The aim of the present study was to assess the anti-obesity effects of fermented kochujang supplement in C57BL/6J mice. Thirty mice were divided into 3 groups; normal diet control group (ND), high fat diet control group (HD), and high fat diet plus kochujang supplemented group (HDK). Results were as follows: 1. Fennented kochujang supplement in high fat diet decreased body weight and epidydimal and back fat weight compared to non-supplement in HD group. 2. Lipid content and blood glucose level were lower in HDK group than HD group. 3. Fermented kochujang supplement increased mRNA level of lipolytic genes such as acyl-CoA synthetase (ACS), carnitine palmitoyltransferase-1 (CPT-1), and uncoupling proteins-1 (UCP-1) expression, whereas decreased mRNA level of adipogenic genes such as acetyl CoA carboxylase (ACC) expression. These findings suggest that fermented kochujang supplement in high fat diet normalized body weight, epididymal and back fat weight, lipid content, and blood glucose levels through controlling lipid metabolism and provides basic information on the control of obesity.
Water-soluble chitin (WSC) was prepared by carefully deacetylating chitins to about $50\%$ of N-acetyl content. Topical formulations based on WSC were prepared and their effects on wound healing were evaluated on a rabbit ear model. Full-thickness, open skin wounds were made on the ears of rabbits and WSC ointments were embedded in the open wounds. The application of WSC ointments significantly accelerated wound healing and wound contraction. The areas of epithelial-ization and granulation tissues in WSC ointment group are remarkably larger than those in control group (no treatment) and in placebo group (treated with ointment-base materials). A large number of grown granulation tissues including dense fibroblast deposition were observed under the thickened epithelium of the wound treated with WSC ointments. The number of inflammatory cells in WSC ointment group was significantly decreased compared with those in control and placebo groups, indicating that WSC would give low stimuli to wounds and prevent excessive scar formation. Neovascularization was the most prominent in WSC ointment group. Wound contraction in WSC ointment group was much larger than those in control and placebo groups. Overall results demonstrate that the topical formulation based on WSC is considered to become an excellent dressing as a wound healing assistant.
For the propose of the quality control of Torilis Fructus, one major compound was isolated by several column chromatograpies and identified by its spectral data as torilin (8,11-dihydroxy-8-angeloyl-11-acetyl-4-guaien-3-one). A quantitative analysis of torilin using HPLC method showed that the average contents at 230 and 254 nm were $7.167{\pm}0.168mg/g\;and\;7.052{\pm}0.160mg/g$, respectively, in 18 samples collected throughout the regions of Korea and China. This method provided a tool for standardization of the commercial Torilis Fructus.
This study evaluated the effects of carnitine supplementation on obesity caused by a high-fat diet in C57BL/6J mice. The mice were fed a normal diet (ND), high-fat diet (HD), or carnitine-supplemented (0.5% of diet) high-fat diet (HDC) for 12 weeks. The results showed that body weight, energy intake, and feed intake were lower in the HDC group than the control groups. Acid-soluble acylcarnitine (A SAC), acid-insoluble acylcarnitine (AIAC), and total carnitine (TCNE) in the serum and liver were significantly higher in the HDC group. Hepatic carnitine palmitoyl transferase-I activity was significantly higher in the HDC group than the control groups. Acyl-coA synthetase (ACS) and carnitine palmitoyl transferase-I (CPT-I) mRNA expression in the liver was highest in the HDC group, however hepatic acetyl-coA carboxylase (ACC) mRNA expression in this group was lowest. Serum leptin levels and abdominal fat weight were lowest in the HDC group. We concluded that L-carnitine supplementation diminished the risk of obesity caused by a high-fat diet.
