Assessment of Attraction Efficiency of By-pass Fishway at Dalseong Weir According to Operating Attraction Waterway (유인수로의 운영에 따른 달성보 인공하도식 어도의 유인효율 평가)

  • In this study, the attraction efficiency of the by-pass fishway installed at Dalseong Weir in Nakdong River was assessed according to operation of the attraction waterway by using River2D which is a two-dimensional physical habitat simulation model. The model was calibrated and validated through the measured water elevation. The attraction efficiency of the fishway was evaluated at the low flow condition because the target fish, Zacco platypus (Z. platypus), have moved frequently up and downstream at the spawning season from April to June. From simulation results, it can be deduced that the attraction efficiency at situation of open attraction waterway is superior to that of close attraction waterway. Also it is shown that velocity field at inner region of the fishway is suitable for migration of the fish.

  • To improve sensitivity of biosensor as yeast two-hybrid detection system for estrogenic activity of suspected chemicals, we tested effects of several combinations of the bait and fish components in the two-hybrid system on Saccharomyces cerevisiae inducted a chromosome-integrated lacZ reporter gene that was under the control of CYC1 promoter and the upstream Gal4p-binding element $UAS_{GAL}$. The bait components that were fused with the Gal4p DNA binding domain are full-length human estrogen receptor ${\alpha}$ and its ligand-binding domain. The fish components that were fused with the Gal4p transcriptional activation domain were nuclear receptor-binding domains of co-activators SRC1 and TIF2. We found that the combination of the full-length human estrogen receptor ${\alpha}$ with the nuclear receptor-binding domain of co-activator SRC1 was most effective for the estrogen-dependent induction of reporter activity among the two-hybrid systems so far reported. The relative strength of transcriptional activation by representative natural and xenobiotic chemicals was well correlated with their estrogenic potency that had been reported with other assay systems.

  • Fluorescence in situ hybridization (FISH) techniques allow the enumeration of chromosome abnormalities and from a great potential for many clinical applications. In order to produce quantitative and reproducible results, expensive tools such as a cooled CCD camera and a computer software are required. We have developed a Chromosome Image Processing System (Chips) using FISH that allows the detection and mapping of the genetic aberrations. The aim of our study, therefore, is to evaluate the capabilities of our original system using a black-and-white video camera. As a model system, three repetitive DNA probes (D18Z1, DXZ1, and DYZ3) were hybridized to variety different clinical samples such as human metaphase spreads and interphase nuclei obtained from uncultured peripheral blood lymphocytes, uncultured amniocytes, and germ cells. The visualization of the FISH signals was performed using our system for image acquisition and pseudocoloring. FISH images were obtained by combining images from each of probes and DAPI counterstain captured separately. Using our original system, the aberrations of single or multiple chromosomes in a single hybridization experiment using chromosomes and interphase nuclei from a variety of cell types, including lymphocytes, amniocytes, sperm, and biopsied blastomeres, were enabled to evaluate. There were no differences in the image quality in accordance with FISH method, fluorochrome types, or different clinical samples. Always bright signals were detected using our system. Our system also yielded constant results. Our Chips would permit a level of performance of FISH analysis on metaphase chromosomes and interphase nuclei with unparalleled capabilities. Thus, it would be useful for clinical purposes.

Characteristics of Fish Community in the Seomjin River and Brackish Area (기수역이 존재하는 섬진강의 어류군집 특성)

  • In this study, to identify ichthyofauna of the Seomjin River, which has no barrage at estuary, and to estimate effect of estuary barrage, fish sampling was conducted at 16 study sites in mainstream of the Seomjin River from 2010 to 2012. A total of 54 species classified into 17 families were collected. Cyprinidae was the dominant family and Zacco platypus (relative abundance, RA: 47.8%) was identified as the dominant species. In total, 17 Korean endemic species (31.5%) were collected, and it is higher than the average endemic rate of Korean peninsula (22.5~25.9%). Although the relative abundance of exotic species (1.1%) was low, it is higher than the past studies (2002: 0.0%, 2009: 0.4%). Dominance index of freshwater region (St.4~St.16) was relatively higher than estuary (St.1~St.3), because Z. platypus showed high relative abundance at freshwater region. But diversity and evenness index showed contradictory results, as estuary was higher than freshwater region. The ratios of sea fish, peripheral freshwater fish and economic fish, and diversity index are higher than the Nakdong River, the Yeongsan River and the Geum River. In conclusion, to conserve the value of the Seomjin River ichthyofauna, continuous monitoring is necessary.

