$\beta$-l, 3-glucans are well known to enhance the immune reactions, resulting in antitumor, antibacterial, antiviral, anticoagulatory and wound healing activities. $\beta$-1, 3-glucans have various activities depending on molecular weight, degree of branching, conformation, water-solubility and intermolecular association. However, the $\beta$-1, 3-glucans linked backbone structure is essential and $\beta$-D-glucopyranosyl units are required for immunopotentiating activities. In this study, we tested the immunophamacological activities of insoluble $\beta$-1, 3-glucan from Agrobacterium sp. R259 KCTC 10197BP and confirmed the following activities: (1) IFN-${\gamma}$ production in PBMCs in the presence or in the absence of PHA, LPS, IL-18, and IL-12; (2) the induction of various cytokines in the spleen and thymus; (3) the adjuvant effect on the antibody production; (4) the cytotoxic and antitumor effects on cell lines and ICR mice. These results strongly suggest that $\beta$-1, 3-glucan from Agrobacterium sp. R259 KCTC 10197BP possesses various immunopharmacologica1 activities.
The ripe fruit of Hass avocado contains one of the highest elvels of cytochrome P-450 protein found in the plant kingdom. To determine whether wounded roots of avocado contain P-450 protein, the roots of avocado were wounded by slicing, and then allowed to incubate in sealed plastic bags, in 0.4M mannitol, and in the solution to make protoplast preparation containing cellulysin and macerase during the specified times. The microsomal proteins were extracted from the samples, separated by SDS-PAGE, and then subjected to Western blot analysis using polyclonal antibodies which are generated against the CYP71A1 protein. wounded roots in sealed bags produced CYP71A1 within 6 hours after cutting, and those in 0.4M mannitol did not produce CYP71A1 even after 72 hours, but those in the protoplast preparation by cellulysin and macerase induced and produced CYP71A was induced in only 24 hours. These results indicate that CYP 71A1 plays a role for wound healing for root tissue o avocado, and would-inducible P-450 protein was not detected in the mannitol solution by preventing a synthesis of ethylene in a liquid state, but the softening of tissues by cellulysin and macerase to make protoplast preparation was involved in an activation of CYP 71A1 even in the liquid state.
본 연구에서는 권선형 유도전동기의 회전자회로에 스위치모드 컨버터를 적용시킨 새로운 슬립전력 회수시스템을 제안하고 이를 분석한다. 제안된 시스템의 해석을 통해 적용된 컨버터의 듀티비에 의해 속도가 제어될 수 있으며, 동시에 기존 시스템의 몇가지 특성을 개선할 수 있음을 보인다. 특히 종래 시스템에서 주된 단점으로 되어 있는 저역률 및 전원전류의 고조파성분을 크게 개선할 수 있을 뿐만 아니라 선형적인 속도조절도 가능하다. 제안된 시스템의 타당성을 보이기 위해 이론 및 실험 결과를 제시하고 이를 설명한다.
Switched Reluctance Motor has doubly salient poles in stator and rotor, windings are wound in just stator and no magnet or windings on the rotor. This configuration is robust mechanically and thermally. The inverter of SRM is more robust than that of induction or brushless DC(BLDC) motor, but still its drive is comparatively expensive for home appliance. To drive the conventional three or four-phase SRM, 6 to 8 power switches are required when asymmetric bridge inverter is employed. Generally, more than 50% of the cost for the SRM drive is allocated to power devices and gate drives. This paper proposed single phase SRM that have both radial and axial air gaps. The stator and rotor were stacked with two types of stampings that have different diameters. This configuration is very effective to increase align inductance(Lmax). The high value of Lmax increases the motor efficiency and power density. The proposed single phase SRM(Claw SRM) can be driven by only two power switches. To show the validity of the proposed idea, the analysis using finite element method(FEM) and experimental works are carried out. The proposed SPSRM can be driven with high efficiency and can be made compactly and inexpensively because of high value of align inductance and less number of switches. For the comparison, we used same stator for three-phase and single phase, and slightly different stator and rotor for proposed single phase SRM(Claw SRM)
Special anesthetic considerations were required for children with acute or chronic liver disease. We experienced a case of dental treatment to control infection under general anesthesia in the 2-year-old girl with liver failure. She was also scheduled for liver tansplantation. Her preanesthetic results of liver function test, electrolytes, and coagulation panel were unstable and out of normal ranges. Uneventful anesthetic induction using isoflurane and atracurium and nasotracheal intubation were carried out. General anesthesia was maintained with isoflurane for 2 hours. Oozing from multiple extraction sites was sustained, so the transfusion of platelet concentration 1 units, fresh frozen plasma 1 unit, and packed red blood cell 1 unit was done. She was recovered without complication but was transferred to pediatric intensive care unit for wound care with her endotracheal tube kept. She was transferred to a ward without noticeable complications next day. So we report this successful case of anesthetic management for dental treatment in a child with liver failure.
