Rapid Determination of Chlorostyrenes in Fish by Freezing-Lipid Filtration, Solid-Phase Extraction and Gas Chromatography-Mass Spectrometry

  • An analytical method has been developed for measuring chlorostyrenes in fish tissue sample. Extraction of chlorostyrenes from fish tissue was carried out by ultrasonication using acetone/n-hexane (5:2, v/v) mixture. Most of the lipids in the extract were eliminated by freezing-lipid filtration, prior to solid-phase extraction (SPE) cleanup. During freezing-lipid filtration, about 90% of the lipids extracted from the fish samples were easily removed without any significant losses of chlorostyrenes. For purification, SPE using Florisil was used for the rapid and effective cleanup. Quantification was performed using gas chromatography-mass spectrometry in the selected ion monitoring mode. Spiking experiments were carried out to determine the recovery, precision, and limits of detection (LODs) of the method. The overall recovery was above 80% in the spiked fish tissue sample at 10 and 100 ng/g levels, respectively. The detection limits for chlorostyrenes were ranged from 0.05 to 0.1 ng/g. This developed method is demonstrated to give efficient recoveries and LODs for detecting chlorostyrenes spiked into fish tissue with high lipid content.

Phytogenic silver nanoparticles (Alstonia scholaris) incorporated with epoxy coating on PVC materials and their biofilm degradation studies

  • The advantages of nano-scale materials (size 1-99 nm in at least in one dimension) could be realized with their potential applications in diversified avenues. Herein, we report for the first time on the successful synthesis of homogeneous epoxy coatings containing phytogenic silver nanoparticles (Ag) on PVC and glass substrates by room-temperature curing of fully mixed epoxy slurry diluted by acetone. Alstonia scholaris bark extract was used to reduce and stabilize the silver ions. The surface morphology and mechanical properties of these coatings were characterized using the techniques like, UV-Vis (UV-Visible) spectrophotometry, X-ray diffraction (XRD), Fourier transform infrared spectrophotometry (FT-IR), Epifluorescence microscopy and scanning electron microscopy (SEM). The effect of incorporating Ag nanoparticles on the biofilm (scale) resistant epoxy-coated PVC was investigated by total viable counts ($CFU/cm^2$) from epoxy coating from (Initial) $1^{st}$ day to $25^{th}$ days. The phytogenic Ag nanoparticles were found to be significantly improving the microstructure of the coating matrix and thus enhanced the anti-biofilm performance of the epoxy coating. In addition, the antimicrobial mechanism of Ag nanoparticles played an important role in improving the anti-biofilm performance of these epoxy coatings.

Liquid Chromatographic Determination of Etofenprox Residues in Foods with Mass-Spectrometric Confirmation

  • BACKGROUND: An official analytical method was developed to determine etofenprox residues in agricultural commodities using high-performance liquid chromatography (HPLC). METHODS AND RESULTS: The etofenprox residue was extracted with acetone from representative samples of five raw products which comprised rice grain, apple, mandarin, cabbage, and soybean. The extract was then serially purified by liquid-liquid partition and Florisil column chromatography. For rice and soybean samples, acetonitrile/n-hexane partition was additionally coupled to remove nonpolar lipids. Reversed phase HPLC using an octadecylsilyl column was successfully applied to separate etofenprox from co-extractives. Intact etofenprox was sensitively detected by ultraviolet absorption at 225 nm. Recovery experiment at the quantitation limit validated that the proposed method could apparently determine the etofenprox residue at 0.02 mg/kg. Mean recoveries from five crop samples fortified at three levels in triplicate were in the range of 93.6~106.4%. Relative standard deviations of the analytical method were all less than 10%, irrespective of crop types. A selected-ion monitoring LC/mass spectrometry with positive atmospheric-pressure chemical ionization was also provided to confirm the suspected residue. CONCLUSION(s): The proposed method is simple, rapid and sensitive enough to be employed in routine inspection or monitoring of agricultural products for the etofenprox residue.

Effects of Yangkyuksanhawtang on the Allergic Contact Dermatitis (양격산화탕(凉隔散化湯)이 Allergy성(性) 접촉피부염(接觸皮膚炎)에 미치는 영향(影響))

