Metabolism of Safrole, a Betel Quid Component, and its Role in the Development of Oral Cancer in Taiwan

  • Chewing betel quid is associated with an increased risk of oral cancer. The betel quid chewed in Taiwan includes the inflorescence of Piper betle, which contains high concentrations of safrole (15 mg/fresh weight). Piper betle leaf is also used in betel quid; however, the concentration of safrole in betel leaf has not been documented. Chewing betel quid may contribute to safrole exposure in man (420 mm in saliva). Using $a^{32}$P-postlabeling method, we have recently demonstrated the presence of stable safrole-like DNA adducts in human oral tissues following betel quid chewing. Safrole is a rodent hepatocar-cinogen, and the real nature of safrole-DNA adducts in human tissues beside oral has not been elucidated. In this paper, we tested the safrole DNA adducts forming potential in human hepatic and oral derived cells by the ${32}^P$-postlabeling technique. The results suggest that oral cancer derived cell OC-2 alone is not able to form safrole-DNA adduct. However, safrole DNA adducts can be detected following I'-hydroxysafrole, a proximate safrole metabolite, treatment. In addition, pretreament of cytochrome P450 inducers also enhanced the formation of previously undetectable safrole DNA adducts. This finding couples with our previous results suggest that oral may serve as a target tissue for safrole, and safrole may be involved in oral carcinogenesis.

  • The solution-state of a reclaimed waste would be known to the method in using an analysis for seepage. But it is not the best method in the huge landfill reclaimed all kinds of the waste at random. Especially in case of the landfill called the Nan Gi-do located along the Han-river, it is difficult to judge the generative seepage to be flowed in to the Han-river. So to plan the effective stabilization on a landfill, it is very useful survey method using the Landsat TM image. Operating a heat-distribution analysis with the Landsat TM image, in case of a landfill not having definite data, we would assume the reclaimed sections of the waste to judge a solution-speed late comparatively such as a industry waste or a harmful waste through the heat change.

  • The purpose of this study is to plan the social space for utilization in rural protestant church. Sindai church is the oldest and has rural characteristics in Chung-Ju. This model is used with the data which were obtained by the paster, believer, inhabitants. The results of this study are as follows ; The movable furnitures have merits of multi-use space in the rural protestant church. This could be possible the various functions for a flexible worship space.

  • Al doped ZnO (AZO) thin films were deposited on Si substrates with rod-shaped-surface by pulsed laser deposition method (PLD). Si-rods were prepared through chemical etching. To analyze the influence on the formation of the rod structure, samples with various chemical etching conditions such as AgNO3/HF ratio, etching time, and solution temperature were prepared. The morphology of Si-rod structures were examined by FE-SEM. Fig. 1 shows a typical structure of n-AZO/p-Si-rod juncions. The fabricated n-AZO/p-Si-rod devices exhibited p-n diode current-voltage characteristics. We compared the I-V characteristics of n-AZO/p-Si-rod devices with the samples without Si-rod structure.

  • A nutrition survey of 274 children, 6 to 12 years old, in the Nan Hyang elementary school, which offers no school lunch program and is situated in Shinllim 7-dong of Seoul, as the location of one of the socioeconomically vulnerable groups, was undertaken in July of 1989, to investigate nutritional status. anthropometric data of subjects surveyed slightly lower results than the Korean standards and further lower results than those of Yun Jung elementary school children of Youido, Seoul. 16.3% of subjects were proven to be anemic according to the hematocrit criterion established by the WHO. Mean urinary urea nitrogen/creatinine ration was 9.5$\pm$4.1. Family environment were positively correlated with anthropometric data and results of biochemical tests.

  • Avian influenza virus (AIV) is recognized as key to the emergence of pandemic influenza for humans; there are growing concerns that AIV H9N2 may become more efficient to transmit to humans in the near future, since the infection of poultry with AIV H9N2 has been common in recent years. In this study, we aimed to produce antisera recognizing the HA and NA proteins of AIV H9N2. Initially, coding sequences corresponding to the N-terminal regions of the HA and NA proteins of the Korean AIV H9N2 (A/Ck/Kr/MS96/96) isolated from a domestic chicken were amplified from the genomic RNA. Following cloning of the amplified cDNA fragments into pGEX4T-1 vector, two GST-fusion proteins (GST-HAln and GST-NAn) were expressed in E. coli BL21 and purified with glutathione sepharose columns; the recombinant GST-HAln and GST-NAn proteins were both used as immunogens in rabbits. The antigenicity of the rabbit antisera was analyzed by immunoblotting of the cell lysates prepared from AIV H9N2-infected MDCK cells. Overall, the recombinant HAln and NAn proteins fused to the C-terminus of GST and the rabbit antisera raised against the corresponding recombinant proteins would provide a valuable reagent for AIV diagnosis and basic research.

Preclinical application of 188Re-Tin colloid for treatment of mouse tumor model with peritoneal effusion

  • Re-188 is an excellent and practical radioisotope produced by W-188/Re-188-generator for therapy. We prepared Re-188-tin colloid for therapy of various diseases and tried to treat peritoneal effusion in animal model. Sarcoma-180 cells were injected into ICR mice to induce peritoneal effusion and the mice were grown for 3 d. Re-188-tin colloids (0.25, 0.5, and 1 mCi/mL per 30 g body weight) were injected into the mice and the mice were grown for 90 d. Planar gamma scintigraphy showed even distribution of Re-188-tin colloid radioactivity. Bax expression was found to be dose dependent to Re-188-tin colloid. Normal saline treated group showed the shortest survival time. Among the treated groups, 0.5 mCi dose group showed the longest survival time. In conclusion, Re-188-tin colloid was prepared successfully and showed the feasibility to use as a peritoneal effusion treatment in mice.

Production of hGM-CSF by transformed rice cell suspension culture

  • Recombinant human GM -CSF was expressed and secreted from transgenic rice cell suspension cultures in its biologically active form. This was accomplished by transforming rice callus tissues with an expression vector, pMYN44. containing the hGM -CSF cDNA. Regulated expression and secretion of hGM -CSF from this vector achieved using the promoter, signal peptide, and terminator from a rice alfa-amylase gene Amy3D. The Amy3D gene is expressed in response to sugar deprivation. The recombinant hGM -CSF was expressed from the transgenic rice cell culture on the sugar-free medium as a yield of about 110 mg/L in the culture filtrate, which was determined by ELISA. Biological activity of hGM-CSF was confirmed by measuring the proliferation of the hGM -CSF dependent TF -1 cells.(This work was supported by a grant from the NRL program of the Korean Ministry of Science and Technology.
