Z-DNA-Containing Long Terminal Repeats of Human Endogenous Retrovirus Families Provide Alternative Promoters for Human Functional Genes

  • Transposable elements (TEs) account for approximately 45% of the human genome. TEs have proliferated randomly and integrated into functional genes during hominoid radiation. They appear as right-handed B-DNA double helices and slightly elongated left-handed Z-DNAs. Human endogenous retrovirus (HERV) families are widely distributed in human chromosomes at a ratio of 8%. They contain a 5'-long terminal repeat (LTR)-gag-pol-env-3'-LTR structure. LTRs contain the U3 enhancer and promoter region, transcribed R region, and U5 region. LTRs can influence host gene expression by acting as regulatory elements. In this review, we describe the alternative promoters derived from LTR elements that overlap Z-DNA by comparing Z-hunt and DeepZ data for human functional genes. We also present evidence showing the regulatory activity of LTR elements containing Z-DNA in GSDML. Taken together, the regulatory activity of LTR elements with Z-DNA allows us to understand gene function in relation to various human diseases.

Determination Grid Cell for Estimation of Radar Relationship (레이더 관계식 산정을 위한 격자망 크기 결정)

  • The purpose of the study is to determine grid cell of radar relationship, and to promote the radar data on hydrology field. The study region is the Chungjudam basin with a drainage area of $6,648km^{2}$ located within the 260 km circle under the DWSR-88C C-band umbrella(Kwanak Mt Radar). Grid cell was produced to extract radar reflectivity and rainfall data of the same time and point using Arc-view software package. The grid cell size is to estimate mean correlation coefficient for $1km{\times}1km,\;2km{\times}2km,\;3km{\times}3km$ grid. The result of mean correlation coefficient showed good result(0.57) for the $1km{\times}1km$ grid cell. The 32 rainfall stations Z-R relationship was estimated in Chungjudam basin.

  • Galaxy cluster is the most important laboratoriy to study the effect of environment on galaxies, one of key questions in astronomy. In the local universe, it is well known that red, passive galaxies are concentrated in the cluster core. However, it is still controversial whether the star formation-density relation at the low redshift is retained in the distant universe. Many surveys have tried to find galaxy clusters at various epochs. However the optical dataset has limitations in finding galaxy clusters at z > 1, since the bulk of stellar emission of z > 1 galaxies is redshifted into the near-IR regime. We used the multi-wavelength data from the UKIDSS DXS (J and K bands), the SWIRE (4 IRAC bands), and the PAN-STARRS (g, r, i, z, y bands) and IMS (J band; Im et al. 2015, in preparation) in the European Large Area ISO Survey North1 (ELAIS-N1) field to search for high redshift galaxy clusters and study the properties of member galaxies. Using the multi-wavelength data, we investigated overdensities of galaxies at 0.2 < z < 1.6 based on the photometric redshift information. We found several superclusters where cluster candidates are concentrated within scales of few tens of Mpc at z ~ 0.9. Interestingly, some of the supercluster candidates consist of galaxy clusters which are dominated by blue galaxies. We will present high redshift galaxy cluster and supercluster candidates in ELAIS-N1 field and galaxy properties in different environments including dense clusters and fields.

A Multi-sensor basedVery Short-term Rainfall Forecasting using Radar and Satellite Data - A Case Study of the Busan and Gyeongnam Extreme Rainfall in August, 2014- (레이더-위성자료 이용 다중센서 기반 초단기 강우예측 - 2014년 8월 부산·경남 폭우사례를 중심으로 -)

  • In this study, we developed a multi-sensor blending short-term rainfall forecasting technique using radar and satellite data during extreme rainfall occurrences in Busan and Gyeongnam region in August 2014. The Tropical Z-R relationship ($Z=32R^{1.65}$) has applied as a optimal radar Z-R relation, which is confirmed that the accuracy is improved during 20mm/h heavy rainfall. In addition, the multi-sensor blending technique has applied using radar and COMS (Communication, Ocean and Meteorological Satellite) data for quantitative precipitation estimation. The very-short-term rainfall forecasting performance was improved in 60 mm/h or more of the strong heavy rainfall events by multi-sensor blending. AWS (Automatic Weather System) and MAPLE data were used for verification of rainfall prediction accuracy. The results have ensured about 50% or more in accuracy of heavy rainfall prediction for 1-hour before rainfall prediction, which are correlations of 10-minute lead time have 0.80 to 0.53, and root mean square errors have 3.99 mm/h to 6.43 mm/h. Through this study, utilizing of multi-sensor blending techniques using radar and satellite data are possible to provide that would be more reliable very-short-term rainfall forecasting data. Further we need ongoing case studies and prediction and estimation of quantitative precipitation by multi-sensor blending is required as well as improving the satellite rainfall estimation algorithm.