Mouse thymosin β-4 (TB4) 유전자를 Escherichia coli에서 intein-tag 융합 단백질로 발현시키고 정제하였다. 재조합 TB4-intein 융합 단백질의 분자량을 10% glycine SDS-PAGE로 확인한 결과, 세포내 soluble 분획에서 60 kDa의 단백질을 얻을 수 있었고, 유도발현 최적 조건은 0.1 mM IPTG, 25℃에서 3시간 동안 유도 발현할 때가 최적임을 확인하였다. TB4 만을 얻기 위해서 유도발현된 TB4-intein을 DTT를 이용해 self-cleavage를 일으킨 다음, chitin bead를 이용한 친화성 크로마토그라피로 정제 후 분자량을 확인 한 결과, 5 kDa으로 확인되었으며, 순도는 95%이상 이었다. 정제한 recombinant TB4가 생물학적 기능을 보유하고 있는지 확인을 하기 위하여, TB4를 농도 별(1~1,000 ng/ml)로 하여 HT1080 cell을 이용한 cell migration을 측정한 결과, 모든 농도에서 recombinant TB4가 화학합성한 TB4 보다 약 20% 이상 높은 활성을 보였으며, recombinant TB4 1 ng/ml의 농도에서 cell migration 활성이 가장 높게 나타났다. Recombinant TB4를 마우스 상처 부위에 5일 동안 매일 처리한 결과(최종 처리 농도 0.5 mg/ml), 화학합성 TB4 보다 recombinant TB4의 상처치유 활성이 약 35% 더 높음을 알 수 있었다. 이상의 결과는 recombinant TB4가 화학합성 TB4보다 cell migration과 상처치유에 훨씬 높은 활성을 나타냄을 보여주고 있다.

Background: Incisional site of surgical operation become transient ischemic state and then occur reoxygenation due to vasodilatation by inflammatory reaction, the productive reactive oxygen species (ROS) give rise to many physiologic results. Apoptosis have major role on elimination of inflammatory cell and formation of granulation tissue in normal wound healing process. Remifentanil can prevent the inflammatory response and can suppress inducible nitric oxide synthase expression in a septic mouse model. After cardiopulmonary bypass for coronary artery surgery, remifentanil can also inhibit the release of biomarkers of myocardial damage. Here we investigated whether remifentanil pretreatment has cellular protective effect against hypoxia-reoxygenation in HaCaT human keratinocytes, if so, the role of apoptosis and autophagy on this phenomenon. Methods: The HaCaT human keratinocytes were exposed to various concentrations of remifentanil (0.01, 0.05, 0.1, 0.5 and 1 ng/ml) for 2 h before hypoxia (RPC/HR group). These cells were cultured under 1% oxygen tension for 24h at $37^{\circ}C$. After hypoxia, to simulate reoxygenation and recovery, the cells were reoxygenated for 12 h at $37^{\circ}C$. 3-MA/RPC/HR group was treated 3-methyladenine (3-MA), autophagy inhibitor for 1h before remifentanil treatment. Cell viability was measured using a quantitative colorimetric assay with thiazolyl blue tetrazoliumbromide (MTT, amresco), showing the mitochondrial activity of living cells. To investigate whether the occurrence of autophagy and apoptosis, we used fluorescence microscopy and Western blot analysis. Results: The viability against hypoxia-reoxygenation injury in remifentanil preconditioning keratinocytes were increased, and these cells were showed stimulated expression of autophagy 3-MA suppressed the induction of autophagy effectively and the protective effects on apoptosis. Atg5, Beclin-1, LC3-II and p62 were elevated in RPC/HR group. But they were decreased when autophagy was suppressed by 3-MA. Conclusions: Remifentanil preconditioning showed the protective effect in human keratinocytes, and we concluded that autophagy may take the major role in the recovery of wound from hypoxia-reoxygenation injury. We suggest that further research is needed about the cell protective effects of autophagy.
