Determination of Optimum Hydrolysis Conditions for Flavonoid Analysis in Plant Leaves (식물체 플라보노이드 성분 분석을 위한 적정 가수분해 조건)

  • Acidic hydrolysis conditions prior to HPLC analysis of plant flavonoids was investigated by response surface methodology (RSM), using leaves of Ginkgo biloba L. Ten hydrolysis conditions using 0.5-2.5 M HCl and 0.5-2.5 hr hydrolysis time were chosen to form a central composite rotatable design (CCRD), and optimization by RSM was achieved by measuring myricetin, quercetin, and kaempferol levels by HPLC. Optimum hydrolysis condition was 1.5 M HCl for 1.5 hr. The levels of flavonoids obtained under the condition were in good agreement with predicted maximum values, with yields of more than 95%. These optimum hydrolysis condition was applied to analysis of flavonoid content in the leaves of Camellia sinensis, Ficus carica, and Sageretia theezans.

Effective Treatment of Wastewater from the Electroplating Plant of Cold-mill by using Microorganism (냉연공장 도금공정에서 발생되는 폐수의 효율적인 미생물 처리에 관한 연구)

  • This research was carried out to establish the effective treatment condition and characteristic of wastewater from the electroplating plant of cold rolling mill by using microorganism. Alkaline wastewater and acidic heavy metal wastewater accounted for 64%, 30%, respectively, of the total wastewater. Highly concentrated thiocyanate was 53890 mg/L as COD and it was 53% of total COD, even though it was 0.03% of wastewater from the electroplating plant. When treating mixed wastewater with microorganism, it was easy to remove when SCN concentrations of mixed wastewater was 200 mg/L or less. While the treatment effect of COD-causing materials was low at the concentration of 400 mg/L or less, it implies that highly concentrated thiocyanate contains a large amount of slowly biodegradable organics. When treating with mixed wastewater, pH was 7.33 at the beginning, but after 8 hours it increased to 7.99. This is caused by ammonia which is generated when SCN of highly concentrated thiocyanate was degraded by microorganism.

The Effects of Turbidity and pH on the Removal of Cryptosporidium and Giardia by Coagulation Process (원수 탁도와 pH 변화가 혼화응집침전 과정에서 원생동물과 탁질 제거에 미치는 영향)

  • The removal of protozoa in the coagulation process was evaluated under the different pH and turbidity using the jar test after the addition of polyaluminium chloride (PAC) as a coagulant. Two well-known protozoa of Cryptosporidium parvum and Giardia lamblia were tested at the same time with turbidity, the critical water quality parameter of the water treatment process. Both protozoa were removed about 1log (and up to 2log) at the optimum injection of PAC. The source water turbidity and pH affected the removal of protozoa and turbidity. At neutral and alkaline pH, 1.3-1.7log removal of protozoa for low turbid water with 5NTU, and 1.6-2.3log removal for high turbid water with 30NTU were achieved. However, at acidic pH, maximum 0.8-1.0log and 1.1-1.2log were removed for low and high turbid water, respectively, at the optimum PAC injection of 15mg/L. The relation of protozoa and turbidity removals were expressed as the 1st order equation (significantly positive relation) in the most of the tested conditions. In addition, the relation of protozoan removals with residual turbidity were also expressed the 1st order equation (significantly negative relation), although the significance of the equations were reduced at acidic pH. Therefore, residual turbidity could be a good index of efficient protozoan removal in the coagulation process, probably except at the low pH condition.

