A Study on the Anti-microbacterial Activity, Anti-inflammatory and Anti-allergic Effects of Samhwangseje gagambang(SHB) (삼황세제가감방(三黃洗劑加減方)의 항균, 항염 및 항알레르기 효과에 대한 실험적 연구)

  • Herbal mixture water extract of (Phellodendron amurense, Scuellaria baiklensis, Spphora flavescens, Lithospermum erythrorhizon, Mellaphis chinesis, Alumite, Zanthoxylum schinifolium, Glycyrrhiza uralensis), which exhibit several beneficial effects including acne and skin diseases, was tested for anti-microbial activity and anti-inflammation effects. The herbal mixture extract showed antimicrobial activity against Stapylococcus epidermis, Propionibacterium acne, and Malassezia furfur. The growth of Stapylococcus epidermis, Propionibacterium acne, and Malassezia furfur was inhibited allergy and LPS induced cyclooxygenase-2(COX-2)gene expression in RAW24.7macrophage. The results indicated th ear swelling and histamine release induced by compound 48/80 were dose-dependently reduced, ranging 11-38% and 11-56%, respectively. Furthermore the extract inhibited the expression of LPS-induced COX-2 proteins and mRNAs without an appreciable cytotoxic effects on RAW264.7 cells. The cytotoxicity of the extract using M7T assay showed the cytotoxicity of 7 and 18% against L929 cell line. Based on these results, it is concluded that the herbal mixture water extract can be applied to the acne and skin diseases therapy.

  • Herbal mixture water extract of (Rheum coreanum Scutellaria baikalensis, Phellodendron amurese), which exhibit several beneficial effects including acne and skin diseases, was tested for anti-microbial activity and anti-inflammation effects. The herbal mixture extract showed antimicrobial activity against Stapylococcus epidermis and Propionbacterium acne. The growth of Stapylococcus epidermis and Propionibacterium acne was inhibited completely by addition of 1.0% of the extract. Also in the present study we examined the mixture extract on compound 48/80 induced allergy and LPS induced cyclooxygenase-2(COX-2) gene expression in RAW264.7 macrophage. The results indicated the ear swelling and histamine release induced by compound 48/80 were dose-dependently reduced, ranging 18-36% and 10-61%, respectively. Furthermore the extract inhibited the expression of LPS-induced COX-2 proteins and mRNAs without an appreciable cytotoxic effects on RAW264.1 cells. The cytotoxicity of the extract using MTT assay showed the cytotoxicity of 6% and 13% against L929 cell line. Based on these results, it is concluded that the herbal mixture water extract can be applied to this acne and skin diseases therapy.

Effects of Lithospermum erythrorhizon extracts on P. acnes induced cytokine gene expression in human monocytes (자초(紫草) 추출물이 P. acnes의 단핵구 세포 사이토카인 유전자 발현에 미치는 영향)

  • Objective : This study was performed to evaluate the effect of Lithospermum erythrorhizon extracts on the inflammatory cytokines gene expression by the bacteria of Propionibacterium acnes (P. acnes) which elicits acne in human monocytes, THP-1 cell line. Experiment : Cytotoxicity of Lithospermum erythrorhizon extracts was analyzed by XTT assay. Real time RT-PCR was applied to analyze the cytokines gene expressions of IL-8, MCP-1 and TNF-$\alpha$. Translocation of transcription factor NF-${\kappa}B$ from cytoplasm into nucleus was observed using immunocytochemistry and confocal microscopy. Results : Lithospermum erythrorhizon extracts did not show cytotoxicity as high as in $1,000\;{\mu}g/ml$ of concentration. Transcription levels of inflammatory cytokines, IL-8, MCP-1 and TNF-$\alpha$ were increased by P. acnes in THP-1 and Lithospermum erythrorhizon extracts decreased the upregulated transcription levels. Lithospermum erythrorhizon extracts significantly inhibited the translocation of NF-${\kappa}B$ into nucleus by P. acnes. Conclusion : This study suggests that Lithospermum erythrorhizon extracts have anti-inflammatory effects on P. acnes treated THP-1 as decreasing the mRNA expressions of IL-8, MCP-1 and TNF-$\alpha$. This anti-inflammatory effect of Lithospermum erythrorhizon extracts may be useful in therapeutic treatments for acne vulgaris.

