Synthesis of 4-Hydroxypiperidine Derivatives and Their Screening for Analgesic Activity

  • Six substituted phenacyl derivatives of 4-hydroxypiperidine were prepared and their structures were confirmed through spectroscopic techniques. These newly synthesized derivatives were also screened for analgesic activity by chemical and thermal methods. Only halogenated phenacyl derivatives demonstrated more or less protection against acetic acid induced writhing in mice where as rest of three derivatives were found inactive when screened by this chemical method. Similarly all the six derivatives were proved inactive by tail flick test.

조각자 에탄올 추출물 및 그 분획의 항염증 및 진통효능 (Anti-inflammatory and Antinociceptive Properties of the Ethanolic Extract of Gleditsia sinensis and its Sub-fractions)

  • Traditionally, the thorns of Gleditsia sinensis LAM. (GS) have been used for the treatment of various types of cancer and heart, skin, vascular and inflammatory diseases. However, there have been no reports on the antinociceptive or antiinflammatory properties of the thorn of GS. The present study was carried out to evaluate the anti-inflammatory and antinociceptive effects of the ethanolic extract of GS (EEGS) and its sub-fractions. The administration of EEGS (500 mg/kg) or its butanolic fraction (50 and 100 mg/kg) reduced the frequency of the acetic acid-induced writhing reflex in mice. In addition, the administration of the butanolic fraction of EEGS (50 and 100 mg/kg) prolonged the latency of reaction at the hot plate in mice. The butanolic fraction of EEGS also inhibited lipopolysaccharide-induced nitric oxide, prostaglandin $E_2$, and tumor necrosis factor-$\acute{a}$ production in the RAW 264.7 cell line. These results suggest that EEGS has anti-inflammatory and analgesic properties and is a potential therapeutic for inflammation and nociception.

Anti-nociceptive and Anti-inflammatory Properties of Ilex latifolia and its Active Component, 3,5-Di-caffeoyl Quinic Acid Methyl Ester

  • The present study was conducted to investigate anti-nociceptive and anti-inflammatory effects of the leaves of Ilex latifolia Thunb (I. latifolia) in in vivo and in vitro. Writhing responses induced by acetic acid and formalin- and thermal stimuli (tail flick and hot plate tests)-induced pain responses for nociception were evaluated in mice. I. latifolia (50 - 200 mg/kg, p.o.) and ibuprofen (100 mg/kg, p.o.), a positive non-steroidal anti-inflammatory drug (NSAID), inhibited the acetic acid-induced writhing response and the second phase response (peripheral inflammatory response) in the formalin test, but did not protect against thermal nociception and the first phase response (central response) in the formalin test. These results show that I. latifolia has a significant anti-nociceptive effect that appears to be peripheral, but not central. Additionally, I. latifolia (50 and $100{\mu}g/mL$) and 3,5-di-caffeoyl quinic acid methyl ester ($5{\mu}M$) isolated from I. latifolia as an active compound significantly inhibited LPS-induced NO production and mRNA expression of the pro-inflammatory mediators, iNOS and COX-2, and the pro-inflammatory cytokines, IL-6 and $IL-1{\beta}$, in RAW 264.7 macrophages. These results suggest that I. latifolia can produce antinociceptive effects peripherally, but not centrally, via anti-inflammatory activity and supports a possible use of I. latifolia to treat pain and inflammation.

  • Loxoprofen-Na (sodium 2-〔4-(2-oxocyclopentylmethyl)pheny)propionate dihydrate) is a potent analgesic drug. We developed loxoprofen-Na plasters to extend duration time of analgesic activity and to reduce side effect on gastrointestinal tract. Analgesic effect of Loxoprofen-Na plasters was investigated. Loxoprofen-Na plaster had good analgesic effect in rat paw pressure test, Tail-flick latency test and acetic acid-induced writhing test. Also, it had anti-inflammatory effect on carrageenan-induced rat hind paw edema. In pharmacokinetic study of Loxoprofen-Na, plasters dosage form showed that plasma drug concentration was prolonged up to 14 hours. So, we can conclude that loxoprofen-Na plasters, when applied on skin, will be a new type of drug for controlling the various local pain or inflammation.

  • Anti-inflammatory and analgesic activities and skin irritation of piroxicam patch were investigated. Piroxicam patch increased the pain threshold in rat hind paw inflamed by carrageenan and inhibited writhing induced by acetic acid in mice. Piroxicam patch also inhibited the carrageenan-induced edema in rat hind paw as well as the increased vascular permeability induced by histamine in rats. In adjuvant arthritis of rats, piroxicam patch showed anti-inflammatory effects. Skin irritation of piroxicam patch was tested in Newzealand White rabbits and evaluated by Primary Irritation Index of Draize. The results from skin irritation test showed that piroxicam patch seemed practically non-irritating. The result from the present study indicates that piroxicam may be useful without serious side effects as anti-inflammatory analgesics in this patch form.

