Karyotypes were established in five Pulsatilla species from Korea : P. cernua, P. davurica, P. koreana, P. chinensis and P. tongkangensis. The somatic chromosome numbers of five species were all 2n=2x=16 with the basic number of x=8. The chromosome complement of P. cernua consisted of 5 pairs of metacentric, 1 pair of submetacentric and 2 pairs of subtelocentric. P. davurica, P. koreana and P. chinensis consisted of 5 pairs of metacentric and 3 pairs of subtelocentric. P. tongkangensis consisted of 5 pairs of metacentric, 2 pairs of submetacentric, and 1 pair of subtelocentric. Karyotype formulas of P. davurica, P. koreana, and P. chinensis were the same as K (2n) = 2x = 16 = 10m + 6st, while those of P. cernua was K (2n) = 2x = 16 = 10m + 2sm + 4st and P. tongkangensis was K (2n) = 2x = 16 = 10m + 4sm + 2st, respectively.
The mototic and meiotic chromosomal characteristics of ICR mice were investigated with G-and C-banding techniques. For the puposes, the chromosomal preparations were made with the modified air-drying method of Imal et al. Chromosomal analysis using testis could be observed mitotic as well as meitotic chromosomal behaviors, and the centromeric regions of all chromosomes including X chromosome were strongly stained in C-banded preparations. Nineteen autosomal bivalents and a single uniequal terminally associated X-Y bivalent in normal cells were observed during the late prophase and the metaphase of the meiosis I. The mean frequencies of previously dissociated X-Y chromosomes in the primary apermatocytes of the control group were 7.45%, but the frequencies of X-Y dissociation in the alkylating agents-treated group were about 3-4 times higher than that in the control group. Application of C-banding in meiotic stages could be certainly distinguish between vibalent type and univalents type of sex chromosomes.

The fragile X syndrome, which is considered to be synonymous with the Martin-Bell syndrome, is a relatively common form of X-linked mental retardation. The syndrome seems to occure in many different ethnic groups and its prevalence among mentally retarded males has been estimated to be in the order of 2 to 6%. The karyotypic hallmark of the syndrome is made up with a pronounced constriction near each tip of the long arm of the X chromosome(fragile site), shown in vitro only under conditions in which thymidylate production is blocked(lowered folate levels). Special culture media are needed to demonstrate this constriction site. Major clinical features associated with the syndrome include macroorchidism, large or prominent ears, significant emotional and behavioral dysfunctions such as hyperactivity, self-injury, lack of eye contact and social interaction, schizophrenia, autism, etc., and speech and language dysfunctions ranging from nonverbal to verbal speech with moderate to severe expressive language delays. Some have minor clinical features in common such as an increase in birth weight high forehead, prognathism, increased head circumference in infancy and childhood which did not persist into adult life. The recent research findings have shown that the fragile X syndrome is associated with infantile autism. Many patients with the fragile X syndrome fulfill the diagnostic criteria for infantile autism. Therefore it is recommendable that any patient with developmental delays and autism or autistic manifestations should have a chromosomal analysis, including fragile X examination. In the present review, historical aspects, incidence, and clinical features are presented. Recent anecdotal reports of the association with autism and the clinical improvement following high dose folic acid treatment will be discussed.
This study was carried out to confirm the chromosome constitution and homoeologous recombination of progenies derived from various cross combination using tetraploid OA interspecific hybrid originated from mitotic chromosome doubling. Based on the chromosome analysis of progenies crossed reciprocally, there were only triploid progenies when crossed with diploid Asiatics as male or female parent. While only tetraploid progenies were produced when crossed tetraploid Asiatics or tetraploid OA hybrid with tetraploid OA hybrid, respectively. However, two types of progenies, that is, diploid and triploid plants, were produced from cross combinations between diploid Oriental hybrid and tetraploid OA hybrid. From the GISH analysis of OA hybrid, it was confirmed that diploid $F_1$ OA hybrid was consisted of 24 chromosomes (12 Oriental and 12 Asiatics) showing authentic OA hybrid. On the other hand, it was notified that triploid plants (3x=36) were consisted of 24 Asiatics lily chromosomes and 12 Oriental lily chromosomes by analysis of backcross progenies derived from either $A{\times}OA$ or $OA{\times}A$ crosses. In cross between tetraploid OA and OA, all the progenies were tetraploid with equal number of chromosomes without any homoeologous recombination, i.e. each 24 chromosomes of Oriental and Asiatics. In 2x-4x ($O{\times}OA$) cross combination, some progenies had 2x=24 chromosomes originated from only Oriental hybrid, and other progenies had 3x=36 chromosomes derived from 24 chromosomes of Oriental hybrid and 12 chromosomes of Asiatic hybrid. Only tetraploid Asiatics chromosomes without any Oriental one were produced in all the progenies from 4x-4x ($AA{\times}OA$) cross combination.

Objective: X inactivation is the silencing one of the two X chromosomes in female mammals for gene dosage on the X-chromosome between female and male. X inactivation is controlled by X inactive-specific transcript (XIST) gene, untranslated RNA. XIST is expressed only from the inactive X (Xi), not expressed from the active X (Xa). The Xist promoter is methylated on the silent Xist allele on the Xa in somatic cells, and less methylated on the Xist-expressing Xi. We investigated the difference of XIST methylation pattern of the promoter and 5'-region of XIST from male (XY) and female (XX) subjects. Methods: The direct quantification of XIST methylation is required for clinical application of normal XX and XY blood. Methylation percentage of eight CpG sites (-1696, -1679, -1475, -1473, -1469, +947, +956, +971) of XIST gene were diagnosed by pyrosequencing. Results: We directly quantitated the methylation percentage of the promoter and 5'-end of XIST by pyrosequencing. The average methylation percentages at CpG6-8 sites (+947, +956, +971) were 45.2% at CpG6, 49.9% at CpG7, and 44.2% at CpG8 from normal female and normal male were 90.6%, 96.7%, 87.8%, respectively. Nether CpG 1-5sites (-1696, -1679, -1475, -1473, -1469) had any effect on XX and XY. Conclusion: This method is sensitive for quantifying the small percentage change in the methylation status of XIST, and may be used for diagnosis.
