Screening of Microorganisms with High Poly (butylene succinate-co-butylene adipate)-Degrading Activity (고활성 Poly(butylene succinate-co-butylene adipate) 분해균의 선발)

  • Microorganisms capable of degrading poly(butylene succinate-co-butylene adipate) (PBSA) were isolated from 40 soil samples such as landfill site soil, cultivating soil and activated sludge soil from 20 different sites in Korea by using the enrichment culture and the clear zone test at $37^{\circ}C$. Based on the 16S rDNA sequences, the isolated bacterium was identified to be Streptomyces sp. PBSA-1. Morphological and cultural characteristics were employed for the identification of the isolated fungi and they were proved to be Aspergillus fumigatus PBSA-2 and Aspergillus fumigatus PBSA-3. The PBSA degradation activity of the isolated microorganisms was enhanced through the serial acclimation in PBSA plate medium. The PBSA degrading microorganisms appeared to be highly active for the PBSA degradation in that 83% of PBSA was degraded by Streptomyces sp. PBSA-l, and 65% and 75% of PBSA was mineralized by A. fumigatus PBSA2 and A. fumigatus PBSA-3 respectively during 40 days of the modified Sturm test.

Evaluation of Corrosion Resistance and Weldability for the Butt Welding Zone of Hot Rolled Clad Steel Plates (열간압연 클래드강의 맞대기용접부 내식성 및 용접성 평가)

  • We have investigated the traits of clad metals in hot-rolled clad steel plates, including the sensitization and mechanical properties of STS 316 steel plate and carbon steel (A516), under various specific circumstances regarding post heat treatment, multilayered welds, and thick or repeated welds for repair. For evaluations, sectioned weldments and external surfaces were investigated to reveal the degree of sensitization by micro vickers hardness, tensile, and etching tests the results were compared with those of EPR tests. The clad steel plates were butt-welded using FCAW and SAW with the time of heat treatment as the variable, a that was conducted at $625^{\circ}C$, for 80, 160, 320, 640, and 1280 min. Then, the change in corrosion resistance was evaluated in these specimens. With carbon steel (A516), as the heat treatment time increased, the annealing effect caused the tensile strength to decrease. The micro-hardness gradually increased and decreased after 640 min. The elongation and contraction of the area also increased gradually. The oxalic acid etch test and EPR test on STS316 and the clad metal showed STEP structure and no sensitization. From the test results on multi-layered and repair welds, it could be concluded that there is no effect on the corrosion resistance of clad metals. The purpose of this study was to suggest some considerations for developing on-site techniques to evaluate the sensitization of stainless steels.

Antifungal Activities of Metarhizium anisopliae against Fusarium oxysporum, Botrytis cinerea, and Alternaria solani (Metarhizium anisopliae의 Fusarium oxysporum, Botrytis cinerea, Alternaria solani 등에 대한 항진균활성)

  • This study was conducted to find out antifungal activities of entomopathogenic fungus, Metarhizium anisopliae, against phytopathogenic fungi, Fusarium oxysporum, Botrytis cinerea and Alternaria solani. M. anisopliae was confirmed its antagonistic effect through mycelial inhibition zone of phytopathogenic fungi by culture filtrate of the antagonist. The filtrate (30%: v/v) inhibited the conidial germination of B. cinerea and F. oxysporum to 21.5% (control: 88.2%) and 53.0% (control: 78.6%), respectively and delayed the start of spore germination about 8hours. Microscopic observations proved that the addition of 10% culture filtrate of M. anisopliae restricted the growth of phytopathogenic fungus, F. oxysporum, to the formation of chlamydospore. From these results, we concluded that an addition effect of the filtrate from M. anisopliae on culturing F. oxysporum was fungistatic.

