Effect of Artemisia capillaris Extracts on Antioxidant Activity and Allergic Dermatitis (인진호(Artemisia capillaris) 추출물의 항산화 활성 및 알러지성 피부염에 대한 효과)

  • The antioxidant activities of 6 solvent extracts of Artemisia capillaris were evaluated in a dintroflurobenzen (DNFB)-induced allergic mouse model. In vitro antioxidant activities were determined using DPPH and the FRAP test. Methanol (DPPH: 85.87%, FRAP: 1.772) and $dH_2O$ (DPPH: 60.69%, FRAP: 3.185) extracts showed the highest antioxidant activities compared with other solvents (ethyl acetate 41.81%, 0.407, hexane 8.37%, 0.328, etc.). In addition, we tested atopic dermatitis (AD)-like skin lesions in mice treated with DNFB. The methanol extract of A. capillaris on the AD-like skin lesions in DNFB-induced atopy inhibited ear thickness increases (47%) and the skin lesions (45%) compared with a positive control (methanol). The results suggest that they have potential as natural antioxidants and allergy-improving substances and that they may be valuable materials in the functional food or cosmeceutical industry.

  • In the culture medium, the three antifungal fractions against P. capsici were separated by Sephadex G-25 column chromatography and Silica-gel chromatography. The substance A in white powder and the substance B in sticky oil were isolated by ethyl acetate : acetone mixture(7 : 3), and the substance C in yellow powder was isolated by chloroform : ethyl acetate mixture(95 : 5). The crude extract by ethyl acetate from the culture medium acidified to pH 2 was known to inhibit completely the growth of P. capsici at the level of $50mgkg^{-1}$. The substance A and B were known to be effective above the level of $5mgkg^{-1}$, and the substance C was effective above the level of $1mgkg^{-1}$. However, at the level of $20mgkg^{-1}$, the efficiency was in the order of A>C>B. It is apparent on a pot-experiment scale that the three substances effectively control Phytophthora blight of the red-pepper plant grown in the soil inoculated with P. capsici.

Anti-Inflammatory Effect of Ligularia fischeri, Solidago virga-aurea and Aruncus dioicus Complex Extracts in Raw 264.7 Cells (곰취(Ligularia fischeri), 미역취(Solidago virga-aurea), 삼나물(Aruncus dioicus) 복합 추출물의 항염증 효과)

  • The objective of this study was to evaluate the skin inflammation effects of three herb mixture extracts, Ligularia fischeri, Solidago virga-aurea and Aruncus dioicus, which are from Ullung island in Korea. Regulatory mechanisms of cytokines and nitric oxide (NO) are involved in the immunological activity of Raw 264.7 cells. Tested cells were pretreated with 70% acetone extracts of Ligularia fischeri, Solidago virga-aurea and Aruncus dioicus (LSA-A) and further cultured for an appropriated time after lipopolyssacharide (LPS) addition. During the entire experimental period, 1, 10, and 100 ${\mu}g/ml$ of LSA-A had no cytotoxicity. In these concentrations, LSA-A inhibited the production of NO and prostaglandin $E_2$ ($PGE_2$), tumor necorsis factor-a (TNF-a), interleukin-1${\beta}$ (IL-1${\beta}$), interleukin-6 (IL-6) expression of inducible NO synthase (iNOS), and cyclooxygenase-2 (COX-2). LSA-A showed a 60% $PGE_2$ inhibition rate at 100 ${\mu}g/ml$. iNOS and COX-2 inhibition activities were 54%, and 65% at 100 ${\mu}g/ml$, respectively. In addition, LSA-A extract reduced the release of inflammatory cytokines including TNF-a, IL-1${\beta}$ and IL-6. These results suggest that LSA-A may have significant effects on inflammatory factors, and may be a potential anti-inflammatory therapeutic agent.

Anti-Allergic Activities of Ultra-fine Powder from Persimmon (감 초미세 분말의 추출물을 이용한 항산화 및 항아토피 활성)

  • Ultra-fine powder (FA) extracts from persimmon were investigated as potential food materials in industrial processes because such extracts have various useful biological activities. We examined the antioxidant and anti-allergic activity of FA extracts using the 1,1-diphenyl-2-picrylhydrazyl (DPPH) and ferric reducing activity of plasma (FRAP) models, and by measuring interleukin 4 (IL-4) expression in spleen cells and in an atopy animal model. FA extracts were prepared using solvents including distilled water and 50% (v/v) acetone. The results showed that FA extracts had potent DPPH and FRAP activities, decreased IL-4 expression in spleen cells, and inhibited ear thickness and inflammation in a mouse animal model. The data indicate that a FA extract could be used as an antioxidant and/or anti-inflammation agent, and in industrial food preparation processes.

