A Study on the Anti-wrinkle Properties of Cosmetics Containing Essential Oil from Chrysanthemum boreale MAKINO (산국 에센셜오일을 함유한 화장품의 주름개선 임상효능에 관한 연구)

  • We recently reported the characteristics of Chrysanthemum boreale MAKINO essential oil (CBMEO) at different harvesting stages and its in vitro antioxidant, whitening, skin regeneration, and antiwrinkle properties. In the present study, we investigated the effectiveness of cosmetic creams containing 0.1% of CBMEO in terms of wrinkle reduction and safety. The participants were 22 women aged 42 to 60 y with natural wrinkles. All the participants applied the CBMEO-containing cream to the eye area for 8 weeks. Visual observations of the skin by a dermatologist were used to assess the safety of the product, and improvements in skin wrinkles (roughness) were analyzed using a skin visiometer. Neither the CBMEO-containing cream nor the placebo caused abnormal reactions, such as erythema or allergies, during the test period. The CBMEO-containing cream-exposed test regions showed a greater reduction in the roughness index as compared with those of the placebo-exposed control regions. The R1, R2, and R3 indices showed a statistically significant decrease in the test regions as compared with the placebo control regions after using the cream for 2 weeks (p<0.05). Therefore, this study demonstrated that CBMEO-containing cosmetics are safe for human use and that they have skin wrinkle improvement properties. In the future, CBMEO may be a promising product for use in the development of functional anti-wrinkle cosmetics.

Studies on skin whitening efficacy and skin permeation using O/W Nanoemulsion system with Resorcinol Dipentyl Ether (레조시놀다이펜틸에터를 함유한 O/W 나노에멀젼의 경피흡수 및 미백 효능 효과 연구)

  • In this study, we investigated the effect on the increase of In vitro skin permeation experiments and In-vivo skin whitening efficacy using a O/W nanoemulsion produced via PIC(Phase Inversion Composition) with 1,3-di(pentyloxyl)benzene. skin permeation experiments of RS-nanoemulsion formulated with selected condition was evaluated compared to mineral oil containing 1,3-di(pentyloxyl)benzene and normal O/W type RS-emulsion. Compared to mineral oil with 1,3-di(pentyloxyl)benzene and RS-emulsion. RS-Nanoemulsion has a statistically significant high percutaneous absorption in terms of index substance, which is 1,3-di(pentyloxyl)benzene. In vivo test were prepared in the system of O/W cream containing RS-nanoemulsion. There was no adverse reactions in both samples. After 8 weeks, the subjects was evaluated by a dermatologist's scoring and Chromameter. In conclusion, the testing product showed statistically improvement (p<0.05) compared to the controlled product and proved its whitening efficacy.

The Study on the Whitening Effect of Ephedra sinica Extract (마황추출물의 미백효과에 관한 연구)

  • In this study, we investigated the application of an extract from Ephedra sinica which has been composed of traditional Korean medicine as a whitening ingredient. The extract of Ephedra sinica which was obtained from the mixture of methanol and water (1:1) the inhibitory effect of tyrosinase. Then, Ephedra sinica was extracted by two different solvents. One was water and the other was methylene chloride. Only, the water extract of Ephedra sinica showed the inhibitory effort of tyrosinase; the anti-tyrosinase activity with $0.2\%$ of the water extract was $60.6\%$. But the extract of Ephedra sinica in methylene chloride fraction showed little inhibitory effect on tyrosinase. The inhibitory effect of the concentrated water extract of Ephedra sinica was tested on L-DOPA auto-oxidation and melanin synthesis in B-16 melanoma. In L-DOPA auto-oxidation, $0.5\%$ of the concentrated water extract showed $87\%$ of inhibition of L-DOPA auto-oxidation and the $0.75\%$ concentrated Ephedra sinica extract in wafer fraction inhibited $98.8\%$ of that. In melanin synthesis of B-16 melanoma, the concentrated water effect of Ephedra sinica inhibited $70.2\%$ or $79.9\%$ of inhibitory effect on that at the concentration of $0.05\%$ or $0.075\%$, respectively. For verifying the skin whitening effect of the concentrated water extract of Ephedra sinica in vivo, we performed the clinical test of that. The research showed the significant skin whitening effect of a cream containing $0.5\%$ Ephedra sinica extract and the statistical analysis showed a significant difference (p < 0.05) between sample (containing $0.5\%$ Ephedra sinica extract) and placebo after 10 weeks.

