솔잎과 쑥 추출물의 기능성 점토 (Studies on the Physiological Functionality of Pine Needle and Mugwort Extracts)

  • 솔잎과 쑥의 기능성 검토를 위한 연구의 일환으로 열수 및 70% acetone으로 처리하여 얻은 추출물의 항변이원성, angiotensin I converting enzyme(ACE) 저해작용, 전자 공여작용, polyphenol oxidase(PPO)에 대한 저해작용, 아질산염 소거작용, 항산화 작용을 조사하였다. 솔잎의 열수 및 70% acetone 추출물 모두 총 polyphenol 성분 중 flavanol형 tannin의 함량이 60% 이상이었고, 간접변이원인 Trp-P-1을 이용한 항변이원성 실험결과 70% acetone 추출물은 처리구는 82.2%로 열수 추출물보다 높은 억제 활성을 나타내었으나 ACE 저해작용은 열수 추출물이 61%로 70% acetone 추출물의 50%보다 저해능이 높았다. 전자공여작용은 두가지 추출물 모두 80% 이상의 높은 효과를 나타내었다. 미나리에서 분리한 PPO 조효소에 대한 70% acetone 추출물의 저해효과는43%로 나타났고 또한 아질산염 소거작용은 산성 부근(pH 3.0)에서 각 추출물 모두 $80{\sim}90%$의 높은 효과를 보였다. 한편 쑥추출물의 경우 항변이원성은 70% acetone 추출물이 84.1%로 솔잎과 유사하였으나 ACE 저해작용은 열수 추출물이 75.1%로 acetone 추출물의 57.6%보다 높았고, 전자공여 작용은 두 처리구 모두 45% 정도였으며, 아질산염 소거작용은 솔잎에 비해 $20{\sim}30%$ 분해능이 떨어지는 것으로 판명되었다 또한 솔잎 열수 추출물을 chloroform, ethyl acetate, butanol을 이용하여 얻은 분획물을 대두유에 첨가하여 저장기간 중 항산화성을 조사한 결과 ethyl acetate 분획물이 가장 효과가 있었다.

Synthesis of Ce-doped In2O3 nanoparticles via a microwave-assisted hydrothermal pathway and their application as an ultrafast breath acetone sensor

  • Acetone, a metabolite detected from the exhaled breath of people doing a diet, can be used for non-invasive monitoring of diet efficiency. Thus, gas sensors with rapid response and recovery characteristics to acetone need to be developed. Herein, we report ultrafast acetone sensors using Ce-doped In2O3 nanoparticles prepared by the one-pot microwave-assisted hydrothermal method. The pure In2O3 sensor shows a high response and fast response time (τres = 6 s) upon exposure to 2 ppm acetone at 300 ℃, while exhibiting a relatively sluggish recovery speed (τrecov = 1129 s). When 20 wt% Ce is doped, the τrecov of the sensor significantly decreased to 45 s withholding the fast-responding characteristic (τres = 6 s). In addition, the acetone response (resistance ratio, S) of the sensor is as high as 5.8, sufficiently high to detect breath acetone. Moreover, the sensor shows similar acetone sensing characteristics even under a highly humid condition (relative humidity of 60%) in terms of τres (6 s), τrecov (47 s), and S (4.7), demonstrating its high potential in real applications. The excellent acetone sensing characteristics of Ce-doped In2O3 nanoparticles are discussed in terms of their size, composition, phase, and oxygen adsorption on the sensing surface.

