한우(韓牛)의 위장관(胃腸管)에 존재(存在)하는 내분비세포(內分泌細胞)의 면역세포화학적(免疫細胞化學的) 연구(硏究) (Immunocytochemical study of the endocrine cels in the gastrointestinal tract of the Korean native cattle)

  • Regional distribution and relative frequency of endocrine cells in ten portions of the gastrointestinal tract of the Korean native cattle were observed by immunocytochemical methods using specific antisera against chromogranin, serotonin, somatostatin, glucagon, bovine pancreatic polypeptide(BPP), motilin, gastric inhibitory polypeptide(GIP), neurotensin, secretin, gastrin and substance P. The results observed are summarized as follows: In the abomasum, chromogranin-, serotonin-, somatostatin-, motilin-, glucagon-, gastrin-, and substance P-immunoreactive cells were found. Chromogranin-and serotonin-immunoreactive cells were more numerous in the fundic region than pyloric region. Somatostatin- and gastrinimmunoreactive cells were numerous in the pyloric region than in the fundic region. In the small intestine, chromogranin-, serotonin-, somatostatin-, glucagon-, BPP-, motilin-, gastrin-, GIP-, neurotensin-, secretin-, and substance P-immunoreactive cells were detected. Chromogranin-, somatostatin-, GIP- and secretin-immunoreactive cells were most numerous in the duodenum, while BPP-, motilin-, glucagon-, neurotensin- and substance P-immunoreactive cells were rarely seen in the small intestine. In the large intestine, chromogranin-, serotonin- and BPP-immunoreactive cells were widely distributed and most numerous in the rectum. Somatostatin-, glucagon- and substance P-immunoreactive cells were rarely seen in the large intestine.

Response of Pancreatic Exocrine Secretion in Sheep Fed Different Type and Amount of Hay

  • Three wethers fitted with silastic catheters for collection of pancreatic juice, and cannulas located in the abomasum and the duodenum were used to investigate the effects of different hay and energy intake on pancreatic exocrine secretion. The wethers were fed Italian ryegrass hay or alfalfa hay at maintenance energy requirement and alfalfa hay ad libitum. High energy intake from alfalfa significantly increased abomasal flow of dry matter and both the concentration and daily secretion of ${\alpha}-amylase $. The high energy intake also tended to increase daily secretion of lipase, trypsin and chymotrypsin through the large volume of pancreatic juice. Compared with Italian ryegrass hay, alfalfa hay at the maintenance decreased abomasal dry matter flow, but increased concentration of ${\alpha}-amylase $ in the pancreatic juice, and tended to increase daily secretion of ${\alpha}-amylase $. The secretion of the other enzymes was not different between the two hays at maintenance intake. These results suggest that the kind of hay could change the concentration of ${\alpha}-amylase $ in the pancreatic juice, and that the intake level of alfalfa hay affects the ${\alpha}-amylase $ concentration and the juice volume secreted from the pancreas.

홀스타인 독우(犢牛)에 발생(發生)한 질산염중독(窒酸鹽中毒) (Nitrate Poisoning Occurred in Holstein Calves)

  • Nitrate poisoning was observed in Holstein calves raising for beef purpose in a dairy-farm of Gyeongbug province. The results observed were summarized as follows: 1. These calves had been fed mainly cornstalk, and a qualitative reaction by "dephenylamine blue test" for nitrate was positive in the serum of the affected calves and in the cornstalk provided. 2. In the clinical signs, the affected calves were suddenly recumbent without premonition and then dyspnea, followed by death. Death also occurred after inappetence, depression, dyspnea, cyanosis of mucous membrane, and terminal anoxic convulsions were observed. 3. In the macroscopical findings, the blood was dark brown (chocolate in color) and more or less poor coagulative. Petechiae of epicardium, severe cloudy swelling of liver, cloudy swelling and hemorrhage of kidney, congestion and hemorrhage of spleen, congestion and edema of lung, and congestion af abomasum and small intestine were observed in these cases. 4. Microscopical changes observed in these cases were peripheral congestion of lobules, centrilobular necrosis and sinusoidal dilatation in liver, hydropic degeneration of hepatic cells, hemorrhage and edema of kidney, necrosis of convoluted-tubular epithelium, and dilatation of Bowman's space and convoluted tubule. There were also congestion and hemorrhage of spleen, hemorrhage and edema of lung, cloudy swelling of myocardium, and congestion and hemorrhage of various organs.

