Purification and Characterization of Anticoagulant Protein from the Tabanus, Tabanus bivittatus

  • Tabanus anticoagulant protein (TAP) was isolated from the whole body of the tabanus, Tabanus bivittatus, using three purification steps (ammonium sulfate fractionation, gel filtration on Bio-Gel P-60, and ion exchange chromatography on DEAE Sephadex gel). The purified TAP, with a molecular weight of 65 kDa, was assessed to be homogeneous by SDS-polyacrylamide gel electrophoresis, and an isoelectric point of 7.9 was determined by isoelectric focusing. The internal amino acid sequence of the purified protein was composed of Ser-Leu-Asn-Asn-Gln-Phe-Ala-Ser-Phe-lle-Asp-Lys-Val-Arg. The protein was activated by $Cu^{2+}\;and\;Zn^{2+}$, and the optimal conditions were found to be at pH $3\sim6\;and\;40\sim70^{\circ}C$. Standard coagulation screen assays were used to determine thrombin time and activated partial thromboplastin time. Chromogenic substrate assays were performed for thrombin and factor Xa activity. TAP considerably prolonged human plasma clotting time, especially activated partial thromboplastin time in a dose-dependent manner; it showed potent and specific antithrombin activity in the chromogenic substrate assay. Specific anti-factor Xa activity in TAP was not detected. Overall, this result suggested that TAP has significant anticoagulant activity on blood coagulation system.

선모(仙茅) 열수(熱水) 추출물(抽出物)의 항혈전(抗血栓) 효능 연구 (Effects of Curculiginis Rhizoma on anti-thrombotic activity)

  • Objectives : An aim of study is to investigate effects of curculiginis rhizoma in vitro (factor Xa (FXa) inhibitor assay, prothrombinase assay, prothrombin time (PT) assay, activated partial thromboplastin time (aPTT) assay) and in vivo experiment (blood clotting time, thromboxane B2 content assay in serum and weight of thrombus by AV-shunt rat model). Methods : We gained a human serum and used serum in vitro study such as factor X activity (FXa) inhibition, prothrombinase inhibition, prothrombin time (PT) and activated partial thromboplastin time. Fifteen SD rats were divided into three groups (intact control group and two experimental group treated with extract of Curculiginis Rhizoma(ECR)). Rats were orally administrated DW (intact control group), 600 mg/kg concertration of ECR and 200 mg/kg concertration of ECR. After one hour, we anesthetized rats and made arteriovenous (AV) shunt rat models to study weights of thrombus, took a hole blood to study content of thromboxane B2 and blood clotting time. Results : In vitro, ECR increased a inhibitory activity of FXa, prothrombinase and aPTT compared than intact control group. Especially ECR made significant increase of FXa and prothrombinase inhibitory activity (p<0.05, p<0.01). And PT were increased in ECR control group compared with intact control group. In vivo, a blood clotting time of experiment group treated with ECR 600 mg/kg were significantly increased compared with that of intact control group (p<0.05) and content of thromboxane B2 was significantly decreased in group treated with ECR 600 mg/kg in seum. The weight of thrombus were significantly reduced in group treated with ECR 600 mg/kg compared with intact control group (p<0.05). But in vivo experiment study, those of group treated with ECR 200 mg/kg were reduced compared with those of intact control group without statistical significance. Conclusions : ECR has a antithromboic activity in internal course with inhibitory activity of FXa and prothrombinase in vitro, it required to research more study for effective compounds.

하수오(何首烏)의 항응고효과에 대한 실험적 고찰 (The study about anticoagulant effect of Polygoni Mutiflori Radix)

  • The purpose of this study is to investigate the anti-coagulant effects of Polygoni Mutiflori Radix extracts. The results were as follows : 1. This study show that Polygoni Mutiflori Radix extracts have Anti-coagulant significant effects on Prothrombin time in vitro test. 2. This study show that Polygoni Mutiflori Radix extracts have Anti-coagulant significant effects on Activated Partial Thromboplastin Time vitro test.

