Establishment of Processing Conditions of Salted Anchovy 1. Changes of Chemical Compositions during Fermentation of Salted Anchovy by Salting Methods (염장 멸치 (Salited Anchovy)의 제조조건 1. 염장방법에 따른 염장 멸치 (salted anchovy)의 제조 중 성분 변화)

  • We investigated the changes of chemical compositions during fermentation of salted anchovy by salting methods for the purpose of establishment of processing condition. Dehydration of anchovy meat occurred remarkably by dry salting compared with that by brine salting and salinity was higher in anchovy by brine salting than by dry salting. Dehydration and salinity were increased in more anchovy fermented at $20^{\circ}C$ than at $5^{\circ}C$. Total nitrogen content was lower in anchovy by brine salting than by dry salting. Amino nitrogen increased remarkably during fermentation of salted anchovy at $20^{\circ}C$, while increased slightly at$5^{\circ}C$. Amino nitrogen showed maximum value on 120 days in dry salting and on 30 days in brine salting at $20^{\circ}C$, respectively. The changes of VBN were similar to the changes of amino nitrogen. The brine salting accelerated hydrolysis of anchovy meat compared with that of dry salting at $20^{\circ}C$, and the hydrolysis were suppressed at $5^{\circ}C$, The POV increased rapidly in dry-salted anchovy than brine-salted anchovy. We suggested that the appropriate processing condition of salted anchovy is to ferment for 5-6 months at $5^{\circ}C$ by addition of $25\%$ salt after pre-salting of raw anchovy.

The Color Measurement and Sensory Evaluation for the Accelerated Fish Sauce Products (속양(速釀) 어장유(魚醬油)에 대한 색(色) 측정(測定) 및 관능검사(官能檢査))

  • The color distribution of fish sauces was studied by using the change in log absorbance per 100nm as a parameter of color tone. A linear relationship was found between the logarithm of absorbance (log A) and wavelength at 450 nm to 650 nm in the color of fish sauces. Change in log A per 100nm $({\Delta}A)$ in the color of the fish sauce products was in the range of 0.55 to 0.59. Absorbance at 450 nm (A 450) of the fish sauce treated with soy sauce koji was high in comparison with A 450 of the fish sauce treated by pronase and control. The color of fish sauces treated with soy sauce koji was dark reddish orange, and was similar to the color of soy sauce. A 450 of every fish sauce increased with the progress of fermentation but ${\Delta}A$of the fish sauces slightly increased at the beginning of fermentation and then decreased at the end of fermentation. In the results of sensory evaluation for the flavor of fish sauce products, the fish sauce product treated with soy sauce koji that 20% salt was added at the first stage was the molt desirable in the strength of flavor, tastefulness and after taste, and the fish sauce was also the best in acceptability.

Study on Roughage Degradation and Adhesion of Rumen Fibrolytic Bacteria by Real-Time PCR (Real-Time PCR 기법을 이용한 반추위 섬유소분해 박테리아의 부착과 조사료 분해에 관한 연구)

  • The comparisons between cellulolytic bacteria adhesion on rice straw and fiber digestion in time course during rumen fermentation were studied in situ. The adhesions of cellulolytic bacteria, F. succinogenes. R. albus and R. flavefaciens, were measured by RT-PCR. When the rice straws were incubated at 0. 2, 4, 8, 12 and 24 hours of the in situ rumen, straw was degraded with increasing speed during the incubation and showed the highest disappearance increasing rate (DM g/h) from 8 to 12 hour. The adhesions of F. succinogenes, R. flavefaciens and R. albus were achieved above 80% in 1 hour of in situ rumen fermentation and then keep adhesive population up after the time of fermentation. When the in situ samples were collected at 0, 5, 10, 30 and 60 min to detect the early stages of adhesion on the rice straws ingested into rumen, the numberous adhesive colony of F. succinogenes, R. flavefaciens and R. albus were detected in 5 min. In case of rice straw treated with 0, 2, 4 and 8% NaOH, all of three cellulolytic bacteria showed the increasing trends of adhesion with increasing DM disappearance of rice straw by higher concentration of NaOH at 12 hour of in situ. However, there were showed respectively difference at 24 hour. The present results gave certain evidence that adhesion of cellulolytic bacteria is definitely achieved in early stage of roughage ingestion into rumen, their colony develop the stable communities on roughage in process of rumen fermentation and then fiber degradation is accelerated.

