  • YBCO superconducting thick films were prepared on Ag wire by electrophoresis in acetone and ethanol with chemically modified suspension. The addition of organic compounds, such as PEG, EG into suspension solution for improving critical current density was investigated. Surface state, deposition condition, pore distribution and cracks were investigated by using SEM photographs. Controlling preparation conditions were studied for reducing these defects. As a results, in acetone solution, the surface crack of samples was decreased with increasing PEG. On the contrary, the surface crack of sample was increasing with increasing the amount of EG. In ethanol solution without I$_2$, which was generally used for an electrolyte, the deposition time was longer than this of acetone. For that reason the sample deposition in ethanol time was needed with enough stirring time for suspending YBCO powder and deposition time.

  • Pyridine and acetone are efficacious inducers of CYP2E1 in both rats and rabbits. The response in the elevation of CYP2E1 levels, changes in CYP2E1 mRNA levels, and enhanced translational processing of hepatic CYP2E1 mRNA during the early phase of CYP2E1 induction by the solvents pyridine and acetone were examined. Time-depen-dent incease in CYP2E1 levels occurred at early times (6-24h) following a single dose of pyridine treatmene, as assessed by Western immunoblot analysis, whereas the levels in CYP2E1 mRNA transiently decreased at 12h post-treatment, returning to the level present in untreated animals.

Yellow Pigment from Rhodospirillum rubrum by Acetone Extraction (Acetone으로 추출한 Rhodospirillum rubrum의 황색색소)

  • A study of general characteristics of acetone extracted pigment from the cell mass of Rhodospirillum rusrum DSM 467 was carried out for the development of natural food colorant. Through visible absorption scanning, it showed seven absorption peaks at 355, 410, 529, 624, 680 and 747nm, and it was shown to be yellow color. In acidic and neutral conditions, the color was yellow, while in the alkaline condition it was greenish yellow. This pigment was stable at pH range between 3.0~10.0, and below 4$0^{\circ}C$. In the presence of light and oxygen, the stability of pigment rapidly degraded and it became unstable in the presence of metal ions such as Cu2+, Al3+ and Fe3+. Through TLC analysis, it was shown to be composed of seven color fractions.

Antioxidant Activities and Antimicrobial Effects of Solvent Extracts from Lentinus edodes (표고버섯(Lentinus edodes) 용매 추출물의 항산화 활성 및 항균 효과)

  • The aim of this study was to investigate the antioxidant and antimicrobial activities of various solvent (acetone, ethyl acetate, and ethanol) extracts from Lentinus edodes. The antioxidant activities were evaluated by measuring total polyphenol and flavonoid contents, 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity. Total polyphenol content and ABTS radical scavenging activity were highest in ethanol extract. ABTS radical scavenging activity of ethanol extract showed the highest value (98.5%), which was similar to that of ascorbic acid (95.7%). The ethyl acetate extract from Lentinus edodes showed relatively high total flavonoid content and DPPH radical scavenging activity. Negative correlations were found between total polyphenol contents and DPPH radical scavenging activities in Lentinus edodes extracts. Antimicrobial activities of the extracts were determined against Bacillus subtilis, Staphylococcus aureus, Micrococcus luteus, Escherichia coli, Pseudomonas aeruginosa, and Enterobacter cloacae by the disc diffusion method. The acetone and ethanol extracts showed moderate antimicrobial activities against almost all tested microorganisms except E. coli and S. aureus, respectively. The ethyl acetate extract showed a significant growth inhibition effect against E. coli, Ent. cloacae, and B. subtilis.

  • To detect bioactive compounds present in unused marine resources, the screening for the 5-lipoxygenase inhitors in Asterina pectinifera, Halocynthia roretzi skin, Nototodarus sloani ink, Anthocidaris crassispina skin, SArgassum horneri, Agarum cribrosum, Odonthalia corymbifera and Desmarestia ligulata was carried out. THe ether and acetone extracts of Sargassum horneri had the strongest antioxygnic activityon lipid oxidation by potato lipoxygenase-1 (one of 5-lipoygenase) among the tested marine samples and their $IC_{50}$ were 0.3 and 1.1g/ml, respectively. The ether and acetone extracts of Asterina pectinifera, the acetone extracts of Halocynthia roretizi, and the acetone extracts of Nototodarus sloani ink had strong inhibitory activity and their $IC_{50}$ were 72.5, 65, 13.3 and $13.3\mu\textrm{g}/ml$, respectively. In addition, the $IC_{50}$ of the acetone extracts of Agarum cribrosum and Desmarestia ligulata, and the ether extracts of Desmarestia ligulata were 15.5, 35 and $30.5\mu\textrm{g}/ml$, respectively. The nonpolar solvent (ether, acetone) extracts of tested marine organism had more antioxigenic effect against 5-lipoxygenase than the polar solvent(water) extracts.

