Purification and Biochemical Properties of Extracellular Phospholipase $A_1$ from Serratia sp. MK1

  • A novel type of extracellular phospholipase $A_1$ was isolated from Serratia sp. MK1 and purified to homogeneity by ammonium sulfate precipitation, anion exchange and gel filtration chromatography. The purified enzyme was a monomer with a molecular mass of about 43, 000 Da. This enzyme showed the highest lipolytic activity toward phosphatidylserine among the phosphoglycerides tested, and preferentially catalyzed the hydrolysis of the ester bond in phosphatidic acid to lyso-phosphatidic acid. Enzyme activity was completely inhibited by the addition of a chelating agent such as EDTA, and inhibited enzyme activity was fully recovered by the presence of $Ca^{2+}$. This implies that the enzyme requires $Ca^{2+}$ for activity. The enzyme was stable up to $70^{\circ}C$ when incubated for 1 h at pH 8.5, and the optimal pH and temperature were 8.5 and $50^{\circ}C$, respectively.

  • Cephalexin synthesizing enzyme (${\alpha}$ amino acid ester hydrolase) was partially purified from the culture broth of Acetobacter turbidans ATCC9325 through ammonium sulfate fractionation, DEAE, CM, and Sephacryl S-200 gel filtration. The enzyme has optimum pH 6.0 and temperature, 40$^{\circ}C$ respectively. From the analysis of reaction mixtures by thin layer chromatographic and high performance liquid chromatographic techniques, it was confirmed this enzyme catalyzed simultaneously the following reactions : 1) Synthesis of cephalexin from D-${\alpha}$-phenylglycine methylester (PGM) and 7-amino 3-deacetoxy-cetoxycephalosporanic acid (7-ADCA) 2) Hydrolysis of cephalexin to form 7-ADCA and phenylglycine (PG) 3) Hydrolysis of PGM to form PG and methanol. Base on the above experimental observations, the reaction model of this enzyme was identical with that of the enzyme from Xanthomonas citri.

Characterization of Glutaryl 7-ACA Acylase from Pseudomonas diminuta KAC-1

  • The glutaryl 7-aminocephalosporanic acid (glutaryl 7-ACA) acylase was purified from Pseudomonas diminuta KAC-1 cells isolated from soil, and characterized. The acylase was purified by procedures including ammonium sulfate fractionation and column chromatographies on DEAE-Sepharose, Phenyl-Sepharose, Q-Sepharose, and Superose 12H/R. The negative acylase was found to be composed of two subunits with molecular masses of approximately 55 kDa and 17 kDa, respectively. The isoelectric point of the enzyme was 4.0. The specific activities of the purified acylase were 8.0 and 7.0 U/mg on glutaryl 7-ACA and glutaryl 7-aminodesacetoxy cephalosporanic acid (glutaryl 7-ADCA), respectively, and $K_m$ values were 0.45 mM for glutaryl 7-ADCA and 0.67 mM for glutaryl 7-ADCA. The enzyme had a pH optimum at 8.0 and a tmperature optimum at $40^{\circ}C$. The acylase catalyzed the synthesis of glutaryl 7-ACA from glutaric acid and 7-ACA as well as the hydrolysis of glutaryl 7-ADCA, although the reaction rate of the synthesis was slower than that of the hydrolysis. In addition, it was found that the enzyme had a glutaryl transferase activity, thereby transferring the glutaryl group from one cephalosporin nucleus to another.

