Plant Regeneration from Hypocotyl Explants of Several Species of Lycopersicon (토마토속 식물의 배축절편 배양에 의한 식물체 재분화)

  • In an attempt to optimize the in vitro-regeneration conditions necessary for the genetic manipulation of tomato species, we examined several hybrid lines and wild species (peruvianum, pimpinellifolium, glandulosum) of Lycopersicon for. their, different regeneration ability. The basal medium used for callus growth and organogenesis was MSB (MS + B5) supplemented with three combinations of TDZ (Thidiazuron) 0.5mg/L+NAA 0.5mg/L, BA 2.0mg/L+NAA0.05 mg/L, and zeatin 3.0 mg/L + IAA 0.02 mg/L. In the genotype of Lycopenicon grandulosum, combination of TDZ and NAA was more effective in inducing shoot and root differentiation than those of BA and NAA or zeatin and IAA. When all genotypes tested were considered, however combination of zeatin and IAA was shown to be the best in shoot regeneration. Result indicate that callus and organogenesis of Lycopenicon species are dependent upon the hormone types and plant genotypes, but MSB medium with zeatin 3.0 mg/L + IAA 0.02 mg/L maybe appropriate for genotype-independent plant regeneration system of Lycopercicon species. We also tried TDZ as a cytokinin source in tomato tissue culture and found it highly significant in tomato regeneration system.

Effect of DIF and Cytokinins on In Vitro Growth of 'Campbell Early' Crapes (Vitis spp.) (주야 온도차[DIF]와 시토키닌이 '캠벨얼리' 포도의 기내생장에 미치는 영향)

  • Effects of difference between day and night temperatures (DIF) and cytokinins on in vitro growth of 'Campbell Early' grape plantlets were investigated, Shoot growth was suppressed by -7DIF (18/$25^{\circ}C$). Effects of BA and TDZ were not affected by DIF. 0DIF (25/$25^{\circ}C$) enhanced the growth with increasing cytokinin level. Kinetin at 10 $\mu$M was the most effective for shoot growth with 87 mm among cytokinins used except for control. Zeatin and kinetin each at 5 $\mu$M significantly increased shoot growth to respective 162 and 110 mm. In 0DIF, two-fold increase of 162 mm was obtained by 5 $\mu$M zeatin. In control, no further shoot proliferation was observed regardless of DIF. In 0 and +7DIF (25/18$^{\circ}C$), 4 shoots were observed with 10 $\mu$M BA. Zeatin, 2iP and kinetin resulted in poor shoot proliferation while BA at 10 $\mu$M resulted in profuse branching in all DIFs.

  • Effect of cytokinins (BA, TDZ, zeatin, 2iP, and kinetin) applied either singly or in combination on in vitro growth of two grape cultivars ('Cabernet Sauvignon' and 'Campbell Early') was investigated as a serial work for mass production of grapevine nursery stocks. In single treatment, shoot growth of two cultivars was most favorable in control. Shoot proliferation was satisfactory with 10 $\mu$M BA regardless of cultivars and cytokinin combinations, followed by TDZ. Other treatments resulted in very poor or no branching. Total explants ready for subculture produced by 10 $\mu$M BA outnumbered those by other treatments. TDZ was also effective. TDZ significantly increased the fresh weight and callus formation while shoot growth was unsatisfactory. Shoot growth response of two cultivars in combined treatments was also most favorable in control as was in single treatments. When TDZ was combined with zeatin, 2iP, and kinetin which failed to induce branching, proliferous branching was induced though the shoot number was behind that of single treatments of BA and TDZ. TDZ was very effective for total number of explants and fresh weight, showing 10-fold increase.

Influence of Plant Growth Regulators on Shoot Multiplication of Thymus quinquecostatus Celak. (백리향 (Thymus quinquecostatus Celak.)의 기내증식에 미치는 식물생장조절물질의 영향)

  • In vitro culture system was established to induce multiple shoots of Thymus quinquecostatus Celak. by investigating the effects of cytokinins. Stem explants were cultures on MS medium supplemented with either five drfferent plant growth regulators or their combinations under light or dark condition. The most effective cytokininsource was the combination of BA 1.0mg/L and TDZ 0.1mg/L for producing shoots (6.05$\pm$1.51), zeatin 2.0mg/L and TEZ 0.1mg/L for elongating shoots (3.27$\pm$0.66cm) under the light condition. In addition the most effective cytokinin was 2-ip 2.0mg/L for producing shoots(5.20$\pm$1.81), zeatin 2.0mg/L for elongating shoots(5.64$\pm$1.24cm)under the dark condition. Overall, the average percent for in vitro shooting was greater than 89.58%.

Somatic Embryogenesis and Adventitious Bud Formation from Zygotic Embryo of Boxthone (Lycium chinense Mill.) (구기자나무(Lycium chinense Mill.) 접합자 배로부터 체세포배 및 부정아 발생)

  • Somatic embryos or adventitious buds were formed from the segments of zygotic embryo of Lycium chinense Mill. on Murashige and Skoog (MS) medium supplemented with auxins (2,4-D, NAA, IAA) and / or cytokinins (zeatin, kinetin, BAP). Embryogenic callus formed on MS medium containing 0.5 mg/L 2,4-D and then differentiated into somatic embryos without transfer to hormone free medium. On the other hand, adventitious buds were formed on medium with 0.01 mg/L 2,4-D and 0.5 mg/L zeatin. Among various parts of zygotic embryos, the morphogenic potential was higher in the cotyledonary region, and the most organogenic potential was found in cotyledon followed by radicle, hypocotyl, and whole embryo. Histologically, bipolar structure of the heart-shaped embryos were confirmed and in adventitious buds only shoot apical meristems were found to exist.

