  • The aim of this study was to investigate the protective effects of selenium and its combination with ${\beta}$-carotene treatments prior to whole-body irradiation in mice.   • In an attempt to better understand the effect of whole body irradiation on the spontaneous motility and oxygen consumption rate of the isolated mouse duodenum, a whole body X-irradiation of 1,000r. was given to albino mouse, and 1) the total length of contraction of isolated duodenum was recorded on kymograph every five minutes for 60 minutes, 2) glucose and 5-hydroxytryptamine(5-HT) were added to the reaction medium of Kreb's-Ringer-bicarbonate buffer(KRB) and response of the isolated duodenum to the drugs was observed, and 3) the oxygen consumption rate $(QO_2)$ of the isolated duodenum as well as the effect of glucose and 5-HT on $QO_2$ were measured by Warburg's standard manometric method and the comparison was made with the control(i.e. normal) group. The results thus obtained are summarized as follows. 1. The spontaneous motility of the isolated duodenum in the irradiated groups showed a significantly elevated pattern for the first 15 minutes comparing with the control. The motility, however, decreased after 15 minutes and remained so in the irradiated groups to the level of the nonirradiated control, but 24 hours post-irradiation group showed a tendency of an increased motility while one hour post-irradiation group showed no difference comparing with the control. 2. Addition of glucose produced generally elevated motility of the isolated duodenum in both irradiated and non-irradiated groups comparing with the control throughout the experiment, but no difference was observed in contractile amplitude between the irradiated and non·irradiated groups. 3. When 5-HT was added to the irradiated group, the contractile amplitude of isolated duodenum was similar to that of the control, and 5-HT alone caused a slight increase of the motility comparing with the control. 4. The oxygen consumption rate $(QO_2)$ of the isolated duodenum was found to be ,slightly increased in one hour post·irradiated group, but similar in 24 hour post·irradiated group comparing with the control. Glucose produced a significant increase of $QO_2$ in all the groups, but 5-HT produced a tendency of decrease of $QO_2$ in all the groups.

Effects of Reduced Glutathione and Ginseng Extract on Non-Protein Sulfhydryl, and Non-Protein Disulfide of Mouse Liver and Blood Following Whole Body X-Irradiation (Reduced glutathione 및 인삼추출액(人蔘抽出液)이 X-선전신조사(線全身照射)를 입은 마우스 간조직(肝組織) 및 혈중(血中) NP-SH 및 NP-SS에 미치는 영향(影響))

  • In an attempt to better understand the radioprotective effect of reduced glutathione(GSH), and to observe a possible radioprotective effect of Ginseng extract, whole body X-irradiation of 1,200 r was administered to the mouse either independently or immediately following the injection of GSH or Ginseng extract to the mouse intraperitoneally. The non-protein sulfhydryl (NP-SH) and non-protein disulfide (NP-SS) levels of the liver, and NP-SH level of NP-SH of the blood of the mouse were measured at 30, 60 and 120 minutes, and results were compared with the normal. The results thus obtained are summarized as follows; 1) The normal values of NP-SH and NP-SS of the mouse liver were $5.90{\pm}0.46\;{\mu}\;mol/gm\;wet\;wt.,\;and\;3.02{\pm}0.42\;{\mu}\;mol/ml$ wet wt., respectively, and the normal value of NP-SH of NP-SH of the mouse blood was $3.98{\pm}1.29\;{\mu}\;mol/ml$ 2) The injection of both GSH and Ginseng extract produced the highest values of NP-SH in the liver at 30 minutes, but a gradual decrease to the normal was observed thereafter. When X-irradiation alone was applied, the liver NP-SH value was lower than the normal at 60 minutes post-irradiation and thereafter. When Ginseng extract was injected immediately prior to X-irradiation, the liver NP-SH was lower than the normal throughout the experiment with the lowest value at 60 minutes. However, the combination of GSH and X-irradiation produced higher than the normal values throughout the entire experiment. 3) The liver NP-SS value was most significantly elevated at 30 minutes after the injection of GSH, hut the recovery to the normal was observed thereafter. The injection of Ginseng extract produced slightly higher liver NP-SS values at 30 and 60 minutes, but the value at 120 minutes was similar to the normal. The single application of X-irradiation resulted in the lower then normal liver NP-SS values throughout the entire experiment. When GSH was injected price to X-irradiation, the liver NP-SS values were higher than the normal at 30 and 60 minutes followed b the recovery to the normal at 120 minutes. The combination of Ginseng extract and X-irradiation showed generally lower liver NP-SS values throughout the experiment. 4) The blood NP-SH showed the higher than the normal values in all the experimental groups except when GSH was injected prior to X-irradiation alone produced e significantly elevated blood NP-SS value at 30 minutes post-irradiation.

