Can discolored dental composites be bleached in depth?

  • Objectives: Previous in vitro studies determined the whitening effects of bleaching products on stained resin composite surfaces. This in vitro study aimed to verify the effectiveness of a whitening system on composite resin previously subjected to pigmentation, specifically examining the depth of whitening effectiveness within the material structure. Materials and Methods: A commercially available nano-filled composite resin was used. Specimens were stained using a coffee-based solution and a 10% carbamide peroxide-based gel was employed as the whitening agent. The pigment's penetration and the effect of the bleaching gel were evaluated by measuring color (CieLab values) from the outer edge to the inner part of the specimens. Color measurements were taken at 14 points, starting from 0.1 mm from the external perimeter up to 3.0 mm. Results: Analysis of variance tests showed a statistically significant difference between the Control Group (CG), Pigmentation Group, and Whitening Group. The whitening agent was effective up to 1.5 mm in depth, with Whiteness index (W) values not statistically different from those of CG up to 0.5 mm in depth. Conclusions: Whitening agents on nano-filled resin composite previously pigmented appear effective in restoring the W to values similar to the original, particularly in the superficial layers of the sample.

노회(蘆薈)(알로에), 자화지정(紫花地丁)의 항산화, 항염증, 주름, 미백에 미치는 영향 (Effects of Aloe and Violae herba Extract on the Anti-oxidant, Anti-inflammatory, Anti-wrinkle and Whitening)

  • Objective : This study was performed to assess the effects of Aloe and Violae herba extracts on skin disease and skin beauty. Methods : Anti-oxidant effects were measured by the scavenging for DPPH radical, xanthine oxidase activity. Anti-inflammatory effects were examined by relations in NO synthesis, IL-$1{\beta}$, IL-6, TNF-$\alpha$, NF-kB, COX-2, MAP kinase. The skin wrinkle formation effects were measured by collagenase and elastase activities. The whitening effects were examined by tyrosinase activities, melanin synthesis in MNT-1 cell. Results : 1. In an anti-oxidant test, Aloe and Violae herba extracts showed high radical scavenging activity. 2. In an anti-inflammatory test, Aloe and Violae herba extracts strongly inhibited the lipopolysaccharide(LPS)-induced nitric oxide(NO) release from the RAW 246.7 macrophage cells. Aloe and Violae herba extracts also inhibited the LPS-induced IL-$1{\beta}$ and COX-2 expressions. The inhibitory effects of Aloe and Violae herba extracts on macrophage activation were via the inhibition of NF-kB, evidenced by transient transfection assay. Furthermore, Aloe and Violae herba extracts weakly inhibited the activation of Jun-N-terminal kinase(JNK) but they did not have any effects on p38 MAP kinase in RAW 264.7 cells. 3. In the skin wrinkle formation assay, Aloe extract strongly inhibited collagenase and elastase, whose activity are tightly related with the wrinkle formation. 4. In the skin whitening assay, Aloe and Viloae herba extracts weakly inhibited tyrosinase activity, however, it was not statistically significant. Besides they did not have any effects on melanin synthesis, indicating that they could not be applicable for skin whitening. Conclusion : This study show that Aloe and Violae herba extracts may play a significant role in skin disease and skin beauty.

당귀와 구기자 에탄올 추출물의 피부 미백작용 (Skin Whitening Effects of Extracts from Angelicae Gigantis Radix and Lycii fructus Ethanol Extracts)

  • Objective : The purpose of this study was to investigate the synergistic effect of Angelicae gigantis Radix (AG) and Lycii fructus (LF) ethanol extracts on skin-whitening effects. Method : LFAG extracts were prepared by extracting with 80% ethanol. The efficacy of LFAG was judged by measurement of cell viability, tyrosinase activity, melanin production, tyrosinase and microphthalmia-associated transcription factor (MITF) expression in B16F10 murine melanoma cells by lipopolysaccharides (LPS) treatment. Results : Each extract (LF or AG) inhibited the tyrosinase activity in a dose-dependent manner. The co-treatment of LFAG extracts ($25{\mu}g/mL$ LF plus $25{\mu}g/mL$ AG) markedly suppressed the LPS-induced cellular tyrosine activity, melanin production, tyrosinase and MMP-1 expression in B16F10 murine melanoma cells. These suppressive effects were synergistically increased by their combination. Conclusions : With these observations, we suggest that the extracts from Lycii fructus and Angelica gigantis Radix could be potent natural materials for whitening skin.

