Effects of Environmental Factors on the Growth of Gametophytes and Young Sporophytes of Eisenia bicyclis (Kjellman) Setchell (대황(Eisenia bicyclis) 배우체와 아포체의 생장에 미치는 환경 인자의 영향)

  • Eisenia bicyclis, a perennial macroalga is a primary producer of in the ocean, It has been identified as a key species that plays a vital role in maintaining the ecosystem stability. Also, it is an important target in marine afforestation projects and useful marine organisms. In addition, E. bicyclis is used as a health food for humans. This study investigated the effect of water temperature, light (photon irradiance), and duration of light (photoperiod) on the growth of gametophytes and young sporophytes of E. bicyclis. The germination and growth of the zoospores of E. bicyclis were examined at five temperatures (5℃, 10℃, 15℃, 20℃ and 25℃), four intensities of photon irradiance (10, 20, 40, and 80 μmol m-2s-1), and photoperiods (14:10 and 10:14 light/dark cycles). The zoospores released from mature plant germinated into the gametophytes under all experimental conditions. The gametophytes were able to grow at water temperature 5℃-25℃ and mature at 10℃-20℃. The optimal range of water temperature for the maturation of the gametophyte was 15℃-20℃. At 25℃, E. bicyclis gametophytes grew rapidly but did not mature. The optimal culture conditions for the growth of young sporophytes grew slowly in low temperature and photon irradiances.

  • This study was carried out to investigate the efficient in vitro mass propagation methods for juvenile sporophytes of Matteuccia struthiopteris. Chopped segments of pinnae, petiole and rhizome were cultured on 1/2MS with 0.1% activated charcoal. Among these explant sources only rhizome segments produced young sporophytes, regenerating vigorously on 1/2 MS medium. Adjusting sucrose concentration to 2% and supplement to $50mgL^{-1}$ $NaH_2PO_4$ in 1/2MS medium proved to be more efficient for plant regeneration. Various combinations of growth regulators such as kinetin, BA, NAA, and IBA were added to the growing media, and the best sporophyte regeneration was obtained by $1{\mu}M$ kinetin. The BA addition resulted in vigorous proliferation of meristematic tissues, but without differentiation to sporophytes. Three types of culture methods, solid using agar, liquid stationary, and liquid shaking culture, were employed with or without activated charcoal. The addition of 0.1% activated charcoal to modified 1/2MS media (2% sucrose, $50mgL^{-1}$ $NaH_2PO_4$, $1{\mu}M$ kinetin, pH 5.8 and 0.8% agar) yielded highest sporophyte regeneration in liquid shaking culture.

Morphological and biochemical differences in three Undaria pinnatifida populations in Korea

  • Twelve morphological characters and the biochemical composition of Undaria pinnatifida f. distans sporophytes growing on the rocky shores of Jindo and Wando and on cultivation ropes in Kijang were measured to determine whether each population could be characterized by morphological features and biochemical composition. The goal of this study was to compare phenotypic variations between populations as they relate to environmental conditions. The sporophytes of the Kijang population were two times longer and 19 times heavier than those at Jindo. Sporophylls of the Jindo U. pinnatifida population were significantly smaller in length, width, frill number, and weight than those at Wando and Kijang. Kijang Undaria plants showed the highest contents of total protein, crude fiber, total amino acids, the amount of essential amino acids, the proportion of total unsaturated fatty acids, and eicosapentaenoic acid. However, the Jindo population showed the greatest content of carbohydrates, lipids, and minerals (Zn and Ca) of the three U. pinnatifida populations. In particular, Zn content of Jindo plants was 30 times greater than that of Kijang plants. Thus, the proximate composition, mineral composition, amino acids, and fatty acids of Undaria pinnatifida plants were distinguishable among the three representative Undaria populations evaluated. These results suggest that morphological and biochemical differences of the three U. pinnatifida populations can be attributed to differences in environmental conditions of their habitats.

Size determination of Ecklonia cava for successful transplantation onto artificial seaweed reef

  • The objective of this study was to determine the optimal blade size and timing to transplant seed-stock of Ecklonia cava Kjellman onto the reef structure. We used the modified artificial stepped reef structure. A total of 14 units (3.0 m length ${\times}$ 3.5 m width ${\times}$ 1.1 m height) were deployed 7-8 m deep under the water to examine the optimal blade size and timing to transplant seed-stock of E. cava onto the structures. Sporophytes of E. cava <1 cm in length were all died within 1 month of transplantation. The blades of 5-10 cm in length which were transplanted in March 2007 survived and grew well on the artificial reefs. Growth rates of 5-10 cm size class were higher than those of longer blade sporophytes (20-30 cm size class, transplanted in April) while the survival rates showed no difference between the classes of blade size. Both classes of 5-10 and 20-30 cm in length grew until July, and a reduction in size had occurred in September. These results indicate the importance of the blade size of E. cava and timing for successful transplantation of the seaweed on artificial reef structures.

