  • Tuberculosis is a highly contagious disease caused by Mycobacterium tuberculosis. It affects about 10 million people each year and is still one of the leading causes of death worldwide. About 2 to 3 billion people (equivalent to 1 in 3 people in the world) are infected with latent tuberculosis. Moreover, as the number of multidrug-resistant, extensively drug-resistant, and totally drug-resistant strains of M. tuberculosis continues to increase, there is an urgent need to develop new anti-tuberculosis drugs that are different from existing drugs to combat antibiotic-resistant M. tuberculosis. Against this background, we aimed to develop new anti-tuberculosis drugs using probiotics. Here, we report the anti-tuberculosis effect of Pediococcus acidilactici PMC202 isolated from young radish kimchi, a traditional Korean fermented food. Under coculture conditions, PMC202 inhibited the growth of M. tuberculosis. In addition, PMC202 inhibited the growth of drug-sensitive and -resistant M. tuberculosis- infected macrophages at a concentration that did not show cytotoxicity and showed a synergistic effect with isoniazid. In a 2-week, repeated oral administration toxicity study using mice, PMC202 did not cause weight change or specific clinical symptoms. Furthermore, the results of 16S rRNA-based metagenomics analysis confirmed that dysbiosis was not induced in bronchoalveolar lavage fluid after oral administration of PMC202. The anti-tuberculosis effect of PMC202 was found to be related to the reduction of nitric oxide. Our findings indicate that PMC202 could be used as an anti-tuberculosis drug candidate with the potential to replace current chemical-based drugs. However, more extensive toxicity, mechanism of action, and animal efficacy studies with clinical trials are needed.

  • The cultivation area of rapeseed (Brassica napus L.) has been increased for oil production and landscaping purpose in Korea. However, as the color of rapeseed flower is very simple, diversified flower color is necessary to improve landscape effect. Interspecific and intergeneric crosses between rapeseed (Brassica napus) and three Cruciferae crops were performed in order to grow diverse flower color of rapeseed. The silique formation rate of interspecific cross rapeseed with cabbage (B. oleracea L) was relatively high (65.8%) and higher than intergeneric cross with rapeseed and radish (Raphanus sativus L.), rapeseed and Orychophragmus, respectively. During silique developing period after artificial pollination, there were many siliques without seeds due to the failure of fertilization. The average number of seed per silique obtained from cross rapeseed and cabbage, rapeseed and radish, rapeseed and O. violaceus were 0.12, 0.4 and 0.12, respectively. The phenotypes of $F_1$ hybrid plants from cross rapeseed and Cruciferae crops were mostly similar to maternal line, but leaf length and leaf width were increased. The interspecific cross of rapeseed and cabbage generated ivory color of flower which is the medium color of parents, and intergeneric cross of rapeseed and O. violaceus created entities with larger flowers which seems to enhance landscape effect. The fatty acid composition of most hybrid seeds intermediated of the two parents for oleic acid, linoleic acid and linolenic acid, content. Whereas hybrid of rapeseed and radish produced less erucic acid than radish parent.

  • This survey was conducted in 2015, following up on theed tthe occurrence of Turnip mosaic virus (TuMV) nationwide in radish and Chinese cabbage fields of 28 cities in South Korea. A total of 152 samples of Raphanus sativus and 29 samples of Brassica rapa, showing virus-like symptoms, were collected. Among these, 107 B. rapa samples and 9 B. rapa samples were positive for TuMV when analyzed by RT-PCR. The TuMV strains found in the two crops showed 99% homology in nucleotide and amino acid sequences of coat protein to each other. Furthermore, their sequences showed 99% homology to the sequences of TuMV isolates R007 (GenBank: KU140420) and R041 (GenBank: KU140421) that were collected in 2014. These results suggested TuMV isolated from radish and cabbage in 2015 were the same strain as the isolates R007 and R041 collected in 2014. A screening test was conducted using these two isolates to select TuMV-resistant B. rapa lines out of 167 B. rapa breeding lines.and identified eight lines resistant to R007 (Kenshin, 279002, 279012, 279064, 279081, MP, C-21, HKC-004) and nine lines resistant to R041 (C-26, HKC-005, 11Su-4, 11Su-5, 11Su-7, 11Su-8, Tian Jin Lv Qing Ma Ye, CNU_141193, Jing Lv 60). Our prior data indicated 4.24% difference in sequences between the two isolates and these can serve as potential tools to develop B. rapa markers to screen for resistance against TuMV strainsin breeding populations.

