Anti-stress Effect of Cholic acid Derivatives in Restraint Stress Induced Rats (구속스트레스를 가한 흰쥐에서 Cholate류의 항스트레스 작용에 관한 연구)

  • This study was done to investigate whether cholic acid derivatives have anti-stress activity and what is a cause of this anti-stress effect. Seven cholic acid derivatives (cholic acid, taurocholic acid, ursodeoxycholic acid, tauroursodeoxychoic acid, chenodeoxy cholic acid, dehydrocholic acid, hyodeoxycholic acid) were used, silymarin and valproic acid were used as positive controls. Stress was induced by restraint immobilization technique plus water immersion (24hrs) and adrenal weight, spleen weight, adrenal ascorbic acid, serum cholesterol, lactate dehydrogenase (LDH), alkaline phosphatase (ALP), adrenal cholesterol, glucose and corticosterone levels were measured as stress indicators. Most cholic acid derivatives markedly decreased the adrenal weight, and TUDCA and DHCA increased the spleen weight. The restraint stress induced increments in serum LDH, ALP and cholesterol were attenuated by most cholic acid derivatives. Cholic acid, taurocholic acid and tauroursodeoxycholic acid only increased the content of adrenal ascorbate. While valproic acid showed an inhibitory effect against stress, silymarin did not. Our findings suggest that most cholic acid derivatives have anti-stress effect and that their anti-stress effect is, in part, related to choleretic activity.

The Comparative Studies for the Anti-stress Effect of Guibitang and Ascorbic Acid on Heating and Swimming Stress (열(熱)Stress 및 유영(游泳)Stress에 대(對)한 귀비탕(歸脾湯)과 Ascorbic Acid의 항(抗)Stress효능(效能) 비교연구(比較硏究))

  • In order to compare the anti-stress effect of Guibitang with that of ascorbic acid, after these medicines were administered to guinea pigs induced by heating and swimming stress, the changes of the weight of body and organ, and content of plasma catecholamines, serum total cholesterol, free cholesterol, triglyceride, protein, glucose and cortisol were measured. The results were as follows : 1. The weight of the body was increased with statistical significance in the groups administered ascorbic acid and ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid on both heating and swimming stress. 2. The weight of spleen decreased with statistical significance in the groups administered ascorbic acid, Guibitang and ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid on heating stress, but in case of swimming stress, the weight of spleen decreased with statistical significance in the groups administered ascorbic acid and ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid. The weight of adrenal decreased with statistical significance in the groups administered ascorbic acid and ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid on heating stress alone. 3. Plasma norepinephrine content decreased with statistical significance not only in the groups administered ascorbic acid and ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid but in the group administered ascorbic acid with Guibitang as compared with in the group administered ascorbic acid on heating stress. In case of swimming stress, norepinephrine decreased with statistical significance in the groups administered ascorbic acid, Guibitang and ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid. 4. Plasma dopamine content decreased with statistical significance only in the group administered ascorbic acid and ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid on both heating and swimming stress. 5. Serum total cholesterol content decreased with statistical significance in the groups administered ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid on heating stress, but in case of swimming stress, it decreased with statistiscal significance only in the group administered ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid. 6. Serum triglyceride content decreased with statistical significance not only in the groups administered ascorbic acid, Guibitang and ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid but in the group administered ascorbic acid with Guibitang as compared with in the group adminstered ascorbic acid on heating stress. In case of swimming stress, triglyceride decrease with statistical significance only in the group administered ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid. 7. Serum glucose content increased with statistical significance in the groups administered Guibitang and ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid on both heating and swimming stress, particulaly in case of swimming stress, in the group administered ascorbic acid with Guibitang, it increased with statistical significance as compared with in the group administered ascorbic acid. 8. Serum cortisol content decreased with statistical significance only in the group administered ascorbic acid and ascorbic acid with Guibitang as compared with in the group administered non ascorbic acid on heating stress.