Chemical Compositions and Antioxidant Activity of Extract from a Extruded White Ginseng (압출성형 백삼추출물의 화학적 조성 및 항산화 활성)

  • Chemical components and antioxidative activities of white ginseng, red ginseng and extruded white ginseng (EWG) were evaluated. Extrusion condition was 20% moisture content, 100 and $140^{\circ}C$ barrel temperature. The results showed that total sugar and acidic polysaccharide contents of white ginseng powder were increased after extrusion treatment of which EWG at $140^{\circ}C$ barrel temperature had higher value than EWG at $100^{\circ}C$ barrel temperature. Free radical scavenging activity of EWG at $140^{\circ}C$ barrel temperature was 80.2 and 45.6% respectively. The butanol fraction of polyphenolic compound and acidic polysaccharide were $27.2{\pm}0.1\;mg/g$ and $217.6{\pm}0.7\;mg/g$, respectively. The ginsenosides were quantified by HPLC and the yield of ginsenoside-Rg3s and Rg3r were achieved by extrusion process.

  • The advent of photonic technologies in the field of communications and data transmission has been heavily increasing the demand in integrated optical (IO) circuits capable of accomplishing not only simple tasks like signal, but also more sophisticated functions like all-optical signal routing or active multiplexing/demultiplexing. In the last decade, sol-gel technology has been widely used to prepare optical materials. Sol-gel processes show many promises for the development of low-loss, high-performance glass integrated optical circuits. However, crack formation is likely to occur during heat treatment in thick gel films. In order to overcome the critical thickness limitation, the organic-modified silicate has been widely used. In this case coating matrices have been prepared from the organo-silanes of T structures, acidic catalyst and the as-prepared gel films have been heat-treated below 200$^{\circ}C$ to avoid the crack formation and the degradation of organic components. However, the films prepared in the acidic condition and the low heat temperature make the films contain high OH groups which is the major optical loss function. In this work, C$\sub$6/H$\sub$5/SiO$\sub$1.5/ films were prepared on silicon substrate by sol-gel method using base catalyst in a PTMS/NH$_4$OH/H$_2$O/C$_2$H$\sub$5/OH system. The sol showed spinable viscosity at 50 wt% of solid content, and neglectable viscosity change with time. The films were crack-free and transparent after curing at 450 $^{\circ}C$, and highly condensed to minimize OH content in C$\sub$6/H$\sub$5/SiO$\sub$1.5/ networks. The effects of heat treatment of the films are characterized on the critical thickness, the chemical composition and the refractive indices by means of SEM, FT-IR, TGA, prism coupler, respectively.

Production of Water-Solubled Pigment from Mycelial Culture of Cordyceps scarabaeicola KEFC-C252 and Its Antimutagenic Effect (Cordyceps scarabaeicola KEFC-C252의 균사체 배양에 의한 수용성 색소의 생산과 색소의 항돌연변이 효과)

  • Cultural conditions for the production of water-soluble pigment from mycelial culture of Cordyceps scarabaeicola KEFC-C252 and antimutagenic activity of the pigment were investigated. To obtain the maximum productivity of the pigment from mycelial culture of C. scarabaeicola KEFC-C252, the optimized medium was made with 1.5% sucrose, 2.5% yeast extract and initial pH 5.5. C. scarabaeicola KEFC-C252 was cultivated to reach the maximum concentration of the pigment at $26^{\circ}C$ for 108 hrs. C. scarabaeicola KEFC-C252 produced about 1.2 g/liter pigment under the optimized condition. The pigment was isolated from the culture filtrate by ethylacetate extraction, acidic precipitation and crystallization. The isolated pigment was scarlet hexagonal column crystal, and the color of the pigment was changed according to pH of the solution. The pigment showed violet in the alkaline water but showed red color in the acidic water. The pigment showed inhibitory activity against mutagenic activity induced by 4-nitroquinoline N-oxide. Furthermore, the pigment showed inhibitory activity against spontaneous mutation on Salmonella typhimurium TA98 and TAlOO.