Physiological and Whitening Effects of Morus alba Extracts

  • Mulberry extracts can be incorporated into skin-whitening products. The compound attributed to lighten the skin is arbutin, a form of hydroquinone that inhibits melanin release by suppressing the tyrosinase enzyme. For the cosmetic applications, the physiological effects of mulberry (Morus alba) extracts were investigated. The water soluble fraction of mulberry contains higher amount of protein (16.28~4.47%) in contrast to fat (1.55~1.41%). In addition, the fraction abundantly contains succinic acid (972.4-275.8 mg/g) and phosphoric acid (1,628.4-121.9 mg/g) in different parts of mulberry. The free radical scavenging ability in water soluble fraction was found to display remarkable effects in comparison with methanol and ethyl acetate fraction. The ethyl acetate-soluble of root and leaf showed remarkable tyrosinase inhibition activity by IC 50 (${\mu}g/ml$). The anticancer activity of methanol fraction obtained from mulberry using human cancer cell lines showed growth inhibition effect (270.14 mg/ml in Calu-6 cells, 295.29 mg/ml in HCT-116, and 332.29 mg/ml in MCF-7 cells, respectively). Based on the results, Morus alba extracts include cosmetic ingredients with antioxidizing and whitening properties.

A mixture of seaweed extracts and glycosaminoglycans from sea squirts inhibits α-MSH-induced melanogenesis in B16F10 melanoma cells

  • Background: In the present study, the skin-whitening effects of a marine-sourced mixture that includes a fucoidanrich extract of Undaria pinnatifida (UPEF), a phlorotannin-rich extract of Ecklonia cava (ECE), and glycosaminoglycans (GAGs) from sea squirt skin were investigated. Methods: The whitening effects of the mixture and its components were evaluated by measuring the inhibition of mushroom tyrosinase and melanin synthesis in alpha-melanocyte-stimulating hormone (${\alpha}$-MSH)-stimulated B16F10 melanoma cells. Results: Each component alone markedly inhibited mushroom tyrosinase in a dose-dependent manner, and in ${\alpha}$-MSH-stimulated B16F10 cells, they inhibited melanin synthesis and were cytotoxic. However, the whitening effects of UPEF, ECE, and GAGs in combination were greater than those of each component alone. A mixture in the ratio of 4:5:1 (UEG-451) showed the strongest activity without cytotoxicity. Further study suggested that UEG-451 inhibits ${\alpha}$-MSH-stimulated melanogenesis in B16F10 cells by downregulating tyrosinase and tyrosinase-related proteins, such as TRP-1 and TRP-2, via the inhibition of MITF expression. Conclusions: These results suggest that mixing the different components at optimum ratios might be an effective way to improve their bioactivities and reduce toxicity and that UEG-451 possesses strong whitening effects that could be used in the cosmetic industry.

단풍잎돼지풀 발효 추출물의 항산화 효과 및 B16F10 세포에서의 미백 활성 검증 (Verification of the Antioxidant Effects and Whitening Activity of fermented Ambrosia trifida L. Extracts in B16F10 Cells)