Korean Red Ginseng extract ameliorates melanogenesis in humans and induces antiphotoaging effects in ultraviolet B-irradiated hairless mice

  • Background: Panax ginseng is a marvelous herbal remedy for all ailments of body. That may be why it is called Panax, which means "cure for all". Melanin is a pigment that gives color to our skin; however, increased melanin production can lead to tumor formation. Human exposure to ultraviolet B radiation has increased extensively owing to the increased sunlight due to global warming. Consequently, a phenomenon called photoaging has been observed for all skin colors and types. As a result of this phenomenon, a set of enzymes called matrix metalloproteinases, which serve as degradation enzymes for extracellular matrix proteins, mainly collagen, is increased, causing depletion of collagen and resulting in early wrinkle formation. Methods: Therefore, in our study, we used the murine melanoma cell line B16/F10 to study the inhibition of melanogenesis by Korean Red Ginseng (KRG) extract in vitro and HRM-2 hairless mice exposed to artificial ultraviolet B to examine the efficacy of KRG in vivo. We prepared a 3% red ginseng extract cream and evaluated its effects on human skin. Results: Our results demonstrated that KRG induced potent suppression of tyrosinase activity and melanin production in B16/F10 cells; moreover, it reduced the transcription and translation of components involved in the melanin production pathway. In the in vivo experiments, KRG potently suppressed the expression of matrix metalloproteinases, reduced wrinkle formation, and inhibited collagen degradation. On human skin, ginseng cream increased skin resilience and skin moisture and enhanced skin tone. Conclusion: Therefore, we conclude that KRG is an excellent skin whitening and antiaging product.

The Processed Radish Extract Melanogenesis in Humans and Induces Anti-Photoaging Effects in Ultraviolet B-Induced Hairless Mouse Model

  • The radish skin and radish greens are an edible part of the radish. But they are removed before eating the radish and used as a byproduct or an animal feed material because of their tough and rough texture. Melanin is a pigment that gives colour to our skin. But increased production of melanin can turn into benign or malignant tumours. These days due to global warming, the amount of Ultra violet (UVB) rays has been extensively increased with sunlight. Due to this, a phenomenon called exogenous photo aging is widely observed for all skin colour and types. As a result of this phenomenon, a set of enzymes called matrix metalloproteinases (MMP's) that serves as degradation enzymes for extracellular matrix proteins mainly collagen is increased, causing depletion in collagen and resulting in early wrinkles formation. Therefore in our study we used the murine melanoma cell line B16/F10 to study the melanogenesis inhibition by Heated radish extract (HRE) in vitro and we used HRM-2 hair less mice exposed to artificial UVB for checking the efficacy of Heated radish extract in vivo. Furthermore, we prepared a 3% Heated radish extract (HRE) cream and checked its effects on human skin. Our results have clearly demonstrated that Heated radish extract (HRE) have potently suppressed the tyrosinase activity and melanin production in B16/F10 cells. It had also reduced the expression of components involved in melanin production pathway both transcriptionally and transitionally. In in vivo studies, HRE had potently suppressed the expression of MMP's and reduced the wrinkle formation and inhibited collagen degradation. Moreover, on human skin, ginseng cream increased the resilience, skin moisture and enhanced the skin tone. Therefore in light of these findings, we conclude that HRE is an excellent skin whitening and antiaging product.