Interaction between Whey and Soybean Proteins (유청 및 대두 단백질의 상호작용)

  • To investigate the interaction between whey and soybean protein, thermal changes of component proteins were analyzed by column chromatography and gel electrophoresis. In the Sephadex G-200 chromatography of the mixture treated at above $80^{\circ}C$, the amount of low molecular weight proteins and high molecular aggregates were increased. This implicated that dissociation of 1ls globulin into subunits and the formation of soluble aggregates between these subunits and whey proteins that contain thiol and disulfide groups. These interaction between soy proteins and ${\beta}-lactoglobulin$, ${\alpha}-lactalbumin$, and proteose-peptone 3 were confirmed by gel electrophoresis. Bovine serum albumin, Immunoglobulin-G(H), Lactoferrin, 1ls-subunits(basic and acidic), and subunit of 7s globulin were also considered to interact each other depending on the condition of the salt solutions.

The Effect of Acidification on Membrane Distillation Process for Strong Nitrogenous Wastewater (산화 전처리가 고강도 질소폐수의 막증류 공정에 미치는 영향)

  • A direct contact membrane distillation (DCMD) was applied to treat strong nitrogenous wastewater of anaerobic digestion supernatant (ADS) and human urine (HU). The ammonia transfer was evaluated in terms of specific ammonia transfer (SAT) value, which is the ratio of total ammoniacal nitrogen divided by the amount of water transferred. The acidification resulted in low SAT values and high quality of produced water. The ammonia transfer control in the acidic condition was stronger for HU than ADS due to higher alkalinity (pH 8.8) and ammonia concentration (5700 mg-N/L) of HU. Acidified HU at pH 4 exhibited a SAT value of 1.64 × 10-5, which was significantly smaller than the SAT value of 3.00 × 10-3 for the original HU. The low pH enhanced the water flux for ADS, but HU showed a steep decrease in water flux due to enhanced fouling. It was considered that the fouling intensity in acidic conditions depends on the characteristics of the wastewater source. The major foulants on the MD membrane were NaCl, CaCO3 and CuSO4 as recognized by the SEM-EDS. Acidified ADS and HU at pH 4 showed relatively high N content of 8.18 % and 28.03 %, respectively, as organic fouling.

Antifungal Activity of EDTA and Combinatory Synergism of EDTA with Polygodial against Saccharomyces cerevisiae (Saccharomyces cerevisiae에 대한 EDTA의 항진균 활성 및 EDTA와 Polygodial 간의 병용 효과)

  • The antifungal activity of ethylenediaminetetraacetic acid (EDTA), an antioxidative food preservative, was investigated against Saccharomyces cerevisiae. At the general condition for antifugal test, EDTA exhibited the minimum inhibitory concentration (MIC) of 200 ${\mu}g/ml$ and the minimum fungicidal concentration (MFC) of 6,400 ${\mu}g/mL$, As seen with the antibacterial activity, the antifungal activity of EDTA was greatly decreased by high inoculum size, acidic medium, and cation ($Ca^{++},\;or\;Mg^{++}$) added to medium. On the other hand, when EDTA was combined with polygodial isolated from natural food spice, they showed strong synergism on the antifungal activity. Further, the diminishing antifungal activity of EDTA by high inoculum size, acidic medium, and cation ($Ca^{++}\;or\;Mg^{++}$) added to medium was considerably improved by the combination with polygodial.

  • Multicrystalline silicon wafers were textured in an alkaline bath, basically using sodium hydroxide and in acidic bath, using mainly hydrofluoric acid (HF), nitric acid $(HNO_3)$ and de-ionized water (DIW). Some wafers were also acid polished for the comparative study. Comparison of average reflectance of the samples treated with the new recipe of acidic solution showed average diffuse reflectance less than even 5 percent in the optimized condition. Solar cells were thus fabricated with the samples following the main steps such as phosphorus doping for emitter layer formation, silicon nitride deposition for anti-reflection coating by plasma enhanced chemical vapor deposition (PECVD) and front surface passivation, screen printing metallization, co-firing in rapid thermal processing (RTP) Furnace and laser edge isolation and confirmed >14 % conversion efficiency from the best textured samples. This isotropic texturing approach can be instrumental to achieve high efficiency in mass production using relatively low cost silicon wafers as starting material.