  • To elucidate the relationship between in vitro flowering and the structure of cytokinins in ginseng (Panax ginseng C.A. Meyer), zygotic embryos, seedlings, and cotyledonary nodes were cultured on MS medium supplemented with 5 $\mu$M of various cytokinins (BA, kinetin, 2-iP, and zeatin) with or without GA$_3$ (5 $\mu$M). The frequency of in vitro flowering was the highest when explants were cultured on the medium containing BA regardless of the kinds of explants, followed by kinetin, 2-iP, and zeatin. Flowering frequency of cotyledonary node explants was significantly increased by the combined treatment of cytokinin and GA$_3$. Flowering frequency was highly correlated with the logP of cytokinins, indicating that the lipophilicity of each cytokinin may involved in the in vitro flowering of ginseng.

Efficient Regeneration of Plants Independent of Exogeneous Growth Regulators Using Tissues in Pepper (Capsicum annuum L.) (고추의 조직을 이용한 생장조절제 처리가 식물체 재분화 효율에 미치는 영향)

  • The study was carried out to develop efficiency of transformantion in pepper plants. We used cotyledons of two native varieties, 'Subicho' and 'Kumtap' to establish some conditions of plant regeneration. Differentiation rate of shoot was higher in 2 to 4 mg/l zeatin and 0.05 mg/l NAA than in other treatments. Meanwhile, differential rate of roots was the highest in 0.5 mg/l NAA.

  • Cytokinins are essential plant hormones that play crucial roles in various aspects of plant growth and development. To better understand the molecular mechanisms of cytokinin action, we identified cytokinin related proteins by a proteomic approach. Proteins extracted from control and trans-zeatin treated Arabidopsis seedlings were separated and analyzed by two dimensional gel analysis. Differentially expressed protein spots were identified with peptide mass fingerprinting based on matrix assisted laser desorption/ionization time-of-flight (MALDI-TOF) mass spectrometry and database searching, We obtained ten up-regulated and one down-regulated proteins upon t-zeatin treatment. The expression of the following proteins was induced; pollen allergen like protein, L-ascorbate peroxidase, tetrapyrrole methylase family protein, SGT1 protein homolog, disease resistance related protein, maternal embryogenesis control protein, paxneb related protein, gluthathione S-transferase and IAA amino acid hydrolase homolog.

Contents of Mineral Elements and Cytokinins in Xylem Sap of Two Oriental Melon Cultivars Affected by Rootstocks (참외품종과 대목종류에 따른 목부액 내의 무기성분 및 시토키닌 함량)

  • Contents of mineral element and cytokinin in the xylem sap of 'Keumdongee' and 'Tongilhwang' oriental melons were compared with those in oriental melons grafted onto 8 rootstocks. The effect of grafting on the fruit quality of oriental melon was also investigated. Flesh firmness varied with rootstocks. Soluble solids contents in the placenta tissue of grafted 'Tongilhwang' were higher than that in the 'Keumdongee'. Electric conductivity of the xylem sap in own-rooted plants was higher in 'Keumdongee' than in 'Tongilhwang', but it increased in 'Tongilhwang' once they were grafted. The sap volume per plant was greater in 'Keumdongee' than in 'Tongilhwang'. The mineral concentrations varied considerably depending on the rootstock used. Xylem sap of grafted oriental melons contained a higher amount of mineral ions, especially $NO_3{^-}$ and $PO_4{^-}$, than did the sap in own-rooted plants. The increase in the mineral levels in sap due to grafting was most apparent in 'Tongilhwang'. Xylem sap from both 'Keumdongee' and 'Tongilhwang' contained trans-zeatin (t-Z), trans-zeatin riboside (t-ZR), and dihydrozeatin riboside (DHZR). Small amounts of isopentenyl adenine (IPA) and isopentenyl adenine riboside (IPAR) were also detected. Trans-zeatin riboside was the most abundant, followed by t-Z. Cytokinin concentration in 'Keumdongee' was not significantly influenced by rootstock type used, although the highest concentration of cytokinins in 'Keumdongee' was obtained with 'Chamtozwa' rootstock. However, the cytokinin concentration in 'Tongilhwang' increased with grafting irrespective of rootstock type used.

Effects of Growth Regulators and Explants on Direct Somatic Embryogenesis in Liquid Culture of Rehmannia glutinosa (지황의 액체배양에서 식물생장조정제와 치상 조직이 직접 체세포배 형성에 미치는 영향)

  • The effects of plant regulators on direct somatic embryogenesis in liquid culture of Rehmannia glutinosa were investigated and the proper explant for direct somatic embryo formation was studied. Direct somatic embryos were induced from leaf segments culture in the MS liquid medium containing 0.5 mg/l of both IAA and NAA, while IBA of 1.0 mg/l was required for the same effect. Many somatic embryos were directly formed at the concentration of 2.0 mg/l cytokinin such as BA, kinetin and zeatin, but somatic embryogenesis was relatively poor at above or below this level. Relatively more somatic embryos were induced in the combination of 1.0mg/l IAA and 2.0mg/l zeatin. Formation of somatic embryos begun after 6 weeks on stem segments, while 7 weeks both on petiole and leaf. However, overall production of somatic embryos after 8 weeks was higher in leaf segment than that of stem segment.