Recent insight and future techniques to enhance rumen fermentation in dairy goats

  • Recent development of novel techniques in systems biology have been used to improve and manipulate the rumen microbial ecosystem and gain a deeper understanding of its physiological and microbiological interactions and relationships. This provided a deeper insight and understanding of the relationship and interactions between the rumen microbiome and the host animal. New high-throughput techniques have revealed that the dominance of Proteobacteria in the neonatal gut might be derived from the maternal placenta through fetal swallowing of amniotic fluid in utero, which gradually decreases in the reticulum, omasum, and abomasum with increasing age after birth. Multi "omics" technologies have also enhanced rumen fermentation and production efficiency of dairy goats using dietary interventions through greater knowledge of the links between nutrition, metabolism, and the rumen microbiome and their effect in the environment. For example, supplementation of dietary lipid, such as linseed, affects rumen fermentation by favoring the accumulation of ${\alpha}$-linolenic acid biohydrogenation with a high correlation to the relative abundance of Fibrobacteriaceae. This provides greater resolution of the interlinkages among nutritional strategies, rumen microbes, and metabolism of the host animal that can set the foundation for new advancements in ruminant nutrition using multi 'omics' technologies.

강제급수(强制給水)가 우근육(牛筋肉)의 함수율(含水率)과 조직학적성상(組織學的性狀)에 미치는 영향(影響) (Effect of Forced Oral Administration of Excessive Tap water on the Water Content and Histological Changes of Some Muscles of Korean Cattle)

  • Experimental studies on the water content of muscles and the histological changes of muscles, digestive tract and kidney, influenced by forced oral administration of excessive tap water, were performed by using 10 Korean bulls. Experimental animals were divided into two groups, five heads for test group and five for control group. All of the bulls used in this experiment were weighing between 250 kg and 300 kg and 3 and 4 years of age. In each test animal, a total of 140 litters of tap water was passed in the forestomach by means of catheterization within 12 to 18 hours. And each of them was slaughtered immediately after the animals showing symptoms of respiratory distress. In control group, the animals were allowed to drink tap water normally. From test and control animals after slaughter, each 10 gm of M. biceps femoris, M. satorius, M. adductor, M. gluteous supercialis, M. iliocostalis lumborum, and M. transversus costarum were taken from definite parts in order to measure water content. In the histopathological studies tissues of rumen, reticulum, omasum, abomasum, duodenum, colon and kidney were taken as wall as the above mentioned muscles. The results obtained were as follows; 1. Increase of water content in the muscles of test group were 4.6% in M. satorius, 4.24% in M. transversus costarum, 4.14% in M. gluteous supercialis, 4.02% in M. adductor, 3.88% in M. biceps femoris, and 2.46% in M. iliocostalis lumborum respectively. The highest increase was found in M. satorius and the lowest in M. iliocostalis lumborum. 2. In test animal, average increasing value of water content in muscles was 3.9% and shown highly significant (p<0.01). 3. On the microscopical findings of carcass, marked watery edema was observed all of the subcutaneous tissues and intermuscular connective tissues. 4. Microscopically, the epimysium, perimysium and endomysium were widened, and the muscle fibers were lacerated. The cells of stratum lucidum in the epithelium of rumen and reticulum were shown marked vacuolization. In the kidney, dilatation of Bowman's spaces and proximal tubles was observed.