NAC는 GSH의 전구물질로, thiol기를 포함하는 항산화제 중 하나로 잘 알려져 있으며, 방사선 조사 시 발생하는 생체 내 영향을 감소시켜 생체 손상의 방호 및 회복에 도움을 주는 방사선 방어제로 이용된다. S. cerevisiae에서 항산화제 NAC를 전처리 함에 따라 이온화 방사선 조사에 따른 효모의 세포사멸 방어효과 및 superoxide dismutase (SOD), catalase, glutathione peroxidase (GPx)와 같은 항산화 효소들의 유전자 발현을 분석하여 NAC의 항산화적 효과를 확인하였다. 효모는 다양한 농도의 NAC 전처리 후 다양한 선량의 이온화 방사선에 조사되었으며, 세포생존율은 세포형성단위(CFU)를 계수해 측정되었고, 항산화 효소의 유전자 발현은 real-time PCR수행 후 분석하였다. 우선적으로 효모에 NAC 처리를 위한 적정농도를 확인하였는데, 35 mM 이상의 NAC 농도에서 효모세포의 성장이 억제 되었다. NAC 전처리는 감마선 조사에 의한 세포사멸을 방어하지 않았으며, 100 Gy 방사선 조사는 항산화 효소들의 유전자 발현을 유도하였다. NAC 전처리 후 항산화 효소들의 유전자 발현은NAC의 농도 증가에 따라 감소하였다. 이러한 결과로,NAC의 높은 농도(35 mM 이상)는 효모세포의 성장을 저해하며, NAC는 이온화 방사선 조사에 따른 세포사멸을 방어할 수 없으나, 생체 내에서 활성산소종을 제거 하여 세포를 보호하는 유용한 항산화제임을 알 수 있었다.
규폐증의 신기능 장애 평가를 위한 기초 자료를 제공하기 위하여, 우리나라 일부 지역의 진폐요양기관에서 규폐증으로 치료중인 환자 58명을 실험군으로 하고 폐결핵으로 6 개월이상 치료중인 환자 40명을 결핵 대조군 그리고 일반사무직 근로자 51명을 건강대조군으로하여, 신기능 장애의 예민한 지표로 알려진 요중 N-ncetyl-$\beta$-D-glucosaminidase (NAG) 활성치를 측정하여 얻은 바를 아래와 같이 요약 한다. 1. 조사대상자 전체의 혈중 요소질소와 혈청 크레아티닌은 대상자 모두에서 참고치내에 있어 임상적으로 신장장애의 소견을 보이지 않았으나, 요중 NAG 활성치는 $7.25{\pm}7.31U/g\;creatinine$으로서 신기능 장애 선별기준 이상의 측정치를 보여 보다 민감한 검사 결과를 나타내었다. 2. 규폐증군의 요중 NAG 활성치는 $11.98{\pm}9.05U/g\;creatinine$으로서 결핵대조군($5.71{\pm}4.33U/g\;creatinine$) 및 건강대조군($3.63{\pm}1.67U/g\;creatinine$)에 비하여 통계학적으로 유의하게 증가되어 있었으나(p<0.05), 결핵대조군과 건강대조군에서는 유의한 차이를 나타내지 않았다(p>0.05). 3. 결핵대조군에서 결핵의 병증에 따라 요중 NAG 활성치가 증가되는 양상을 보였으나 통계학적으로 유의 하지않았고(p>0.05), 치료약제중 신장애 유발 항결핵제를 사용한 대상자에서 요중 NAG 활성치는 유의하게 증가되어 있었다(p<0.05). 4. 규폐증군에서 결절의 크기가 클수록 그리고 신장애 유발 항결핵제를 사용한 자 및 발병기간이 짧았던 자의 경우 요중 NAG 활성치가 증가되는 양상을 보였으나, 통계학적으로 유의한 차이는 아니었다(p>0.05). 5. 신독성 항결핵제 사용자를 제외한 요중 NAG 활성치는 건강대조군과 결핵대조군에서 각각 3.63U/g creatinine, 3.60U/g creatinine으로 차이를 보이지 않았으나, 규폐증군에서는 10.90U/g creatinine 으로 현저히 증가되어 있었다. 이상의 결과로 규폐증에서는 일반적인 건강진단 항목상의 신기능 검사에 이상소견을 보이지 않은 경우라도 신장기능의 변화가 동반될 수 있으며, 요중 NAG 활성치를 분진 폭로 근로자에게 적용하면 산업보건학적 측면에서 근로자의 건강관리를 위하여 매우 유익한 자료가 될 것으로 생각한다.
