겨울철 양식 능성어의 바이러스성뇌망막증(VER) 감염사례 (A Case Study of Mortality Caused by Viral Encephalopathy and Retinopathy (VER) in Cultured Sevenband Grouper, Epinephelus septemfasciatus during Winter)

  • 2015년 겨울, 우리나라 남해안 소재 육상 양식장에서 사육 중이던 능성어의 약 30%가 폐사하였다. 수온 $9{\sim}12^{\circ}C$의 겨울철에 발생한 능성어 폐사는 신경괴사성바이러스 (NNV) 감염에 의한 바이러스성뇌망망증 (VER)으로 밝혀졌다. 시험어는 비정상적인 유영, 배를 위로한 채 수면을 떠 다니는 현상, 체색흑화, 안구백탁 및 부레의 과도한 팽창 소견을 보였다. 원인 바이러스는 일반적으로 고수온기 능성어에 감염되는 RGNNV genotype이 아닌, 냉수성 어류에서 분리되는 BFNNV genotype으로 밝혀졌으며, NNV 가장 근연관계가 높은 Pacific cod betanodavirus (PCNNV)와 99.7~100% 상동성을 보였다. 병리조직학적 관찰 결과, 바이러스성뇌망망증 (VER)의 특징적인 병변인 뇌조직의 공포화 및 괴사를 수반하였다. 이 연구는 수온 $12^{\circ}C$ 이하의 겨울철에 능성어에 감염된 BFNNV genotype에 대한 첫 보고이다.

춘천의 가정에서 미세분진 ($PM_{2.5}$)과 입자상 다환방향족탄화수소에 대한 계절적 노출 변동 (Seasonal Variations of Human Exposure to Residential Fine Particles ($PM_{2.5}$) and Particle-Associated Polycyclic Aromatic Hydrocarbons in Chuncheon)

  • Lately human exposure to fine particles smaller than $2.5{\mu}m$ in aerodynamic diameter ($PM_{2.5}$) has become a great concern in Korea due to their possible cause of elevated mortality, lung function decrements, and more frequent hospital admissions for asthma. This study was conducted to investigate seasonal variations of human exposure to residential $PM_{2.5}$ and particle-associated polycyclic aromatic hydrocarbons (PAHs). Ten homes in Chuncheon, Korea were visited for continuous 72 hour sampling of $PM_{2.5}$ in the living rooms using a MiniVol Portable Sampler from December 22, 2002 to November 3, 2003. During the same period, outdoor $PM_{2.5}$ samples were collected on the top of the Natural Sciences Building of Kangwon National University which is located in the middle of the ten households. Samples were analyzed for $PM_{2.5}$ mass concentrations and six selected PAHs. In two smoking homes, the highest $PM_{2.5}$ concentrations were measured ranging from 51.1 to 69.7 {\mu}g/m^3$ on average in all seasons, indicating smoking is a very important contributor to the elevation of indoor particle concentrations. Seasonal comparison showed that indoor particle concentrations were higher than outdoor ones except winter. Total PAH concentrations in smoking homes were highest in winter among the seasons primarily due to low ventilation rate, followed by the outdoor site and nonsmoking homes. BaP toxic equivalents (TEQs) were calculated for five PAHs. The TEQ for smoking homes in winter was highest followed by the outdoor site in winter. It is concluded that smoking and ventilation rate are two important contributors to the elevation of indoor $PM_{2.5}$ and PAH concentrations.

우리나라 고속도로에서 야생동물 로드킬에 관한 시공간 추이 분석 (Spatial and Temporal Patterns on Wildlife Road-kills on Highway in Korea)