  • Purpose: The present study was to investigate the effect of music therapy on anxiety, nausea, and vomiting in patients undergoing total body irradiation (TBI) for hematopoietic stem cell transplantation (HSCT). Method: A untreated control group with pretest and posttest was used in this study. A total of 35 patients receiving TBI for HSCT were recruited from a medical center at a university in Korea. The music therapy taking for 15-20 minutes per a time was performed twice a day for three days while the patients in the experimental group were undergoing the TBI. Results: The scores of state anxiety, nausea and vomiting tended to increase in the control group as compare with those in the experimental group. The patients in the control group had been administerd antiemetics more often than those in the experimental group. The levels of systolic and diastolic blood pressure and the pulse rates in the experimental group tended to decrease after the experiment. However, there was no statistically significant differences in anxiety, nausea, vomiting, blood pressure, and pulse between the groups. Conclusion: It is suggested to repeat this study with a enough sample size.

Effect of Whole-body gamma-Irradiation on Blood Picture in Mice (Cobalt-60 gamma선(線) 전신조사(全身照射)가 생쥐의 혈액상(血液像)에 미치는 영향(影響))

  • In order to investigate the effect of cobalt-60 gamma irradiation on the blood picture, the mice were subjected to single whole-body gamma irradiation externally. In this experiment 42 of 12 weeks old white mice were used and animals were allotted to group of three. On of chose groups served as control and the others as test groups that were exposed to single dose of 370 Rads (group I) and 500 Rads (group II). The exposure dose rate were averaged 33 Rads per Minute. The results obtained in this experiment were as follows: 1. Erythrocyte counts in both test groups dropped to the lowest level at 4 weeks postirradiation, returning towards the control level, but in recovery, group I showed rather prumpt compared with group II. Erythrocyte count in group II was significantly lower than that of group I. 2. Total leukocyte counts dropped to the lowest loved at 2 weeks postirradiation and group I recovered to normal level at 4 weeks postirradiation. Recovery group II was very slow and it reached to the control level on 12 weeks postirradiation. Leukocyte count was significantly different between group I and II. 3. Neutrophil count was increased to the highest level at 2 weeks postirradiation, with recovery to normal value after 8 weeks in group I and 12 weeks in group II, respectively. Neutrophil count was significantly different between group I and II. 4. Lymphocyte count dropped to the lowest level at 2 weeks postirradiation, with recovery to normal average after 8 weeks in group I and 12 weeks in group II, respectively. Lymphocyte count was significantly different between group I and II. 5. The other leukocyte count was increased after the irradiation and recovered at 12 weeks postirradiation in both test groups. The other leukocyte count was significantly different between group I and II.

Radioprotective Effect of Propolis on the Blood Corpuscle of a Mouse by SEM after X-irradiation on the Whole Body (전신 X-선 조사 후 마우스의 대동맥 혈구에서 SEM을 이용한 프로폴리스의 방사선방어효과 연구)

  • After x-ray 5Gy radiation on the whole body of a mouse using a linear accelerator, its leucocyte, erythrocyte, and platelet were observed by SEM. Also, after injecting propolis into the abdominal cavity, the radio-protective effect of blood corpuscles was studied. The observation of micromorphology in blood corpuscles revealed that the number of leukocyte, erythrocyte, and thrombocyte decreased in the experimental group and the lump got together in blood corpuscles after 10 and 20 days. In RBC, crack or break on the surface and poikilosperocytes were observed. In the irradiation group, the size of leucocytes was smaller than that in control group and the number of villus at the verge substantially decreased. The blood corpuscles in the propolis group, however, had the similar results to control group.

