Effect of Molecular Weights of Polyethyleneimine on the Polyplex Formation with Calf Thymus DNA

  • In this study we investigated the spectral properties, including electric absorption, circular and linear dichroism (CD and LD), and fluorescence emission, of DNA in a DNA-branched polyethyleneimine (BPEI) polyplex at various polymer molecular weights (M$\_$w/) and BPEI-amine-to-DNA-phosphate ratios (N/P ratios). All BPEIs exhibited a common N/P dependence in their absorption and CD spectra. At N/P ratios < 1.0, we observed some hyperchromism in the absorption spectrum, red-shifts in CD bands, and decreases in LD intensity and fluorescence intensity of intercalated ethidium. At intermediate N/P ratios, complete collapse of all spectra occurred. As the N/P ratio increased further, the polyplex dissolved in water. From its characteristic CD spectrum obtained under these conditions, we conclude that the DNA exists in a B-like form. The fluorescence and LD intensities never recovered even at high N/P ratios- which indicates that the dissolved polyplex possesses positive charges and the DNA in the polyplex is condensed despite its B-form CD spectrum. The N/P range in which the absorption and CD signals collapsed was wider when the BPEIs M$\_$w/ decreased. In the case where the BPEIs M$\_$w/ was 0.8 k, recovery of the absorption and CD spectral properties at a high N/P ratio was never achieved, which suggests that the molecular weight of the polymer plays an important role in its dissolution at a high N/P ratio.

  • We have investigated the spectral properties of DNA, including its electric absorption, circular and linear dichroism (CD and LD), and fluorescence emission, in the DNA-linear polyethyleneimine (LPEI) and DNA-branched polyethyleneimine (BPEI) complexes at various polymer concentrations. The spectral properties of both complexes are similar. We observed a relatively moderate change in the absorption and CD spectra at low amine/DNA phosphate (NIP) ratios (< 0.5), followed by a drastic collapse within the N/P range from 0.8 and 1.0. The absorption and CD spectra recovered as the N/P ratio increased to ca. 1.2. In contrast, the LD and emission of ethidium intercalated between the DNA bases decreased almost linearly at N/P ratios between 0.0 and 1.0. These spectra never recovered at higher N/P ratios. We believe that the moderate changes in the spectrum at low N/P ratios occurred because of electrostatic interactions between DNA and BPEI, while the collapsed spectra at N/P ratios between 0.5 and 1.5 occurred because of condensation/aggregation of the DNA. Considering the structure of the polymers, we suggest that the secondary amino group of LPEI and all three amino groups of BPEI are equally involved in DNA condensation.

Calculation of Energy Dependent Neutron Correction Coefficient Ratios of Natural Rhodium in Energy Region from 0.003 to 100 eV

  • In the neutron capture experiment and calculation, the neutron absorption and scattering are very important. Especially these effects are conspicuous in the resonance energy region and below the thermal energy region. In the present study, we obtained energy dependent neutron absorption ratios of natural rhodium in energy region from 0.003 to 100 eV by MCNP-4B Code. The coefficients for neutron absorption was calculated for several types of thickness. In the lower energy region, neutron absorption is larger than higher region, because of large capture cross section (1/v). Furthermore it seems very different neutron absorption in the large resonance energy region. These results are very useful to decide the thickness of sample and shielding materials.

  • Unlike many laboratory-scale studies on absorption of organic compounds (VOCs), limited pilot-scale studies have been reported. Accordingly, the present study was carried out to examine operation parameters for the effective control of a hydrophilic VOC (methyl ethyl ketone, MEK) by applying a circular pilot-scale packed-absorption system (inside diameter 37 cm ${\times}$ height 167 cm). The absorption efficiencies of MEK were investigated for three major operation parameters: input concentration, water flow rate, and ratio of gas flow-rate to washing water amount (water-to-gas ratio). The experimental set-up comprised of the flow control system, generation system, recirculation system, packed-absorption system, and outlet system. For three MEK input concentrations (300, 350, and 750 ppm), absorption efficiencies approached near 95% and then, decreased gradually as the operation time increased, thereby suggesting a non-steady state condition. Under these conditions, higher absorption efficiencies were shown for lower input concentration conditions, which were consistent with those of laboratory-scale studies. However, a steady state condition occurred for two input concentration conditions (100 and 200 ppm), and the difference in absorption efficiencies between these two conditions were insignificant. As supported by an established gas-liquid absorption theory, a higher water flow rate exhibited a greater absorption efficiency. Moreover, as same with the laboratory-scale studies, the absorption efficiencies increased as water-to-gas ratios increased. Meanwhile, regardless of water flow rates or water-to-gas ratios, as the operation time of the absorption became longer, the pH of water increased, but the elevation extent was not substantial (maximum pH difference, 1.1).

