 랫드 48마리를 군당 8마리씩 6군으로 음성 대조군과 골관절염 대조군, 체중당 diclofenac 2 mg 투여군, 계피 추출물 25 mg과 50 mg, 100 mg 투여군으로 각각 분류하여 수술 1주 후부터 12주동안 투여하였다. 연골 소실과 관절의 불안정성은 계피 추출물과 diclofenac 투여군이 골관절염 대조군과 비교할 때 유의하게 감소하였다(p < 0.05). 병리조직학적 평가에서 연골의 퇴행은 계피 추출물 투여량에 따라 용량 의존적으로 개선됨이 확인되었다(p < 0.01). 계피 추출물 투여군에서 관절구조 퇴행성 변화의 감소와 Safranin-O 염색 정도의 용량 의존적인 증가를 보여 연골 퇴행이 억제됨을 확인하였다. Diclofenac이 골관절염 진행에 있어 활성화된 caspase-3와 절단된 poly(ADP-ribose) 중합효소에 유도된 세포자멸사 표지율 증가에 별 다른 영향을 주지 않았지만 계피 추출물 투여군에서는 이들 세포자멸 표지자에 대한 반응세포는 유의하게 감소되었다(p < 0.05). 그러나, diclofenac과 계피 추출물 투여군은 5-bromo-2-deoxyuridine의 섭취에는 영향을 주지 않았다. 이러한 결과에서 계피추출물이 항염활성과 항세포 자멸 활성을 통해 관절연골에 보호 효과가 있음을 보였다.

H2O2로 유도된 C6 신경교세포의 세포사멸에 대한 감초 추출물과 감초 활성물질의 보호효과 (Protective Effects of Glycyrrhiza uralensis Radix Extract and Its Active Compounds on H2O2-induced Apoptosis of C6 Glial Cells)

  • Background: Glycyrrhiza uralensis Radix (GR) is a crude drugs used in Asian countries that has been reported to prevent the progression of neurodegenerative diseases such as Alzheimer's disease. The present study examined whether GR and its active compounds, glycyrrhizic acid (GA) and isoliquiritigenin (IL), exerted protective effects on $H_2O_2$-induced oxidative damage in C6 glial cells. Methods and Results: We exposed C6 glial cells to hydrogen peroxide ($H_2O_2$) for 24 h and investigated the cellular response to GR and its active compounds by evaluating cell viability, reactivie oxygen species (ROS) production, and apoptosis-related protein expression. GR successfully mitigated the reduced cell viability and ROS production induced by $H_2O_2$ in C6 glial cells, IL and GA significantly increased the cell viability and decreased ROS production. In addition, IL and GA down-regulated apoptotic Baxdependent caspase-3 activation, but each compound exerted different mechanisms, i.e., IL dose-dependently decreased ROS production and, GA up-regulated anti-apoptotic Bcl-2 expression. Conclusions: These results demonstrated that GR and its active components, IL and GA, exhibit potential for use as natural neurodegenerative agents for the modulation of apoptosis in C6 glial cells.

Suppression of Autophagy and Activation of Glycogen Synthase Kinase 3beta Facilitate the Aggregate Formation of Tau

  • Neurofibrillary tangle (NFT) is a characteristic hallmark of Alzheimer's disease. GSK3β has been reported to play a major role in the NFT formation of tau. Dysfunction of autophagy might facilitate the aggregate formation of tau. The present study examined the role of GSK3${\beta}$-mediated phosphorylation of tau species on their autophagic degradation. We transfected wild type tau (T4), caspase-3-cleaved tau at Asp421 (T4C3), or pseudophosphorylated tau at Ser396/Ser404 (T4-2EC) in the presence of active or enzyme-inactive GSK3${\beta}$. Trehalose and 3-methyladenine (3-MA) were used to enhance or inhibit autophagic activity, respectively. All tau species showed increased accumulation with 3-MA treatment whereas reduced with trehalose, indicating that tau undergoes autophagic degradation. However, T4C3 and T4-2EC showed abundant formation of oligomers than T4. Active GSK3${\beta}$ in the presence of 3-MA resulted in significantly increased formation of insoluble tau aggregates. These results indicate that GSK3${\beta}$-mediated phosphorylation and compromised autophagic activity significantly contribute to tau aggregation.