  • Objectives: The objective of this study was to teach patients clinically useful external application of acne. Methods: We investigated the effects and manufacturing methods of the medical herbs published in the woman's monthly magazines according to "Zhong-Yao-Da-Ci-Dian(中藥大辭典)". Results: We investigated that Saururus chinensis Baill.(三白草), Houttuynia cordata Thunb.(魚腥草), Coix lachrymajobi L.(薏苡仁) were discovered 3 times, Brassica oleracea L.(甘藍), Raphanus sativus L.(래복), Daucus carota L. var. Sativa DC.(홍래복), Oryza sativa L.(米皮糠), Spinacia oleracea L.(파래), Cucumis sativus L.(黃爪) 2 times, and Glycyrrhiza uralensis Fisch.(甘草), Gallus gallus domesticus Braisson.(?子白), Aloe vera L.(노회), Phascolus radiatus L.(綠豆), Prunus persica Batsch.(桃葉), Lycopersicon esculentum Mill.(番茄), Taraxacum mongolicum Hand.-Mazz.(蒲公英), Prunus armeniaca L.(杏仁), Musa paradisiaca L. var. Sapientum O. ktze.(香蕉) and Malus pumila Mill.(평과) were discovered once in woman's monthly magazine during last two years. According to the effects, we classificated four groups which were defervescence(淸熱), subsidence of edema(消腫), pyorrhea(排膿) and gloss(滋潤) group. Anti-inflammatory Effects of Chitosan-phytochemical Conjugates against Propionibacterium acnes-induced Inflammation (Propionibacterium acnes에 의해 유도된 염증에 대한 Chitosan-phytochemical Conjugates의 항염증 효과)

  • Propionibacterium acnes infection in skin tissue often causes acne vulgaris, commonly characterized by inflammatory papules, pustules, and nodules. Chitosan and its derivatives possess strong anti-inflammatory effects. In this study, the anti-inflammatory activity of chitosan-phytochemical conjugates on P. acnes-infected human skin keratinocytes (HaCaT) was evaluated. We designed a model of P. acnes-induced inflammation in viable HaCaT cells. Nitric oxide (NO), an inflammatory marker, was successfully elevated by P. acnes infection in HaCaT cells in a dose-dependent manner. Furthermore, the levels of NO were reduced by treatment with chitosan-phytochemical conjugates (chitosan-caffeic acid, -ferulic acid and -sinapic acid) in a dose-dependent manner. Among these conjugates, chitosan-caffeic acid exhibited the strongest NO suppression in HaCaT cells infected with P. acnes. The results obtained in this study suggest that chitosan-phytochemical conjugates could be used as a potential therapeutic agent against acne vulgaris.

Antibacterial Activity against Pathogens of Acne and Tyrosinase Inhibitory Activity of Extract and Fractions from Bark of Prunus sargentii (산벚나무 수피 추출물 및 용매 분획물의 여드름 원인균에 대한 항균활성과 Tyrosinase 저해 활성)

  • In this study, we investigated on antioxidative activity, antibacterial activity against pathogens of acne and tyrosinase inhibitory activity in 75% EtOH extract and its fractions from bark of Prunus sargen tii. The total polyphenol and flavonoidcontent of the EtOAc fraction were found to be 378.0 mg/g and 67.5 mg/g as the highest content. In the measurement of DPPH radical scavenging ability, EtOAc and BuOH fraction were exhibited stronger scavenging ability than the other fractions and 75% EtOH extract. In antibacterial activity by disc diffusion assay against pathogen of acne, antibacterial activity of the EtOAc fraction and 75% EtOH extract was stronger than the other fractions. Especially, the EtOAc fraction was the highest effective fraction in the antibacterial activity. Moreover, tyrosinase inhibitory activity of EtOAc fraction, BuOH fraction and 75% EtOH extract was higher than the other fractions. In particular, tyrosinase inhibitory activity of the EtOAc fraction showed higher activity than ascorbic acid used as positive control.

Antibacterial and Nitric Oxide Production Inhibitory Activities of Prunus sargentii Branches Extract and Its Fractions against Pathogens of Acne (산벚나무 가지 추출물 및 용매 분획물의 Nitric Oxide 생성 억제 효과와 여드름 원인균에 대한 항균활성)

