Cloning, Expression, and Characterization of UDP-glucose Pyrophosphorylase from Sphingomonas chungbukensis DJ77

  • The bacterium Sphingomonas chungbukensis DJ77 produces the extracellular polysaccharide gellan in high yield. Gellan produced by this bacterium is widely used as a gelling agent, and the enzyme UDP-glucose pyrophosphorylase (UGP) is thought to play a key role in the gellan biosynthetic pathway. The UGP gene has been successfully cloned and over-expressed in E. coli. The expressed enzyme was purified with a molecular weight of approximately 32 kDa, as determined by a SDS-polyacrylamide gel, but the enzyme appears as ca. 63 kDa on a native gel, suggesting that the enzyme is present in a homodimer. Kinetic analysis of UDP-glucose for UGP indicates $K_m$ = 1.14 mM and $V_{max}$ = 10.09 mM/min/mg at pH 8.0, which was determined to be the optimal pH for UGP catalytic activity. Amino acid sequence alignment against other bacteria suggests that the UGP contains two conserved domains: An activator binding site and a glucose-1-phosphate binding site. Site-directed mutagenesis of Lys194, located within the glucose-1-phosphate binding site, indicates that substitution of the charge-reversible residue Asp for Lys194 dramatically impairs the UGP activity, supporting the hypothesis that Lys194 plays a critical role in the catalysis.

Antitumor Activities of Red Ginseng Acidic Polysaccharide(RGAP) as an Immunomodulator

  • A red ginseng acidic polysaccharide(RGAP) with immunomodulating antitumor activities was isolated from Korean red ginseng, The molecular weight of RGAP was estimated to be 12-450 kDa by gel filtration chromatography, RGAP was found to increase survival rate and to inhibit of tumor growth significantly in a dose dependent manner in mice transplanted with tumor cells. RGAP significantly promoted nitric oxide(NO) production from peritoneal macrophages bothin vivo and in vitro. Western blot analysis exhibited a newly synthesized inducible nitric oxide synthase(iNOS) protein band in the RGAP treated group. It seems likely that immunomodulating antitumor activities of RGAP are mainly mediated by NO production of macrophage. RGAP was further purified by ultrafiltration and anion exchange chromatography on DEAE-sepharose, followed by gel filtration on Sephacryl S-300 to give an active fraction(GFP) with stronger NO production in murine macrophages. GFP increased survival rate ten times compared to RGAP in male ICR mice transplanted with sarcoma 180 and also showed more potent tumoricidal activities of natural killer cells than RGAP. Sugar $composition(mol\%)$ of GFP was found to be arabinose:rhamnose:xylose:galacturonic acid:mannose:galactose:glucose(10:9:1:25:8:20:27) by GC/MS. The results suggest that clinical trials of RGAP in immunotherapy against cancer are highly feasible.

The Physiological Activity of Crude Polysaccharide Solvent Extracted from Herbal Medicine Mixture (생약복합물 용매추출물과 조다당획분의 생리활성)

  • After ethanol (BM-E and RW-E) and hot-water (BM-HW and RW-HW) extracts were fractionated from two herbal mixtures (BM and RW), their physiological activities were investigated. All extracts consisted of more than 50% of neutral sugar, with their total polyphenol levels higher than flavonoid levels. Radical scavenging activities of EtOH extracts remained significantly higher compared to that of hot-water extracts, and in particular, RW-E showed consistently higher antioxidant activity than BM-E. When anti-inflammatory activities of the extracts were evaluated by LPS-stimulated RAW 264.7 cells at 10~500 μg/mL non-cytotoxicity doses, BM-E showed significantly higher levels of TNF-α, IL-1β, IL-6, and nitric oxide inhibitory activity than those of hot-water extracts and RW-E. Murine peritoneal macrophage cells were shown to be enhanced in crude polysaccharides (BM-CP and RW-CP fractionated from BM-HW and RW-HW) compared to hot-water extracts and polysaccharide K (PSK, positive control). Especially, RW-CP exhibited higher activity than BM-CP, and component sugar analysis showed that BM-CP mainly contained galacturonic acid, glucose, arabinose, galactose, and xylose (34.5%, 33.9%, 16.1%, 7.1%, and 6.3%, respectively), whereas RW-CP showed different measurements (29.5%, 59.2%, 5.0%, 4.5%, and 0.2%). In conclusion, two herbal mixtures could contain varying sets of physiological activities dependent on different extraction and fractionation methods.