  • 본 연구는 단풍잎돼지풀 발효 추출물의 항산화 및 미백 효과를 검증하여 화장품 소재로서 활용가능성을 확인하고자 하였다. 항산화 효과를 측정하기 위해 전자공여능 측정과 ABTS 라디칼 소거능을 측정하였다. 단풍잎돼지풀 발효 추출물의 전자공여능 측정 결과 1,000 ㎍/ml 농도에서 68.4%의 효과를 보였으며, ABTS 라디칼 소거능 측정 결과 같은 농도에서 58.7%의 효과를 나타내었다. 미백효과를 Tyrosinase의 효소 억제 활성에 의해 측정한 결과, 1,000 ㎍/ml 농도에서 32.4%의 저해활성 효과를 보였다. 세포 차원에서 미백효과를 측정하기 위해 단풍잎돼지풀 발효추출물의 세포 생존율을 멜라노마 세포에서 측정하였다. 그 결과, 100 ㎍/ml 농도에서 85.2%의 생존율을 보였으며, 독성을 보이지 않는 농도인 100 ㎍/ml 농도 이하에서 western blot을 진행하였다. 단풍잎돼지풀 발효 추출물의 단백질 발현억제 효과를 25, 50, 100 ㎍/ml의 농도에서 western blot으로 측정하였으며, 양성대조군으로 β-actin을 사용하였다. 그 결과, 100 ㎍/ml 농도에서 MITF, TRP-1, TRP-2, Tyrosinase인자들은 각각 51.14%, 55.4%, 38.6%, 83.77%의 효과를 나타내었다. 결론적으로 단풍잎돼지풀 발효 추출물의 항산화 및 미백효과가 검증되었으며, 화장품 천연물 소재로서 활용가능성을 확인하였다.

Melanin 생성 인자 억제 효과를 통한 Mangifera indica L. Peel의 미백효과 연구 (A Study on the Whitening Effect of Mangifera indica L. Peel Extracts through Inhibition of Melanin Synthesis Factor)

  • 본 연구의 목적은 애플망고 껍질의 열수 및 70% 에탄올 추출물의 미백 효과를 검증하고자 하였다. 애플망고 껍질의 열수 및 에탄올 추출물의 미백 효과를 측정하기 위해 tyrosinase 저해 활성을 측정한 결과, 최종 농도인 1,000 ㎍/ml에서 열수 추출물은 9%, 에탄올 추출물은 35%의 저해 효과를 보였다. 세포를 통해 미백효과를 측정하고자 멜라노마세포인 B16-F10을 이용해 세포 생존율을 MTT assay를 사용하여 측정하였다. 세포 생존율 측정 결과, 100 ㎍/ml 농도에서 각각 95.64%, 103.36%의 세포 생존율을 나타내었다. 이후 실험은 세포 생존율이 95% 이상 나타난 농도인 100 ㎍/ml 이하의 농도에서 실험을 수행하였다. 미백 효과는 멜라닌 합성에 관여하는 인자의 단백질 및 mRNA 발현을 측정하여 결정하였다. 단백질 발현은 western blot을 이용하여 측정하였으며, 그 결과 MITF, tyrosinase, TRP-1 및 TRP-2에 대한 단백질 발현은 100 ㎍/ml 농도에서 열수 추출물에 의해 59%, 65%, 26%, 18% 감소하였고, 에탄올 추출물에 의해 64%, 40%, 18%, 52% 감소하였다. MITF, Tyrosinase, TRP-1 및 TRP-2의 mRNA 발현은 RT-PCR을 통해 확인하였으며, 그 결과 100 ㎍/ml에서 열수 추출물에 의해 27%, 44%, 40%, 22% 감소하였고, 에탄올 추출물에 의해 9%, 51%, 11%, 52% 감소하였다. 따라서 애플망고 껍질 추출물이 미백 효과가 있음을 확인하였고, 천연물 소재로서의 이용가치가 높을 것으로 사료되어진다.

산약의 멜라노마 세포(B16F10)에서 MITF, TRP-1, TRP-2, Tyrosinase, PKA, ERK 발현 억제 효과 (Inhibitory Efficacy of Dioscoreae Rhizoma on MITF, TRP-1, TRP-2, Tyrosinase, PKA and ERK Expression in Melanoma Cells (B16F10))

