Investigation of Packaging of Fresh Jujube (Zizyphus jujuba) on Quality during Storage (농가에서 활용가능한 생대추의 저장조건 연구)

  • The quality of fresh jujube (Zizyphus jujuba) fruit rapidly declines in the postharvest stage and thus the effective storage conditions for agricultural warehouses were investigated in this study. Fresh jujubes (~10 kg) were stored using five different storage methods: plastic box without further packaging as a control (C-PL), plastic box with hanji packaging (H-PL), plastic box with functional film packaging (F-PL), cardboard box with hanji packaging (H-CB), and cardboard box with functional film packaging (F-CB). The storage boxes were stored in a refrigerator ($0^{\circ}C$ and 80~90% RH), equipped with an ethylene gas scavenging device for 9 weeks and then the quality characteristics were analyzed. The control storage conditions of jujubes showed a substantial decline in quality with decreases in moisture content, hardness, and sugar content as well as increases in decay ratio and redness. Among the tested storage conditions, F-CB was most effective in terms of maintaining the quality of jujube fruit while maintaining firmness, weightlessness, ripening degree from redness and visible appearance. In conclusion, effective storage and packaging conditions during storage enabled maintenance of fruit quality of jujubes for 9 weeks.

Studies on the Changes of Chemical Components of Dried Jujube (Zizyphus jujuba MILLER) during Storage (대추 (Zizyphus jujuba MILLER)의 저장중(貯藏中) 화학성분변화(化學成分變化)에 관(關)한 연구(硏究))

  • In order to find out changes of chemical components related to browning of dried jujube, four varieties were subjected to the proximate analysis. Boeun, one of the major varieties in Korea, was sun-dried and stored for 12 months and analyzed periodically for one year. Browning, titrable acidity, pH, contents of ascorbic acid, organic acid, total amino acid, free amino acid, free sugar, hydroxymethylfurfural (HMF) and phenolic compounds were determined and compared with those of fresh jujube. The results obtained are summarized as follows; 1. In comparison with other similar fruits, jujube was high contents of ascorbic acid (62-79mg%) and carbohydrate (22-28%) excluding fiber. 2. Browning was increased in both steam-treated and nontreated plot together as storage period was prolonged. 3. Ascorbic acid content of fresh jujube was as high as 297.4mg% DB, but it was decreased to 20.2mg% DB, after 12 months storage. Therefore, loss of ascorbic acid was very great up to 93% of its original content. 4. Five kinds of organic acid, oxalic, succinic, fumaric, malic and citric acid were identified as major organic acids. It was interesting that only fumaric acid content was increased while others decreased during storage. 5. Seventeen kinds of amino acid were identified in the analysis of total amino acid content. Major amino acids were found to be proline, threonine, glutamic acid and lysine. During 12 months storage, 30% of original total amino acid was decreased and this was mostly accounted for free amino acids. 6. Threonine, proline, alanine and valine were identified as free amino acids which showed 85% decrease after 12 months storage. 7. Free sugars of jujube were composed of fructose, glucose and sucrose. They showed 24% decrease after 12 months storage, Especially sucrose was not detected after 4 months storage. 8. HMF of dried jujube was isolated and identified. Its content was increased up to great extent as storage period prolonged. 9. Caffeic, ferulic and P-coumaric acid were identified as jujube phenolic compounds which were significantly decreased during storage. In conclusion, non-enzymatic browning was thought to be more important than enzymatic reaction in the dried jujube. Amino-carbonyl reaction, ascorbic acid oxidation and reaction between phenolic compounds and sugar or nitrogen compounds were observed to be related to this browning.

Withes Broom of Jujube Tree, Zizyphus jujube Mill, Var. inermis Rehd.(Part.3) (대추나무 미친병에 관한 연구 3)

  • Over a period of 1962-64, a transmission-experiment of witchess' broom of jujube tree by stem-grafting was conducted. When stem-grafting of sound scions upon diseased roots or diseased scions upon sound roots were carried out, disease transmission of high rate was witnessed; 99% in the former and, in the latter, 62% of the stocks which saw union by callussing and had new shoots. Even when the diseased scions by stem-grafting or the diseased buds by budding upon sound stock died away, the transmission rate was 21% in stem-grafting and 14% in budding which seems to show that, when tissues of diseased plants and stocks are kept contacted over a certain period, the disease transmission occurs. And when the recovered scions taken from once diseased tree were grafted upon diseased roots, the transmission rate was 100 % and therefore it is presumed that the immunity could not be acquired even under the assumption of complete recovery from the disease. In stem-grafting of the diseased scions upon sound roots, 98% of the scions which were stored in the cellar, overwintered and grafted in spring was diseased, whereas the disease rate of the scions which were cut and grafted in spring was only 33%. It was particularly noteworthy that 90% of the scions in the former case and only 3% in the latter case were diseased as of June 18th approximately 2 months after the actual grafting and then the latter advanced to 33% with the passage of time. It appears that the pathogen in branches and shoots of the diseased trees standing outdoors become inactivated or diminished during winter. Through its symptom, pathological change in tissue, and easy transmission of the disease via stem-grafting, it seems certain that the pathogen of the witches broom disease in jujube tree is a virus.

