Identification of Chinese Yam Necrotic Mosaic Virus(ChYNMV) infecting Chinese yam(Dioscorea opposita Thunb. cv. Jang-Ma) in Korea.

  • Chinese yam(D. opposita Thunb. cv. Jang-Ma) plants showing necrotic mosaic symptom were collected from their growing fields in Andong, Korea. Electron microscoptic examination of negatively stained preparations showed filamentous particles of 660nm in length. The virues purified partially were used to isolate Viral RNA as template for RT-PCR to amplify the CP gene with ChYNMV specific and oligo dT primers. Amino acids sequeces revealed that the viruses shared 99.3% similarity with ChYNMV(AB044386) which was known as the member of macluravirus. So the viruses from Chinese yam(D. opposita Thunb. cv. Jang-Ma) plants were identified as ChYNMV. Comparing the amino aced sequences of ChYNMV strains with other macluraviruses such as CdMV, NLV and MacMV revealed that N-terminal was the most variable region and conserved regions were present within the genus Macluravirus.

Dormancy-related Change in Endogenous ABA, Batatasin, and Sugar in Stored Tuber and Bulbil of Chinese Yam

  • Endogenous ABA and batatasins were first quantified in the stored tubers and bulbils of the Chinese yam, Dioscorea opposita Thunb. cv. 'Tsukune' by GC-MS with comparison of internal standard, and sugar content was also analyzed by HPLC with comparison of external standard. Endogenous ABA content in stored tubers and bulbils was rapidly decreased as storage period prolonged. ABA content of the bulbils was always higher than that of tuber during storage periods. All batatasins of stored tubers and bulbils at 30 days after storage decreased steadily compared to 120 days after storage. On the contrary, batatasin-Ⅲ of the stored tuber and bulbils was only decreased at 60 days after storage. In Dioscorea opposita Thunb. cv. 'Tsukune' like D. japonica, D. alata, and D. bulbifera, may be controlled by endogenous batatasins and ABA, In these compounds, batatasins rather than ABA might be closely related to dormancy-inducing compound during dormancy of the Chinese yam, Dioscorea opposita Thunb. cv. Tsukune.

Effect of Proton Beam Radiation on Bulbil Yield and Gibberellins of Chinese Yam (Dioscorea opposita Thunb.)

  • The study was carried out to evaluate the affect of proton beam radiation on production of bulbil and tuber including change of endogenous gibberellins, of Dioscorea opposita Thunb. The yield of bulbils and tubers from non- and irradiated D. opposita Thunb at doses of 5, 10, 15 and 20 Gy were determined. Endogenous gibberellins were also quantified by GC/MS analysis. D. opposita tubers irradiated at 15 Gy produced higher bulbil production than non-irradiated plants. Enlarged bulbil (above size diameter 4 mm) was significantly increased at 15 Gy. Bioactive endogenous $GA_4$ was dominant in bulbils and tubers irradiated with proton beam rather than $GA_1$. Major gibberellins biosynthetic pathways in bulbils and tubers of D. opposita plants were non C-13 hydroxylation route. From the results of this study, 15 Gy proton beam radiation was suggested as an optimal dose that can produce high amounts of bulbil for mass production of D. opposita plant.

둥근마(Dioscorea opposita Thunb.)의 괴경비대 및 성분특성 (Tuber Enlargement and Chemical Components of Yams (Dioscorea opposita Thunb.))

  • 둥근마(Dioscorea opposita L.) 계통 중 품질이 우수하고 이용 가치가 기대되는 단파마를 우리나라에서 재배시 그 가능성과 제 기관의 생육특성을 밝히기 위하여 실시하였다. 두 D. opposita의 괴경의 수량성 및 성분특성을 비교하여 재배 가능성을 시험하였다. 둥근마의 괴경형성은 정식 60일 정도에서 일어나, 그 후 지속적으로 비대 생장이 일어나 160일까지 비대한 후 등숙하였다. 200일째 단마의 괴경중이 512g으로 둥근마 498g보다 약간 높았으나 건물중은 108/127g으로 둥근마가 높아 모양과 건물중이 우수하였다. 둥근마의 일반성분을 측정한 결과 단백질 함량에서 단마$(2.10\%)$보다 $3.62\%$로 높았으며 섬유, 지방의 함량이 낮았다. 수분함량은 $ 64.53\%로 단마$(79.38\%)$보다 건물량이 높았다. 둥근마의 경도를 보면 2787.6로 단마의 4946.9 보다 약 2배정도 낮고, 색도 중 명도(L)는 둥근마가 77.37으로 단마보다 높았다. 둥근마의 괴경으로부터 추출한 물질은 diosgenin의 표준물질과 일치하는 피크를 얻었으며, retention time은 약 5.9에서 나타났다. 둥근마의 괴경에서 diosgenin은 $3.32\%$의 함량을 나타낸 반면 단마에서는 $2.61\%$로 낮은 함량치를 나타냈다.

