Citrinin (CIT) is a toxic secondary metabolite produced by fungi belonging to the Penicillium, Aspergillus, and Monascus spp. This toxin has been detected in many agricultural products. In this study, a strain Y3 with the ability to eliminate CIT was screened and identified as Cryptococcus podzolicus, based on the sequence analysis of the internal transcribed spacer region. Neither uptake of CIT by cells nor adsorption by cell wall was involved in CIT elimination by Cryptococcus podzolicus Y3. The extracellular metabolites of Cryptococcus podzolicus Y3 stimulated by CIT or not showed no degradation for CIT. It indicated that CIT elimination was attributed to the degradation of intracellular enzyme(s). The degradation of CIT by C. podzolicus Y3 was dependent on the type of media, yeast concentration, temperature, pH, and initial concentration of CIT. Most of the CIT was degraded by C. podzolicus Y3 in NYDB medium at 42 h but not in PDB medium. The degradation rate of CIT was the highest (94%) when the concentration of C. podzolicus Y3 was $1{\times}10^8cells/ml$. The quantity of CIT degradation was highest at $28^{\circ}C$, and there was no degradation observed at 3$5^{\circ}C$. The study also showed that acidic condition (pH 4.0) was the most favorable for CIT degradation by C. podzolicus Y3. The degradation rate of CIT increased to 98% as the concentration of CIT was increased to $20{\mu}g/ml$. The toxicity of CIT degradation product(s) toward HEK293 was much lower than that of CIT.
The abatement of methane emission from ruminants is an important global issue due to its contribution to greenhouse gas with carbon dioxide. Methane is generated in the rumen by methanogens (archaea) that utilize metabolic hydrogen ($H_2$) to reduce carbon dioxide, and is a significant electron sink in the rumen ecosystem. Therefore, the competition for hydrogen used for methanogenesis with alternative reductions of rumen microbes should be an effective option to reduce rumen methanogenesis. Some methanogens parasitically survive on the surface of ciliate protozoa, so that defaunation or decrease in protozoa number might contribute to abate methanogenesis. The most important issue for mitigation of rumen methanogenesis with manipulators is to secure safety for animals and their products and the environment. In this respect, prophylactic effects of probiotics, prebiotics and miscellaneous compounds to mitigate rumen methanogenesis have been developed instead of antibiotics, ionophores such as monensin, and lasalocid in Japan. Nitrate suppresses rumen methanogenesis by its reducing reaction in the rumen. However, excess intake of nitrate causes intoxication due to nitrite accumulation, which induces methemoglobinemia. The nitrite accumulation is attributed to a relatively higher rate of nitrate reduction to nitrite than nitrite to ammonia via nitroxyl and hydroxylamine. The in vitro and in vivo trials have been conducted to clarify the prophylactic effects of L-cysteine, some strains of lactic acid bacteria and yeast and/or ${\beta}$1-4 galactooligosaccharide on nitrate-nitrite intoxication and methanogenesis. The administration of nitrate with ${\beta}$1-4 galacto-oligosaccharide, Candida kefyr, and Lactococcus lactis subsp. lactis were suggested to possibly control rumen methanogenesis and prevent nitrite formation in the rumen. For prebiotics, nisin which is a bacteriocin produced by Lactococcus lactis subsp. lactis has been demonstrated to abate rumen methanogenesis in the same manner as monensin. A protein resistant anti-microbe (PRA) has been isolated from Lactobacillus plantarum as a manipulator to mitigate rumen methanogenesis. Recently, hydrogen peroxide was identified as a part of the manipulating effect of PRA on rumen methanogenesis. The suppressing effects of secondary metabolites from plants such as saponin and tannin on rumen methanogenesis have been examined. Especially, yucca schidigera extract, sarsaponin (steroidal glycosides), can suppress rumen methanogenesis thereby improving protein utilization efficiency. The cashew nutshell liquid (CNSL), or cashew shell oil, which is a natural resin found in the honeycomb structure of the cashew nutshell has been found to mitigate rumen methanogenesis. In an attempt to seek manipulators in the section on methane belching from ruminants, the arrangement of an inventory of mitigation technologies available for the Clean Development Mechanism (CDM) and Joint Implementation (JI) in the Kyoto mechanism has been advancing to target ruminant livestock in Asian and Pacific regions.

