Screening and Classification of Actinomycetes Producing $\alpha$-Amylase Inhibitors and the Isolation, their Kinetic Studies of $\alpha$-Amylase Inhibitors ($\alpha$-Amylase 저해제 생산 방선균의 선별과 분류 및 $\alpha$-Amylase저해제의 분리와 Kinetics 연구)

  • To find microorganisms of producing $\alpha$-amylase inhibitors, actinomycetes were isolated from soil samples that were collected at different locations in Korea and screened for enzyme inhibitory activity. A strain of these microbes had a high inhibitory activity and was identified as one of the genus Streptomyces by morphological, biochemical and physiological studies according to the methods of the International Streptomyces Project (ISP). The medium used consisted of 3 % corn starch, 0.2% yeast extract and 0.8% peptone (pH 7.0). When this strain was aerobically cultured in the medium on a rotary shaker, the highest inhibitory activity was obtained after four days. This inhibitor had inhibitory activities on various $\alpha$-amylases and glucoamylase, but not on $\beta$-amylase.

Improvement of Aspergillus niger 55, a Raw Corn Meal Saccharifying Enzyme Hyperproducer, through Mutation and Selective Screening Techniques (옥수수 生 전분 당화 효소 高 생산성 변이주 개발)

  • Mutation experiments were performed to select the mutant of Aspergillus niger 55, which had lost almost all the ability to produce transglucosidases but retained that of high productivity of raw meal saccharifying enzyme, by means of successive induction with N-methyl-N'-nitro-N-nitrosoguanidine(MNNG), ultraviolet(UV) light, and ${\gamma}$-rays. Also, we used the mutant enrichment techniques, such as liquid culture-filtration procedure and differential heat sensitivity of conidia, in order to increase the possibility of obtaining a mutant. The glucoamylase productivity of mutant PFST-38 was 11 times higher than that of the parent strain. The mutant PFST-38 was morphologically identical to the parent strain, except for the size of conidia, the tendency to form conidia and the lenght of conidiophore. Asp. niger mutant PFST-38 apeared to be useful for the submerged production of the raw corn meal saccharifying enzyme.

The Effect of Estragole Identified and Extracts from Agastache rugosa O. Kuntze on the Fungal Growth and Metabolism (진균류의 증식과 대사에 미치는 방아(Agastache rugosa)추출물과 Estragole의 효과)

  • The extracts from Agastache rugosa O. Kuntze, their chloroform and hexane fractions, and estragole identified from hexane fraction were tested to investigate the effects on the growth and metabolic activities of several true fungi. The fungi used were: Aspergillus oryzae KFCC 890, Aspergillus niger KCCM 11240, Saccharomyces cerevisiae IAM 4597, Saccharomyces ellipsoideus PNU 2215. The growth of S. Cerevisiae by treatment of water extract(1%), hexane fraction (0.05%), and estragole (0.05%) were inhibited 93%, 50%, and 33% respectively, and S. ellipsoideus was also inhibited markedly with delaying the alg phase maximum 12 hrs. The growth of A. oryzae was inhibited by treatment of extracts and fractions. The echanol production by S. cerevisiae was increased more than two times in the highest value around 42 hrs incubation by water extract, but chloroform fraction inhibited its production. The glucoamylase actibities by A. niger were strongly inhibited by hexane and chloroform fractions (0.05%). The invertase activity by S. cerevisiae using estragole (0.05%) reached to 57.5% of control group. S. cerevisiae treated with the estragole was damaged the cell wall and cell membrane, leaked the protoplasm, and observed broken pieces of cell.

A Study on Sugars in Korean Sweet Rice Drink "Sikhye"(II) -Enzymatic Analysis of Isomaltooligosaccharides and Rice Residue- (식혜의 이소말토올리고당에 관한 연구(II) -효소적 분석-)

  • Isomaltooligosaccharides in Sikhye were digested with enzyme (30unit/ml) of $\alpha$-amylase, $\alpha$-glucosidase and glucoamylase from Aspergillus awamori, sweet potato $\beta$-amylase and human salivary $\alpha$-amylase at 37$^{\circ}C$ for 1 hour, respectively. These amylases acted on these saccharides to give hydrolysis products with less than 20% of degree of hydrolysis, except the case of glucoamylase with 62% of high degree of hydrolysis. $\alpha$-Glucosidase plus human salivary $\alpha$-amylase hydrolyzed it to attain the hydrolysis value up to 25%, but further increment of hydrolysis was not observed. Rice residue in Sikhye has similar sugar composition and structure, judging from sugar analyses by the enzymatic hydrolysis. These results suggest that isomaltooligosaccharides and rice residue in Sikhye can be a growth factor for Bifidobacterium and dietary fiber which is useful for human health.

Preparation and Quality of Uncooked-Colored Wine Using Black Rice (흑미를 이용한 무증자 유색주의 제조와 품질)

  • In order to study the quality of uncooked-colored wine by using polished black rice(PBR) and glucoamylase, the mixture ration of PBR ws investigated. The growths of yeast and lactic acid bacteria, pH and alcohol concentration of the colored wine prepared by adding PBR in the range of 20 to 100% were higher than those of polished rice only, whereas the contents of residual sugars, total free amino acids and fusel oil of the colored wine were lower. The colored wine prepared by 20% PBR and that of over 80% PBR showed a light red color and a dark red, respectively. The optimal addition ratios of PBR evaluated by palatability of color and flavor, and sensory overall quality was 40 to 60%.