  • The high-purity purification of 2,6-dimethylnaphthalene (2,6-DMN, 10.43 wt%) from the concentrate of DMN isomers recovered from light cycle oil (LCO) through distillation-extraction combination was examined by a crystallization operation. To select the most suitable crystallization solvent for purification of 2,6-DMN, several conventional solvents, which have been employed commercially as crystallization solvents for high-purity performance, were tested, through measurement of purity and yield of 2,6-DMN. The solvents used were acetone, cyclohexanone, ethanol, ethyl ether, ethyl acetate, isopropyl ether, methanol, n-hexane, n-heptane, pyridine, THF, toluene, and a mixture of methanol and acetone. The mixture of 60 vol% methanol and 40 vol% acetone (M/A = 1.5) was found to be suitable for purification of 2,6-DMN in the concentrate of DMN isomers based on purity and yield. Increasing the operation temperature and the volume ratio of solvent (M/A = 1.5) to feed (concentrate of DMN) resulted in improving the purity of 2,6-DMN, whereas the yield decreased. The crystal recovered by crystallization run using the concentrate of DMN isomers contained about 76 wt% 2,6-DMN. Furthermore, for recovery of high-purity 2,6-DMN, crystal containing 76 wt% 2,6-DMN was crystallized. As a result, crystal with 99.7 wt% 2,6-DMN was recovered with 40% yield.

The nature of non-specific inhibitor and natural agglutinin for goose erythrocytes in chicken serum (계혈청(鷄血淸)에 들어 있는 뇌염(腦炎)바이러스에 대한 비특이물질 및 거위 혈구(血球)에 대한 응집소의 성상(性狀)에 관한 연구(硏究))

  • (1) The non-specific inhibitors (NSI) in normal chicken sera were active against all the tested group A and group B arboviruses, but the group B arbovirus were more sensitive than group A arboviruses. (2) The titres of the NSI were distributed nearly uniformly among chickens from seven different age groups to group A arboviruses. In contrast, the NSI titres to group A arboviruses were found to increase with age. (3) No significant difference could be demonstrated between acetone-ether extraction and kaolin adsorption for removal of the NSI in normal chicken sera. (4) After heating, the NSI titres in chicken sera were increased for both group A and group B arboviruses. (5) After heating the sera at $80^{\circ}C$ and $100^{\circ}C$, kaolin adsorption was less efficient for removing the NSI than it, was in unheated serum. Acetone-ether extraction of the NSI was unimpaired after heating at $80^{\circ}C$ but was less efficient after heating at $100^{\circ}C$. (6) The NSI activity was found mainly in the first peak (IgM) and diffused to a part of second peak (IgG) by fractionation of chicken serum by gel filtration through Sephadex G200. After zonal centrifugation of chicken serum in a linear ten to 40 percent sucrose gradient all of the NSI activities were found on the top of the centrifugal tubes. These properties of large molecular size and low density indicated that the NSI in chicken serum were probably lipoproteins. (7) The natural agglutinins for goose erythrocytes in chicken sera were partially destroyed by acetone-ether extraction but not by kaolin adsorption, and were efficiently adsorbed with ten percent goose erythrocytes. No difference of the NA titre was demonstrated with diluents of different pH. (8) The NA in chicken serum was found to possess the properties of IgM by gel filtration through Sephadex G200 and zonal centrifugation in linear ten to 40 percent sucrose gradient.