Fish Fauna of the Main Channel in the Nakdong River (낙동강 본류의 어류상)

  • Longitudinal distribution of the fish fauna of the 20 main channel and tributary sites in the Nakdong River was investigated from July 1999 to December 2000. A total of 4,791 individuals of fisHes were collected from three to seven sampling programs of all study sites and 48 species of 17 families were collected. Dominant family was Cyprinidae (25 species, relative abundance (RA): 91.8%). Zacco platypus (RA: 18.0%) was found to be the most abundant inhabitant. Subdominant species were Opsariichthys bidens (17,1%), Squalidus chankaensis tsuchigae (13.2%), Hemibarbus labeo (9.9%), and Z. temmincki (8.2%). Among the total species, 12 species were identified as endemic species (4 families, 24.5% of 1,172 individuals). Four exotic fishes were collected (Carassius cuvieri, 25 indv.; Micropterus salmoides, 214 indv.; Lipomis macrochirus, 14 indv.; Oreochromis niloticus, 6 indv.). M. salmoides was distributed in mid to lower part of the Nakdong River. As the result of cluster analysis, sampling sites were divided into three groups (upper, middle and lowerpart). Site below Gumi (St. 10) were considered as the lower part of the Nakdong River. Combined with previous surveys, about 80 species are present in the main channel and tributaries of the Nakdong River. Since the middle of 1990s, population density of the M. salmoides rapidly increased in the stagnant lower part of the river(RA 8.8% in lower 5 sites). The feeding strategy of M. salmoides was "generalized" on Daphnia sp. (<100 mm), and "specialization" about Cladocera (<150 mm) and Insecta(<200 mm).

Comparison on Ecological Index Characteristics between Zacco platypus and Zacco koreanus by Stream order in Korea (하천차수에 따른 피라미와 참갈겨니 개체군의 생태지표 특성 비교)

  • We collected fishes at the 12,873 sites in stream order (1~7) from 2008 to 2016. In the results, two populations(Zacco platypus and Zacco koreanus) represented distributional differences in the stream order gradient, and correlation analysis showed that the two populations had a potential competitive relationship. The ecological characteristics of the fish except for the Z. platypus and the Z. koreanus, according to the gradient of stream order showed a pronounced gradient in the species such as intermediate species, sensitive species, carnivores, herbivores and omnivores. The two populations showed a high correlation between intermediate species, sensitive species of the tolerance guild and omnivores, insectivores of the trophic guild. Fish assessment index (FAI) was negative relation with stream order. According to the stream order gradient, the two populations showed different correlations with FAI.

Length-weight Relation and von Bertalanffy's Growth Model of Zacco koreanus Population Distributed in the Tributaries of the Nakdong River (낙동강 지류에 분포하는 참갈겨니 개체군의 전장과 체중 관계 및 von Bertalanffy의 성장모델)

  • Populations of Zacco koreanus, distributed in four different tributaries of mid-upper reach Nakdong River were investigated to analyze a length-weight relation and von Bertalanffy's growth model. Fish sampling was conducted by common method (cast net and kick net) during March to October 2005. Fishes caught in the field were identified immediately, and then individuals of Zacco koreanus were preserved in 10% formalin to further measure their total length and weight in the laboratory. As the results of the equation based on length-weight relation, values of parameter b on the population of all tributaries were greater than 3.0 and the value on Bohyeon Stream was the maximum (3.26), indicating that the fish in the stream became more rotund as the length increases. In the mean time, we examined Brody growth constant (k) induced by the von Bertalanffy's growth model, and we found more steady state population in Wi (-0.18) and Byeongbo (-0.21) Streams than in fan (-0.38) and Bohyeon (-0.37) Streams. The findings would be used to assess local water environment on tributaries of the Nakdong River with understanding of ecological characteristics on the population of Zacco koreanus, as well as provide us fundamental information on domestic study of fish ecology.