천초근 에탄올 추출물의 항산화 효능 및 대장암 세포 억제 효과 (Anti-colorectal Cancer and Anti-oxidant Activities of Rubiae radix Ethanol Extract in vitro)

  • 본 연구는 꼭두서니의 뿌리인 천초근의 물 추출물과 에탄올 추출물을 이용하여 대장암 세포에 대한 암세포 성장 억제 및 사멸효과가 있는지 알아보고자 수행하였다. ERA(천초근 에탄올 추출물)은 폴리페놀($45.77{\pm}2.03mg/g$)과 플라보노이드($22.82{\pm}1.33mg/g$)를 함유하고 있었으며, $H_2O_2$에 의해 증가된 ROS(reactive oxygen species)를 억제하는 효과를 나타냈지만 WRA(천초근 물 추출물)은 효과가 없었다. 또한 ERA는 $500{\mu}g/m{\ell}$의 농도로 대장암 세포주(HCT-116)에 처리했을 때 세포사멸을 유도할 뿐만 아니라 caspase-3 단백질 활성화, DNA fragmentation 및 apoptotic cell death를 일으키는 것으로 확인되었다. 이는 ERA가 HCT-116 세포주에 대해 apoptosis(세포자멸사)를 통해 항암효과를 나타내는 것으로 생각되지만 다른 연구결과들과 비교하였을 때 농도 대비 효능이 미미하다. 따라서 천초근 에탄올 추출물에 대장암 세포의 성장을 억제하는 유효성분을 분석하여 그 효능을 탐색하는 추가실험이 필요할 것으로 생각된다.

Protective effects of perilla oil and alpha linolenic acid on SH-SY5Y neuronal cell death induced by hydrogen peroxide

  • BACKGROUND/OBJECTIVE: Oxidative stress plays a key role in neuronal cell damage, which is associated with neurodegenerative disease. The aim of present study was to investigate the neuroprotective effects of perilla oil (PO) and its active component, alpha-linolenic acid (ALA), against hydrogen peroxide $(H_2O_2)$-induced oxidative stress in SH-SY5Y neuronal cells. MATERIALS/METHODS: The SH-SY5Y human neuroblastoma cells exposed to $250{\mu}M$ $H_2O_2$ for 24 h were treated with different concentrations of PO (25, 125, 250 and $500{\mu}g/mL$) and its major fatty acid, ALA (1, 2.5, 5 and $25{\mu}g/mL$). We examined the effects of PO and ALA on $H_2O_2$-induced cell viability, lactate dehydrogenase (LDH) release, and nuclear condensation. Moreover, we determined whether PO and ALA regulated the apoptosis-related protein expressions, such as cleaved-poly ADP ribose polymerase (PARP), cleaved caspase-9 and -3, BCL-2 and BAX. RESULTS: Treatment of $H_2O_2$ resulted in decreased cell viability, increased LDH release, and increase in the nuclei condensation as indicated by Hoechst 33342 staining. However, PO and ALA treatment significantly attenuated the neuronal cell death, indicating that PO and ALA potently blocked the $H_2O_2$-induced neuronal apoptosis. Furthermore, cleaved-PARP, cleaved caspase-9 and -3 activations were significantly decreased in the presence of PO and ALA, and the $H_2O_2$-induced up-regulated BAX/BCL-2 ratio was blocked after treatment with PO and ALA. CONCLUSIONS: PO and its main fatty acid, ALA, exerted the protective activity from neuronal oxidative stress induced by $H_2O_2$. They regulated apoptotic pathway in neuronal cell death by alleviation of BAX/BCL-2 ratio, and down-regulation of cleaved-PARP and cleaved caspase-9 and -3. Although further studies are required to verify the protective mechanisms of PO and ALA from neuronal damage, PO and ALA are the promising agent against oxidative stress-induced apoptotic neuronal cell death.

Protective effect of ginsenoside Rh3 against anticancer drug-induced apoptosis in LLC-PK1 kidney cells

  • Background: Ginsenosides are active components of Panax ginseng that exert various health benefits including kidney protection effect. The medicinal activity of ginsenosides can be enhanced by modulating their stereospecificity by heat processing. Ginsenosides Rk2 and Rh3 represent positional isomers of the double bond at C-20(21) or C-20(22). Methods: The present study investigated the kidney-protective effects of ginsenosides Rk2 and Rh3 against cisplatin, a platinum based anticancer drug, induced apoptotic damage in renal proximal LLC-PK1 cells. Results: As a result, ginsenoside Rh3 shows a stronger protective effect than that shown by Rk2. Cisplatin-induced elevated protein levels of phosphorylated c-Jun N-terminal kinase (JNK), extracellular signal-regulated kinase (ERK), p38, and cleaved caspase-3 decreased after cotreatment with ginsenoside Rh3. The increase in the percentage of apoptotic LLC-PK1 cells induced by cisplatin treatment also significantly reduced after cotreatment with ginsenoside Rh3. Conclusion: These results demonstrate that inhibition of the JNK and ERK mitogen-activated protein kinase signaling cascade plays a critical role in mediating the renoprotective effect of ginsenoside Rh3.