  • The negative impacts of roads on wildlife mortality have been well documented, and one of the most significant impact is wildlife-vehicle collisions (WVCs) in most countries throughout the world. While road impacts on wildlife are a truly global concern with a large socio-economic cost, few researches in Korea have been quantified road-kill occurrence on highways or identified extensively seasonal and geographic patterns of this phenomenon. Therefore, we analyzed highway mortality of wild mammals in Korea using database from five years of nation-wide data on WVCs, and estimated road-kill density by standardizing on per km and per $10^3$ vehicle basis. During 2008 through 2012, a total of 10,940 wildlifes were reported killed on highways, with an average of 2,188 cases per year. There were 2,376 road-kills in 2012, and this equates to 0.01 road-kills per km per week or one road-kill every 88.5 km per week. For time of day, road-kills occurred more frequently in the early morning (05:00-08:00, 38.3%), and day of week did not have a significant influence in any individual year. The road-kill was highest in the spring (March- May, 33.0%) and least in the winter (December-February, 16.1%), and the most frequently killed native species were of Korean water deer (79.7%), raccoon dog (12.7%), Korean hare (3.1%), and leopard cat (1.2%). The overall standardized kill-rate (number/10 km/1,000 vehicles/month) in 2012 was 0.057 with highest on Dangjinyeongdeok highway (0.476), followed by Yeongdong (0.274), Sooncheonwanju (0.233), Iksanpohang (0.187), and Joongang (0.150). This study highlights that the frequency of WVCs are prevalent throughout the highways in Korea. Further work is needed to determine whether such a level of mortality is sustainable from an ecological point of view.

Effects of Water Temperature Change on the Hematological Responses and Plasma Cortisol Levels in Growing of Red Spotted Grouper, Epinephelus akaara

  • This study was conducted to determine the stress response [ethological (operculum movement number (OMN)), hematological (hematocrit and hemoglobin), biochemical (glucose, cortisol and glutamic oxaloacetic transaminase (GOT))] in red spotted grouper, Epinephelus akaara during exposure of different water temperature in winter season. This species (Total length, $18.56{\pm}0.34cm$) previously maintained in water temperature of $15^{\circ}C$ were transferred to 15, 20 and $25^{\circ}C$. During experimental period (7 days), OMN, hematocrit (Ht), glucose and GOT values were significantly high in $15^{\circ}C$ when compared to 20 and $25^{\circ}C$. Hemoglobin value was also increased at $15^{\circ}C$, but no significant differences. There was no differences in cortisol levels among the temperature groups. No fish mortality was observed during the experimental period. From these results, $15^{\circ}C$ is likely more stressful to red spotted grouper than $20^{\circ}C$ and $25^{\circ}C$. These observations confirm that red spotted grouper adapts better to temperatures between 20 and $25^{\circ}C$ during the winter season.

대규모 교란현상 후 형성된 대형갈조류 감태(Ecklonia cava) 개체군의 계절적 변동 및 회복 양상 (Recovery Pattern and Seasonal Dynamics of Kelp Species, Ecklonia cava Population Formed Following the Large-scale Disturbance)

  • 대규모 교란현상 이후 형성된 대형갈조류 감태 개체군의 계절적 변동과 회복양상을 이해하기 위해서 감태의 형태학적 특성, 가입, 사망률, 밀도와 생물량을 2013년 6월부터 2015년 6월까지 조사하였다. 감태의 전체 길이, 가장 긴 측엽의 길이와 개체당 무게는 뚜렷한 계절적 경향성을 보였다. 줄기부의 길이는 겨울부터 봄까지 증가하였으나, 여름부터 가을까지는 변화가 없었다. 이러한 결과는 감태의 형태적 변화가 주로 엽상부의 변화에 의해 나타난다는 것을 의미한다. 감태의 성장은 수온이 $15{\sim}18^{\circ}C$인 겨울부터 봄까지 매우 활발하게 일어나지만, $20^{\circ}C$ 이상의 수온에서는 저해되는 것으로 나타났다. 가입은 실험이 진행되는 기간 동안 봄부터 여름에 걸쳐 매우 낮았다. 그러나 2015년 4월 생육밀도의 감소로 인해 감태의 피도가 매우 낮아진 시점에 대량의 가입이 발생하였으며, 이것은 감태의 가입이 계절적 요인 보다는 공간과 차광효과 같은 물리적 요인에 의해 조절된다는 것을 시사한다. 어린 개체는 부착기질의 불안정성으로 인해 매우 높은 사망률을 보였다. 태풍 '볼라벤'에 의한 교란 이후 34개월이 되는 2015년 6월에 이르러 개체군의 구조가 교란 이전 수준으로 회복되었다. 따라서 대규모 교란현상 이후 감태 개체군이 교란 이전의 개체군 구조로 회복되는 데에는 3년 이상의 시간이 소요되는 것으로 판단된다. 이 연구는 해중림을 구성하는 대형갈조류의 관리, 복원 및 보호 전략을 수립하는데 매우 귀중한 생태학적 자료를 제공할 것이다.