Laminariae Thallus has been known to be effective for the treatment of obesity. Te show the effectiveness of Laminariae Thallus in a more scientific way, Laminariae Thallus extract was prepared and evaluated in high fat diet rats by measuring the changes of body weight and lipid metabolism as described briefly below. 200 g of crushed Laminariae Thallus was extracted with methyl alcohol. The extract was evaporated under reduced pressure to give 14.4g. For 10 weeks, control group rat were ingested high fat diet, while the test group rat were fed with a fat diet pius Laminariae Thallus extract. Normal group was fed with normal diet. 150 mg of Laminariae Thallus extract per 1 kg of body weight was added to the diet in a test group rats. Control group rats on a high fat diet gained weight significantly, whereas the test group rats on a high fat diet plus Laminariae Thallus extract gained weight less. Significant increase of liver weight caused in a high fat diet was also inhibited by the Laminariae Thallus extract treatment. Total lipid, triglyceride, phospholipid and total cholesterol levels of serum in the high fat diet rats were remarkably increased, whereas their levels on a high fat diet plus Laminariae Thallus extract were less increased. While serum HDL-cholesterol levels was remarkably decreased in a high fat diet, its level was less decreased in a high fat diet plus Laminariae Thallus extract. Furthermore, we observed that the activities of hepatic acetyl-CoA carboxylase and fatty acid synthetase increased in a high fat diet, while their activities in a high fat diet plus Laminariae Thallus extract were getting back nearly to the normal levels of normal diet rats. These result showed that the obesity caused by a high fat diet was effectively inhibited by a Laminariae Thallus extract. Our results also showed that the abnormal lipid metabolism caused by a high fat diet was effectively cured by adding Laminariae Thallus extract.
N-아세틸-ㅣ-페닐 알라닐-ㅣ-페닐 알라닌 메틸 에스테르(APhPhMe)와 N-아세틸-ㅣ-페닐 알라닌메틸에스테르(APhMe) 그리고 N-아세틸-ㅣ- 페닐알라닐-l-알라닌메틸에스테르(APhAlMe)를 모델화합물로 사용하여 단백질 변성을 연구하였다. APhPhMe는 압력이 증가하면 물에서의 용해도가 증가하였고 APhMe는 반대로 크게 감소하였지만 APhAlMe는 측정한 온도에서 거의 변화가 없었다. APhPhMe의 부피 변화는 20, 30, 40, 50$^{\circ}C$에서 각각 -0.9, -1.47, -1.09, -1.52 ml/mole이었고 APhMe는 20, 30, 40$^{\circ}C$에서 +6.0, +7.0, +7.5 ml/mole이었지만 APhAlMe는 세 온도 모두 0에 가까왔다. 이러한 결과를 소수성 상호작용과 펩티드 결합에 의한 수소결합으로 설명하였다. 즉 펩티드 결합만 존재하거나, 비교적 큰 소수성 작용기 1개와 펩티드 결합 1개가 동시에 있을 경우에는 펩티드 결합만 존재하거나, 비교적 큰 소수성 작용기 1개와 펩티드 결합 1개가 동시에 있을 경우에는 펩티드 결합사이의 수소결합이 소수성 상호 작용보다 우세하였고, 펩티드 결합과 소수성 작용기가 동시에 1개씩 증가할때는 소수성 상호작용이 우세한 것 같다.
New analogues of Sildenafil (Viagra${\circledR}$)were identified in the imported Chinese herb drinks, which have been added illegally for affecting penile erectile dysfunction. In order to elucidate the chemical structures of Sildenafil analogues, separation and purification were carried out and followed by analysis of the routine 1D and 2D NMR spectroscopy and FAB mass spectrometry. Based on analytical data one compound was maned as Homosildenafil contains ethylpiperazine switched with methylpiperazine of Sildenafil and the other one was called an Hongdenafil contains ethylpiperazine switched with methylpiperazine and acetyl group instead of sulfonyl group of Sildenafil. As results, Homosildenafil and Hongdenafil were put on the list as new inspection items of the imported food for protectiong public health in Korea.