Effects of Extracts from Dried Yam on Antioxidant and Growth of Human Cancer Cell Lines (건조 마 추출물의 항산화 및 인체 암세포 증식 억제 효과)

  • We investigated the inhibitory effects of solvent extracts from dried yam on $H_2O_2$-induced oxidative stress and growth of cancer cell lines (HT1080 human fibrosarcoma and HT-29 human colon cancer cells). Yam (Dioscoreacea) has been recognized as a healthy food due to its various biological activities, such as anti-obesity, anti-constipation, anti-proliferation, and anti-mutagenic activities, as well as its ability to decrease blood glucose and cholesterol levels. In order to determine the protective effect on $H_2O_2$-induced oxidative stress, DCFH-DA (dichlorodihydrofluorescin diacetate) assay was conducted. Acetone with methylene chloride (A+M) extract of dried yam appeared to reduce the levels of intracellular reactive oxygen species (ROS) with dose responses. Among the fractions, 85% aq. methanol fraction showed the highest protective effect on production of lipid peroxides. Inhibitory effects of A+M and methanol (MeOH) extracts on the growth of HT1080 and HT-29 cancer cells increased in a dose dependent manner. The treatments of n-hexane, 85% aq. methanol and n-butanol fractions (${\geqq}0.5$ mg/ml concentrations) significantly inhibited the growth of both cancer cells (p<0.05). From these results, 85% aq. methanol fraction showed inhibitory effects on cellular oxidation and growth of human cancer cells, suggesting that this fraction may contain active compounds of dried yam.

Lowering the Bitterness of Enzymatic Hydrolysate Using Aminopeptidase-active Fractions from the Common Squid (Todarodes pacificus) Hepatopancreas (살 오징어(Todarodes pacificus) 간췌장으로부터 aminopeptidase 활성 획분의 쓴맛 개선 효과)

  • Aminopeptidase-active fractions from crude extract of the hepatopancreas of a common squid (Todarodes pacificus) were obtained using acetone (AC; 30-40%) and ammonium sulfate precipitation (AS; 60-70% saturation), anion exchange (AE-II; 0.2 M NaCl) and gel filtration chromatography (GF-I; 30-50 kDa), respectively. The debittering capacity of GF-I fraction based on the aminopeptidase activity (89.2 U/mg), recovery (56.6%) and sensory evaluation (1.0) was better than that of other fractions. Release of amino acids increased as incubation time was increased, and the bitterness of the enzyme reaction mixtures decreased. Incubation with the GF-I fraction for 24 h resulted in the hydrolysis of several peptides, as revealed by reverse-phase HPLC profiles. Peaks 3, 5 and 6 showed the decreased area (%), whereas peaks 1, 2 and 4 showed the increased area. The GF-I fractions were found to be suitable for reducing bitterness in protein hydrolysates by catalyzing the hydrolysis of bitter peptides.

Analgesic Effects of the Combination of Aconitum Ciliare Tuber with Honey in the Rat Models of Peripheral Neuropathic Pain (초오(草烏) 봉밀(蜂蜜) 혼합물(混合物)이 백서(白鼠)의 말초신경병증성 통증 억제에 미치는 영향)

  • Objectives : We have studied to know analgesic effects of the combination of Aconitum Cilliare Tuber with honey in the models of peripheral neuropathic pain. Methods : Neuropathic pain model was made by ligating left 5th lumbar spinal nerve. After 3 days, combination of Aconitum Ciliare Tuber and honey extract was administrated each alternate day. Administration was divided three groups, that is NP-OA1(0.06 mg/ml), NP-OA2(0.24 mg/ml), and NO-OA3(0.96 mg/ml). After that, we examined the withdrawl response of neuropathic rats legs by von Frey filament and acetone stimulation. And also we examined c-Fos, glutamic oxaloacetic transaminase(GOT), glutamate-pyruvate transaminase(GPT) and change of weight. Results : Mechanical allodynia in NP-OA1 groups were significantly decreased compared with the control group. Cold allodynia in all experimental groups were no significant differences with the control group. c-Fos protein expression on the central grey, all experimental groups were lower than that of control groups. But, there were no statistically significant differences. Change of weight in all experimental groups were significantly increased compare with the control group. In blood serum GOT in NP-OA1, NP-OA2 groups were significantly decreased compare with the control group. In blood serum GPT in all experimental groups were no significant difference with the control group. Conclusions : We had noticed that the combination of Aconitum Ciliare Tuber and honey decreased mechanical allodynia in the model of neuropathic pain compared with the control group and it has not efficacy in elevation of GOT, GPT and weight loss etc., the element of which becomes damage to liver. This study can be proposed that Aconitum Ciliare with Honey may be applicable to neuropathic pain in clinic. But it is reliability not that cold allodynia and c-Fos expression have effectively to control pain. Therefore we have to follow up about that.