Somatic chromosome counts and karyotype analyses were carried out for eight taxa of Korean Allium sect. Sacculiferum. The basic chromosome number of sect. Sacculiferum was x = 8, and they could be cytologically divided into two groups, that is, a diploid group (2n = 2x = 16) containing A. thunbergii var. thunbergii, A. thunbergii var. deltoides, A. thunbergii var. teretifistulosum, A. deltoidefistulosum, A. longistylum, A. linearifolium and A. taqueti, and a tetraploid group (2n = 4x = 32) with only A. sacculiferum. All observed chromosomes were classified into metacentric, submetacentric and subtelocentric. The metacentric ones appeared in all treated taxa. One or two pairs of submetacentric chromosomes were observed in most taxa except A. sacculiferum, the unique taxon with subtelocentric chromosomes. All taxa had a pair of homologous chromosomes with satellites, and the B-chromosomes found in A. thunbergii var. thunbergii, A. deltoidefistulosum, A. sacculiferum and A. longistylum, were metacentric or telocentric. The karyotypes of A. longistylum and A. linearifolium were firstly investigated in this study. In conclusion, the somatic chromosome numbers and karyotypes for members of the sect. Sacculiferum were valuable characters in identifying taxa, investigating interspecific relationships and delimiting taxa. In addition, A. thunbergii var. teretifolium, an invalid name (homonym), was renamed as A. thunbergii var. teretifistulosum H. J. Choi & B. U. Oh.
The somatic chromosome numbers among the various taxonomic characters about 17 taxa in Korean Aster and its allied taxa were investigated to review accurate scientific name and taxonomic rank. The somatic chromosome numbers of the treated taxa were invariable in same taxa, but variable among different taxa. The treated taxa were divided into two types by basic chromosome numbers, one type was x=9, the other x=8 and x=9 type was subdivided by polyploidy. The somatic chromosome numbers of Aster altaicus var. uchiymae, A. hyatae, Kalimeris chejuensis were reported firstly in this study, and based upon somatic chromosome numbers and leaf morphology, the plants, idenified as Aster pinnatifidus in Korea was considered variant of Kalimeris incisa.
The purpose of this study was to analyze the interspecific relationships of Chenopodium album and its related taxa collected in Korea. The 18S-26S ribosomal DNA (45S rDNA) loci were detected directly on mitotic chromosomes by fluorescence in situ hybridization (FISH) and the chromosome numbers were examined using aceto-orcein methods. The chromosomal numbers of Chenopodium album var. album and C. album var. centrorubrum were 2n = 6x = 54, whereas for C. album var. stenophyllum, this number was 2n = 4x = 36. The basic chromosome number was x = 9. The biotin labeled 18S-26S rDNA probe exhibited eight yellow fluorescent signals on the metaphase chromosome of C. album var. album and var. centrorubrum respectively, while two yellow signals of C. album var. stenophyllum were noted. All of the signals on the chromosomes were located at the terminal regions. The chromosome number and FISH findings suggest that C. album var. centrorubrum is merged into var. album and that it is clearly distinguished from C. album var. stenophyllum.
The human Y chromosome is strictly paternally inherited and does not X-Y crossing over during male meiosis in most of its length. Although this region came to be known as the non-recombining region Y (NRY), it was renamed as male-specific region Y (MSY) due to abundant recombination. The MSY is a mosaic of heterochromatic sequences and three classes of euchromatic sequences: X-transposed, X-degenerated and ampliconic. The X-transposed sequences exhibit 99% identity to the X chromosomal sequences. The X-degenerate sequences are remnants of ancient autosomes from which the modem X and Y chromosomes evolved. Eight palindromes of the ampliconic comprise one-quarter of the euchromatic DNA of the male-specific region of the human Y chromosome. They contain many testis-specific genes and typically exhibit 99.97% intra-palindromic (arm-to-arm) sequence identity. The arms of these palindromes must have subsequently engaged in gene conversion, driving the pair arms to evolve it concert. Averages of approximately 600 nucleotides per newborn male have undergone Y-Y gene conversion, which has had an important role in the evolution of multi-copy testis gene families in the MSY.
OTF9-63 (OTF9) and P19S1O1A1 (P19) embryonal carcinoma (EC) cells were examined for their ability to produce the readivation of inactive X chromosomes from somatic cells. They were hybridized with various somatic cells and resulting HATr EC-somatic cell clones were analysed for their morphology, chromosomal replication pafterns and expression proffies of X-linked and distantiy located genes, Hprt and Pgk-1. The results demonstrated that 0RF9 cells could reactivate the inactive X chromosome whereas P19 cells could not. In adition, EC-somatic cell hybrids tended to reduce the number of sex chromosomes in long-term culture, resulting m 1:2 ratio of sex chromosomes to autosomes The use of EC cell hybrids provides an experimental system for studying the mechanism(s) of the X-reactivatio that is initiated and maintained from meiotic prophase of oogenesis to early embryogenesis.