Artificial Reestablishment of the Kelp and Red Algal Symbiosis

  • A type of symbiosis was previously described from nature in which the gametophytes of Laminariales were endophytic in filamentous red algae. Here we reconstruct this symbiosis for the first time in laboratory culture using zoospores of the kelp, Undaria pinnatifida, and the red alga, Aglaothamnion oosumiense. Zoospores of U. pinnatifida readily attached to A. oosumiense. In 48 h these spores germinated and the initial germ tube penetrated into the host cell wall leaving only an empty zoospore wall outside the host. Within ten days, four to five-celled endophytic gametophytes were present. Zoospores of Laminaria religiosa which were also inoculated into cultures of A. oosumiense rarely attached to the red alga and never became endophytic. Within ten days the free-living gametophytes of L. religiosa on cover slips became fertile and produced young sporophytes. These observations demonstrate the ability of U. pinnatifida to become endophytic, and show differences in host specificity among kelp species.

Differential Gene Expression in a Red Alga Gracilaria textorii(Suringar) Hariot (Gracilariales, Florideophyceae) between Natural Populations

  • The bio-molecules involved in defense mechanisms can be used as efficient biomarkers for physiological changes in organisms caused by both of internal and external stress. Thus, the expression level of genes which encoding such molecules serve as critical 'early warning system' for environmental assessment as well as health diagnosis of biological organisms. In this study, Cytochrome P450, Heat shock protein 90, Ubiquitin and ${\beta}$-actin gene were isolated for the first time from a red alga Gracilaria textorii. The quantitative differential gene expression analyses of three genes, GteCYP1A, GteHsp90 and Gte-UB, were carried out in G. textorii sporophytes collected from two different localities, polluted Sujeong (Masan, Korea) and potentially unpolluted Danggeum (Daemaemuldo Is., Korea). The transcripts of all three tested genes were highly expressed in the Sujeong population. The results suggest: 1) the Sujeong site was more polluted than the Danggeum site; 2) G. textorii could be applicable to marine environment monitoring in coastal regions.

  • Kjellmaniella crassifalia Miyabe is one of the valuable seaweed crops cultured in the Korean coastal waters of the East Sea. Unfortunately recent environmental changes have adversely influenced its growth, prompting the need to develop techniques for species restoration. To obtatin biological information, the growth of the species was monitored for one year. The maximum blade length, $110{\pm}45cm$, was attained in July, and zoospores were released in November. The optimum culture conditions allowing juvenile sporophytes to grow to young blades consisted of $20{\mu}mol/m^2s\;at\;10^{\circ}C$ under which the blade length grew to $1,732.5{\pm}143.3{\mu}m$ in 42 days. Sporangial sori started to form in September, reaching 85% maturity in November and 89% maturity in December. Examining the growth of the species according to depth, the individuals growing at 5 meters grew the most, reaching a blade length of $75.9{\pm}18.9cm$ in August.

  • This study was conducted to investigate the optimal conditions for spore germination, prothallus propagation, sporophyte formation, and seedling growth in Polystichum braunii (Spenn.) $F{\acute{e}}e$. The rate of spore germination and early prothalium development was high in Knop (41.2%), which had low mineral content. The optimal medium for prothallus propagation and sexual organ formation was 2MS medium (2% sucrose). Among the various mixtures of cultivation soil (bedding soil, peat moss, perlite, and decomposed granite), a mixture of bedding soil and decomposed granite at a ratio of 2:1 (v:v) had a positive effect on sporophyte formation (276.3 ea/$7.5m^2$). The most efficient conditions for promoting the growth of sporophytes were pots filled with only bedding soil.

  • Adventitious shoots were induced from pinnae, petiole and rhizome in Pteris cretica 'Wilsonii' in order to develop the efficient mass propagation method, using in vitro culture. Only homogenized rhizome segments could regenerate young sporophytes. Efficient regeneration of multiple shoots was obtained on the one-eighth strength MS medium containing 1% sucrose, and $50mg{\cdot}L^{-1}$ $NaH_2PO_4$. To achieve higher rate of regeneration from rhizome segments, rhizome segments were exposed to growth regulators for 2 months and then subcultured on hormone-free medium. The greatest shoot regeneration was obtained by $1{\mu}M$ kinetin with $5{\mu}M$ NAA. BA was effective in formation of GGB (kind of meristems), but they showed low shoot regeneration rate. Plants obtained from present experiments were transplanted to examine good environmental conditions for acclimation. Juvenile plants obtained by the one-eighth strength MS medium showed highest survival rate and vigorous growth at the seedling stage.