  • Objective : To study the effectiveness of Yangkyuksanhawtang against Allergic Contact Dermatitis I observed the change of cutaneous shape, histochemistry, immunohistochemistry, and the distribution of apoptotic cells. materials and methods : I divided 4-month-old rats into three groups of 10, which are a contrastive group of having applied Acetone olive oil only, ACD group to have intentionally activated Allergic Contact Dermatitis by DNCB and YST group to give medication of Yangkyuksanhawtang extract. And I observed each group of mice after 24, 48 and 72 hours. results : 1. With the result of Contact hypersensitivity assay, YST group shows appreciably less ear swelling than ACD group. 2. Comparing YST and ACD groups to each other regarding general change of skin, YST group shows less hyperplasia of epidermis, less migration of inflammatory cells and less damage of epidermis than ACD group. 3. Regarding the change of collagen fiber, ACD group has appeared to be low in number of collagen fiber while YST shows similarity with the contrastive group. 4. In dermis YST group has showed lower number of mastocyte than ACD group and is granulated type. 5. In dermis YST group has showed less MAC-1, IL-1 , $IL-2R-\;{\alpha}$ G, ICAM-1 and VCAM-1 than ACD group. 6. The distribution of apoptotic cells has appeared littler in YST group than in ACD 7. Among signal molecule of apoptosis Bcl-2 has distributed more in YST group than ACD group and Bax and Fas has distributed less in YST group than ACD group.

Antioxidative Activities and Quantitative Determination of Gallotannins from Barks of Acer ginnala Maxim (신나무 수피로부터 Gallotannin 화합물의 항산화 활성 및 함량분석)

  • Activity guided isolation of 80% acetone extract from the barks of Acer ginnala Maxim. yielded five gallotannins [6-galloyl-1,5-anhydroglucitol (ginnalin B) (1), acertannin (3,6-digalloyl-1,5-anhydroglucitol) (2), methyl gallate (3), acertannin (2,6-digalloyl-1,5-anhydroglucitol) (4) and gallic acid (5)]. All of these isolated compounds from Acer ginnala(1-5) were firstly isolated from Acer ginnala Maxim. And contents of compounds from barks of Acer ginnala (Comp. 1: 0.73$\pm$0.002%, Comp. 2: 0.48$\pm$0.001%, Comp. 3: 0.66$\pm$0.002%, Comp. 4: 1.05$\pm$0.002% and Comp. 5: 0.29$\pm$0.001%) were evaluated by HPLC analysis. And, in order to evaluate anti-oxidative activities on Comp. 1-5 isolated from Acer ginnala, DPPH radical scavenging activity was measured in vitro. All of these isolated compounds from Acer ginnala exhibited potent DPPH radical scavenging activities.

  • From crude drug preparation(Soshiho-Tang) ginseng sapogenins were identified by TLC and $ginsenoside-Rb_1$ was determined quantitatively by HPLC. Panaxadiol, pandaxatriol, acid-hydrolysates of ginseng saponin, were identified by TLC with benzene/acetone(4 : 1, v/v). Rf values of which were measured as 0.26 and 0.14, respectively. The content of $ginsenoside-Rb_1$ was determined by HPLC on $Lichrosorb-NH_2$ column with $CH_3CN/H_2O/n-BuOH$(80 : 20 : 10, v/v). Its recovery rate in the extract granules, was as relatively low as $19.8{\pm}1.4%$ compared to the content in raw red ginseng.

Studies on Solvent Extraction and Flotation Technique Using Metal-Dithizone Complexes(II). Determination of Trace Elements in Water Samples by Solvent Sublation

  • The preconcentration and determination of trace elements in water samples were studied by a solvent sublation utilizing dithizonate complexation. After metal dithizonates were formed, trace amounts of cadmium, cobalt, copper and lead were floated and extracted into small volume of a water-immiscible organic solvent on the surface of sample solution and determined in the solvent directly by GF-AAS. Several experimental conditions as formation condition of metal-dithizonate complexes, pH of solution, amount of dithizone, stirring time, the type and amount of surfactants, N2 bubbling rate and so on were optimized for the complete formation and effective flotation of the complexes. And also four kinds of light solvents were compared each other to extract the floated complexes, effectively. After the pH was adjusted to 4.0 with 5 M HNO3, 8.0 mL of 0.05% acetone solution of dithizone was added to 1.00 L water sample. The dithizonate complexes were flotated and extracted into the upper methyl isobutylketone (MIBK) layer by the addition of 2.0 mL 0.2% ethanolic sodium lauryl sulfate solution and with the aid of small nitrogen gas bubbles. And this solvent sublation method was applied to the analysis of real water samples and good results of more than 85% recoveries were obtained in spiked samples.

Isolation and Identification of Antimicrobial Active Substances from Rhodiola sachlinensis (홍경천(Rhodiola sachlinensis)에서 항균성 물질의 분리 및 동정)

  • The antimicrobial substances from Rhodiola sachalinensis were extncted, isolated and identified. The highest level of antimicrobial activity and its yield were obtained in methanol extract. The minimum inhibition concentrations of the methanol extract were 500 $\mu\textrm{g}$mL on agar plate and 100 $\mu\textrm{g}$mL in broth media for four gram positive and four gram negative microbials. The methanol extract was fractionated by n-hexane, chloroform, ethyl ether, ethyl acetate, and butanol, orderly. The separate was developed on the TLC plate with different solvent system ratio of chloroform and methanol. Nine substances were isolated from chloroform and methanol mixture(9:1, v/v). Among them, three isolates showed antimicrobial activity. Three substances separated by HPLC were identified by GC/MS(EI) spectrum and $^1$H, /sup13/C-NMR spectrum. They were gallic acid, (-)-epicatechin and kaempferol. The antimicrobial activities of each substances were shown gallic acid, (-)-epicatechin, kaempferol orderly.