Stability of pipethanate hydrochloride in aqueous solution

  • In the present investigation, an attempt has been made to apply the methods of classical chemical kinetics to the hydrolytic reaction of pipethanate hydrochloride. By successively keeping all but one variable essentially constant, it has been possible to resolve the overall effect of the individual contributing factors. Since nearly all commercial pipethanate preparations are formulated with antacid, studies were made at several constant hydrogen ion concentration ranging pH 0.4 to 7.5. Rate measurement was also carried out in temperature ranging from $25^{\circ}C$ to $60^{\circ}C$. The hydrolysis of pipethanate is found to be of first order with respect to pipeethanate concentration over an experimental range of hydrogen ion concentration (pH 0407.5). The apparent activation energy(Ea) at pH 7.5 is 18.30 Kcal/mole and the frequency factor is $1.1408 {\times}10^{9}sec^{-1}$. The rate of the hydrolysis has a minimum at pH 2.5-3.5. In this region the half-life of pipethanate was about15.3 days at $60^{\circ}C$. The catalytic effect of water was found to be $K_{H_2O}$ = $3.16{\times}10^{-5}min^{-1}$ at $60^{\circ}C$. The catalytic constants of the hydroxyl ions and hydrogen ions at $60^{\circ}C$ were also found to be $K_{OH}$ = $4.5519{\times}10^{-5}min^{-1}$ and $K_{H}+$ = $1.1568{\times}10^{-2}min^{-1}$, respectively. This reaction appears to be primarily base catalyzed hydrolysis and pipethanate is relatively reluctant toward acid catalyzed hydrolysis. A positive primary salt effect was noted in the solution of phpethanate at pH 7.5 and at $60^{\circ}C$.

  • The hydrolysis rate of insecticidal buprofezin(IUPAC : tert-butylimino-3-isopropyl-5-phenylperhydro-1,3,5-thiadiazin-4-one) in the range of pH 2.0 and 12.0 have been examined in 15%(v/v) aqueous dioxane at $45^{\circ}C$. The hydrolysis mechanism of buprofezin is proposed from the pH-effect, solvent effect(${\ell}{\gg}m$), thermodynamic parameter(${\Delta}H^{\neq}$=11.12 $Kcal{\cdot}mol^{-1}$ &, ${\Delta}S^{\neq}=5.0e.u.$), rate equation and hydrolysis product, l-isopropyl-3-phenyl urea. General acid catalyzed hydrolysis and specific acid catalyzed($k_{H3O+}$) hydrolysis through $A-S_{E}2$ and A-2(or $A_{AC}2$) reaction mechanism with orbital-control reaction proceed below pH 8.0 and above pH 9.0, the nucleophilic addition-elimination, $Ad_{N}-E$ mechanism via tetrahedral($sp^{3}$) intermediate is initiation by general base catalyzed($k_{H2O}$) reaction. Buprofezin was more stable in alkaline ($k=10^{-8}sec.^{-1}$) than acid solutions from the sigmoid pH-rate profile. And the half-life($t=\frac{1}{2}$) of hydrolysis reaction in neutral aqueous solution(pH 7.0) at $45^{\circ}C$ was about 3 months.

  • In this study, biosugar production by the red-algae Eucheuma cottonii was investigated using dilute sulfuric acid-catalyzed hydrolysis and data analysis by response surface methodology. This approach yielded 25.8 g/l total reducing sugar under the conditions of $160.1^{\circ}C$, 1% (v/v) sulfuric acid, and 13.1 min. The sugar concentration showed a linear inverse correlation with the combined severity factor (CSF) of the reaction conditions. CSF was calculated as $log(t{\cdot}e{xp}[(T_H-T_R)/14.75])-pH$, where t is the coupling reaction time, $T_H$ is the target temperature, and $T_R$ is the reference temperature ($100^{\circ}C$). In addition, levulinic acid production showed a linear positive correlation with CSF. E. cottonii may represent a useful feedstock for sugar production in the field of bioenergy.

Comparison of Characteristics of Acid-catalyzed Hydrothermal Fractionation for Production of Hemicellulose Hydrolyzate from Agricultural Residues (농경잔류물로부터 헤미셀룰로오스 가수분해물 생산을 위한 산촉매 열수 분별공정의 특성 비교)