허브 추출물의 항응고 활성 검색 (Screening of Anticoagulant Activities in Extracts from Edible Herbs)

  • 허브를 비롯한 식용되는 채소류로부터 항응고 활성을 검색하기 위하여 93종을 대상으로 냉수(Fr. I), 메탄올(Fr. ll), 열수(Fr. lll)추출물을 각각 조제하였다. 내인성 경로에 작용하는 항응고 활성은 수용성 획분인 Fr. I과 Fr.lll를 대상으로 activated partial thromboplastin time(APTT)으로 검색한 결과 양파, 마늘, 정향, 쑥, 울금 등이 높은 활성을 보였으며, 외인성 경로와 공통경로에 작용하는 항응고 활성은 저분자임을 감안하여 Fr. I과 Fr. II를 대상으로 각각 prothrombin time(PT) 과 thrombin time(TT)으로 검색한 결과 민트, 이탈리안 시즈닝, 로즈메리, 타라곤, 울금, 백리향, 와사비는 공통경로에 항응고 활성을 나타내였으나 외인성 경로에 단독으로 항응고 활성을 나타내는 시료는 없었다. 1, 2차 검색 결과 가장 높은 항응고 활성을 보인 정향(Eugenia caryophyllate, clove)을 대상으로 산지를 달리하여 항응고 활성을 검색한 결과 홀랜드산 정향의 활성이 가장 높았다.

목이버섯 추출물로부터 항혈전물질의 정제와 항혈전효과 (Purification of Antithrombotic Material from Auricularia auricular-judae Extracts and Its Antithrombotic Activity)

  • 목이버섯으로부터 항혈전활성과 항혈소판응집활성을 지니는 물질을 추출하여 추출물의 혈행개선활성을 조사하였다. 건조 목이버섯을 0.1 N NaOH, methanol, ethanol 등의 용매를 사용하여 추출하여 각 추출물의 항혈전활성을 activated partial thromboplastin time, thrombin time과 prothrombin time 값으로 측정한 결과 methanol 용해성 분획이 각각 100, 124, 54 s로 조사한 분획 중에서 가장 높은 활성을 나타내었다. 활성 분획을 DEAE-Sepharose CL6B ion exchange column chromatography와 Sephacryl 400-HR gel permeation column chromatography로 정제하였으며 활성물질은 분자량이 150 kDa 이상인 다당류이며 mannose가 주요 구성당으로 되어 있는 xyloglucomannan의 복합다당체인 것으로 확인되었다. 정제된 다당류의 항혈전활성은 thrombin 활성을 저해하기 때문인 것으로 해석되었다.

Antiplatelet and antithrombotic activities of purpurogallin in vitro and in vivo

  • Enzymatic oxidation of pyrogallol was efficiently transformed to an oxidative product, purpurogallin (PPG). Here, the anticoagulant activities of PPG were examined by monitoring activated partial thromboplastin time (aPTT), prothrombin time (PT), and the activities of thrombin and activated factor X (FXa). And, the effects of PPG on expression of plasminogen activator inhibitor type 1 (PAI-1) and tissue-type plasminogen activator (t-PA) were evaluated in tumor necrosis factor (TNF)-${\alpha}$ activated human umbilical vein endothelial cells (HUVECs). Treatment with PPG resulted in prolonged aPTT and PT and inhibition of the activities of thrombin and FXa, as well as inhibited production of thrombin and FXa in HUVECs. In addition, PPG inhibited thrombin-catalyzed fibrin polymerization and platelet aggregation. PPG also elicited anticoagulant effects in mice. In addition, treatment with PPG resulted in significant reduction of the PAI-1 to t-PA ratio. Collectively, PPG possesses antithrombotic activities and offers a basis for development of a novel anticoagulant.