Research about Hyperspectral Imaging System for Pre-Clinical testing of Small Animal (소형동물 전임상실험을 위한 하이퍼스펙트럼 영상장비 연구)

  • In this study we have developed a hyperspectrum imaging system for highly sensitive and effective imaging analysis. An optical setup was designed using acoustic optical tunable filter (AOTF) for high sensitive hyperspectrum imaging. Light emitted by mercury lamp gets split in to diffracted and undiffracted beams while passing though AOTF. GFP transfected HEK-293 cell line was used as a model for in vitro imaging analysis. Cells were first, analyzed by fluorescence microscope followed by flow cytometric analysis. Flow cytometric analysis showed 66.31% transfection yield in GFP transfected HEK-293 cells. Various images of GFP transfected HEK-293 cell were grabbed by collecting the diffracted light using a CCD over a dynamic range of frequency of 129-171 MHz with an interval of 3 MHz. Subsequently, for in vivo image analysis of GFP transfected cells in mouse, a whole-body-imaging system was constructed. The blue light of 488 nm wavelength was obtained from a Xenon arc lamp using an appropriate filter and transmitted through an optical cable to a ring illuminator. To check the efficacy of the newly developed whole-body-imaging system, a comparative imaging analysis was performed on a normal mouse in presence and absence of Xenon arc irradiation. The developed hyperspectrum imaging analysis with AOTF showed the highest intensity of green fluorescent protein at 153 MHz of frequency and 494 nm of wavelength. However, the fluorescence intensity remained same as that of the background below 138 MHz (475 nm) and above 162 MHz (532 nm). The mouse images captured using the constructed whole-body-imaging system appeared monochromatic in absence of Xenon arc irradiation and blue when irradiated with Xenon arc lamp. Nevertheless, in either case mouse images appeared clearly.

Differential Functions of Caffeine and Ascorbic Acid in $\gamma-Irradiated$ Male Mice

  • Radioprotection is of practical importance for the normal tissues of tumor patients subject to radiotherapy, people with planned or accidental exposure to radiation, and the public and radiation workers. Since oxygen enhances radiation - induced biological damage, antioxidants should be related with the function as a radioprotectors. Ascorbic acid or caffeine is an essential component and antioxidant in the diet of humans and a small range of other mammals. The present study investigates functional radioprotection of caffeine and ascorbic acid against gamma radiation in irradiated C57BL/6N mice. Eight-week-old male C57BL/6N mice were irradiated with 6.5 Gy. A caffeine treated group was administered with $80mg\;kg^{-1}$ body weight by intraperitoneal injection, a single treatment 1 hr before irradiation. Ascorbic acid was administered $330\;mg\;L^{-1}$ in drinking water through all the experimental period. According to time schedules, animals were sacrificed by cervical dislocation. And the samples were collected 2 weeks after whole- body gamma irradiation. The caffeine treated group showed lower decrement of body and organ weights than the other experimental groups. The qualitative analysis of circulating testosterone in serum was performed by means of radioimmunoassay (RIA). The normal level of circulating testosterone was maintained by the treatment of caffeine and ascorbic acid. The change of weight of body and organ and the appearance of seminiferous tubules were improved by an effect of caffeine or ascorbic acid against irradiation. Taken together, caffeine and ascorbic acid protects impairment of spermatogenesis against gamma radiation and may act as a radio-protector.

Analysis of the Spatial Dose Rates According to the Type of Radiation Source Used in Multi-bed Hospital Room (다인병실에서 이용되는 방사선원의 종류에 따른 공간선량률 분석)

  • Medical radiation offers significant benefits in diagnosing and treating patients, but it also generates unnecessary radiation exposure to those nearby. Accordingly, the objective of the present study was to analyze spatial dose rate according to types of radiation source term in multi-bed hospital rooms occupied by patients and general public. MCNPX was used for geometric simulation of multi-bed hospital rooms and radiation source terms, while the radiation source terms were established as whole body bone scan patients and imaging using a portable X-ray generator. The results of simulation on whole body bone scan patients showed $3.46{\mu}Sv/hr$ to another patient position, while experimental results on imaging using a portable X-ray generator showed $1.47{\times}10^{-8}{\mu}Sv/irradiation$ to another patient position in chest imaging and $2.97{\times}10^{-8}{\mu}Sv/irradiation$ to another patient position in abdomen imaging. Multi-bed hospital room, unnecessary radiation generated in the surrounding patients, while legal regulations and systematic measures are needed for radiation exposure in multi-bed hospital rooms that are currently lacking in Korea.