  • An innovative biopolymer known as the Rhizobian gum has been developed in France, which shows some dramatic refreshing effect on the skin. The origin of this innovative project takes its source in the natural environment, and in particular the natural environment of the roots of sunflowers and wheat, where a symbiotic bacterium has been discovered. It is a Rhizobium bacterium, which is hosted by the roots, and which is able to synthesize a specific polymer showing a dramatic water binding capacity. This polymer is in particular synthesized in period of drought, and its biological role is to concentrate the small amount water present in the soil in order to take it available for the root, which becomes then able to absorb it. This vital mechanism allows the plant to survive despite a severe climatic environment. This basic research has been conducted in collaboration whit the French National centre of scientific Research (CNRS), and has lead to the isolation of the Rhizobium bacteria. Rhizobian gum is a branched biopolymer consisting in the repetition of a polysaccharide unit of 3 molecules of glucose, 3 molecules of galactose and 1 molecule of glucuronic acid, whit one pyruvate group an average 1.6 acetyl groups. The fresh effect of Rhizobian gum is a strong sensorial impact that 100 % of the consumers are able to perceive, and which is judged very pleasant by most of them. In addition to this, a large majority of consumers are perceived, and which is judge very pleasant by most of them. In addition to this, a large majority of consumers also feel a very pleasant relaxing sensation. Smoothness and softness are also felt by most consumers and qualified positively by most of them. These qualities guarantee a strong impact on today's consumers.

The analysis of useful components in Flammulina velutipes fruit body, Flammulina velutipes mycelium and Cordyceps militaris mycelium (팽이버섯 자실체, 팽이버섯 균사체 및 동충하초 균사체의 유용성분 분석)

  • Flammulina velutipes fruit body, Flammulina velutipes mycelium and Cordyceps militaris mycelium were analyzed for their proximate composition, protein-bound polysaccharide, nucleic acid and amino acids. The content of ash and crude fiber in F. velutipes fruit body were higher than F. mycelium and C. militaris mycelium. C. militaris mycelium showed the highest crude fat content while F. velutipes fruit body had lowest. Nitrogen free extract content of the samples varied from 56.8% in F. velutipes fruit body to 61.9% in F. velutipes mycelium. The compositions of total protein and total free sugars of protein-bound polysaccharide were found to be significant differences for all samples. Nucleic acid related compounds were identified the 5'-GMP, 5'-XMP, 5'-IMP in all samples. The content of total nucleic acids were high in the orders of F. velutipes myclial (286.71 mg%), F. velutipes fruit body(187.36 mg%) and C. militaris mycelial(76.85 mg%). The highest content of 5'-GMP was found in F. velutipes fruit body. The most nucleic acid of F. velutipes mycelial and C. militaris mycelial were the 5'-XMP. As for the analysis of total amino acids, seventeen amino acids were identified by HPLC and the major amino acid was glutamic acid in all samples. The content of total amino acids were high in the orders of F. velutipes fruit body(19,919 mg%), F. velutipes mycelium(19,018 mg%) abd C. militaris mycelium(18,965 mg%). We determined the developing new food product such as amino acid drink and amino acid containing food using extracts of Flammulina velutipes fruit body, Flammulina velutipes mycelium and Cordyceps militaris mycelium.