Comparison of Physiological Activities of Radish Bud (Raphanus sativus L.) according to Extraction Solvent and Sprouting Period (추출용매 및 발아시기에 따른 무순 추출물의 생리활성 비교)

  • This study extracted radish bud (Raphanus sativus L.) and investigated its nitrite scavenging activity, superoxide dismutases (SOD)-like activity, tyrosinase inhibition activity, xanthine oxidase inhibition activity, and angiotensin-converting-enzyme (ACE) inhibition activity according to extraction solvent and sprouting period. For nitrite scavenging activity, each extract recorded its highest level of 81.44~89.71% at pH 1.2. Radish bud extracts on sprouting days 4 and 8 showed greater scavenging activities than those on sprouting day 12 at pH 1.2 and pH 4.0. There were differences in scavenging activity according to extraction solvent based on water extract exhibiting improved scavenging activity. Ethanol extract recorded scavenging activity of 16.12% at pH 6.0, which was similar to those of ethanol and methanol radish bud extracts on sprouting day 12. SOD-like activity of radish bud extracts was in the range of 4.57~27.05%. For comparison purposes, SOD-like activity of L-ascorbic acid was 52.15%, which was higher than that of radish bud extracts. Acetone and methanol extracts showed high SOD-like activities on sprouting day 8. SOD-like activity of radish bud extracts on sprouting day 12 significantly decreased to 4.57~15.59%. Radish bud extracts recorded good tyrosinase inhibitory activities on sprouting 8 and 12, whereas methanol extracts recorded the greatest tyrosinase inhibitory activity at 62.65~84.89%. Radish bud extracts recorded xanthine oxidase inhibition activity of 21.26~29.52% on sprouting day 4, and acetone extracts showed the highest level of xanthine oxidase inhibition activity. Xanthine oxidase inhibitory activity tended to decrease with sprouting period compared early on. ACE inhibitory activity was in the range of 12.48~51.78% according to sprouting period and extraction solvent. Ethanol extracts on sprouting day 8 showed the highest ACE inhibitory activity of 51.78%. These results will hopefully contribute to research into the identification of materials and development of products for natural functional foods.

Studies of Xanthium strumarium Extract Suppressing Th17-cell Differentiation and Anti-dermatitic Effect in BMAC-induced Atopy Dermatitis of NC/Nga Mice (창이자 추출물이 아토피 피부염 유발 생쥐의 비장 세포 Th17의 세포분화 억제에 따른 아토피 피부 상태에 미치는 영향)

  • Xanthii fructus which is well known as "Chang-ihjah" in Korea is the dried fruit of Xanthium strumarium L. (or Xanthium sibiricum PATR. Ex WIDD., Asteraceae. XS). Water extract of this fruit has been used for treatment of various inflammatory diseases such as tympanitis, allergic rhinitis, or ozena as alternative therapy material usually by oral administration in far Eastern countries including Korea. In this study, the effect of XS extract (XS-E) or XS-30% acetone fraction layer (XS-30% AFL) on the differentiation of $CD4^+$ T cells isolated from NC/Nga mouse and the production of IL-17 was investigated. The experimental results showed that $100\;{\mu}g$/mL of XS-E could decrease the production of IL-17 by $CD4^+$ Th17 cells by 2 fold and only $20\;{\mu}g$/mL of XS-30% AFL could inhibit 3.5 fold. The amount of IL-17A and IL-22 mRNA determined by real-time PCR was decreased remarkably when XS-E or XS-30% AFL was treated on $CD4^+$ Th17 cells(p<0.01, p<0.001). The amount of IL-17A protein determined by ELISA was also decreased remarkably(p<0.05, p<0.001). To study the effect of XS-E or XS-30% AFL on the proliferation of Th17 cells, $CD4^+$ T cells of a NC/Nga mouse was firstly differentiated by rIL-6/TGF-$\beta$ and then stimulated by rIL-23. The control group of Th17 cells were doubled every each day, while those of XS-E or XS-30% AFL treated group were shown to be delayed remarkably by these extracts. In conclusion, XS can inhibit the differentiation of Th17 cells of NC/Nga mouse and the production of IL-17 successfully, which may be a beneficial result for the treatment of atopic skin dermatitis.