All living organisms use numerous signal-transduction pathways to sense and respond to their environments and thereby survive and proliferate in a range of biological niches. Molecular dissection of these signalling networks has increased our understanding of these communication processes and provides a platform for therapeutic intervention when these pathways malfunction in disease states, including infection. Owing to the expanding availability of sequenced genomes, a wealth of genetic and molecular tools and the conservation of signalling networks, members of the fungal kingdom serve as excellent model systems for more complex, multicellular organisms. Here, we employed Cryptococcus neoformans as a model system to understand how fungal-signalling circuits operate at the molecular level to sense and respond to a plethora of environmental stresses, including osmoticshock, UV, high temperature, oxidative stress and toxic drugs/metabolites. The stress-activated p38/Hog1 MAPK pathway is structurally conserved in many organisms as diverse as yeast and mammals, but its regulation is uniquely specialized in a majority of clinical Cryptococcus neoformans serotype A and D strains to control differentiation and virulence factor regulation. C. neoformans Hog1 MAPK is controlled by Pbs2 MAPK kinase (MAPKK). The Pbs2-Hog1 MAPK cascade is controlled by the fungal "two-component" system that is composed of a response regulator, Ssk1, and multiple sensor kinases, including two-component.like (Tco) 1 and Tco2. Tco1 and Tco2 play shared and distinct roles in stress responses and drug sensitivity through the Hog1 MAPK system. Furthermore, each sensor kinase mediates unique cellular functions for virulence and morphological differentiation. We also identified and characterized the Ssk2 MAPKKK upstream of the MAPKK Pbs2 and the MAPK Hog1 in C. neoformans. The SSK2 gene was identified as a potential component responsible for differential Hog1 regulation between the serotype D sibling f1 strains B3501 and B3502 through comparative analysis of their meiotic map with the meiotic segregation of Hog1-dependent sensitivity to the fungicide fludioxonil. Ssk2 is the only polymorphic component in the Hog1 MAPK module, including two coding sequence changes between the SSK2 alleles in B3501 and B3502 strains. To further support this finding, the SSK2 allele exchange completely swapped Hog1-related phenotypes between B3501 and B3502 strains. In the serotype A strain H99, disruption of the SSK2 gene dramatically enhanced capsule biosynthesis and mating efficiency, similar to pbs2 and hog1 mutations. Furthermore, ssk2, pbs2, and hog1 mutants are all hypersensitive to a variety of stresses and completely resistant to fludioxonil. Taken together, these findings indicate that Ssk2 is the critical interface protein connecting the two-component system and the Pbs2-Hog1 pathway in C. neoformans.
본 연구에서는 적효모 X. dendrorhous KCTC 7704로부터 여러 ${\beta}$-ionone 내성 변이균주를 선별하였다. 야생균 KCTC 7704는 ${\beta}$-ionone 0.021 mM 농도에서 생육이 현저히 저하되었지만, NTG처리 후 ${\beta}$-ionone 0.1 mM 농도에서 선별된 변이균주는 ${\beta}$-ionone 0.15 mM에서도 70% 이상의 상대 생육율을 나타내는 매우 강한 ${\beta}$-ionone 내성을 갖고 있었다. 여러 ${\beta}$-ionone 농도에서 선별한 변이균주들을 $20^{\circ}C$에서 4일간 회분식 발효로 배양하여 그 특성을 조사하였다. 선별된 가장 우수한 변이균주는 야생균주보다 카로티노이드 생성능이 2.3배 향상(즉 $1.2{\mu}g$ of total carotenoids per mg of dry cells)되었으며 유기산과 같은 대사산물은 거의 생성하지 않았다. 여러 탄소원 들에 대한 비교 발효특성 연구 결과 과당이나 자당을 사용했을 때봐 비교하여 포도당 배지에서 최종 균체농도 및 총 카로티노이드 생성량이 많았다. 포도당이 제한되는 연속발효(dilution rate $0.04h^{-1}$) 실험을 통하여 pH의 영향을 조사한 결과 균체농도 및 총 카로티노이드 생성은 pH 4.0 조건하에서 최적인 것을 알 수 있었다.
발효식품이 유해곰팡이에 의한 발암물질(aflatoxin)생성에 미치는 억제 효과에 관한 연구의 일환으로 유산균 및 유산균 발효유가 Asperillus Parasiticus ATCC 15517의 성장과 aflatoxin 생성에 미치는 영향을 실험하였다. 발효유를 일정 농도별로 첨가한 YES 배지에서 Asperillus Parasiticus를 배양말기에 대조군에 비하여 건조 균체량, 배양물의 pH, 그리고 alftoxin 생성량 등이 낮게 나타났다(p<0.05). Aflatoxin B1은 48.6~58.1% 각 감소되었으며 G1은 29.8~34.2%가 감소되었다. 이 발효유의 발효에 사용된 유산간균(lactobacillus casei)과 Asperillus Parasiticus를 변형 APT 배지에 혼합 배양한 결과 Asperillus Parasiticus 단독 배양의 경우에 비하여 균체량이 배양 5일째까지는 현저하게 억제되었으나 배양 말기에는 유의한 차이를 보이지 않았다. 또한 배양 말기에 단독 배양의 경우보다 pH가 훨씬 감소되고 (p<0.05) aflatoxin의 생성량도 감소되었다. 이로부터 발효유는 유해곰팡이인 Asperillus Parasiticus의 성장과 aflatoxin 생성을 억제시키는 효과를 가짐을 알 수 있으며, 이는 발효에 관여한 미생물의 경쟁뿐만 뿐만아니라 유산균의 대사산물에 의한 영향으로 보여진다.