Characteristics of Cow Milk and Goat Milk Yogurts Fermented by Streptococcus thermophilus LFG Isolated from Kefir (Kefir에서 분리한 Streptococcus thermophilus LFG를 이용한 우유 및 산양유 요구르트의 품질 특성)

  • This study was carried out to investigate the characteristics of goat and cow milk yogurts containing high-exopolysaccharide fermented by Streptoccous thermophilus LFG isolated from kefir. The pH of cow milk yogurt was higher than that of goat milk yogurt. The contents of lactic acid was greater in goat milk yogurt (743.9-1043.8 mg/100 g) than in cow milk yogurt (441.6-709.9 mg/100 g). The numbers of survival lactic acid bacteria were the greatest in goat milk yogurt fermented by Str. thermophilus LFG. Viscosity was greater in cow and goat milk yogurts cultured by Str. thermophilus LFG than in yogurts by Str. thermophilus TH3. Syneresis of yogurt fermented by Str. thermophilus LFG was 9.6-16.1% and 28.2-31.8% in yogurt fermented by Str. thermophilus TH3 after 10 d storage at $4^{\circ}C$. Flavor compounds identified from goat milk were acetone, ethylbutanoate, ethyl-3-methylbutyrate, ethyl-2-butenoate and ethylhexanoate, and those from cow milk were ethylbutanoate, acetone, 2-heptanone and acetoin. Flavor compounds detected from goat milk and cow milk yogurts were acetic acid, butanoic acid, butanol, diethylcarbinol, acetone, diacetyl, decane, 2-methyl-3-pentanone, hexanal, 2-heptanone, acetoin, benzaldehyde, dimethyldisulfide, and dimethyltrisulfide. In sensory evaluation, overall preference and texture values were higher in goat milk yogurt fermented by Str. thermophilus LFG than in cow milk yogurts and the yogurt fermented by mixed culture resulted in the highest score.

Genetic Parameters of Milk β-Hydroxybutyric Acid and Acetone and Their Genetic Association with Milk Production Traits of Holstein Cattle

  • This study was conducted to estimate the genetic parameters of ${\beta}$-hydroxybutyrate (BHBA) and acetone concentration in milk by Fourier transform infrared spectroscopy along with test-day milk production traits including fat %, protein % and milk yield based on monthly samples of milk obtained as part of a routine milk recording program in Korea. Additionally, the feasibility of using such data in the official dairy cattle breeding system for selection of cows with low susceptibility of ketosis was evaluated. A total of 57,190 monthly test-day records for parities 1, 2, and 3 of 7,895 cows with pedigree information were collected from April 2012 to August 2014 from herds enrolled in the Korea Animal Improvement Association. Multi-trait random regression models were separately applied to estimate genetic parameters of test-day records for each parity. The model included fixed herd test-day effects, calving age and season effects, and random regressions for additive genetic and permanent environmental effects. Abundance of variation of acetone may provide a more sensitive indication of ketosis than many zero observations in concentration of milk BHBA. Heritabilities of milk BHBA levels ranged from 0.04 to 0.17 with a mean of 0.09 for the interval between 4 and 305 days in milk during three lactations. The average heritabilities for milk acetone concentration were 0.29, 0.29, and 0.22 for parities 1, 2, and 3, respectively. There was no clear genetic association of the concentration of two ketone bodies with three test-day milk production traits, even if some correlations among breeding values of the test-day records in this study were observed. These results suggest that genetic selection for low susceptibility of ketosis in early lactation is possible. Further, it is desirable for the breeding scheme of dairy cattle to include the records of milk acetone rather than the records of milk BHBA.

Increasing Effect on Storage Stability of Rosemary Extracts used for Various Solvent System on Seasoning Oils (향미유 제품에 대한 추출 용매별 로즈마리 추출물의 저장 안정성 향상 효과)

  • As for red pepper seasoning oil (RPSO), seasoning oil (SO) and pine needle oil (PNO), various organic solvent extracts from rosemary powder and tocopherol are treated as control group. At this time, amounts that are treated were all 1,000 ppm. It was observed by AV (acid value), POV (peroxide value) and carbonyl compounds content of the stored samples during 3 months at 60${\pm}$2$^{\circ}C$ incubation. Tocopherol was shown to be pro-oxidant than the antioxidant in all seasoning oil samples. Icreasing effect of storage stability of chloroform/MeOH extract was the most superior one. Final result of icreasing effect of storage stability from the determinated data was as follows. The storage stability of solvent system by AV and POV analysis was in the increasing order of chloroform/MeOH extract> ethyl alcohol extract>hot water extract>ethyl acetate extract>acetone extract>none treating group> tocopherol treating group, POV was chloroform/MeOH extract>ethyl alcohol extract ${\geq}$ ethyl acetate extract> acetone extract ${\geq}$ hot water extract>none treating group>tocopherol treating group and by carbonyl compound content analysis was in the increasing order of chloroform/MeOH extract>ethyl acetate extract>ethyl alcohol extract>hot water extract>acetone extract>none treating group>tocopherol treating group.