  • The temperature rise of the mortars while are being irradiated by ultrasonic waves and immersed in a liquid medium, are measured. The mixing ratios of sand to cement are varied for the different mortars. The results of the experiment are as follows, 1. The temperature rise of the mortar is decreased as themixing ration (S/C) is increased. 2. Thd temperature rise of the specimen is increased as its length is increased. 3. The surface conditions, either smooth or rough, may not have much influence on the temperature rise of the mortar. 4. The initial slope of temperature rise may not have much effect of the viscosity of liquid. The results, describes above, appear well coincide with the theory that the temperature rise is attributed mainly to the absorption heating. Since the absorption heating of the mortars varies with the mixing ratios of sand to cement, the strength of them would be estimated by means of irradiation of ultrasonic waves on the specimen.

  • This work proposes a simple method to use the added absorption per person values to predict the expected values of the acoustical conditions in occupied classrooms. This method is based on the effects of the values of added absorption per person on the unoccupied total absorption values of the classrooms and on other room acoustical parameters. The total sound absorption in an unoccupied classroom can be calculated from measured reverberation times in the classroom. The expected occupied absorption can be calculated using equation which was obtained in a previous study (Choi, 2017) by fitting a linear regression line to a plot of total occupied absorption versus the corresponding unoccupied total absorption values measured in 12 university classrooms. The ratios of occupied-to-unoccupied sound absorption are used to predict increments in the values of acoustical parameters that result when occupants are added to the rooms.

Simulation of $H_2O/LiBr$ Triple Effect Absorption Systems with a Modified Reverse Flow

  • In this study, a modified reverse flow type, one of the triple effect absorption cycles, is studied for performance improvement. The cycle simulation is carried out by using EES(Engineering Equation Solver) program for the working fluid of $H_2O/LiBr$ solution. The split-ratios of solution flow rate, UA of each component, pumping mass flow rate of solution are considered as key parameters. The results show that the optimal SRH (split ratio of high side) and SRL (split ratio of low side) values are 0.596 and 0.521, respectively. Under these conditions, the COP is maximized to 2.1. The optimal pumping mass flow rate is selected as 3 kg/s and the corresponding UAEV A is 121 kW/K in the present system. The present simulation results are compared to the other literature results from Kaita's (2002) and Cho's (1998) triple effect absorption systems. The present system has a lower solution temperature and a higher COP than the Kaita's modified reverse flow, and it also gives a higher COP than the Cho's parallel flow by adjusting split ratios.

Estimation of Neutron Absorption Ratio of Energy Dependent Function for $^{157}Gd$ in Energy Region from 0.003 to 100 eV by MCNP-4B Code

  • Gd-157 material has very large neutron capture cross section in the thermal region. So it is very useful to shield material for thermal neutrons. Futhermore, in the neutron capture experiment and calculation, the neutron absorption and scattering are very important. Especially these effects are conspicuous in the resonance energy region and below the thermal energy region. In the case of very narrow resonance, the effect of scattering is to be more considerable factor. In the present study, we obtained energy dependent neutron absorption ratios of natural indium in energy region from 0.003 to 100 keV by MCNP-4B Code. The coefficients for neutron absorption was calculated for circular type and 1 mm thickness. In the lower energy region, neutron absorption is larger than higher region, because of large capture cross section (1/v). Furthermore it seems very different neutron absorption in the large resonance energy region. These results are very useful to decide the thickness of sample and shielding materials.

A New All-optical Flip-flop Based on Absorption Nulls of an Injection-locked FP-LD

  • A new all-optical flip-flop (AOFF) method based on the absorption nulls of an injection-locked Fabry-Perot laser diode (FP-LD) in transverse magnetic (TM) mode is proposed and experimentally demonstrated. For the set and reset operations of the AOFF, injection locking and the destructive minus of beating in transverse electric (TE) mode are used. The absorption nulls on the TM mode are modulated according to the operations, and then non-inverted (Q) and inverted (${\bar{Q}}$) outputs can be obtained simultaneously. Thanks to the use of several absorption nulls, the proposed AOFF can achieve multiple outputs with extinction ratios of more than 15 dB. Even though the experiment is demonstrated at 100 Mbit/s, the results of previous experiments using the injection of a CW holding beam imply that the operation speed can increase to 10 Gbit/s.

Strong Absorption of Cations into a Cation Exchange Resin in Concentrated HClO$_4$

  • The absorptions of Fe(Ⅲ), Tb(Ⅲ), Tl(Ⅰ), Ce(Ⅲ), Th(Ⅳ), and $UO_2^{2+}$ ions into the Dowex 50W-X2, 100-200 mesh resin were investigated by spectrophotometry to understand the abnormal strong absorption behavior of cations to cation exchange resins in concentrated HClO4. The distribution coefficients increase in the order : Tl(Ⅰ) < Fe(Ⅲ) < Tb(Ⅲ)∼Ce(Ⅲ) < $UO_2^{2+}$< Th(Ⅳ) and the order is interpreted in terms of the ratio of charge-to-ionic radius. The mole ratios of increment of $ClO_4^-$ ion absorption to metal ion absorption showed the same order as the distribution coefficients, which indicates that the electrostatic association between $ClO_4^-$ ion and metal ion plays a major role in the strong absorption.
