Wnt/β-Catenin Signaling Pathway Is Necessary for the Specification but Not the Maintenance of the Mouse Retinal Pigment Epithelium

  • β-Catenin (Ctnnb1) has been shown to play critical roles in the development and maintenance of epithelial cells, including the retinal pigment epithelium (RPE). Ctnnb1 is not only a component of intercellular junctions in the epithelium, it also functions as a transcriptional regulator in the Wnt signaling pathway. To identify which of its functional modalities is critically involved in mouse RPE development and maintenance, we varied Ctnnb1 gene content and activity in mouse RPE lineage cells and tested their impacts on mouse eye development. We found that a Ctnnb1 double mutant (Ctnnb1dm), which exhibits impaired transcriptional activity, could not replace Ctnnb1 in the RPE, whereas Ctnnb1Y654E, which has reduced affinity for the junctions, could do so. Expression of the constitutively active Ctnnb1∆ex3 mutant also suppressed the development of RPE, instead facilitating a ciliary cell fate. However, the post-mitotic or mature RPE was insensitive to the loss, inactivation, or constitutive activation of Ctnnb1. Collectively, our results suggest that Ctnnb1 should be maintained within an optimal range to specify RPE through transcriptional regulation of Wnt target genes in the optic neuroepithelium.

  • The polymer blends of waste polyvinyl chloride (RPVC) and waste polyethylene(RPE) were prepared by melt mixing, and their morphology and tensile properties were evaluated after the copolymers having an ethylene group in backbone and ester group in side position were added as comptatibilizers. The blend compositions were varied as follows ; RPVC/RPE 85/15 wt%, where RPVC formed a continuous phase : 50/50, mid composition : 15/85, RPE a continuous phase. The blends revealed a very low compatibility between component polymers because they showed domain sizes greater than $10\mu\textrm{m}$ over all compositions, especially the worst compatibility around mid composition. The blends showed higher compatibility when ethylene vinylacetate copolymer(EVA) and ethylene ethylacrylate-graft-methyl methacrylate copolymers(EEA-MMA) were added.

  • Retinal pigment epithelium (RPE) plays an important role in maintaining the visual function and the degeneration of the RPE causes several retinal degeneration disease. In order to fabricate the suitable carrier for RPE transplantation, the hybrid poly(lactide-co-glycolide) (PLGA) film with hesperidin was prepared. Hesperidin has an anti-inflammatory and antioxidant characteristics. ARPE-19 was seeded on hesperidin/PLGA film and then, cell proliferation was determined by the MTT assay, and cell adhesion and cell morphology were confirmed by SEM. Also, RT-PCR was performed to confirm the expression of the specific genes, and AEC immunohistochemical staining was performed to determine the expression of RPE65. As a result, we confirmed that attachment, proliferation and phenotype maintenance of RPE cells were more excellent on hesperidin/PLGA film than PLGA film, thereby we were able to confirm the potential applications of hesperidin/PLGA film as tissue engineering carrier for regeneration of retina.

Exposure to Ozone and TVOCs during Shipyard Welding and the Adequacy of RPE (조선소 용접작업 중 오존 및 TVOCs의 노출평가와 사용 호흡보호구의 적합성)