Spatio-temporal Fluctuations with Influences of Inflowing Tributary Streams on Water Quality in Daecheong Reservoir (대청호의 시공간적 수질 변화 특성 및 호수내 유입지천의 영향)

  • The objectives of this study were to analyze the longitudinal gradient and temporal variations of water quality in Daecheong Reservoir in relation to the major inflowing streams from the watershed, during 2001~2010. For the study, we selected 7 main-stream sites of the reservoir along the main axis of the reservoir, from the headwater to the dam and 8 tributary streams. In-reservoir nutrients of TN and TP showed longitudinal declines from the headwater to the dam, which results in a distinct zonation of the riverine ($R_z$, M1~M3), transition ($T_z$, M4~M6), and lacustrine zone ($L_z$, M7) in water quality, as shown in other foreign reservoirs. Chlorophyll-a (CHL) and BOD as an indicator of organic matter, were maximum in the $T_z$. Concentration of total phosphorus (TP) was the highest (8.52 $mg\;L^{-1}$) on March in the $R_z$, and was the highest (165 ${\mu}g\;L^{-1}$) in the $L_z$ on July. Values of TN was the maximum (377 ${\mu}g\;L^{-1}$) on August in the $R_z$, and was the highest (3.76 $mg\;L^{-1}$) in the $L_z$ on August. Ionic dilution was evident during September~October, after the monsoon rain. The mean ratios of TN : TP, as an indicator of limiting factor, were 88, which indicates that nitrogen is a surplus for phytoplankton growth in this system. Nutrient analysis of inflowing streams showed that major nutrient sources were headwater streams of T1~T2 and Ockcheon-Stream of T5, and the most influential inflowing stream to the reservoir was T5, which is located in the mid-reservoir, and is directly influenced by the waste-water treatment plants. The key parameters, influenced by the monsoon rain, were TP and suspended solids (SS). Empirical models of trophic variables indicated that variations of CHL in the $R_z$ ($R^2$=0.044, p=0.264) and $T_z$ ($R^2$=0.126, p=0.054) were not accounted by TN, but were significant (p=0.032) in the $L_z$. The variation of the log-transformed $I_r$-CHL was not accounted ($R^2$=0.258, p=0.110) by $I_w$-TN of inflowing streams, but was determined ($R^2$=0.567, p=0.005) by $I_w$-TP of inflowing streams. In other words, TP inputs from the inflowing streams were the major determinants on the in-reservoir phytoplankton growth. Regression analysis of TN : TP suggested that the ratio was determined by P, rather than N. Overall, our data suggest that TP and suspended solids, during the summer flood period, should be reduced from the eutrophication control and P-input from Ockcheon-Stream should be controlled for water quality improvement.

A Study on the Radar Reflectivity-Snowfall Rate Relation for Yeongdong Heavy Snowfall Events (영동 대설사례의 레이더 강설강도 추정 관계식에 관한 연구)

  • Heavy snowfall events have occurred frequently in the Yeongdong region but understanding of these events have trouble in lack of snowfall observation in this region because it is composed of complex topography like the "Taebaek mountains" and the "East sea". These problems can be solved by quantitative precipitation estimation technique using remote sensing such as radar, satellite, etc. Two radars which are able to cover over Yeondong region were installed at Gangneung (GNG) and Gwangdeoksan (GDK). This study uses radar and water equivalent of snow cover to investigate the characteristics of radar echoes and the $Z_e-R$ relations associated with the 10 Yeongdong heavy snowfall events during the last 5 years (2010~2014). It was found that the heights which the probability of detection (POD) of snow detection by GNG radar is more than 80% are 3,000 m and 1,500 m in convective cloud and stratiform cloud, respectively. The vertical gradient of radar reflectivity is less decreased in convective cloud than stratiform cloud. However, POD by GDK radar are lower than 80% at all layers because the majority of Yeondong observational stations are more than 100 km away from GDK radar site. Furthermore, we examined $Z_e-R$ relation from the 10 events using GNG radar and compared the "a" and "b" obtained from these examinations at Sokcho (SC) and Daegwallyeong (DG). These "a" and "b" are estimated from radar echo at 500 m (SC) and 1,500 m (DG). The values of "a" differ in their stations such as SC and DG are 30~116 and 6~39, respectively. But "b" is 0.4~1.7 irrespective of stations. Moreover, the value of "a" increased with surface air temperature. Therefore, quantitative precipitation estimation in heavy snowfall events by radar echo using fixed "a" and "b" is difficult because these values changed according to those precipitation characteristics.