Use of Transgenic and Mutant Animal Models in the Study of Heterocyclic Amine-induced Mutagenesis and Carcinogenesis

  • Heterocyclic amines (HCAs) are potent mutagens generated during the cooking of meat and fish, and several of these compounds produce tumors in conventional experimental animals. During the past 5 years or so, HCAs have been tested in a number of novel in vivo murine models, including the following: lacZ, lacI, cII, c-myc/lacZ, rpsL, and $gpt{\Delta}$ transgenics, $XPA^{-/-}$, $XPC^{-/-}$, $Msh2^{+/-}$, $Msh2^{-/-}$ and $p53^{+/-}$ knock-outs, Apc mutant mice ($Apc^{{\Delta}716}$, $Apc^{1638N}$, $Apc^{min}$), and $A33^{{\Delta}N{\beta}-cat}$ knock-in mice. Several of these models have provided insights into the mutation spectra induced in vivo by HCAs in target and non-target organs for tumorigenesis, as well as demonstrating enhanced susceptibility to HCA-induced tumors and preneoplastic lesions. This review describes several of the more recent reports in which novel animal models were used to examine HCA-induced mutagenesis and carcinogenesis in vivo, including a number of studies which assessed the inhibitory activities of chemopreventive agents such as 1,2-dithiole-3-thione, conjugated linoleic acids, tea, curcumin, chlorophyllin-chitosan, and sulindac.

  • Volatile compounds in salt fermented anchovy on the market were analyzed by vacuum simulta neous distillation solvent extraction/gas chromatography/mass spectrometry/olfactometry(V SDE/ GC/MS/O) and aroma extract dilution anlaysis(AEDA). Predominant odorants(Log3FD$\geq$8) in sample were ethyl methylbutanoate(candy like/sweet) and 2 ethyl 3,5 dimethylpyrazine(nutty/baked potato like). Besides these compounds, 6 odorants such as ethyl 3 methylbutanoate(sweet/floral/ candy like), 3 methylbutanal(dark chocolate like), (Z) 4 heptenal(rancid/fish like), (methylthio) propanal(soy sauce /baked potato like), (E,Z) 2,6 nonadienal(melon /cucumber like) and (E,E) 2,4 decadienal(fatty/cooked soybean like) were potent in odor value of salt fermented anchovy. Seven amino acids having high taste value in sample were glutamic acid, aspartic acid(sour and umami taste), lysine, alanine(sweet), histidine, valine, and methionine(bitter).

Ecological diagnosis of the Gongjicheon water system using length-weight relationship and condition factor(K) of population of the Zacco platypus (피라미 개체군의 length-weight relationship 및 condition factor(K)를 이용한 공지천수계의 생태적 진단)

  • The purpose of this study is to investigate the dynamics of the Zacco platypus population among streams in the Gongjicheon water system, Korea from December 2010 to October 2011. In this study, fish fauna was collected 27 Species belonged to 9 Families. The legal protection species, such as a natural monument and endangered species was not collected. Korean endemic species was collected 6 species including to Z. koreanus. The ratio of Korean endemic species was 22.6%, and it was lower than 51.3% that average of Korean endemic species in the Hangang water system. About these status, we considered that Gongjicheon water system had lost of the unique characteristics of the Hangang water system by the anthropogenic disturbances. The result of analysis to length-weight relationship and condition factor(K) of Z. platypus population in the Gongjicheon water system, the regression coefficient value(b) to indication of growth degree of the population was 3.04 and the slope of the condition factor(K) to indication of corpulency had positive value, respectively. These results seems to be that the Z. platypus population is maintained to a little unstably. In addition, the values of the degree of growth and corpulence were very low than the other natural streams in the Hangang water system, so it considered that the many artificially stress factors are occurred in this study area. These results are similar to Fish Assessment Index(FAI) in the assessment for health of aquatic ecosystem. Therefore, we are considered to require precise investigation and sustained monitoring for the restorations of stream ecosystem in the Gongjicheon water system.
