Glutaraldehyde-Mediated Synthesis of Asparaginase-Bound Maghemite Nanocomposites: Cytotoxicity against Human Colon Adenocarcinoma Cells

  • Drugs processed using nanobiotechnology may be more biocompatible, with sustainable and stabilised release or action. L-asparaginase produced from fungi has many advantages for treatment of lymphocytic leukemia with lesser side effect. In the present work, maghemite nanobiocomposites of fungal asparaginase were produced using glutaraldehyde-pretreated colloidal magnetic nanoparticles. Formation of nanobiocomposites was observed using laser light scattering and confirmed by UV-visible spectrophotometry with the absorption peak at 497 nm. The specific asparaginase activity was increased from 320 U/mg with crude asparaginase to 481.5 U/mg. FTIR analysis confirmed that primary amines are the functional groups involved in binding of asparaginase on magnetic nanoparticles. The average size of the produced nanobiocomposite was found in the range of 30 nm to 40 nm using histogram analysis. The magnetic nanobiocomposite of asparaginase synthesised using glutaraldehyde showed 90.75% cytotoxicity against human colon adenocarcinoma cell lines. Hence it can be used as an active anticancer drug with an augmented level of bioavailability.

  • This paper is about automatic design of measuring master gears. Master gears are usually thought of as gears of extreme accuracy level, but are better defined as gages to check the meshing action of production gears. This is usually not recognized because most mechanical gages are associated with static measurements rather than having the form of machine elements used in a functional check involving machine motion. In this paper the interface that allows beginners to design easily and quickly is provided. The addition and modification of data is easy and the reduced design lead time is feasible with the program even though users don't know much about program since it is developed with Visual Lisp and DCL.

Regulation of Nrf2-Mediated Phase II Detoxification and Anti-oxidant Genes

  • The molecular mechanisms by which a variety of naturally-occurring dietary compounds exert chemopreventive effects have been a subject of intense scientific investigations. Induction of phase II detoxification and anti-oxidant enzymes through activation of Nrf2/ARE-dependent gene is recognized as one of the major cellular defense mechanisms against oxidative or xenobiotic stresses and currently represents a critical chemopreventive mechanism of action. In the present review, the functional significance of Keap1/Nrf2 protein module in regulating ARE-dependent phase II detoxification and anti-oxidant gene expression is discussed. The biochemical mechanisms underlying the phosphorylation and expression of Keap1/Nrf2 proteins that are controlled by the intracellular signaling kinases and ubiquitin-mediated E3 ligase system as well as control of nucleocytoplasmic translocation of Nrf2 by its innate nuclear export signal (NES) are described.

Brain Reward Circuits in Morphine Addiction

  • Morphine is the most potent analgesic for chronic pain, but its clinical use has been limited by the opiate's innate tendency to produce tolerance, severe withdrawal symptoms and rewarding properties with a high risk of relapse. To understand the addictive properties of morphine, past studies have focused on relevant molecular and cellular changes in the brain, highlighting the functional roles of reward-related brain regions. Given the accumulated findings, a recent, emerging trend in morphine research is that of examining the dynamics of neuronal interactions in brain reward circuits under the influence of morphine action. In this review, we highlight recent findings on the roles of several reward circuits involved in morphine addiction based on pharmacological, molecular and physiological evidences.

Prostaglandin $F_2{\alpha}$ Controls Reactive Oxygen Species in Bovine Corpus Luteum

  • Luteolysis is a cyclical regression of the corpus luteum in many non-primate mammalian species. Prostaglandin $F_2{\alpha}$($PGF_2{\alpha}$) from the uterus and ovary induces functional and structural luteolysis in bovine. The action of $PGF_2{\alpha}$ is mediated by $PGF_2{\alpha}$ receptor located on the luteal steroidogenic and endothelial cell membranes. $PGF_2{\alpha}$ plays an important role in regulating nitric oxide production in endothelial cells of the bovine corpus luteum. Nitric oxide production and nitric oxide synthase activity are stimulated and induced by $PGF_2{\alpha}$ in luteal endothelial cells. Moreover, the reactive oxygen species inhibits progesterone secretion in bovine luteal cells and induces apoptosis. Thus, the interaction between $PGF_2{\alpha}$ and reactive oxygen species provides important aspects in physiology of the corpus luteum forfunctional and structural luteolysis.