A Study on Anti-Stress Activities of Cholic Acid Derivatives (담츱산류의 항스트레스 효능에 관한 연구)

  • This study was done to investigate whether cholic acid derivatives have anti-stress activity in various stress models. Two cholic acid derivatives, ursodeoxycholic acid (UDCA) and tauroursodeoxycholic acid (WDCA), were used. physical, psychological, chemical and environmental stress models were performed. Adrenal weight, serum glucose levels and ALP activity were elevated in restraint stress model, but this elevation was prevented by UDCA treatment. Moreover, UDCA and TUDCA inhibited exploratory and spontaneous movements in oscillation stress model. In alcohol-induced stress model, TUDCA improved rotarod performance. UDCA and TUDCA significantly reduced the involution of lymphoid organs and the increment of WBC counts in cold stress model. These findings suggest that choric acid derivatives have antistress effects in various stress models.

  • This study aimed to compare the anti-stress effect of Guibitang with Ascorbic Acid on the guinea pigs in heat or forced swimming stress. the experimental animals were heated in hot room$(34{\pm}2^{\circ}C)$ for 2 hours in a day during 7 days. Forced swimming stress was loaded by forcing guinea pigs to swim in a oval tub for 2 minutes in a day during 7days. And administered 1.0mg/100g of Ascorbic Acid or 86.0mg/100g of Guibitang extract formonce befor stress. 1. In brain, the contents of norepinephrine in group of the heat of forced swimming stress, in case administered Ascorbic Acid and in case administered Guibitang extract with Ascorbic Acid significantly decreases as compared with these in case administered non Ascoric Acid. 2. In brain, the contents of epinephrine in group of the forced swimming stress, in case administered Guibitang extract with Ascorbic Acid significantly decreases as compared with these in case administered non Ascorbic Acid. 3. In brain, the contents of dopamine in group of the heating stress, in case administered Guibitang extract with Ascorbic Acid significantly decreases as compared with these in case administered non Ascorbic Acid, and these in group of the forced swimming stress in case administered Ascorbic Acid and in case administered Guibitang extract without Ascorbic Acid and in case administered Guibitang extract with Ascorbic Acid significantly decreases as compared with these in case administered non Ascorbic Acid.

Global Analysis of Gene Expression upon Acid Treatment in Arabidopsis thaliana

  • To obtain global gene expression profiles of Arabidopsis thaliana by acid stress, seedlings were subjected to low pH stress. Using Affymetrix AH1 chips covering 24,000 genes, we analyzed gene expression patterns. Fifty-four genes were up-regulated, and 38 were down-regulated more than 3-fold after 2 h of acid stress (pH 3.0). Several defense and abiotic stress-related genes were recognized among the up-regulated genes and peroxidase and extensin genes were identified among the down-regulated genes. After 12 h treatment, relatively fewer genes showed changed expression, indicating that plants seem to adjust themselves to this abiotic stress. Most of the up-regulated genes are already known to be involved in abiotic stress responses and pathogen attacks, especially wounding. However, down-regulated genes for the members of extensins and peroxidases are specific to the acid treatment. These results suggest that acid treatment turns on genes involved in stress responses, especially in wounding and turns off genes very specific for the acid stress.

Anti-stress Effects of Ursodexycholic Acid on the Restraint Stress in Rats (흰쥐에서 구속스트레스에 대한 우루소데옥시콜린산의 항스트레스 효과)

  • Effects of restraint stress and its modulations by ursodeoxycholic acid(UDCA) were evaluated on some biochemical and biophysical parameters in rats. Restraint stress induced elevations in blood alkaline phosphatase (ALP). cholesterol (CHOL), aspartate transaminase (GOT), alanine transaminase (GPT), lactate dehydrogenase (LDH) levels. It was also caused adrenal hypertrophy, decrease in weight of spleen and contents of ascorbic acid in stressed rats. As a results, stress indicators such as spleen, ascorbic acid, GOT, GPT, LDH were fastly changed after imposing stress, but those such as ALP, CHOL, adrenal were induced relatively later. UDCA was tested if it has an inhibitory effect against 18-hr restraint induced stress. UDCA lowered ALP, CHOL, LDH level and also effectively elevated the ascorbic acid contents in 25 mg/kg dosage of UDCA. In organ weights. the restraint stress induced increases in spleen and adrenal were attenuated by UDCA in 50 mg/kg dosage. However. stress-induced GOT and GPT levels were unaffected by UDCA.