  • Objectives: The purpose of this study is to assess the exposure to ozone and total volatile organic compounds(TVOCs) generated during welding work at a shipyard and recommend respiratory protective equipment(RPE) adequate against these hazards. Method: Ozone was collected for about 30 minutes at two-minutes intervals using a direct reading instrument, specifically an ozone analyzer(Serinus 10, Ecotech, Australia). TVOCs were collected for about 30 minutes at three-minute intervals using a portable GC (Alpha 115, Synspec BV, the Netherlands), and were determined simultaneously by area sampling at the welding plume closest to the welder's breathing zone. The total measurements were 162 for ozone($CO_2$ welding 47, TIG 60, stick 55), and 136 for TVOCs($CO_2$ 65, TIG 50, stick 21). Based on these measurements, a literature survey was conducted to assess the adequacy of RPE. Results: Relative to Korean OEL, measurements above STEL 0.2 ppm were 23.4% for $CO_2$, 63.3% for TIG and 14.5% for stick welding. There were significant differences(p=<0.0001) among welding types. Compared with ACGIH peak exposure of 0.4 ppm for ozone, which is not applied in Korea, $CO_2$ welding exceeded it by 10.6%, TIG by 40.0% and stick by 7.3%. Although it was not feasible to compare them directly since there are no Korean OEL, TVOCs had very high levels similar to the concentrations before moving into a new apartment and about 10-20 times the indoor air quality recommendations for some individual measurements. Conclusions: As ozone removal RPE has been recommended in welding environments for a long time(Lunau, 1967), this fact was demonstrated based on the results of the on-site work environment measurements(ozone and TVOCs). In conclusion, for all welding at a shipyard, gas/vapor and particulate combination RPE are recommended. If this is not possible, it should at least be present for TIG welding.

Evaluation of the effects of miniscrew incorporation in palatal expanders for young adults using finite element analysis

  • Objective: The aim of this study was to evaluate the stress distribution and displacement of various craniofacial structures after nonsurgical rapid palatal expansion (RPE) with conventional (C-RPE), bone-borne (B-RPE), and miniscrew-assisted (MARPE) expanders for young adults using three-dimensional finite element analysis (3D FEA). Methods: Conventional, bone-borne, and miniscrew-assisted palatal expanders were designed to simulate expansion in a 3D FE model created from a 20-year-old human dry skull. Stress distribution and the displacement pattern for each circumaxillary suture and anchor tooth were calculated. Results: The results showed that C-RPE induced the greatest stress along the frontal process of the maxilla and around the anchor teeth, followed by the suture area, whereas B-RPE generated the greatest stress around the miniscrew, although the area was limited within the suture. Compared with the other appliances, MARPE caused relatively even stress distribution, decreased the stress on the buccal plate of the anchor teeth, and reduced tipping of the anchor teeth. Conclusions: The findings of this study suggest that the incorporation of miniscrews in RPE devices may contribute to force delivery to the sutures and a decrease in excessive stress on the buccal plate. Thus, MARPE may serve as an effective modality for the nonsurgical treatment of transverse maxillary deficiency in young adults.

Comparison of characteristics during backward walking according to various stride frequencies in underwater and ground environments

  • Objective: The purpose of this study was to investigate the relationship between heart rate (HR), self-awareness of exercise intensity (rating of perceived exertion, RPE), and 5-meter walk test (5MWT) of persons affected by stroke during backward walking according to the preferred stride frequency (PSF), PSF+3 and PSF+6 conditions. Design: Cross-sectional study. Methods: A total of 11 persons with stroke (9 males, 2 females) participated voluntarily. All patients underwent backward walking under the PSF, PSF+3, and PSF+6 conditions in underwater and ground environments, and each condition was performed for 5 minutes. The HR, RPE, and walking speeds were measured during walking, and the measured values from underwater and ground environments were compared. Results: The HR and RPE in the ground environment were significantly increased (p<0.05), and although the 5MWT showed an increase in speed, it was not significant. The HR and RPE in the underwater environment were also significantly increased (p<0.05), however, although the 5MWT results was increased, it was not significant. The HR and RPE were significantly increased in the PSF+6 condition (p<0.05). Conclusions: The results of this study showed that backward gait training underwater can provide an appropriate exercise intensity for stroke survivors and suggests that exercises performed in an underwater environment is more effective compared to the ground environment.