  • Objectives : The purpose of this study was to research the whitening effects and developing by cosmetics of the extract fromDioscoreae Rhizoma, which is one of the most popular health-promoting herb in herbal medications.Methods : We performed tyrosinase inhibition assay, reverse transcription-polymerase chain reaction (RT-PCR) and western blot for whitening effects. Also we measured MTT assay for cell viability.Results : The results were obtained as follows : For whitening effect, tyrosinase inhibition rate of extract fromDioscoreae Rhizomashowed more than 42.28% at 1,000 ㎍/㎖ concentration. Cell toxicity effect on melanoma cells (B16F10) of extract fromDioscoreae Rhizomashowed 81.97% with toxicity at 50 ㎍/㎖ concentration. So we were measured at a concentrations of 5, 10 and 50 ㎍/㎖ in all experiments involving cell. In addition, whitening related mRNAs including microphthalmia associated transcription factor (MITF), tyrosinase related protein-1 (TRP-1), tyrosinase related protein-2 (TRP-2), tyrosinase were reduced byDioscoreae Rhizoma. We also foundDioscoreae Rhizomatransiently decreased protein kinase A (PKA) which is known to be upstream to the down regulation of MITF and tyrosinase. But phosphorylation of extracellular signal related kinase (pERK) were increased byDioscoreae Rhizoma. These results imply thatDioscoreae Rhizomadecrease melanogenesis via ERK activation and subsequent down regulation of MITF and tyrosinase.Conclusions : Therefore, all these findings suggested the potent usage ofDioscoreae Rhizomaas materials of functional cosmetics by confirming whitening activity related with melanin content.

Comparison of Coffee Stain Removal Effects of Commercial Whitening Toothpaste in Sound and Demineralized Teeth In Vitro

  • Background: The size of the tooth whitening market and toothpaste market is increasing worldwide. The purpose of this in vitro study is to confirm and compare the coffee stain removal effects of commercial whitening toothpaste in sound and demineralized teeth, respectively. Methods: A total of 112 flat permanent bovine teeth specimens were manufactured. Half of the surface of the specimen was coated with an acid-resistant varnish and deposited in an artificial demineralizing solution for 65 hours. The varnish applied to half of the specimen was removed and deposited in a coffee solution for 96 hours to induce coloring. Two control and five experimental group toothpastes for teeth whitening were selected and the main components were investigated. Toothbrushing was performed 50, 100, and 150 times for each toothpaste group. A total of four images were obtained: before the start and after 50, 100, and 150 times of brushing to obtain the lightness (L*) values of the sound and the demineralized tooth surfaces. The difference in the average value between toothpaste groups at each treatment period was analyzed by one-way ANOVA. The difference in the L* average value according to the number of the brushing was analyzed by repeated measure ANOVA. Results: All toothpastes in the seven groups contained abrasive agents and had different ingredients for each product. Compared to before brushing, the L* value changed significantly in all toothpaste groups after brushing 50 times (p<0.05). This was common in both the sound and demineralized teeth surfaces. Demineralized teeth had significantly lower L* values at all brushing times than that in sound teeth (p<0.05). Conclusion: The effect of whitening teeth was different for each toothpaste. Demineralized teeth were more likely to cause coloration than sound teeth, and the coloration was not removed well.

루틴 대사체의 미백 효능 (Potent whitening effects of rutin metabolites)

  • 본 연구는 rutin 및 rutin 대사체인 DHPAA, HPAA, DHT, HVA가 버섯 유래 tyrosinase 활성과 마우스 피부표피 세포주인 JB6 P+ 세포의 증식에 미치는 영향에 대해 살펴보았다. HVA가 버섯유래 티로시나제 저해활성이 가장 우수한 것으로 관찰되었으며, DHPAA, HPAA, rutin, DHT 순서로 저해효능을 나타내었다. DHT를 제외한 rutin 및 rutin 대사체들 모두 $100{\mu}M$ 까지 세포 독성이 관찰되지 않은 것으로 관찰되었으며, DHT는 50 또는 $100{\mu}M$에서 세포증식을 저해하였으나 그 이하의 농도에서는 세포독성을 유도하지 않았다. 따라서 본 연구결과, rutin 대사체 모두 세포 독성이 없는 수준에서 버섯 유래 티로시나제 활성을 효과적으로 억제하였으므로, 앞으로 미백제제로 활용될 수 있을 것으로 기대된다.