Effect of Organic Gemanium, Oligosaccharide and Starters on Fermentation of Fresh Kimchi Juice (김치즙액의 발효에 미치는 유기게르마늄, 올리고당 및 Starter의 영향)

  • Changes in pH, titratable acidity, CO$_2$production, reducing sugar, and lactic acid bacteria of fresh kimchi juice supplemented with combinations of 3 lactic acid bacteria and germanium(Ge)-132 or fructooligosaccharide(FO) during fermentation at 30$^{\circ}C$ were investigated to assess the potential for extending the shelf life and enhancing the functional properties in kimchi. In kimchi juice containing Ge-132, sample(I) (inoculated with a mixture of bacteriocin-producing SNF-13 strain and E. faecium) exhibited that the amounts of organic acid and evolved CO$_2$gas were lower than those of the other starter samples(II-IV). The growth of lactic acid bacteria naturally present in kimchi juice, particularly Lb. plantarum and Leu. mesentroides, may be inhibited due to competition of the isolated SNF-13 strain and E. faecium by Ge-132. During fermentation of kimchi juice containing FO sugar, the contents of organic acid and evolved CO$_2$gas On juice broth with 4 starters were predominantly higher than those of control and Ge-132 groups, and then the growth of lactic acid bacteria originated from kimchi ingredients was thought to be markedly accelerated. Our results indicated that functional properties like the extension of shelf life and increase of biological activity in kimchi were enhanced by adding Ge-132 and bacterium-producing lactic acid bacterium, which were resistant to organic acid and stimulated by Ge-132.

Influence of Vibration on Freezing and Fermentation of Watery Kimchi (진동이 물김치의 동결과 발효에 미치는 영향)

  • Vibration at frequency of 20 Hz and 0.18 g of acceleration was applied to 600 g watery kimchi contained in a glass jar of 1 L at subfreezing temperature (-3 and $-6^{\circ}C$) and $10^{\circ}C$ in order to see its effect on freezing and fermentation behaviors, respectively. The vibration at the subfreezing temperature delayed the freezing process and contributed to maintaining small ice crystal slurry in subsequent frozen storage. The vibration at $10^{\circ}C$ accelerated the acid and carbon dioxide production from the watery kimchi, which may be beneficial in shortening ripening time and attaining fresh cool taste.

Quality of Accelerated Salt-fermented Anchovy Sauce Prepared with Fig (무화과를 이용한 속성발효 멸치액젓의 품질)

  • To facilitate fermentation of liquid anchovy sauce, 3% unripe figs, ripe figs, or fig leaves were added to the sauce and fermented at $25^{\circ}C$ for 60 days. The anchovy sauce prepared with ripe figs showed higher protease activity and better red-pupple color than others. Amino-nitrogen content in anchovy sauce treated with ripe figs was attained to 600 mg% within 10 days. Total free amino acid content in the product with fig addition showed about twice higher, 4543.21 mg%, than the control, 2255.29 mg%. Hypoxanthin was found as a major components of nucleotide and their related compounds in the fermented anchovy sauce. However, using the unripe figs and fig leaves impart greenish color and grass taste to the product.

Standardization of Ingredient Ratios of Wooung (Burdock, Arctium lappa, L) Kimchi (우엉김치 재료배합비의 표준화)

  • This study was conducted to standardize ingredient ratios of wooung kimchi. The ingredient ratios of model wooung kimchiwere determined by the survey in Pusan and Kyungnam province and using the literatures including cooking books. Several kinds of wooung kimchi were prepared by adjusting the ingredient ratios fo the model wooung kimchi within standard deviation. The wooung kimchi with different ingredient ratios were fermented for 6 days at 15$^{\circ}C$. The chemical, microbial and sensory properties of the wooung kimchi were investigated. There was little change in pH but the counts of lactic acid bacteria were decreased, as the ratio of pickled anchovy juice became high. The wooung kimchi adding 9.4% pickled anchovy juice obtained high score in appearance and overall acceptability. The counts of lactic acid bacteria were increased in wooung kimchi adding 5% red pepper powder, and there was obtained better result in appearance, texture and overall acceptability than the other groups. The activity, reducing sugar and counts of lactic acid bacteria were increased, as the ratio of glutinous rice paste became high. The wooung kimchi including 6% glutinous rice paste showed the highest score in overall acceptability. The fermentation process of wooung kimchi accelerated, as the ratio of garlic became high. The wooung kimchi adding 3% garlic showed good appearance and acceptability. In addition to these, the addition of 1.3% ginger ehhanced the appearance, texture and overall acceptability fermented anchovy juice, 5.0% red pepper powder, 6.0% glutinous rice paste, 3.0% crushed garlic and 1.3% crushed ginger.