  • This study was carried out to evaluate the effects of row cover materials supported by flexible FRP frames with three kinds of non-woven fabric, and five kinds of net in leaf vegetable cultivation. Leaf radish (Raphanus sativus L.), Chinese cabbage (Brassica rapa L. ssp. pekinensis) were sown on May 21, 2003, and leaf lettuce (Lactuca sativa L.) was planted at the same date in Agricultural Research Center for Climate Change, Jeju, Korea. Net coverings showed higher light transmittance than in non-woven fabric coverings. The average air and soil temperatures increased by $0.3{\sim}1.0^{\circ}C$ in net coverings and $0.5{\sim}3.0^{\circ}C$ in non-woven fabric coverings, respectively. There was no difference in relative humidity among the covering materials. All the vegetables showed higher growths and yields in non-woven coverings than in non-covering. The yields increased 81% in leaf lettuce, 58% in leaf radish, and 93% in Chinese cabbage, respectively. In addition, the insect damages were considerably reduced in the row covering plots than in the control, especially in non-woven fabric coverings.

  • The red radish (Raphanus sativus L.; RR) sprout is a plant of the cruciferous family. In this study, we elucidated the effect of the water extract of RR sprout (RRSE) against ${\alpha}$-amylase, ${\alpha}$-glucosidase, and pancreatic lipase enzyme activity and adipogenesis in 3T3-L1 preadipocytes. ${\alpha}$-amylase, ${\alpha}$-glucosidase, and pancreatic lipase enzyme activity was inhibited in a concentration-dependent manner by RRSE treatment. RSSE also abolished adipocyte differentiation and lipid and triglyceride accumulation without cytotoxicity in 3T3-L1 adipocytes. In addition, RRSE modulated the expression of the proteins related to adipogenic transcription factors: peroxisome proliferator-activated receptor (PPAR)${\gamma}$, sterol regulatory element-binding protein 1 (SREBP-1), and CCAT/enhancer binding protein (C/EBP)${\alpha}$. RRSE also suppressed expression of the proteins responsible for lipid synthesis, transport, and storage: adiponectin, fatty acid synthesis (FAS), perilipin, and fatty acid bind protein-4 (FABP4). This study showed that RRS treatment has the potential to inhibit obesity by controlling the expression of adipogenic transcription factors and adipogenic proteins.

  • In order to investigate the compression and decompression properties of cucumber, radish, garlic, ginger and potato, edible parts of samples were prepared to size of ${\Phi}\;5\;mm{\times}H\;5\;mm$, and force deformation relationship during application and removal of force were observed. Compositions of sample and cell characteristics were measured, and correlations between them were investigated. Deformation rate was large in initial stage of compression and decreased afterward, but the reverse trends were observed in the decompression. The time and deformation to 9 N were large of 5.30 sec and 1.344 mm in potato, and small of 4.62 sec and 0.896 mm in garlic, respectively. Force(y)-deformation(x) curve between compression and decompression were clearly showed hysteresis loop and relationship of x and y were as follows: y=esp (a+b log(x)). The maximum work was $3.888{\sim}5.099{\times}10^{-3}\;J$ for potato in compression and $2.09{\times}10^{-3}\;J$ for garlic in decompression. Irrecoverable work were large as $77{\sim}96%$ in cucumber, radish and potato, and small as $36{\sim}42%$ in garlic. Compression deformation were large as $1.016{\sim}1.344\;mm$ in potato, and small as $0.656{\sim}0.896\;mm$ in garlic. Degree of elasticity were large as $0.756{\sim}0.777$ in garlic, and small as $0.301{\sim}0.465$ in radish and potato. Compression and decompression characteristic values were showed high correlation with moisture, viscosity of juice, ceil size, density and regularity.