Properties of Starches in Chinese Yam, Dioscorea oppsita Thunb. Irradiated with Proton Beam

  • The survival rate and thermal, crystal, and physicochemical properties of starches in chinese yam tubers irradiated to proton beam were determined. Survival rate was decreased with increased proton beam irradiation. Amylose content of D. opposita starches from different proton beam ranged from 13.2% to 17.8%. D. opposita starch at 5 Gy showed the highest ${\Delta}H_{gel}$ values (12.0 J/g) while D. opposita starch at 25 Gy showed the lowest values (10.1 J/g). Several parameters such as PKV (peak viscosity), HPV (Hot peak viscosity) and CPV (Cooling peak viscosity) decreased with the increase in irradiation dose. The degrees of crystallinity of the D. opposita starches at 5, 10, 15, 20 and 25 Gy were 37.2, 38.3, 38.9, 39.2 and 39.9%, respectively. It might be deduced that proton beam irradiation causes changes of starch, especially at high dose irradiation.

The Effects of Growth Regulators and Medium Strength on the Shoot and Bud Formation from the Shoot Apex of Chinese Yam (Dioscorea opposita Thunb)

  • Plantlet regeneration from the shoot apex was studied in three different genotypes of the chinese yam (Dioscorea opposita Thunb) cv. Jnagma and Danma, Dunggunma. The effects of plant growth regulators and inorganic salts concentration of the culture medium on bud induction and shoot growth were examined. The combinations of 0.2 mg/L BAP + 0.2mg/L kinetin, 0.01mg/L NAA + 0.2 mg/L kinetin and a single treatment of 0.2mg/L BAP were equally effective for bud and shoot formation from the shoot apices in the three cultivars. Auxin (2,4-D, NAA) treatment enhanced calli formation from the cultured apices. Also, the shoot apices of the cv. Dunggunma produced more callus and buds on the culture medium (MS) containing 0.05mg/L NAA and 0.5-1.0mg/L SAP. Lower salt strength of medium inhibited shoot elongation but did not have much effect on the shoot and bud induction from the shoot apices. These results will be useful to obtain disease-free plants of the Chinese yam.

  • YMV-K strains were purified from D. oppostita Thunb. tv. Dung-Gun-Ma showing mosaic symptom on their leaves. YMV-K strains were filamentous particles of 780nm in length and induced cytoplasmic disorder such as inclusion body formation. Nucleotide and amino acid sequences of 5'-UTR, Pl and CP of YMV-K strains shared 80.8, 64.7 and 98.3% identity respectively to JYMV J1 in the mean value. Purification of YMV-K strains according to JYMV purification method(S. Fuji) was conducted to product antiserum. With antiserum against YMV-K strains, the Various diagnosis methods such as IC-RT-PCR, DIBA, RIPA and indirect-ELISA were used to detect YMV-K strains in Chinese yam plant.

Effects of the Extract Yam Powder Addition on Yogurt Properties during Storage

  • Physicochemical and sensory properties of Dioscorea opposita Thunb. (yam) powder-added yogurts (1, 3, 5, and 7%, w/v) were examined when stored at $4^{\circ}C$ for 16 d. When the yam concentration increased, the pH increased with 16 d storage. The lactic acid bacteria counts in all yogurt samples showed an increase during storage period, and the highest counts were appeared with 7% yam powder addition. The dramatic decrease was found in viscosity with higher concentrations of yam powder-added yogurt samples. No change was found in allantoin and diosgenin contents for 16 d storage periods. A significantly strong bitterness and astringency and decreased viscosity were found with 5 and 7% yam powder addition. However, less than 3% yam powder-added yogurt samples did not show a significant difference compared with the control (p>0.05) and overall acceptability score was the highest in 3% yam powder-added yogurt throughout the storage. The present study indicated that the concentrations (1 and 3%, w/v) of yam powder could be used to produce yam powder-added yogurt without significant adverse effects on physicochemical, microbial and sensory properties, and enhance functional components from the supplementation.

생마 저온부패 원인세균의 분리 및 부패균의 특성 (Isolation and Characterization of Yam-Putrefactive Psychrotrophic Bacteria from Rotted Yam)

  • 본 연구는 부패 생마로부터 저온성 부패균을 분리, 동정하고 분리균의 생육특성과 다양한 효소의 부패관련성을 조사하여, 생마 저온 장기저장 및 생마 가공식품 개발을 위한 기초자료를 제시할 목적으로 수행되었다. 저온 장기저장중의 부패생마로부터 서로 다른 13종의 저온세균을 분리, 동정하였으며, 다양한 온도에서 생마 절편을 이용한 부패력 측정 결과 분리균주 중 YAM-10및 YAM-12균주가 저온부패에 직접적으로 관련됨을 확인하였으며, 이들은 각각 Pseudomonas cepacia 및 Pseudomonas rhodesiae 동정되었다. 이들 균주는 20$^{\circ}C$에서도 우수한 amylase, CMCase, xylanase 활성을 나타내었으며 , 특히 amylase 활성은 생마 부패에 중요한 역할을 하는 것으로 판단되었다. YAM-10 및 YAM-12균주는 양파절편을 부패시키지는 못하였으며, 4$\sim$12$^{\circ}C$의 온도에서도 생육가능하며, pH 5 이하 및 pH 10 이상에서는 생육이 급격히 억제되었다. 현재 생마의 저온 장기저장 및 부패억제를 위한 저온성 Pseudomonas sp.의 제어와 생마부패에 관련되는 효소들의 특성과 저해에 대한 연구 및 저장성, 관능성이 강화된 산장, 염장 등을 통한 생마 가공식품 개발이 진행 중이다.