Development of a Hot Water Boiler System with a Rice Hull Furnace (왕겨 연소기(燃燒機)를 이용(利用)한 온수(溫水)보일러 시스템 개발(開發) (I) -실험적(實驗的) 연구(硏究)-)

  • This study was performed to develop a hot water boiler system with small scale automatic rice hull furnace for the multi-purpose use in the farm. For the experiment a prototype hot water boiler system with rice hull furnace was fabricated, which was equipped with automatic hull feeder, igniter and ash removal device. Optimum operational conditions of the prototype: system were analyzed. The results arc summarized as follows. 1. The temperature measured right above the burning surface should be higher than $500^{\circ}C$ combustion. 2. The top zone of the combustion chamber was the most suitable location of the thermocouple to pick up the control temperature for the automatic operation of the rice hull furnace. 3. The content of carbon monoxide in the flue gas was increased with the filling height of burning material but it was less than 0.3 percent in volume in this experiment. When the filling height was expressed as the ratio of rice hull feed rate to the volume of the combustion chamber above the burning surface, the optimum ratio was about $150kg/m^3-h$. 4. The combustion efficiency of the prototype was higher than 95 percent when the feed rate was 1.1 to 2.3 kg/h and moisture content of rice hull was 22.4 percent (w.b.) or less. 5. It was estimated that the optimum operational conditions of the system were 1.3 to 2.0 kg/h in feed rate, 70 to 100 percent in excess air and 500 to $510^{\circ}C$ in control temperature. 6. The efficiency of coil heal exchanger increased with a decrease in feed rate of rice hull. When the rice hull feed rates were 1.1, 1.7 and 2.3 kg/h, the efficiencies of coil heat exchanger were about 34, 30 and 25 percent and heat transfer rates were 5.7, 7.6 and 8.8 MJ/h, respectively. When the flat plate heat exchanger was used in addition to the coil heat exchanger, the efficiency of the heat exchanger system increased to 48 percent.

  • Bacillus licheniformis KMU-3 shown a strong antifungal activity was isolated from Swedish forest soils. B. licheniformis KMU-3 produced a maximum level of antifungal substance under incubation aerobically at $24^{\circ}C$ for 24 hours in LB broth containing $1.0\%$ sodium acetate, $1.0\%$ ammonium sulfate at 180 rpm and initial pH adjusted to 8.0. Chloroform extraction of culture broth was confirmed inhibitory zone by plate assay and Rf value 0.49 substance by thin layer chromatography (TLC) represented high antifungal activity against Rhizoctonia solani AG-1. This substance also exhibited against Rhizoctonia solani AG-4, Colletotrichum orbiculare, Colletotrichum gloeosporioides, Cladosporium cucumerinum, Fusarium oxysporum, and Fusarium graminearum.

  • In this study, an attempt for formulating a new and simplified rectangular finite element having only four corner nodal points is made to analyze the elastic-plastic large deformation behaviour up to the ultimate limit state of plates with initial imperfections. The present finite element contains the geometric nonlinearity caused by both in-plane and out-of-plane large deformation because for very thin plates the influence of the former may not be negligible. Treatment of expanded plastic zone in the plate thickness direction of the element is simplified based upon the concept of plastic node method so that the elastic-plastic stiffness matrix of the element is derived by the simple matrix operation without performing complicated numerical integration. Thus, a considerable saving of the computational efforts is expected. A computer program is also completed based on the present formulation and numerical calculation for some examples is performed so as to verify the accuracy and validity of the program.

  • In the course of search for multifunctional microbial inoculants, three Bacillus strains (BS11-1,BS11-2,BS11-3) with biological control and biofertilizing effects were selected. In this study, their ability for solubilization of insoluble phosphate, production of indole-3-acetic acid (IAA), siderophore, and hydrolytic enzymes, and antagonism against phytopathogenic fungi were estimated. All strains produced IAA and siderophore depending on culture time and produced a visible clear zone on agar plate containing 0.5% carboxylmethyl cellulose as a carbon source. Also, these strains exhibited antifungal activities against phytopathogenic fungi, Botrytis cinerea, Cylindrocarpon destructans, Fusarium oxysporum, Rhizoctonia solani, and Phytophthora capsici.