Digestion and Nitrogen Utilization by Sheep Fed Diets Supplemented with Processed Broiler Litter

  • In vivo digestion and metabolism trials were conducted with 10 wethers equipped with ruminal, abomasal, and ileal cannulae to evaluate digestion of ensiled broiler litter (EBL), deepstacked broiler litter (DBL), and composted broiler litter (CBL). Wethers were fed a low protein (6.3% CP) basal diet alone or supplemented to 10.3% CP with EBL, DBL, CBL or soybean meal (SBM). All diets were formulated to be isoenergetic (56% TDN, DM basis). Apparent digestibilities of DM, OM, and ADF were not affected (p<0.05) by diet, but digestibility of CP was improved (p<0.05) by N supplementation. Apparent digestibility of CP was lower (p<0.05) for diets supplemented with CBL and DBL than for diets supplemented with SBM and EBL. Ruminal $NH_3$ concentration was 20 to 24 mg/dl at 2 h after feeding litter-supplemented diets compared with 13 mg/dl for SBM. Abomasal N, $NH_3$ N, and nonammonia N flows were increased (p<0.05) by N supplementation, whereas microbial N flow was not influenced (p<0.05) by diet. Compared with SBM and EBL, undegraded dietary CP flow to the abomasum tended to be greater (p<0.1) when wethers were fed DBL and CBLsupplemented diets. Retention of N (g/d) also was greater (p<0.05) due to greater (p<0.05) N intake and lower (p<0.05) urinary N excretion when wethers were fed diets supplemented with litter (especially EBL) vs. SBM. Overall, characteristics of ruminal fermentation and digestion indicated that broiler litter N was utilized efficiently by wethers, but ensiling may be preferable to deepstacking or composting.

EFFECT OF SOYBEAN EXTRUSION ON NITROGEN METABOLISM, NUTRIENT FLOW AND MICROBIAL PROTEIN SYNTHESIS IN THE RUMEN OF LAMBS

  • Soybeans were dry extruded at three different temperatures (125, 135 and $145^{\circ}C$) for 30 s. Four lambs fitted with cannulae in the rumen and abomasums were used in a balanced $4{\times}4$ Latin square design. Lambs were fed at 2 h intervals for 12 times a day with automatic feeder to maintain steady state conditions in digestive tract. A dual-phase marker system was used to estivate ruminal flow rate of both liquid and solid digesta. Objectives of this study were to determine the effect of extrusion temperature of raw soybean on the ruminal liquid and solid dilution rate, nitrogen digestion and flow at the abomasum and availability of amino acid in lambs. There were no significant effects of extrusion on liquid and solid dilution rate, and liquid volume. Ruminal liquid flow rate was not influenced by extrusion and ranged from 389 to 435 ml/hr. Extrusion had no influence on ruminal OM digestion and flow rate to the abomasums. Dietary N flow to the abomasums increased (p < 0.05) as extruding temperature increased. Extruding temperature had a significant effect (p < 0.05) on flow of N escaping ruminal degradation and ranged from 34.91 to 57.38%. Microbial N synthesized/kg OMTDR ranged from 27 to 37 g and highest with $145^{\circ}C$ ESB diet. Extrusion decreased the amount of degradable amino acid in the rumen and increased the supply of amino acid to the lower gut, especially with 135 and $145^{\circ}C$ ESB diets.

The Effect of Energy Supplementation on Intake and Utilisation Efficiency of Urea-treated Low-quality Roughage in Sheep II. Rumen Kinetics and Acetate Clearance Rate

  • Inadequate supply of glucose or glucogenic substrates to the body tissues can affect metabolism of absorbed acetogenic metabolites from the gut and therefore, influence feed intake in ruminants. This study investigated the effect of energy supplementation on rumen kinetics in the gut, and the acetate clearance rate in the body tissues of sheep fed low quality basal roughage. A basal diet consisting of urea-treated mixture of wheaten chaff and barley straw (3:1 DM) containing 22.2 g N/kg DM was used. Four Merino cross wethers weighing $45{\pm}4.38\;kg$ fitted with permanent rumen and abomasal cannulae were allocated to four treatments in a $4{\times}4$ LSD. The treatments were basal diet ($E_0$), or basal diet supplemented with sucrose (112.5 g/d) administered intraruminally ($E_R$), abomasally ($E_A$), or via both routes (50:50) ($E_{RA}$). There was no difference (p>0.05) in the rumen liquid kinetics parameters between the four dietary treatments. However, there was a trend of animals supplemented with sucrose wholly or partly through the abomasum having lower faecal DM and therefore poor pellet formation, and low pH. Although the glucogenic potential of the fermentation products absorbed from the rumen was increased (p<0.001) by intra-ruminal supplementation with sucrose ($E_R$ and $E_{RA}$), there was no significant difference (p<0.05) in acetate clearance rate between the four dietary treatments.