  • Background: In this study, we investigated the antibacterial and nitric oxide (NO) production inhibitory activities of 75% ethanol extract of Prunus sargentii branches and its fractions against acne pathogens. Methods and Results: The antibacterial activity against acne causing pathogens was determined using the disc diffusion assay. The ethyl acetate fraction showed higher activities against Propionibacterium acnes, Staphylococcus aureus and Staphylococcus epidermidis than those shown by other fractions. In the DPPH radical and NO scavenging assays, the butanol fraction showed strong DPPH radical and NO scavenging abilities. These activities were related to the total polyphenol and flavonoid contents of butanol fraction. On the other hand, the chloroform and ethyl acetate fractions exhibited the highest NO production inhibitory activity in Lipopolysaccharide (LPS)-stimulated Raw 264.7 cells compared to those exhibited by other fractions. Conclusions: The extract and its ethyl acetate fraction from the branches of P. sargentii exhibited antibacterial activity and could be used as functional materials in antimicrobial related fields. Moreover, the chloroform and ethyl acetate fractions are potential antiinflammatory agents and butanol fraction acts as an effective radical scavenger.

  • Safflower is called as the 'beneficial flower' because 'it helps human health', and it was introduced as red flower in Tonguibogam due to the red color of floral leaf. From old times, it has been used for the material of cloth and rouge. Recently, polyphenol compound, the main ingredient of safflower, known as anti-aging and anti-oxidizing material in the healthy food industry becomes the emerging hot topic. This study aims to confirm by DDT (Disk Diffusion Test) assay, MTT assay, and NF-${\kappa}$B Luciferase activity inhibition assay in vitro that polyphenol compound, which is the main ingredient of safflower, has the anti-microbial efficacy to inhibit the growth of acne germs that make troubles for the teenagers or middle aged. Also it aims to evaluate its clinical efficacy on the acne skin, utilizing the facial cleansing cosmetic form of soap sample. This study can contribute to take a major step forward to the development of cosmetic soap for acne in the cosmeceutical industry.

An Expert Survey for Developing the Pattern Diagnosis Instrument of Acne (여드름 변증 유형 도구 개발을 위한 전문가 설문조사 연구)

  • Objective : The purpose of this study was to develop a Pattern Diagnosis Instrument of Acne in traditional Korean Medicine. Methods : Data was collected by structured survey papers from 20 professors of The Korean Oriental Medical Ophthalmology & Otolaryngology & Dermatology Society(Delphi method). We investigated importance rates of the symptoms of each Pattern in the survey papers, then calculated Weighted Value of the symptoms. Results : Nine objective symptoms and nine subjective symptoms were selected by investigated importance rate in the survey from 20 professors of The Korean Oriental Medical Ophthalmology & Otolaryngology & Dermatology Society. In Wind-Heat pattern(WHP, 風熱型), inflammatory lesion of acne was the most important symptom. Oily skin, nodule and effect of menstruation are the most important symptoms in Dampness-Heat pattern(DHP, 濕熱型), Phlegm-Stasis pattern(PSP, 痰瘀型) and Disharmony of the thoroughfare and conception vessels pattern(DTCVP, 衝任不調型) each. Conclusions : The results might provide backgrounds and methods for clinical care of acne and follow-up study.

External Application of Herbal Medicines for Acne Vulgaris: A Systematic Review and Meta Analysis

  • Aim of the study: The objective of this systematic review is to critically evaluate the evidence of the effectiveness and safety of external application of herbal medicines (EAHM) for acne vurgalis (AV). Methods: English, Chinese and Korean language databases were searched up to May 2018. Randomized clinical trials (RCTs) that reported the effects of EAHM for AV were included and analysed. Results: A total of 10 randomized trials with 656 AV patients were identified. A meta-analysis of two RCTs indicated that EAHM had a significant effect on improving primary outcome 'global assessment' compared with placebo (mean difference (MD) = -2.62, confidence interval (CI) = -4.84 to -0.40, p = 0.02). Furthermore, data extracted from two RCTs showed that EAHM significantly reduce primary outcome 'inflammatory lesion count of acne' (MD = -1.25, CI = -1.68 to -0.83, p < 0.00001) and 'non-inflammatory lesion count of acne' (MD = -1.32, CI = -1.75 to -0.90, p < 0.00001). No significant difference was observed between groups in secondary outcome 'sebum of skin' (MD = -0.21, CI = -0.53 to 0.11, p = 0.20) and 'patient-reported changes in symptom' (relative risk (RR) = 2.56, CI = 0.43 to 15.22, p = 0.30). No severe adverse events (AEs) were found and no treatment was stopped due to AEs of EAHM. Conclusions: EAHM seems to have affirmative effects, but quality of evidence, and non-standardized use of EAHM make our conclusion weak. Our suggestion is rigorously designed RCTs and standardization of EAHM are required in the future.