  • 케토시스 질병의 지표 형질인 milk ${\beta}$-hydroxybutyratc acid (BHBA), milk acetone, 그리고 사료효율의 지표형질 에너지보정유량 및 젖소의 주요 경제 형질인 산유량에 대한 유전력과 이들 간의 유전적 관계를 통하여 젖소의 케토시스 저항성 증진 가능성을 확인하기 위하여 본 연구를 수행하였다. 국내 젖소 14,397두에서 수집된 산유능력 검정자료 75,072건의 기록과 혈통기록 44,954두의 기록을 다형질 임의회귀 검정일 선형 모형을 이용하여 분석하였다. 케토시스 지표형질들의 유전력은 범위 0.06 ~ 0.15, 평균 0.08 (${\pm}0.03$)로 추정되었으며, 케토시스 지표형질들 간에 표현형 상관은 범위 0.73 ~ 0.90, 평균 0.78 (${\pm}0.04$), 유전 상관은 0.93 ~ 0.98 평균은 0.97 (${\pm}0.01$), milk BHBA, milk acetone과 산유량의 표현형 상관은 범위 -0.18 ~ -0.05 평균 -0.08 (${\pm}0.027$), 범위 -0.05 ~ -0.23, 평균 -0.1 (${\pm}0.041$), 유전상관은 -0.55 ~ 0.05, 평균 -0.16 (${\pm}0.18$), -0.62 ~ -0.04, 평균 -0.24 (${\pm}0.17$)로 각각 추정이 되었다. mill BHBA, milk acetone과 ECM의 표현형 상관관계는 0.056 (${\pm}0.023$), -0.04 (${\pm}0.029$), 유전적 관계는 평균 0.023 (${\pm}0.08$), 0.09 (${\pm}0.067$)로 각각 추정되었다. 따라서 산유능력 검정자료 수집 시 부가적으로 수집되는 케톤체를 이용하면 케토시스 저항성 증진을 할 수 있을 것으로 사료된다.

꾸지뽕나무 열매 추출물의 항산화 활성 (Antioxidative Activity of Extrcts from Fruit of Curdrania tricuspidata)

  • The comparative activities of acetone, ethanol, and aqueous fractions extracted from fruit powder of Cudrania tricuspidata by different temperature were tested by in vitro experimental models; peroxidation of linoleic acid and autooxidation of rat hepatic and renal microsomes by using thiobarbituric acid (TBA) for assay of free malondialdehyde production, and scavenging activities of free radicals by DPPH (α, α'-diphenyl-β-picrylhydrazyl). In DPPH method, acetone fraction extracted at 30℃ showed the highest free radical scavenging activities and acetone fractions extracted at 30℃ and 60℃ and ethanol fraction extracted at 30℃ showed stronger than BHT (butylated hydroxitoluene) although used ten-fold lower concentrations. In thiocyanate method used linoleic acid an inhibitory effects of all fractions showed higher than control treatment. TBA method used linoleic acid showed the highest antioxidative activity in acetone fraction extracted at 30℃ and 60℃. an inhibition activity against lipid peroxidation in hepatic microsomes of rats showed the highest at acetone faction extracted at acetone fraction among extracted fractions was shown to be the most potent antioxidative properties and this action was more potent in fractions extracted at 30℃ than those extracted at 60℃.

시베리아산 전나무 바이오매스의 아세톤 용매분해반응 (Acetone Solvolysis of a Siberian Spruce Biomass)

  • In this study, thermochemical degradation by acetone solvolysis reaction of siberian spruce wood was investigated for a temperature range of $200{\sim}400^{\circ}C$. The liquid products by acetone solvolysis from siberian spruce wood produced various kinds of aliphatics, cyclic compounds and aromatics included phenols etc. Combustion heating value of liquid products by acetone solvolysis conversion processes was in the range of $8,010{\sim}8,180cal/g$. The energy yield in acetone solvolysis of siberian spruce wood was as high as 74.2% after 40min of reaction at $400^{\circ}C$. The liquid products from the thermochemical conversion of siberian spruce wood could be used as high-octane-value fuels and fuel additives.

Effects of Petrolatum and Glycerin on Acetone Damaged Canine Skin Barrier

  • The purpose of this study is to investigate whether the effects of topically applied petrolatum and glycerin on the barrier repair of acetone-induced skin damage in 6 beagle dogs. To confirm the effects of petrolatum and glycerin on acetone disrupted skin models, we performed to evaluate the characteristics of transepidermal water loss and SC hydration and scanning electron microscopic observations. TEWL and SC hydration measurements were carried out 3, 6, 12, 24, 48h after applying petrolatum and glycerin during recovery from acute disruption. Our results showed that there were some different effects between petrolatum and glycerin on the acetone damaged skin such as barrier function repair process and SC hydration status. The results indicate that the significant improvement could be observed in glycerin apply more than petrolatum after acetone damages, and further study will be required.