호장근 추출물의 항혈전 효능 및 혈소판 응집 억제작용 (Antithrombotic Effect and Antiplatelet Activity of Polygonum cuspidatum Extract)

  • 항혈전 효능을 탐색하기 위하여 한약재의 호장근 추출물로 혈전 용해능 활성과 혈액 응고시간 지연효과 즉 PT(prothrombin time), APTT(activated partial thromboplastin time)와 혈소판 응집억제 활성 등에 대해 항혈전 효능을 평가하였다. 혈전용해도를 측정하는 fibrin plate가 용해되어 형성된 투명환의 넓이를 측정하는 실험을 진행한 결과 혈전 용해도가 농도의존적으로 탁월한 효능을 나타내었다. 혈액응고 cascade에 미치는 영향을 알아보기 위해 혈액 응고시간 지연 및 단축 효과를 확인하고자 APTT와 PT에 미치는 영향을 조사한 결과 PT의 경우 대조군보다 단축됨을 보였다. APTT의 경우 농도의존적으로 우수한 지연효과를 보였다. 혈소판의 응집에 따라 형성되는 두 전극 사이에 형성된 전기적 저항의 변화로 나타나는 실험을 시행한 결과 호장근의 ADP로 유도한 결과에서 뛰어난 응집억제 활성을 보였다. 따라서 호장근을 향후에 혈전 질환의 치료제 개발에 효과적으로 이용될 수 있을 것으로 사료된다.

목초액의 혈소판 응집억제를 통한 혈행개선 작용에 관한 연구 (Improvement of Haemostasis Mediated by Anti-Platelet Activities by Plant Vinegar)

  • We investigated the effects of plant vinegar on platelets and blood coagulation system. Plant vinegar inhibited in vitro platelet aggregation in a concentration dependent manner, when platelets were activated by thrombin and collagen. In addition, plant vinegar showed inhibitory effects on the serotonin secretion induced by thrombin in a concentration dependent manner. However, treatment with plant vinegar to platelets did not induce any cytotoxicity, as determined by the release of lactate dehydrogenase. Plant vinegar did not change the coagulation parameters such as activated partial thromboplastin time (aPTT) and prothrombin time (PT) using rat citrated plasma. In vivo study revealed that, treatment with plant vinegar prolonged the bleeding time from mouse tail. All these results suggest that plant vinegar might improve blood hemostasis mediated via anti platelet activities.

Anticoagulant activities of curcumin and its derivative

  • Curcumin, a polyphenol responsible for the yellow color of the curry spice turmeric, possesses antiinflammatory, antiproliferative and antiangiogenic activities. However, anticoagulant activities of curcumin have not been studied. Here, the anticoagulant properties of curcumin and its derivative (bisdemethoxycurcumin, BDMC) were determined by monitoring activated partial thromboplastin time (aPTT), prothrombin time (PT) as well as cell-based thrombin and activated factor X (FXa) generation activities. Data showed that curcumin and BDMC prolonged aPTT and PT significantly and inhibited thrombin and FXa activities. They inhibited the generation of thrombin or FXa. In accordance with these anticoagulant activities, curcumin and BDMC showed anticoagulant effect in vivo. Surprisingly, these anticoagulant effects of curcumin were better than those of BDMC indicating that methoxy group in curcumin positively regulated anticoagulant function of curcumin. Therefore, these results suggest that curcumin and BDMC possess antithrombotic activities and daily consumption of the curry spice turmeric might help maintain anticoagulant status.

홍삼류의 섭취가 사람 혈소판의 응집반응 및 혈중 지질에 미치는 영향 (Effects of Intaking of Red Ginseng Products on Human Platelet Aggregation and Blood Lipids)

  • Thrombogenesis and atherosclerosis are mainly caused by platelet aggregation, blood coagulation, and hyperlipidemia. Platelet aggrelation, activated platelet thromboplastin time (APTT) were measured as indexes of blood coagulation and lipid contents in the subjects who have taken red ginseng products (e.g. water extract, tea, drink etc.) for 4 to 5 years. The platelet aggregation in the red ginseng-taking group was significantly decreased, as compared with the non-red ginseng-intaking group, when platelets were stimulated by 100 $\mu\textrm{g}$/ml of collagen (P<0.01). The atherogenic index and the ratio of triglyceride to HDL-cholesterol in blood, the risk factors of atherosclerosis, were decreased in the subjects of ginseng group, compared with that in control group. APTT was also prolonged to greater extent in ginseng group than in control group. These results suggest that long-term intake of ginseng products may help to prevent the risks of thrombogenesis and atherosclerosis.