  • Studies were conducted on the nature of allelopathic effect of the substances in the waste of ginkgo leaves from pharmaceutic factory. In the first step, to find out whether there was any allelophatic effect, young seedlings of radish and rice were grown in the water (crude) extract of ginkgo leaf waste and in different liquid/lquid partitioned fractions of EtOAc at pH 9, EtOAc at pH 3, and BuOH. As second step, attempts were made to isolate and identify the allelophatic substance in different liquid/liquid partitioned fractions using GC/MS and NMR techniques. The water (crude) extract of ginkgo leaf waste retarded the growth of radish seedlings under 10% concentration. In case of rice seedlings, the water extract of ginkgo leaf extract showed adverse effect on the growth when combined with $3.3{\times}10^{-6}M$ gibberellin A3. All of the liquid/liquid fractions of crude extract showed strong retardation of seedling growth of radish and rice at the concentration of 1%. Allelophatic substance was isolated from the crude extract using liquid/liquid partition, column chromatography and HPLC techniques. The analytical results of isolated componet using GC/MS and NMR proved that the allelophatic substance in the ginkgo leaf wastes is catechol; one of phenol compounds. Based on the experiences current study, a practical method for the testing of allelophatic effect of crude extract of some materials was proposed. In this method, rice seeds were allowed to sprout until the length of coleoptile to reach 0.5 mm. Such seedlings were submerged in the solution containing supposedly allelophatic substance and the length of shoot and root was measured 3 days after treatment.

  • The gaseous product of nitrogen cycle, nitrous oxide ($N_2O$) is a potent greenhouse gas whose Global Warming Potential (GWP) is about 300 times greater than $CO_2$. The dynamics of $N_2O$ emission are controlled by such environments and soil conditions. The main aim of this study is to investigate variations of $N_2O$ emission and its controlling factors with different land-use patterns in Haean basin. A forest, a radish field and a rice paddy were selected as three different land-use patterns. Their $N_2O$ emissions were measured every month during a growing season. We also collected soil samples with seasons and analyzed soil characteristics including inorganic nitrogen content. $N_2O$ emission was greatest at the radish field likely due to anthropogenic nitrogen addition by fertilization. Soils of forest and rice paddy also contained inorganic nitrogen originated from organic matter. However, the spatial variation was great and it looks that nitrogen cycle and $N_2O$ production were slower than that of radish field. Intensive observation and control of fertilization would be requiredto adjust $N_2O$ emission from agriculture soils.

  • This research was carried out to study the seed dormancy of radishes which were used for summer vegetation. The differences of dormancy of cultivars, maternal effects, the effect of full day length to mother plants, and the effects of chemical treatments for breaking the seed dormancy were considered for this study. These results sere summarized as follows; 1. The germination percentage of 7 cultlvars sown just after harvest were as follows;SP70-:0, PS11-:37.5, GJ75-:75.5, JD127-:77.5, YH85-:81.0, M55-:96.5, and JC73-:98.0%. 2. The dormany of $F_1$ seed of radish was severe when SP70-was used for mother plant. 3. Germination behavior was different by the maturity of plants grown for seed production. The dormancy of seed harvested from immature field-grown plants sown Nov. 10 was deeper than mature plants sown Mar. 22. 4. The germination ratio of tire seed of SP70-radish was increased by treatment of full day length to tire mother plant during full period of growing or since first flowering stage, while that was 0% by treatment since yellow ripening stage. 5. GA 200 ppm+thiourea 0.2% treatment was most effective for breaking of radish seed dormancy.

  • This study used crop data from statistics yearbooks in Naju and climate data from Gwangju weather station to investigate whether climate changes have had significant impact on crops. The sample crops are rice, barley, pear, radish, Korean cabbage and red pepper. The results showed that the changes in temperature have shifted crop phonology and affected crop growth. The rice and barley heading date were advancing and had negative correlation with average temperature over 30days before average heading date. The number of rice grains per unit area $(m^2)$ were decreasing while the number of barley grains per unit area $(m^2)$ were increasing because average temperature during grain filling period of rice (barley) was increasing (decreasing). Therefore, decreasing (increasing) yields of rice (barley) can be predicted by global warming. The sprouting, flowering and full flowering date of pear were advancing. The sprouting date of pear had negative correlation with average temperature from February to March and the flowering and full flowering date of pear had negative correlation with average temperature from February to April. The brix and weight of pear were increased and were most sensitive to August and September average temperature. An earlier blossom of pear trees holds the danger of damage by late frosts. The plant length of radish and chinese cabbage were decreasing and negatively influenced by maximum temperature on September. The fruit set numbers of red pepper were increasing recently and had positive correlation with minimum temperature on August. The growth of radish and Korean cabbage will be poor, but the growth of red pepper will be good by rising temperature.