본 연구는 기존에 과실주 발효 시, non-Saccharomyces 효모를 혼합 발효하여 향기성분을 증가시키는 기술을 확대하여 단독 및 혼합 발효로 제조한 탁주 술덧의 향기성분을 증가시킨 후 증류 과정을 통해 주질을 향상시킬 수 있다는 이론을 검증하고자 한 연구이다. 연구를 진행하기 전, 예상한 것처럼, 혼합 발효 증류식 소주에서 과실향을 나타내는 저분자 ester 계열의 휘발성 향기성분들의 함량이 증가하는 것이 입증되었으며, H. uvarum 계열의 효모를 사용한 경우, HU SJ69 혼합 발효구는 S. cerevisiae 단독 발효구와 비슷한 아미노산 함량을 나타냈으나, HU S6 혼합 발효구의 경우, 아미노산 함량이 감소하는 것으로 나타났다. 관능검사 결과, 휘발성 향기성분이 증가한 HU S6 혼합 발효구에서 SC NY21을 사용한 다른 증류식 소주들과 비교하여 유의적으로 큰 차이를 나타내지는 않았지만, 조금 더 높은 향미 점수를 얻은 것을 통해, 향후 non-Saccharomyces 효모의 혼합 발효를 통한 증류식 소주의 품질을 향상시킬 수 있는 기초연구로써 중요한 의미를 가진다고 판단된다. 또한, H. uvarum 계열 효모의 경우에는 국내에서는 식품 원료로 등재되지 않았기 때문에, 향후 산업적 이용을 위한 안전성 검증 등의 연구가 추가되어야 할 것으로 사료된다.
가능한 한 대량의 균주를 테스트하여 고생산성을 지닌 균주를 신속히 선별하기 위해서는 소규모 액상배양 방법의 확립이 필수적인데, 강력한 지질저하제인 monacolin-K의 생산균주인 균사형성 Monascus의 경우 포자나 균사체의 형성이 활발하지 않은 배양 생리적 특성으로 인해 소규모 (miniature) 액상배양이 매우 어려운 문제점이 있다. 본 연구에서는 monacolin-K 고생산성 균주개량의 효율성을 큰 폭으로 증가시키고자, 기존의 플라스크 액상배양 방법을 소규모화시킨 tube 배양 방법을 개발함으로써 단기간에 보다 많은 양의 균주를 테스트하고자 하였다. 이차대사산물인 monacolin-K의 생합성은 각각의 배양단계에서의 생산균주의 배양형태가 중요한데, 특별히 최종 생산배양에서 생산성 증가에 심각한 영향을 미치는 배양형태인 직경 1 mm 이하의 pellet 모양을 유도하기 위해서는 성장배양 시에 반드시 고농도의 균사모양이 유도되어야 하는 것으로 관찰되었다. 50 ml tube (7 ml의 조업부피)를 이용하는 소규모 액상 성장배양의 경우 solid seed 배양 단계에서의 포자형성배지의 조성을 통계적 방법을 통해 최적화하고, 또한 성장배지 성분에 brown rice 가루 20 g/L를 첨가할 때, 원하는 균사모양의 배양형태가 유도됨을 확인할 수 있었다. 한편 7 ml의 조업부피의 tube를 이용한 소규모 성장배양 방법의 개발로 인해, 기존의 flask 배양을 이용할 때보다 단기간에 훨씬 많은 변이주의 생산성을 조사하게 되어 균주개량 속도를 큰 폭으로 향상시킬 수는 있었으나, 선별된 개별 균주의 monacolin-K의 생산성은 오히려 전체적으로 감소하는 경향을 보여 주었다. 이러한 결과는 소규모 배양방법을 확립하기 위해 새로이 개발한 포자형성배지와 성장배지의 조성변화로 인해 기존에 확립된 생산배지에서는 생산균주의 monacolin-K 생합성 능력이 최대로 발휘되지 않았기 때문에 발생한 것으로 판단되었다. 따라서 본 연구를 통해 확립된 소규모 성장배양에 적합한 최적의 생산배지 조성을 확립하고자 통계적 방법인 Plackett-Burman design을 적용하여 monacolin-K 생산성 향상에 영향을 주는 glycerol, malt extract, yeast extract, brown rice의 4가지 성분들을 최종 선별하였고, 또한 이들의 최적농도 결정을 위해 반응표면분석 실험을 수행하였다. 이와 같이 결정된 최적 생산배지를 사용하여 생산배양을 수행한 결과, monacolin-K의 생산성이 2배 이상 증가하고, 배양 형태 또한 용존산소와 영양분의 전달이 매우 용이한 직경 1mm 이하의 pellet 모양을 유지함을 확인할 수 있었다.