Ethanol Production from Xylulose by Saccharomyces cerevisiae (효모에의한 Xylulose로부터의 에탄올 생산)

  • Xylose represents a major component of cellulosic materials. This paper describes patterns of ethanol fermentation by Saccharomyces cerevisiae from xylulose, which is an isomer of xylose. Special emphasis was placed on the effects of xylulose concentration and growth temperature on cell growth and ethanol yield. The maximum specific growth of $0.087 1/hr^{-1}$ was obtained at an initial xylulose concentration of 5 g/1. The ethanol yield was propotional to initial xylulose concentrations. A xylulose concentration of 16 g/l resulted in the maximum ethanol yield of 0.49 g EtOH/g xylulose, which corresponds to 90% of a theoretical value. It is interesting to nota that xylulose metabolism was accelerated by the presence of glucose as a carbon source.

Preparation of Wine Using Wild Yeast from Dried Omija and Optimal Nutritional Requirements for Alcoholic Fermentation (건조 오미자에서 분리된 야생 효모로 와인 제조 및 알코올 발효 시 영양요구성 조사)

  • This study was conduced to ferment high quality wine by using Omija fruit. Dry Omija farmed and dried in the Moonkyung area was used in this study. The Omija was soaked in 10~40 folds of distilled water to extract water-soluble components and the fluid was filtered after soaking for 6 hours at $50^{\circ}C$. Strains of alcoholic yeast were isolated respectively from spoiled Omija extract. Isolated alcoholic yeasts, OM-1 and OM-2, showed a round to ellipsoidal shape and formed white or milky white colonies on a solid YM medium. Two yeasts produced 10.33~11.23% alcohol from Omija extract adjusted to $10^{\circ}Brix$ with sugar. Their abilities to ferment alcohol were higher than those of other yeast strains belonging to Saccharomyces cerevisiae such as KCTC 7296 (standard strain of Korean Biological Resources Center), Makgeolli yeast, or beer yeast. The isolates OM-1 and OM-2 showed similar abilities in alcohol fermentation. However, the wine fermented by OM-2 got a better sensory score especially with color. Growth of OM-2 was significantly accelerated by addition of a 0.1% urea and 0.02% mineral mixture. A vitamin mixture was effective for the growth only when urea was added as well.

Effects of Extrusion-Texturization on Defatted Soy Flour for Soy Sauce Fermentation (압출성형기에 의한 전처리가 탈지대두분의 장유 제조특성에 미치는 영향)

  • Extruded defatted soy flour (DSF) with twin screw extruder was compared with steam treated DSF for soy sauce fermentation. Independent variables of response surface methodology (RSM) for extrusion were barrel temperature $(145{\sim}165^{\circ}C)$, feed moisture content $(25{\sim}35%)$, and feed rate $(20{\sim}30\;kg/hr)$. Extrusion conditions for production of the extrudates having highest water absorption capacity, lowest bulk density and highest expansion ratio were obtained by regression analysis of each dependent variable. Electron photomicrographs revealed significant increase of porosity inside the koji made from extruded DSF. Therefore, growth of fungi inside the koji made from extrudates of DSF was accelerated to achieve higher activities of protease, ${\alpha}-amylase$, and glucoamylase compared to the koji made from steamed DSF. Although amino acid composition of soy sauces prepared from two different DSF did not show any significant difference, the content of free amino acids in soluble nitrogen compounds was higher $(13%{\pm}2)$ in soy sauce made from extruded SDF. During the whole period of fermentation color of soy sauce made from extrudates of DSF was darker. Sensory evaluation did not reveal any serious off-odor and off-taste in the soy sauce fermented from extruded DSF.