Mitigation of motions of tall buildings with specific examples of recent applications

  • Flexible structures may experience excessive levels of vibration under the action of wind, adversely affecting serviceability and occupant comfort. To ensure the functional performance of a structure, various design modifications are possible, ranging from alternative structural systems to the utilization of passive and active control devices. This paper presents an overview of state-of-the-art measures that reduce the structural response of buildings, including a summary of recent work in aerodynamic tailoring and a discussion of auxiliary damping devices for mitigating the wind-induced motion of structures. In addition, some discussion of the application of such devices to improve structural resistance to seismic events is also presented, concluding with detailed examples of the application of auxiliary damping devices in Australia, Canada, China, Japan, and the United States.

Antinocicepetive Effects of 3,4-Dicaffeoyl Quinic Acid of Ligularia fischeri var. spiciformis

  • The plant Ligularia fischeri var. spiciformis (Compositae) is a candidate for available functional foods. It has been used to treat diabetes mellitus and rheumatoid arthritis. We have reported the isolation of a new eremophilanolide named 6-oxoeremophilenolide and cytotoxic intermedeol together with the isolation of hydrophilic constituents, chlorogenic acid, 3,4-di-O-caffeoylquinic acie (3), and 5-O-[1-butyl]-3,4-di-O-caffeoylquinic acid. Compound 3 was again isolated by combination of silica gel- and ODS column chromatography for the anti-nociceptive action. Compound 3 and 4 were assayed in hot plate- and writhing tests in the rat. Although the three derivatives of caffeic acid exhibited significant anti-nociceptive effects at 10 mg/kg dose (i.p.),(activity potency: 4>3). These results suggest that compound 3 is responsible for at least rheumatoid arthritis, and caffeic acid moiety is the active moiety of dicaffeoylquinic acid.

항산화 및 미백화장품 원료로서의 땅콩새싹 추출물에 관한 연구 (A Study on Peanut Spouts Extract as the Anti-oxidant Activity and the Skin Whitening Cosmetic Ingredients)

  • To investigate the effect of peanut sprout extract on skin care, we measured anti-oxidant activity and whitening action. As a result of measuring DPPH radical scavenging activity to examine independent anti-oxidation of peanut sprout extract, there was strongly scavenging activity. Fluorescent material DCF-DA was used to measure hydrogen peroxide created in RAW 264.7 cells, and all concentration dependently decreased ROS production. As a result of measuring nitric oxide to examine anti-inflammation of peanut sprout extract, there was strongly inhibited nitric oxide production in RAW 264.7 cells. Tyrosinase activation was found to inhibited dose-dependant. Melanin production was also prevented dose-dependant. Therefore, it is expected to be used effectively in development of functional cosmetic materials.

패턴 생성기의 PLD 회로설계에 관한 연구 (A Study on the PLD Circuit Design of Pattern Generator)

  • 일반적으로 반도체 소자의 집적도가 증가함에 따라 기능적 오류 검사 시간이 급격하게 증가하며, 이러한 문제를 해결하기 위해 제조공정에서 패턴 발생기의 사용은 필수적이다. 본 논문에서는 반도체 소자의 기능적 오류를 검사 하기 위한 패턴 발생기의 PLD(Programmable Logic Device) 회로를 설계하였다. 이러한 모든 사항은 시뮬레이션을 통하여 회로의 동작과 기능을 검증하였으며, 만족할만한 결과를 얻었다.

Complete genome sequence of Bacillus coagulans CACC834 isolated from canine

  • Bacillus coagulans CACC 834 was isolated from canine feces, and its potential probiotic properties were characterized by functional genome analysis. Whole-genome sequencing of B. coagulans CACC 834 was performed using the PacBio RSII platforms. The complete genome assembly consisted of one circular chromosome (3.1 Mb) with guanine (G) + cytosine (C) content of 47.1%. Annotation revealed 3,181 protein-coding sequences (CDSs), 30 rRNAs, and 83 tRNAs. Gene associated 11% of the genes were involved in replication, recombination, and repair. We also annotated various stress-related, acid resistance, bile salt resistance and adhesion-related domains in this strain, which likely provide support in exerting probiotic action by survival under gastrointestinal tract. These results add to our comprehensive understanding of B. coagulans and suggest potential mammal-related industrial applications.