본 연구는 자연 상태로는 뿔이 발생하지 않는 암사슴에서 인공적으로 뿔 발생을 유도하기 위하여 동물의 조직을 괴사시키는데 사용되는 CaCl2 용액을 뿔 발생 예정부위인 좌우 양측 두개 골막에 주사하여 뿔 발생 여부 및 CaCl2 농도와 주사량이 뿔 발생 유도에 미치는 영향을 구명하고자 하였다. 공시축은 엘크 암사슴 5두이다. 엘크 암사슴에 15%용액 2ml 및 3ml를 처리한 시험구에서는 육경과 뿔 발생이 모두 유도되지 않았으며, 15%용액 4ml 처리구와, 30%용액 1.5ml, 2ml, 3ml구 및 50%용액 1ml, 2ml 구에서는 육경과 뿔 발생이 모두 유도되었다. 뿔 생산량은 30%용액 1.5ml 처리구에서 1,290g로 가장 많았으며, 주사량이 증가할수록 뿔 생산량이 줄어드는 경향을 보였다. 발생한 가지수는 15%용액 4ml 처리구는 2개, 30%용액 1.5ml, 2ml 및 3ml 처리구는 각각 5개, 2.3개 및 1개였으며, 50%용액 1ml 및 2ml 처리구는 각각 3개 및 1개로 농도 및 주사량과 가지수는 관련이 없었다. 오히려 초기 가지수의 발생은 주사 횟수 및 주사방법의 차이에 의한 상처부위의 모양에 따라 결정되는 경향을 보였다. 이상의 결과에서 30% CaCl2 용액이 엘크 암사슴의 육경 및 뿔 발생과 그 생산량 및 가지수에 가장 적합한 농도였다.
Purpose: Adipose tissue-derived stem cells(ADSC) has an osteoconductive potential and demineralized bone matrix(DBM) is an osteoinductive material. A combination of DBM and ADSC wound probably create osteoinductive properties. The purpose of this study is to determine the effect of the combination of DBM and ADSC mixture on healing of rat calvarial defect. Methods: Thirty adult male Sprague-Dawley rats were randomized into 3 groups(n=10) as 1) Control, 2) DBM alone, 3) DBM with ADSC mixture. DBM with ADSC mixture group has had a 3-day preculture of ADSC from groin fat pad. An 6 mm critical size circular calvarial defect was made in each rat. Defect was implanted with DBM alone or DBM with ADSC mixture. Control defect was left unfilled. 6 and 12 weeks after the implantation, the rats were sacrificed and the defects were evaluated by histomorphometric and radiographical studies. Results: Histomorphometric analysis revealed that DBM with ADSC mixture group showed significantly higher bone formation than DBM alone group(p<0.05). Although radiographs from DBM alone group and DBM with ADSC group revealed similar diffuse radiopaque spots dispersed throughout the defect. Densitometric analysis of calvarial defect revealed DBM with ADSC mixture group significantly higher bone formation than DBM alone(p<0.05). There was correlation of densitometry with new bone formation(Spearman's correlation of coefficient=0.804, 6 weeks, 0.802, 12 weeks). Conclusion: The DBM with ADSC mixture group showed the best healing response and the osteoinductive properties of DBM were accelerated with ADSC mixture. It will be clinically applicable that DBM and ADSC mixture in plastic and reconstructive surgery, such as alveolar cleft and congenital facial deformities that bone graft should be required.
양막은 다양한 생체재료로 이용되어 왔으며 생체적합성과 환부의 치료효능이 우수한 것으로 알려져 있다. 본 연구에서는 양막을 갈아 양막스폰지를 제조하고 중간엽줄기세포를 배양한 뒤 누드마우스이식을 통하여 콜라젠스폰지와 비교하여 골형성유도능에 대한 연구를 실시하였다. 연구결과 세포의 부착과 증식면에서는 두 시료가 유사하였으나, 체내에 이식한 결과 양막스포지군에서 좀 더 많은 콜라젠 분비와 칼슘이 침착되었음을 확인하였다. 그리고 면역화학염색 결과 골형성 시 필요한 오스테오칼신과 오스테오넥틴이 좀 더 발현된 것을 관찰할 수 있었다. 따라서 양막은 골형성유도를 향상시킬 수 있는 가능성이 있음을 알 수 있었다.