  • For the well observed 16 globular clusters with known metal abundance (Z), the helium abundances (Y) and ages are determined by various methods, and the relations between Y, Z and age are examined. The luminosity $L_{RR}$ of RR Lyrae stars is known to be dependent of evolutionary models and pulsation theory in the sense that the pulsation theory and horizontal branch (HB) models yield the anticorrelation between $L_{RR}$ and Z whereas main sequence (MS) and red giant branch (RGB) models yield the direct correlation between them. Similarly the anticorrelation between Y and Z is obtained from the HB models and pulsation theory whereas the direct correlation between them is obtained when the RGB model is applied. The current evolutionary models yield the anticorrelation between Z and age of clusters whenever the direct correlation between Y and Z holds. However when the anticorrelation between Y and Z is applied for age determination, the similar age of clusters is obtained as shown by Sandage (1982b). The ages, which are determined by the fitting of C-M diagrams to isochrones in the ($M_v$, B-V)-plane, suggest the two different chemical enrichment processes, which could be accounted for by the disk-halo model for the chemical evolution of the Galaxy (Lee and Ann 1981). Also it is known that the R-method is very useful for Y-determination and the derived Y's show the increasing rate of $\frac{{\Delta}Y}{{\Delta}Z}{\simeq}0.5$ which is comparable to the observed value of $\frac{{\Delta}Y}{{\Delta}Z}{\simeq}0.3$ from HII regions and planetary nebulae by Peimbert and Torres-Peimbert (1976). In this case, the age-metallicity relation of globular clusters could be explained by the disk-halo model.

Patterns of Plasma Fatty Acids in Rat Models with Adenovirus Infection

  • Adenoviral vectors are among the most promising vectors available for human gene therapy. However, the use of recombinant adenoviral vectors, including replicationcompetent adenovirus (RCA), raises a variety of safety concerns in relation to the development of new therapies based on gene therapy. To examine how organic compounds change in rat plasma following the injection of adenovirus, $\beta$-galactosidase expressing recombinant adenovirus (designated rAdLacZ) or RCA, we investigated the content of fatty acids (FAs), which are important biochemical indicators in pathological conditions. Pattern recognition analysis on the level of FAs in rat plasma is described for the visual discrimination of adenovirus infection groups from normal controls. Plasma FAs from four control rats (normal group), and from four rats with rAdLacZ infection and six rats with RCA infection (the two abnormal groups), were examined by gas chromatography-mass spectrometry in selected ion monitoring modes as their tert-butyldimethylsilyl derivatives. In total, 20 FAs were positively detected and quantified. The results of the Student's t-test on the normal mean of two abnormal groups, the levels of three FAs (p<0.05) from rAdLacZ group and eleven FAs (p<0.05) from RCA group were significantly different. When star symbol plotting was applied to the group mean values of 20 FAs after normalization to the corresponding normal mean values, the resulting eicosagonal star patterns of the two infected groups were distorted into similar shapes, but were distinguishable from each other. Thus, these approaches will be useful for screening and monitoring of diagnostic markers for the effects of infection following the use of adenoviral vectors in gene therapy.

ULTRAVIOLET COLOR - COLOR RELATION OF EARLY-TYPE GALAXIES AT 0.05

  • We present the ultraviolet (UV) color-color relation of early-type galaxies (ETGs) in the nearby universe (0.05 < z < 0.12) to investigate the properties of hot stellar populations responsible for the UV excess (UVX). The initial sample of ETGs is selected by the spectroscopic redshift and the morphology parameter from the SDSS DR 7, and then cross-matched with the GALEX far-UV (FUV) and near-UV (NUV) GR6 data. The cross-matched ETG sample is further classified by their emission line characteristics in the optical spectra into quiescent, star-forming, and active galactic nucleus categories. Contaminations from early-type spiral galaxies, mergers, and morphologically disturbed galaxies are removed by visual inspection. By drawing the FUV-NUV (as a measure of UV spectral shape) versus FUV-r (as a measure of UVX strength) diagram for the final sample of -3700 quiescent ETGs, we find that the "old and dead" ETGs consist of a well-defined sequence in UV colors, the "UV red sequence," so that the stronger UVX galaxies should have a harder UV spectral shape systematically. However, the observed UV spectral slope is too steep to be reproduced by the canonical stellar population models in which the UV flux is mainly controlled by age or metallicity parameters. Moreover, 2 mag of color spreads both in FUV-NUV and FUV-r appear to be ubiquitous among any subsets in distance or luminosity. This implies that the UVX in ETGs could be driven by yet another parameter which might be even more influential than age or metallicity.