Anti-inflammatory and Antinociceptive Effects of Methanol Extract from the Fomes fomentarius

  • As an attempt to search for bioactive natural products exerting antiinflammatory activity, we have evaluated the anti-inflammatory and antinociceptive activities of the methanol extract from the Fomes fomentarius (MEFF). MEFF (50, 100 mg/kg/day, p.o.) significantly reduced an acute paw edema induced by carrageenan in rats. When analgesic activity was measured by acetic acid-induced writhing test and hot plate test, MEFF showed a dose-dependent inhibition in animal models. In addition, MEFF potently inhibited the LPS-induced production of NO, $PGE_2$ and TNF-$\alpha$production of macrophages. (omitted)

Investigations of Pet Ether Extract of Caesalpinia Pulcherrima (L.) Swartz Leaves Extract on Analgesic, Anti-inflammatory, and Antipyretic Properties

  • The pet ether extract of Caesalpinia pulcherrima, leaves was studied for its antinociceptive, anti-inflammatory and antipyretic property. The extract at doses of 50 and 200 mg/kg, p.o., significantly (p<0.05) reduced the number of writhing induced by acetic acid and inhibited the late phase (20-30 min) in formalin test in mice. The extract failed to increase the pain threshold level in tail immersion test in mice. In carrageenan induced paw edema in rats and in acetic acid induced increase in vascular permeability test in mice, the extract (50-600 mg/kg, p.o.) failed to produce any significant activity. While in cotton pellet granuloma test, the extract at doses of 200 and 600 mg/kg (p.o.) significantly (p<0.05) reduced the granuloma formation and was comparable to reference drug, dexamethasone. In ethylpheylpropiolate ear edema test 0.5 mg and 1 mg/ear application of extract significantly (p<0.05) inhibited ear edema. In yeast induced hyperthermia in rats, the extract did not produce any reduction in temperature. The results suggest that the extract acts peripherally to produce analgesic action and anti-inflammatory activity through steroidal mechanism.

갈근 추출물에 의한 염증성 Cytokine 생성 억제 및 Prostaglandin E2 활성 저해에 관한 연구 (Inhibition of Inflammatory-cytokines Production and Prostaglandin E2 Activity by Puerariae Radix Extracts)

  • 갈근(Puerariae Radix)의 $70\%$ ethanol 추출물 중 ethyl acetate 분획물(EPR)은 염증성 cytokine을 처리한 마우스 대식세포 및 토기 연골조직세포에서 염증의 발현과 관련된 NO 생성 저해효과를 보였고, 관절조직의 주요 성분 중 하나인 proteoglycan의 분해 억제효과와 관절조직 분해효소인 MMP-9의 활성이 억제되었다. 또한, 통증유발물질인 프로스타글란딘의 유의성 있는 감소를 보여 통증억제 효과가 있음을 확인하였을 뿐만 아니라 초산 유발 진통 효과테스트인 동물 모델에서도 효과적으로 통증을 억제함을 확인하였다. 즉, 갈근의 $70\%$ ethanol 추출물 중 ethyl acetate 분획물(EPR)은 독성의 문제뿐만 아니라, 소염, 진통 효과 및 연골 조직세포의 분해를 억제하는 다양한 효과를 나타내는 장점을 지니고 있어 관절염 치료제의 훌륭한 후보약재가 될 것으로 기대된다.

팥배나무의 진통 효과 (Anti-nociceptive Effects of Sorbus alnifolia)

  • In this study, we evaluated the anti-nociceptive activities of Sorbus alnifolia. To investigate the anti-nociceptive properties of the methanolic extract of Sorbus alnifolia (MSA), we conducted several tests using various experimental mouse pain models. Herein, MSA significantly delayed the latency time and writhing motion in the hotplate test and acetic acid test, respectively. These result indicated that MSA has an ability to manage both peripheral and central nociception. We could further confirm the analgesic effects of MSA by performing formalin test. In combination test using naloxone, a non-selective opioid receptor antagonist, analgesic activity of MSA was partly antagonized by naloxone, but not completely, indicating that the MSA acts as a partial opioid receptor agonist. Out results suggest that the S. alnifolia may be possibly used as valuable anti-nociceptive agent.