토양에서 분리한 곤충병원성세균 Bacillus thuringiensis 의 혹명나방과 벼애나방에 대한 실내살충효과검정 및 생물학적 특성에 미치는 영향 (Pesticidal Activities and Effect on Its Biological Characteristics of Bacillus thuringiensis Strains from Soil against Rice Pests, Cnaphalocrosis medinalis and Maranga aenescens)

  • 벼에 발생하여 잎을 가해하여 피해를 주는 혹명나방(Cnaphalocrosis medinalis)과 벼애나방(Naranga aenescens)의 곤충병원성미생물을 이용한 친환경적 방제를 위해, 국내토양에서 분리된 Bacillus thuringiensis 균주들을 대상으로 생물검정을 수행하였다. 실험에 이용한 Bt 53균주 중, 약 18개 균주가 혹명나방에 90% 이상의 살충률을 나타내었고, 13개 균주가 벼애나방에 살충활성을 보였는데, 두 종 모두에 90% 이상의 살충률을 보이는 균주도 CAB141균주를 비롯해 11균주나 확인되었다. 이러한 균주들은 혹명나방에 대한 실내방제실험결과, 96%이상의 방제효과를 나타내기도 하였다. 또한 Bt 포자현탁액에 노출된 유충의 용화율, 용길이, 우화율이 감소되는 경향이 나타났는데, 혹명나방의 경우, 번데기 길이에서도 정상개체와 처리 후 생존개체가 현저한 차이를 나타내었다.

Experimental infection of white spot syndrome virus (WSSV) in wild crab, Gaetice depressus

  • To confirm the possibility of a wild marine crab, Gaetice depressus, as a carrier for white spot syndrome virus (WSSV) and to develop an alternative experimental model for WSSV in winter season, the susceptibility of the crab to WSSV was assessed by artificial challenge and subsequently tested for infection by PCR assay. The results revealed that the crabs were as highly susceptible as penaeid shrimps. WSSV caused 100% mortality in G. depressus within 16 days after intramuscular injection. The presence of WSSV in the moribund crabs was confirmed by PCR and was found in gills and muscle tissue. These results suggest that G. depressus can be naturally infected by WSSV via moribund shrimps, and can act as a potential carrier of WSSV. In addition, G. depressus can be used as an alternative experimental animal for WSSV.

  • Mycoplasmal pneumonia of swine(MPS) cause by Mycoplasma hyopneumoniae has been recognized as a serious impediment to swine production due to chronic respiratory disorder which result in the weight loss and decreased feed conversion. The disease causes a great economic losses in pig industry by characterizing with high morbidity, low mortality, growth retardation and low feed efficiency. The present study was conducted to investigate the titers of antibody against M hyopneumoniae from the regional and seasonal groups of the slaughtered pigs by enzyme-linked immunosorbent assay(ELISA). The result have shown that the average seropositive rate of M hyopneumoniae infection was 84.6% . The regional seropositive rate in Korea showed 87.4% in Kyonggj, 83.4n in Kangwon, 89.2% in Chungnam and 77.6% in Chungbuk area, respectively. Also the seasonal seropositive rate was appeared as 78.6% in spring,90.1% in summer, 76.9% in autumn and 83.8% in winter, respectively.

Serous Fat Atrophy of a Reticulated Giraffe (Giraffa camelopardalis)

  • A female reticulated giraffe (Giraffa camelopardalis), 20-month-old, 342 kg, died at Seoul Zoo on January 2, 2009 after a stressful episode of chronic diarrhea. Given the appearances postmortem, it was strongly suspected that the giraffe suffered from malnutrition for a long time. Typical appearances of serous fat atrophy were shown on most fat tissues of body organs such as heart, bone, liver, mesentery and kidney. In this study, the sudden death that had been known as "peracute mortality syndrome" was clearly identified to have resulted from a lack of understanding the Browser's diet and general failure in giraffe husbandry. Individualized care and high quality hay must be provided to compensate higher consumption of metabolic energy and to prevent animal loss in winter season.