Physiological Responses of Oxygen-Tolerant Anaerobic Bifidobacterium longum under Oxygen

  • In order to investigate what kind of response anaerobic bifidobacteria has on oxygen stress, five oxygen-tolerant bifidobacteria were isolated from human fecal samples. All were temporarily identified as Bifidobacterium longum through an analysis of carbohydrate utilization patterns and cellular fatty acid profiles. In the presence of oxygen, the lag phase became extended and the cell growth was suppressed. Bifidobacterial cell was able to remove dissolved oxygen in an early stage of growth and to overcome oxygen stress to a certain extent. The cell became long n size and showed a rough surface containing many nodes which were derived from abnormal or incomplete cell division. Cellular fatty acid profiled changed remarkably under a partially aerobic condition, so that the carbon chain of cellular fatty acid became short. All the dimethyl acetals originated from plasmalogen were reduced, any cyclopropane fatty acid, 9, 10-methyleneoctadecanoic acid ($C_{19:0}cyc9,10$), was increased remarkably. Oxygen stress induced a 5.5 kD protein in B. longum JI 1 of the oxygen-teolerant bifidobacteria, that was named Osp protein, and its N-terminal amino acid sequence was as follows: unknown amino acid-Thr-Gly-Val-Arg-Phe-Ser-Asp-Asp-Glu. Therefore, the oxygen-tolerant bifidobacteria seemed to defend against oxygen stress byincreasing the content of short fatty acid and cyclopropane fatty acid, and induction of an oxygen stress protein, but not the plasmalogen.

  • In this study, we are interested in comparing the protein profiles of acid-shocked and control cells of S. mutans isolated from Korean children with caries. The results of 2D gel electrophoresis showed that twelve proteins are up-regulated when the cells were grown under 20 mM lactic acid stress in the exponential phase. Up-proteins under acid stress were estimated a major key of the survival and proliferation of S. mutans in low pH environments. These proteins are estimated generally associated with three biochemical pathways: glycolysis, alternative acid production and branched-chain amino acid biosynthesis.

  • Glutathione and ascorbic acid have been shown to fulfill many essential functions in animal and plant growth, development, defence and protection against oxidative damage. Effects of glutathione and ascorbic acid were examined in transgenic N. tabacum cells producing hGM-CSF to determine the effects of the vitamins on growth and cell viability. In lag phase, cell viability was preserved by glutathione and ascorbic acid. Therefore, recombinant protein productivity was increased. The purpose of present study is to investigate the role of antioxidants in cold stress-induced apoptosis in plant suspension cells. Cold stress lowered cell viability and increased total genomic DNA fragmentation. Supplementing the cell cultures with glutathione and ascorbic acid inhibited cold stress-induced decrease in cell viability and increase in total genomic DNA fragmentation.

  • Nonalcoholic steatohepatitis(NASH) is serious metabolic disease related to fatty acid. According to "two hit theory", fatty acid-induced oxidative stress is important factor to progress nonalcoholic steatohepatitis from steatosis. In this study, we evaluated stearic acid induced oxidative stress in human hepatocarcinoma SK-Hep-1 cell. Cell viability, reactive oxygen species (ROS) production, glutathione (GSH), malondialdehyde and expression of antioxidant enzymes were determined at various time-points and concentrations of stearic acid. At 0.2 mM, non-toxic concentration, of stearic acid, production of ROS was significantly increased at 24 hours and the level of GSH was significantly decreased. Expression of superoxide dismutase-1 and 2 was slightly increased in 0.2 mM stearic acid at 24 hours. These results represent that the non-toxic concentration of stearic acid resulted in oxidative stress, suggesting that stearic acid may play a critical role in development of steatohepatitis.