Immune Enhancement of Polysaccharide from Submerged Culture with Phellinus linteus in the Medium Supplemented with Ginseng Extract (수삼추출물 첨가 혼합배지에서 조제된 상황 균사체 심부배양물 다당획분의 면역활성 증진)

  • Crude polysaccharide (CP) was fractionated from the submerged culture (containing both mycelia and culture broth, SC) with Phellinus linteus (PL) in mushroom complete medium (MCM) supplemented with ginseng extract ($65^{\circ}$Bx, GE) to enhance the immune activity. PL-GE-15-CP from SC cultivated in MCM supplemented with GE-15% (v/v, a ratio of MCM volume to GE) showed significantly higher macrophage stimulation (1.45 fold of the saline control at $100{\mu}g$/mL) than PL-GE-5 and 10-CP with GE-5 and 10%, or PL-CP from SC without GE. The potent intestinal immune system modulating activity through Peyer's patch was also obtained by PL-GE-15-CP (1.46 fold). When PL-GE-15-CP further fractionated on DEAE-Sepharose CL-6B (Cl- form), PL-GE-15-CP-II was the significantly higher than others from PL-GE-15-CP or PL-CP on macrophage stimulation, interleukin (IL)-12 production and intestinal immune system modulation (1.54, 3.96 and 1.56 fold, respectively). PL-GE-15-CP-II also had higher anti-metastatic activity against colon 26-M3.1 carcinoma cell (57.3% inhibition of tumor control, $200{\mu}g$/mouse) rather than PL-CP-II. This active fraction (PL-GE-15-CP-II) mainly contained neutral sugar (82.45%) and uronic acid (12.99%), and component sugar analysis showed that PL-GE-15-CP-II consisted mainly of uronic acid, Ara, Man, Gal and Glc (molar ratio of 0.52:0.97:0.63:1.00:0.54). Furthermore, the activity of GE culture was higher compared with culture without GE, indicating that GE helped to enhance the immune activity of P. linteus; also, it is assumed that the polysaccharide plays an important role in immune enhancement.

Physicochemical Properties of Rice Kernels Affected on Palatability (식미에 영향을 미치는 쌀 곡립의 이화학적 특성)

  • This study was investigated the relationship between palatability and physicochemical properties of rice kernels from two rice cultivars, including Gopumbyeo and Palgongbyeo. Major fatty acids of rice were oleic(35% in Gopum, 33.2% in Palgong, respectively) and linoleic acids (42.9% in Gopumbyeo, 40.7% in Palgongbyeo, respectively). There were significant differences in composition of amino acids and content of sugar from non-starch polysaccharide between two rice cultivars. However, no difference was found in mineral contents between Gopumbyeo and Palgongbyeo. In Rapid Visco Analyzer examination, pasting temperature and breakdown of Gopumbyeo were lower than those of Plagongbyeo. In X-ray diffraction patterns of starches separated from two rice cultivars, traditional A type and there was no difference in crystalline of rice starch. We could also obtain the results that Goupumbyeo, known to have good eating quality, had higher palatability value than Palgongbyeo.

Changes in the Non-cellulosic Monosaccharides of Cell Wall Polysaccharides of Persimmon Fruits during Maturation and Postharvest (감과실의 성숙과 추숙중의 세포벽 다당류의 비섬유성 단당류의 변화)

  • Glucose, arabinose and total non-cellulosic neutral sugar contents of alcohol-insoluble substance were increased during maturation of persimmon, but arabinose, galactose and total non-cellulosic neutral sugar contents were decreased in soft persimmon. The main non-cellulosic neutral sugars of cell wall were galactose, arabinose and glucose. Arbinose and galactose contents were decreased during maturity and this tend was remarkable in soft persimmon. Pectic fraction contained $70{\sim}82%$ of uronic acid, and galactose, arabinose and uronic acid of pectic fraction were decreased. The main non-cellulosic neutral sugars of hemicelluloses were glucose, xylose, and galactose. Galactose was decreased during maturation and postharvest, and contents of non-cellulosic neutral sugar were decreased in soft persimmon.

  • As a separation membrane for water-ethanol mixtures, alginic acid has been used which is hydrophilic polysaccharide and has excellent bonding capacity with divalent metal ions. Pervaporation characteristics of the alginic acid membrane were examined. The membrane was crosslinked with metal ions for the improvement of mechanical strength and chemical affinity. And its pervaporation characteristics were investigated. The first group(I A) metal complexed membrane cannot be used because of their brittleness and excessive swelling in low concentrations of ethanol solution. But the permeation characteristics of other metal complexed membrane were more improved than that of the alginic acid membrane because of their contraction of the membrane and hydrophilic property of metal ion.