Extraction and Purification of Agar from Gelidium amansii (우뭇가사리로부터 한천의 추출 및 정제)

  • The effect of different treatments on the quality and yield of purified agar produced from Gelidium amansii has been studied, and the extraction condition ol agar produced from G. amansii has been examined. The contents of ash, sulfate in agar produced from g. amansii collected from different places were $2.63\~2.92\%\;and\;1.38\~1.78\%$, respectively. Yields and gel strength of agar produced from G. amansii collected from different places were $31.6\~46.8\%\;and\;496\~887g/cm^2$, respectively. It was effective to extract agar at $120^{\circ}C\;for\;2\~3hrs$. Agar was purified by D.W. washing, EDTA washing, chitosan treatment, CPC treatment, PEG treatment, ethanol precipitation, acetone precipitation and propanol precipitation. The mineral contents of agar produced from Gelidium amansii were Na (2934ppm), Ca (2472ppm), Mg (2259ppm), K (2527ppm), P1(81.1ppm), Fe (66.4ppm), Al (71.7ppm), Zn (29.7ppm) and Pb (ND: not detected), respectively.

Protection and Detoxification Effects of Oriental Herb Extract Mixture on TCDD-Induced Oxidative Stress (TCDD로 유발된 Oxidative Stress에 대한 생약재 추출물의 방어 및 해독효과)

  • This study was carried out to investigate the protective effects and detoxification of oriental herb extracts mixture (Saururus chinensis, Taraxacum platycarpum, Ulmus macrocarpa, Glycyrrhiza glabra, Rhynchosia nulubilis) on TCDD-induced oxidative stress. Thirty five male rats were divided into 5 groups: normal control group received saline; vehicle control group received DMSO and acetone; only TCDD-treated group received multiple intraperitoneal injection of TCDD ($100{\mu}g/kg$) and saline; post-treated group of OHEM (400 mg/kg/day) received oral administration for 5 weeks after TCDD treatment; and pre-treated group of OHEM (400 mg/kg/day) received oral administration for 6 weeks from 1 week before TCDD treatment. The elevated serum activities of alanine transaminase (AST), aspartate transaminase (ALT), alkaline phosphatase (ALP), lactate dehydrogenase (LDH) and atherorganic index due to TCDD were significantly decreased by the treatment of OHEM (p<0.05); the pre-treatment of OHEM was especially effective. Hydropic lesions and cytoplasmic vacuolizations in the liver of TCDD-treated rats were inhibited by the treatment of OHEM. Also, OHEM treatment reduced edemas in small intestine villus of TCDD-treated rats. These results suggest that OHEM from various oriental herb extracts might be a useful protective and detoxification agent against TCDD.

Induction of Apoptosis by Immature Prunus salicina Lindl. cv. Soldam (피자두(Prunus salicina Lindl. cv. Soldam) 미숙과의 apoptosis 유도 효과)

  • Apoptosis of Prunus salicina Lindl. cv. Soldam, which possesses hematopoiesis, osteoporosis prevention, and antimutagenic effects, at different growth stages was evaluated. Cytotoxic effect of acetone extracts of immature fruits against various tumor cell lines was higher than that of mature fruits, particularly in hormone-independent human breast cancer, MDA-MB-231 cell line. Immature fruit extract increased expression level of pro-apoptotic protein Bax and reduced that of anti-apoptotic protein Bcl-2, and stimulated caspase-3 activity in MDA-MB-231 cells. Results suggest immature fruit of P. salicina Lindl. cv. soldam to be natural source for development of functional food and medical agents to prevent human breast cancer.