Assessment of the pigeon (Columba livia) retina with spectral domain optical coherence tomography

  • Background: To assess the normal retina of the pigeon eye using spectral domain optical coherence tomography (SD-OCT) and establish a normative reference. Methods: Twelve eyes of six ophthalmologically normal pigeons (Columba livia) were included. SD-OCT images were taken with dilated pupils under sedation. Four meridians, including the fovea, optic disc, red field, and yellow field, were obtained in each eye. The layers, including full thickness (FT), ganglion cell complex (GCC), thickness from the retinal pigmented epithelium to the outer nuclear layer (RPE-ONL), and from the retinal pigmented epithelium to the inner nuclear layer (RPE-INL), were manually measured. Results: The average FT values were significantly different among the four meridians (p < 0.05), with the optic disc meridian being the thickest (294.0 ± 13.9 ㎛). The average GCC was thickest in the optic disc (105.3 ± 27.1 ㎛) and thinnest in the fovea meridian (42.8 ± 15.3 ㎛). The average RPE-INL of the fovea meridian (165.5 ± 18.3 ㎛) was significantly thicker than that of the other meridians (p < 0.05). The average RPE-ONL of the fovea, optic disc, yellow field, and red field were 91.2 ± 5.2 ㎛, 87.7 ± 5.3 ㎛, 87.6 ± 6.5 ㎛, and 91.4 ± 3.9 ㎛, respectively. RPE-INL and RPE-ONL thickness of the red field meridian did not change significantly with measurement location (p > 0.05). Conclusions: Measured data could be used as normative references for diagnosing pigeon retinopathies and further research on avian fundus structure.

  • In the discussion of digital mobile communication systems the speech coder based on the high quality low bit rate is an essential part of topics to overcome the limited availability of radio spectrum, which will enhance the communication services. In this paper we present the implementation and performance evaluation of 13kbps RPE LTP speech coder. An implementation of a real time full duplex coder with 75% of DSP loading rate using a single DSP chip has been shown, and also the fixed point simulations for H/W implementation has been performed. Finally, analysis result for relative bit importance of each transmitting parameter has been shown for channel coding.

  • The polymer blends of waste poly(vinyl chloride) (RPVC) and waste polyethylene (RPE) were prepared by melt mixing. Various ethylene-vinyl acetate copolymers (EVA) and ethylene-methacrylic acid Na salt copolymer (ionomer) were used as compatibilizer. Their morphologies and mechanical properties were evaluated as a funtion of mixing sequence and time. EVA with high vinyl acetate contents showed a rapid increment of tensile properties when small amount was added. Tensile properties of the blends were gradually increased with the addition of ionomer. Morphologies of RPVC/RPE blends were analyzed by scanning electron microscopy. FT-IR data showed that EVA was a good compatibilizer in RPVC/RPE blend compared to ionomer. Mechanical properties of the blends were highly enhanced when RPVC and compatibilizer were mixed and first RPE was added later.

Differential Roles of Tubby Family Proteins in Ciliary Formation and Trafficking

  • Cilia are highly specialized organelles that extend from the cell membrane and function as cellular signaling hubs. Thus, cilia formation and the trafficking of signaling molecules into cilia are essential cellular processes. TULP3 and Tubby (TUB) are members of the tubby-like protein (TULP) family that regulate the ciliary trafficking of G-protein coupled receptors, but the functions of the remaining TULPs (i.e., TULP1 and TULP2) remain unclear. Herein, we explore whether these four structurally similar TULPs share a molecular function in ciliary protein trafficking. We found that TULP3 and TUB, but not TULP1 or TULP2, can rescue the defective cilia formation observed in TULP3-knockout (KO) hTERT RPE-1 cells. TULP3 and TUB also fully rescue the defective ciliary localization of ARL13B, INPP5E, and GPR161 in TULP3 KO RPE-1 cells, while TULP1 and TULP2 only mediate partial rescues. Furthermore, loss of TULP3 results in abnormal IFT140 localization, which can be fully rescued by TUB and partially rescued by TULP1 and TULP2. TUB's capacity for binding IFT-A is essential for its role in cilia formation and ciliary protein trafficking in RPE-1 cells, whereas its capacity for PIP2 binding is required for proper cilia length and IFT140 localization. Finally, chimeric TULP1 containing the IFT-A binding domain of TULP3 fully rescues ciliary protein trafficking, but not cilia formation. Together, these two TULP domains play distinct roles in ciliary protein trafficking but are insufficient for cilia formation in RPE-1 cells. In addition, TULP1 and TULP2 play other unknown molecular roles that should be addressed in the future.