Analysis of Acetone Absorption Spectra Using Off-axis Integrated Cavity Output Spectroscopy for a Real-time Breath Test

  • We analyzed the absorption spectra of acetone in the 3.37 ㎛ mid-infrared range using the off-axis integrated cavity output spectroscopy technique to develop a real-time, in-line breath analysis device. The linear relationship between acetone concentration and absorption increase was confirmed as 0.32%/ppm, indicating that the developed device allows for a quantitative analysis of acetone concentration in exhaled breath. To further confirm the feasibility of using our device for breath analysis, we measured the acetone concentration of human breath samples at the sub-ppm level.

Acetone Enhancement of Cumene Hydroperoxide-supported Microsomal Cytochrome P450-dependent Benzo(a)pyrene Hydroxylation

  • In vitro effects of acetone on cytochrome P450 (P450)-dependent benzo(a)pyrene (B(a)P) hydroxylation supported by cumene hydroperoxide (CuOOH) or NADPH/$O_2 $ systems were studied using 3-methylcholanthrene-pretreated rat liver microsomes. The maximal rate of B(a)P hydroxylation at constant concentration ($80\;{\mu}M)$ of the substrate was observed in the presence of $30\;{\mu}M$ CuOOH. However, at concentrations higher than $30\;{\mu}M$ CuOOH the hydroxylation rates were rapidly decreased. In contrast to CuOOH, at a concentration of $200\;{\mu}M$ NADPH, B(a)P hydroxylation rate reached a plateau. At concentrations higher than $200\;{\mu}M$ NADPH, the rates of substrate hydroxylation were maintained at the maximal rate with no inhibition. Acetone at 1% (v/v) enhanced both CuOOH- and NADPH/$O_2$-supported B(a)P hydroxylation at the optimal concentrations of the cofactors. At concentrations higher than 1% (v/v) acetone, substrate hydroxylation was sterero specific under the support of these two cofactors; it was strongly enhanced with $30\;{\mu}M$ CuOOH, but rather inhibited in the $200\;{\mu}M$> NADPH/$0_2 $ system. The lipid peroxidation rate induced during CuOOH-supported P450-dependent B(a)P hydroxylation was increased as CuOOH concentrations were increased. Acetone in the concentration range of 2.5~7.5%(v/v) inhibited lipid peroxidation during CuOOH supported B(a)P hydroxylation. The finding that CuOOH-supported B(a)P hydroxylation is greatly enhanced by acetone suggests that acetone may contribute more to the activation of oxygen (for the insertion of oxygen into the substrate) in the presence of CuOOH than with NADPH/$O_2$. Acetone may also contribute to the partial inhibition of destruction of microsomal membranes by lipid peroxidation.

Suppression of the Methyl Radical Loss from Acetone Cation within (CH3COCH3)n{CH3COCH3}+ Clusters

  • We have investigated the photophysics of the acetone radical cation in the vacuum ultraviolet energy region by multiphoton ionization combined with time-of-flight mass spectrometry in a cluster beam. We have found that the loss of methyl radical from the acetone radical cations is remarkably suppressed at 10.5 eV when they are solvated by a few neutral acetone molecules. The cluster ion mass spectra obtained by nanosecond and picosecond laser pulses reveal that there are intermolecular processes, occurring in several tens of picoseconds, which are responsible for the survival of the acetone cations in clusters. This remarkable solvation effect on the yield of the methyl radical loss from the acetone cation can be rationalized by the intracluster vibrational energy redistribution and the self-catalyzed enolization which compete with the methyl radical loss process.

  • An experimental investigation is conducted to measure phase change temperature and supercooling when acetone is added to TMA 30 wt% clathrate during cooling process in heat source. Also rate of volume change is investigated when acetone is added to TMA 30 wt% clathrate during the cooling process in heat source -8$^{\circ}C$. The results show that phase change temperature is about 4.5~5.5$^{\circ}C$ when acetone is added to TMA 30 wt% clathrate during the cooling process for heat sink temperature of -6, -7$^{\circ}C$ and -8$^{\circ}C$. Supercooling is repressed about 2~1$0^{\circ}C$ when 0.08 wt% acetone is added to it and rate of volume change is decreased about 2.9% when 0.1 wt% acetone is added for the heat sink temperature of -8$^{\circ}C$.