A Study on the Prediction of Thermally-Induced Residual Stress and Birefringence in Quenched Polystyrene Plate Including Free Volume Theory (자유 체적이론을 고려한 급냉 폴리스티렌판에 발생하는 잔류응력과 복굴절 형성에 관한 연구)

  • The residual stress and birefringence in injection-molded plastic parts can be divided into the flow-induced residual stress and birefringence produced in flowing stage, the thermally-induced residual stress and birefringence produced in cooling stage. However, the physics involved in the generation of the thermally-induced residual stress and birefringence still remains to be understood. Because polymer experiences viscoelastic history near the glass-transition temperature it is hard to model the entire process. Volume relaxation phenomenon was included to predict the final thermally-induced residual stress and birefringence in quenched plastic parts more accurately. The present study focused on comparing the predicted values far thermally-induced residual stress and birefringence with and without volume relaxation behavior (or free volume theory) under free and constrained quenching conditions. As a result, tile residual stress remained as a tensile stress at the center and as a compressible stress near the surface for the free quenching cases. In contract the residual stress remained as a compressible stress at the center and as a tensile stress near the surface fur the constrained quenching cases. The residual birefringence remained as minus values at the center and as plus values near the surface for the free quenching cases. Interestingly the residual birefringence showed minus values in entire zone for the constrained quenching cases. In the prediction of birefringence only the case including free volume theory showed the correct result for the distribution of birefringence in thickness direction.

  • Three-dimensional natural convective heat transfer in a vertical channel with a protruding single module was investigated experimentally. The particular interest was in the removal of thermal energy from the module by convective heat transfer. Hence radiative and conductive heat losses were estimated by using thermocouples and heat flux sensor respectively. The flow fields in the channel were visualized by means of a smoke-method. Also, local temperatures were measured by thermocouples inside the channel, along the vertical wall and module surface. It is found that convective heat transfer was promoted at the lower comer of the module and was decreased at the upper comer due to a recirculation zone. A general correlation of the critical channel ratios was found as a function of Rayleigh number. For the range of $8.28{\times}10^3<Ra^*_c<3.48{\times}10^6$, a useful correlation for the mean Nusselt number was proposed as a function of modified channel Rayleigh number.

Comparative assessment of antibacterial activity of different glass ionomer cements on cariogenic bacteria

  • Objectives: Glass ionomer cements (GICs), which are biocompatible and adhesive to the tooth surface, are widely used nowadays for tooth restoration. They inhibit the demineralization and promote the remineralization of the tooth structure adjacent to the restoration, as well as interfere with bacterial growth. Hence, the present study was conducted to assess and compare the antimicrobial activity of three commercially available GICs against two cariogenic bacteria. Materials and Methods: An agar plate diffusion test was used for evaluating the antimicrobial effect of three different GICs (Fuji IX, Ketac Molar, and d-tech) on Streptococcus mutans (S. mutans) and Lactobacillus acidophilus (L. acidophilus). Thirty plates were prepared and divided into two groups. The first group was inoculated with S. mutans, and the second group was inoculated with L. acidophilus. These plates were then incubated at $37^{\circ}C$ for 24 hours. Zones of bacterial growth inhibition that formed around each well were recorded in millimeters (mm). Results: The zones of inhibition for Fuji IX, Ketac Molar, and d-tech on S. mutans were found to be $10.84{\pm}0.22mm$, $10.23{\pm}0.15mm$, and $15.65{\pm}0.31mm$, respectively, whereas those for L. acidophilus were found to be $10.43{\pm}0.12mm$, $10.16{\pm}0.11mm$, and $15.57{\pm}0.13mm$, respectively. Conclusions: D-tech cement performed better in terms of the zone of bacterial inhibition against the two test bacteria, than the other two tested glass ionomers.