  • The objective of this work was to investigate the feasibility of acid-catalyzed hydrothermal fractionation for maximum solubilization of the hemicellulosic portion of two typical agricultural residues. The fractionation conditions converted into combined reaction severity (CS) in the range of 1.2-2.9 was used to establish a simple reaction criteria at glance. The hemicellulosic sugar yield of 56.6% was shown when rice straw was fractionated at the conditions at the conditions; 160 ℃ of temperature 0.75% (w/v) of H2SO4, 20 min of reaction time, 1:15 solid/liquid ratio. The hemicellulosic sugar yield of 83.0%, however, was achieved when barley straw was fractionated at the conditions at the conditions; 150 ℃ of temperature 0.75% (w/v) of H2SO4, and 15 min of reaction time, 1:10 solid/liquid ratio. For barley straw, acid-catalyzed hydrothermal fractionation could be effectively performed. After the fractionation process, the remaining fractionated solids were 48.5% and 57.5% from raw rice and barley straws, respectively. The XMG contents in the solid residues decreased from 17.3% and 17.6% to 6.0% and 2.6%, which corresponded to 16.7% and 8.5% on the basis of the raw straws, respectively. In another way, only 5.6% of cellulose and 8.5% of XMG were lost due to excessive decomposition during the acid-catalyzed hydrothermal fractionation of barley straw, compared to cellulose and XMG losses of 6.4% and 26.6% in rice straw. Hemicellulosic sugars from the rice straw were considered more over-decomposed due to the somewhat higher reaction severity at the acid-catalyzed hydrothermal fractionation.

  • For the development of new synthetic method for unnatural amino acid esters, N-aryl phenylglycine Ο-alkyl esters 4a∼i were synthesized through base-catalyzed hydrolysis of 1,5-diphenylhydantoins 1a∼b and Ο-alkylation in 16∼87% yield. An efficient and practical route for final 4a∼i was that the starting materials 1a∼b were heated in dil-methanol for 30 minute using sodium hydroxide or potassium hydroxide and evaporated. In addition, reaction mixture were refluxed for 1 h in DMF. All synthetic process from hydantoin to N-aryl phenylglycine Ο-alkyl esters 4a∼i could be carried out in one-pot without isolation of intermediates.

  • EHT and CNDO/2 calculations have been performed to determine conformations of acetamides and diacetamides, and of their protonated forms. Results show that: protonation is always favored on the cis position with respect to N due to greater attractive potential between $H^+$ and N; the trans-trans conformer of diacetamides gives the most preferred protonated form although the cis-trans conformer is the most stable one for the unprotonated diacetamides. Protonation on a carbonyl oxygen is predicted to increase both charge and orbital controlled $S_N$SN reactivities of the protonated carbonyl carbon due to increases in positive charge and AO coefficient of ${\pi}$-LUMO of the carbon atom. In the acid catalyzed hydrolysis of diacetamides therefore it appears highly probable that the rate determining attack by a water molecule occurs at the carbon of the protonated carbonyl group and the carbonyl carbon-nitrogen bond scission follows subsequently. This mechanism is consistent with that generally accepted for the hydrolysis of amides in dilute acid solution but disagrees with that proposed by Laureut et al., for acid hydrolysis of N-acetyl-lactams.

Purification and Characterization of Lipase from Trichosporon sp. Y-11and Its Use in Ester Synthesis of Unsaturated Fatty Acids and Alcohols

  • A 28-kDa extracellular lipase (pI 8.7) was purified to homogeneity from the culture supernatant of Trichosporon sp. Y- 11 by mmonium sulfate precipitation, DEAE-Sephadex A-50, Bio-Gel P-30, CM- Sephadex C-50, and Bio-Gel P- 10 chromatographies. The purified enzyme exhibited a specific activity of $2,741{\;}{\mu}mol/min/mg$ based on the hydrolysis of triolein, and the optimal hydrolysis activity was dentified at pH 8.0 and $40^{\circ}C$. The enzyme activity was inhibited by $Ag^+$ and enhanced by $Fe^{2+}$, $Fe^{3+}$, $Mg^{2+}$, $Mn^{2+}$, and $Li^{+}$. The enzyme activity exhibited for the hydrolysis of both tributyrin and trilinolein. The ester synthesis of unsaturated fatty acids with various alcohols catalyzed by the purified lipase in a nonaqueous medium or microaqueous system was also investigated. The esterification activity of the lipase increased with an increase of the carbon chain length in the alcohol. The synthesis rate of linoleic acid and oleyl alcohol was the highest with an optimal temperature and pH of $40^{\circ}C$ and 8.0, respectively. The water content and agitation also affected the esterification activity of the lipase.