우측 겸부를 통한 제4위전위증 교정 수술과 제1위 절개 수술의 병행 (Operation of Abomasal Displacement and Foreign Body Removal in the Rumen through the Right Flank Celiotomy)

  • Twenty one cows in Goyang and Paju cities were referred due to displacement of the abomasum and foreign body in the rumen. Omentopexy and rumenotomy through a right flank celiotomy were performed for treatment of abomasal displacement and the foreign body removal in the rumen. The right paralumbar fossa is clipped and prepared surgically. Local anesthesia is instituted by performing inverted L block. The abdomen was entered through 25 to 30 cm vertical incision in the right paralumbar fossa starting 4 to 5 cm ventral to the transverse processes of the lumbar vertebrae. A 14-gauge needle with rubber tubing attached is inserted to relieve the gaseous pressure and to facilitate further exploration and manipulation. The rumen was gently pulled out of the abdominal cavity and incision was made at the omentum. Rumenotomy was done and retrieved the foreign body. After the rumen was rinsed with sterile saline, the rumen wall was closed by a Lembert suture technique. The omentum was closed by a simple continuous suture. Right flank omentopexy was performed for the surgical correction of abomasal displacement. Recovery results among 21 cows included 9 excellent, 5 good, 2 fair and 5 bad. It was considered that operation of abomasal displacement and foreign body removal in the rumen through right flank celiotomy was a good surgical technique to reduce expenses, surgical pain, and surgery time.

Somatotropic Axis and Nutrition in Young Ruminants around Weaning Time

  • The somatotropic (GH-IGF-I) axis consists of many hormonal and nutritional factors that control GH release from the somatotrophs in the anterior pituitary. The GH-releasing substances are GHRH and GHS (GHRP or ghrelin), while the GH release-inhibiting substances are somatostatin (SRIF), insulin-like growth factor-I (IGF-I), leptin and glucocorticoids. However, there is evidence showing that nutrition is involved in the control of the somatotropic axis. In addition, weaning is a drastic event for neonates because their alimentary and endocrine circumstances are changed due to the switch, even if gradual, from a liquid milk diet to one composed of such solids as hay and grains. The biological role of ghrelin is one of the hormonal factors that have been focused on ever since ghrelin was discovered at the end of the last century. A 27-amino acid peptide that is mainly synthesized and released from the abomasum epithelium, ghrelin has not been fully evaluated in relation to the somatotropic axis of the ruminant. It has also proven difficult even to investigate the cellular mechanisms of ghrelin action, because this hormone exerts animal-species-dependent actions via a complex set of intracellular signaling pathways. This is also the case for the action of leptin. Another substance, IGF-I, shows a partial inhibitory action on GH secretion in the ruminant. The effect of nutrition is also different among animal species. This is evident by the fact that undernutrition suppresses the circulating GH levels in rodents, but increases it in ruminants and humans. Recently, weaning has been shown to change the postprandial GH responses in ruminants; milk feeding increases, but hay and concentrate feeding suppress, the postprandial circulating GH levels. Even if the postprandial GH level is increased, the ghrelin level is decreased by milk feeding. Macronutrients also possess stimulatory and inhibitory actions on GH secretion in vivo and in vitro. These findings indicate the complexity of the control mechanisms of the somatotropic axis. In the present review, we summarize recent findings on the factors controlling the axis of the ruminant.