Host range and Bionomics of the Rhombic Marked Leafhopper, Hishimonus sellatus Uhler(Homoptera: Cicadelliae) as a Vector of the Jujube Witches-Broom Mycoplasma (대추나무빗자루병 매개충 "마름무늬매미충(Hishimonus sellatus Uhler)"의 기주범위 및 생태에 관한 연구)

  • The study was to investigate the bionomics, host plants, and oviposition preference of Hishimonus sellatus Uhler. It has 5 generations under natural conditions and the peak of the 3rd generation was observed about mid and late August. Its average developmental durations were 80 days in spring, 69 days in summer, and 77.8 days in autumn. The lengths of each stage were 0.8mm in eggs, 0.9mm in 1st instar, 1.4mm in 2nd instar, 2.1mm in 3rd instar, 2.5mm in 4th instar, 3.2mm in 5th instar, 4.1mm in female, and 3.8mm in male. Hishimonus sellatus overwintered as egg in Morus alba, Humulus japonicus, and Zizyphus jujuba begining mid October, and attacked the shoot of M. alba and H. japonicus about mid and late May, migrated to the Zizyphus jujuba from late June to early July. Female oviposites 32~62 eggs into epidermis of shoot, vagina and vein during their life. The preferred host plants of H. sellatus were Humulus japonicus, Morus alba, Zizyphus jujuba, and ligustrum obtusifolium. Highly preferable oviposition site was H. japonicus, M. albal, Z. jujuba, and L. obtusifolium, etc. On audlt longevity, the host plants as H. japonicus, M. alba, and Z. jujuba were 43$\pm$2 days and A. brevipedenculata, C. mimosoides, L. obtusifolium, V. rosa, A. sinicus and, A. graveolens were more than 25 days, and other host plants were less than 20 days.

Fluorescence Microscopic Diagnosis of Mycoplasma Infections in Jujube, Mulberry and Periwinckle Plants (형광현미경적 기법에 의한 대추나무, 뽕나무 및 일일초의 마이코플라스마 감염진단)

  • Attempts were made to evaluate the efficacy of three fluorochromes, i.e., DAPI (4'-6-diamidino-2-phenylin-dole-2HCl), aniline blue and quinacrine(quinacrine mustard dihydrochloride) for the detection of mycoplasma infections in jujube (Zizyphus jujuba), mulberry (Mows alba) trees and periwinckle (Catharanthus roseus) plant by fluorescence microscopy. Stem sections from these plants infected with mycoplasma-like organisms (MLO) produced distinct fluorescence in the phloem when stained with DAPI, aniline blue or quinacrine, while fluorescence was absent in the healthy plants. The use of these fluorochromes provided simple and efficient techniques for the diagnosis of MLO infections. IOf the three fluorochromes tested, DAPI was found to be most efficient.

Changes in Phytoplasma Densities in Witches' Broom-Infected Jujube Trees over Seasons

  • The relative density of phytoplasmas in witches'broom (WB)-infected jujube trees was investigated using compatitive polymerase chain reaction (PCR). During dormant and defoliating seasons, the densities of phytoplasmas were about the same in roots and twigs. In early growing season, the density showed the highest rates in roots, then in twigs and in petioles. however, the density was highest in petioles and the lowest in roots during actively growing season. Throughout the year, root samples did not show any serious fluctuation compared with that of t2wigs and petioles. Density was lowest during actively growing season in root samples. In contrast, petiole sample densities varied to a great extent depending on the season, very high during actively growing season, but very low during the early growing season, In twig samples, the densities were very high and almost the same in both defoliating and dormant seasons. Among the parts of the trees, phytoplsma density was the most stable in root samples throughout the year. The highest densities of phytoplasmas were about the same in all tree parts. These results suggest that the phytoplasmas may overwinter not only in roots but also in twigs, and that multiplication rate of phytoplsma becomes very high right after the early growing season.

Physical Properties of Jujube(Zizyphus jujuba Miller) and Jujube Branches (대추 및 대추가지의 물리적 특성)