Quality Characteristics of wheat Nuruk and Optimum Condition of Liquid Starters for Aspergillus sp. (Aspergillus 속 곰팡이를 이용한 액체종국 제조 및 밀누룩의 품질특성)

  • This study focus was primarily the development of liquid starters for Aspergillus oryzae and Aspergillus niger prepared with wheat bran as a low cost culture medium. For the preparation of the liquid media wheat bran was added at rates of 0, 5, 10, 15 and 20% and the Aspergillus sp. strains were then inoculated to these prepared broths. The results indicated that the more that wheat bran was contained in the medium, the more mycelia was produced for A. oryzae and A. niger. The highest enzyme activities were obtained with a 10~15% adding rate of wheat bran for both strains. Changes in the enzyme activities of the liquid starters during various incubation times (0, 24, 48, 72 and 96 hrs), indicated that the highest enzyme activities were seen between 48 and 72 hrs of culture. In addition, a comparative study was carried out on the production of enzymes using wheat as a substrate in nuruk, with liquid starter made from fermented agents according to the same concentrations used with the wheat bran. The pH, acidity, amino acidity, reducing sugar content and enzyme activity (${\alpha}$-amylase, glucoamylase, acidic protease) of wheat nuruk made with liquid starter were compared with those of wheat nuruk made with solid starter. The results suggest that the liquid starter is superior in both cases.

Optimal Preparation of Saccharified Rice Solution for Bifidobacterium Fermentation (비피더스발효를 위한 쌀당화액 제조공정의 최적화)

  • This study was aimed at the development of the rice product by fermentation of saccharified rice solution with Bifidobacterium. To optimize the preparation of saccharified rice solution for Bifidobacterium fermentation, various pretreatment conditions were established. Grinding for 30seconds by an impact mill was more efficient than any other grinding schemes tested. The preheating before gelatinization showed a positive effect for efficient saccharification, and its optimal conditions were at $60^{\circ}C$ for 45 min. The optimum gelatinization conditions were at $100^{\circ}C$ for 40 min. The optimum levels of enzymes for saccharification of rice were 0.135 unit/g rice powder for ${\alpha}-amylase$ and 3.375 unit/g rice powder for glucoamylase, respectively. The physico-chemical properties of the fermented product by a fastidious Bifidobacterium showed a great potential for a functional rice product. However, an improvement on its flavor was required, which might be achieved by the addition of various fruits and vegetables.

Studies on Degrees of Kojic Mycelial Penetration and Some Characteristics of Kojic Enzymes in Moromi (청주국(淸酒麴)의 균사침투 상태(狀態)와 국효소(麴酵素)의 주류중(酒謬中) 동태(動態)에 관(關)한 연구(硏究))

  • In order to make the koji in which kojic mycelium penetrate deeply, some koji making conditions were investigated, thereafter kojies were maked on a large scale, and kojic enzymes in moromi were in vestigated. After 30% of brown rice was polished out, moisture, crude protein, and crude fat was decreased by 9%, 26% and 26% respectively, and starch value was increased by 9%. The optimum conditions for the koji in which kojic mycelium penetrate deeply were found as below. Koji making time was 40 hrs., moisture of ${\alpha}-rice$ was 40%, relative humidity during the first half of koji making time was 98%, and also the relative humidity during the second half of koji making thme was 80% and inoculum size was $1.0{\times}10^4$ spores/g ${\alpha}-rice$. 23-27% of ${\alpha}-amylase$ was inactivated and 35-70% of that was adhered during the moromi fermentation. 14% of glucoamylase was inactivated and 74-92% of that was adhered during the moromi fermentation. 13-14% of acid protease(pH 3.0) was inactivated and 70-73% of that was adhered during the moromi fermentation. Remarkable enzymatic differencies in moromi were not found between the kojies in which kojic mycelium penetrate deeply and not.

Effect of Nicotine on the Various Enzymes' Activity (효소활성에 미치는 니코틴의 영향)

  • Nicotine, the main alkaloid of tobacco, showed different effect according to the enzyme. Among investigated enzymes, protease was inactivated remarkably by nicotine and the mode of inhibition was examined. $\alpha$-amylase and $\beta$-amylase were not affected, and cellulase and glucoamylase were inactivated partially when the concentration of it was over 1.0% , but protease was inhibited powerfully by nicotine The inhibition of protease by nicotine was performed almost in the initial stage of reaction, and was not so much affected by temperature, and was reversible. The inhibition type of protease by nicotine appeared as a Mixed-type inhibition.

Restoration of Traditional Korean Nuruk and Analysis of the Brewing Characteristics

  • In this study, a total of 58 different kinds of nuruk (a traditional Korean fermentation starter) were prepared, including 46 kinds of restored nuruk from ancient documents. Each nuruk was evaluated by analysis of its saccharification power, and the enzyme activities of glucoamylase, ${\alpha}$-amylase, ${\beta}$-amylase, protease, and ${\beta}$-glucanase. The range of saccharification power (sp) of the restored nuruk ranged between 85 and 565 sp. The diastatic enzymes, ${\alpha}$-amylase, ${\beta}$-amylase, and glucoamylase, were significantly correlated to the saccharification power value; conversely, ${\beta}$-glucanase and protease did not have a correlative relationship with saccarification power. In addition, their brewing properties on chemical and organoleptic aspects of traditional alcoholic beverage production were compared. Each raw and supplementary material contained in nuruk showed its own unique characteristics on Korean alcoholic beverage brewing. For the first time, in this study, the traditional Korean nuruk types mentioned in ancient documents were restored using modernized production methods, and also characterized based on their brewing properties. Our results could be utilized as a basis for further study of traditional alcoholic beverages and their valuable microorganisms.