  • 본 연구는 대추 수확기개발을 위한 기초연구로써 대추의 열매, 잎, 잎줄기의 물리적 역학적 특성을 조사.분석하였으며, 그 결과를 요약하면 다음과 같다. .대추 열매의 평균 장경, 경, 부피, 무게, 탈과력은 각각 32.02 mm, 23.9 mm, 10.0$\times$$10^{-6}$m$^3$, 5.2~12.8g, 5.7 N으로 나타났다. .대추무게가 증가하면 탈과력은 증가하고, F/W는 감소하는 경향을 나타났다. .대추나무가지의 평균 탄성계수는 7.01$\times$$10^{8}$ N/m$^2$였고, 가지의 굵기가 증가하면서 감소하였다. .대추나무가지의 평균 비틀림 강성은 5.2$\times$$10^{7}$ N/m$^2$으로 나타났으며, 가지의 굵기가 증가하면서 음의 지수 함수적으로 감소하는 경향을 나타냈으나, 데이터가 일정한 범위에 산재해 있어 가지의 굵기에 따른 강성계수변화 특성은 일정한 경향으로는 나타나지 않았다. .대추 잎줄기의 평균 무게, 잎수, 길이가 각각 0.7g, 6.6개, 12.2 cm로 나타났다. .대추 잎의 평균 잎면적, 무게, 탈리력은 각각 13 $cm^2$, 0.2g, 4.4 N으로 나타났다. .종말속도는 대추 열매의 무게가 증가하면서 증가하고, 대추잎의 무게, 면적이 증가할수록 감소하는 경향을 나타냈으며, 잎줄기의 종말속도는 잎줄기 무게 혹은 잎줄기에 매달린 잎의 수와는 거의 영향을 받지 않는 것으로 나타났으며, 잎줄기의 길이가 증가할수록 약간씩 증가하는 경향을 나타냈다.

Anti-Influenza Activity of Betulinic Acid from Zizyphus jujuba on Influenza A/PR/8 Virus

  • Betulinic acid, a pentacyclic triterpene isolated from Jujube tree (Zizyphus jujuba Mill), has been known for a wide range of biological and medicinal properties such as antibacterial, antimalarial, anti-inflammatory, antihelmintic, antinociceptive, and anticancer activities. In the study, we investigated the antiviral activity on influenza A/PR/8 virus infected A549 human lung adenocarcinoma epithelial cell line and C57BL/6 mice. Betulinic acid showed the anti-influenza viral activity at a concentration of $50{\mu}M$ without a significant cytotoxicity in influenza A/PR/8 virus infected A549 cells. Also, betulinic acid significantly attenuated pulmonary pathology including increased necrosis, numbers of inflammatory cells and pulmonary edema induced by influenza A/PR/8 virus infection compared with vehicle- or oseltamivir-treated mice in vivo model. The down-regulation of IFN-${\gamma}$ level, which is critical for innate and adaptive immunity in viral infection, after treating of betulinic acid in mouse lung. Based on the obtained results, it is suggested that betulinic acid can be the potential therapeutic agent for virus infection via anti-inflammatory activity.

Blockade of vascular angiogenesis by Aspergillus usamii var. shirousamii-transformed Angelicae Gigantis Radix and Zizyphus jujuba

  • The matrix metalloproteinases (MMP) play an important role in tumor invasion, angiogenesis and inflammatory tissue destruction. Increased expression of MMP was observed in benign tissue hyperplasia and in atherosclerotic lesions. Invasive cancer cells utilize MMP to degrade the extracellular matrix and vascular basement membrane during metastasis, where MMP-2 has been implicated in the development and dissemination of malignancies. The present study attempted to examine the antiangiogenic activity of the medicinal herbs of Aspergillus usamii var. shirousamii-transformed Angelicae Gigantis Radix and Zizyphus jujube (tAgR and tZj) with respect to MMP-2 production and endothelial motility in phorbol 12-myristate 13-acetate (PMA)- or VEGF-exposed human umbilical vein endothelial cells (HUVEC). Nontoxic tAgR and tZj substantially suppressed PMA-induced MMP-2 secretion. In addition, $25{\mu}g/mL$ tAgR and tZj prevented vascular endothelial growth factor-stimulated endothelial cell transmigration and tube formation. The results reveal that tAgR and tZj dampened endothelial MMP-2 production leading to endothelial transmigration and tube formation. tAgR and tZj-mediated inhibition of endothelial MMP may boost a therapeutic efficacy during vascular angiogenesis.

Optimization of Acetic Acid Fermentation for Producing Vinegar from Extract of Jujube (Zizyphus jujuba Mill.) Fruits (대추 식초 음료 생산을 위한 대추 추출액 발효 조건 최적화)

  • The optimum conditions for producing vinegar from Jujube (ziziphus jujuba) juice using Acetobacter aceti were exploited by employing the response surface methodology (RSM). In addition to the initial concentration of ethanol, which is known to be a significant factor affecting acetic acid fermentation, the effects of initial concentration of Jujube juice, A. aceti concentration, pH, and temperature on acetic acid fermentation were also investigated. Out of these factors, the effects of the initial concentration of jujube juice and inoculation amount of A. aceti were determined to be negligible based on statistical analysis. By employing the face-centered experimental design in RSM, the optimum conditions for acetic acid fermentation were exploited for achieving maximum acidity and acetic acid production. The coefficients ($R^2$) of the derived equations from the response surface regression were 0.71 and 0.78 for acidity and acetic acid production, respectively. The maximum production of acetic acid was expected to be 52.76 mg/mL from 25% jujube extract at $21.75^{\circ}C$ with 7.69% alcohol content.