Hypopigmentary Effect of Milk Thistle Extract Silymarin (엉겅퀴 추출물 실리마린의 피부 미백효과)

  • We found that silymarin exhibited the inhibitory effect on melanogenesis in a spontaneously immortalized mouse melanocyte cell line, Mel-Ab. Silymarin is a standardized extract obtained from the dried seeds of milk thistle (Silybum marianum Gaertn.). Silymarin significantly prevented melanin production in a dose-dependent manner with an $IC_{50}$ value of 28.2 ${\mu}g/mL$ without effects on cell viability. Also, silymarin inhibited tyrosinase activity in melanocyte, while it did not affect the catalytic activity of cell-free tyrosinase. Furthermore, Western blot analysis indicated that silymarin decreased the expression of tyrosinase protein. Silybin A/B and isosilybin A/B were also able to inhibit melanin production and tyrosinase expression in protein level. Double blind study on the clinical efficacy of a cream containing 2 % silymarin showed that silymarin have a significant skin whitening effect. Therefore, this study suggests that silymarin may be useful as a natural skin whitening agent.

Use and Satisfaction on Cosmetics of Middle- and Old-Aged Women with Well-being Pursuit Disposition (중.노년 여성의 웰빙 추구 성향에 따른 화장품 사용 및 만족도)

  • The purpose of this study is to analyze the use and satisfaction on cosmetics of middle- and old-aged women with well-being pursuit disposition. A survey was conducted from January 20 to February 10 in 2006. The statistical analysis methods used to analyze the 393 middle- and old- aged female subjects were frequency analysis, reliability analysis, factor analysis, cluster analysis, crosstab, and multiple regression analysis. First, middle- and old-aged women were using functional cosmetics for wrinkle and whitening as well as skin lotion, moisture lotion and nutritive cream. They applied basic skin make-up only or partial make-up on the eyes and lips. Second, the well-being pursuit disposition factors were determined to be eco-friendly pursuit disposition, economical pursuit disposition, and health pursuit disposition. The well-being pursuit disposition factors had an effect on satisfaction of basic cosmetics. While the economical pursuit disposition factor had an effect on satisfaction of colored cosmetics. Third, the clusters from well-being pursuit disposition were determined to be wellbeing unconcerned group, eco-friendly concerned group, economical concerned group, and health concerned group. The proportion of subjects applying basic and colored cosmetics was high in health concerned group.

A study on the development of hydrocortisone certified reference material in cosmetic cream using isotope dilution-mass spectrometry (동위원소희석-질량분석법을 이용한 화장품 크림 중 히드로코르티손 인증표준물질 개발 연구)

  • Steroids have a temporary skin improvement and whitening effect by controlling vasodilation, but they also cause side effects when used for a long time. Therefore, steroids were designated and managed as raw materials that cannot be used in cosmetics in Korea. However, steroids are continuously being detected in cosmetics, causing social issues. In this study, we developed a certified reference material (CRM) for the determination of steroids such as hydrocortisone in cosmetics. A cream-type cosmetic CRM was manufactured and subsequently certified following the guidelines outlined in ISO Guide 35. Homogeneity, short-term stability, and long-term stability were evaluated using isotope dilution mass spectrometry (ID-MS). The certified values were determined by using NIST's primary reference material to ensure traceability. From now on, we intend to supply the certified reference material as a cosmetic CRM to national and international companies, as well as research institutes after certification as certified reference material from KOLAS and registering on COMAR.

Evaluation of the Potential of Cellobiose as a Material for Whitening Cosmetics based on Autophagy and Melanin Production Efficacy in Melanocytes (셀로비오스의 미백화장품 소재 가능성 평가를 위한 멜라닌 세포에서 자가포식 및 멜라닌 생성 효능 연구)

  • Cellobiose is a dissacharide constituted by two glucose units joined by a β-('1,4') glycosidic bond that is produced by the decomposition of cellulose. This product exists naturally in plants and has been utilized in different industries as a food sweetener, and as a cosmetic and pharmaceutical material. In this study, the potential of cellobiose as a whitening cosmetic product was evaluated by analyzing autophagy induction and the inhibition of melanin production. A cytotoxicity test conducted in the human melanin-producing cell line MNT-1 with increasing concentrations of cellobiose revealed that this compound did not cause cytotoxicity at 20 mg/mL or less. Based on this, autophagy was firstly evaluated by immunostaining with the autophagy marker microtubule-associated protein 1 light chain 3 (LC3) after treatment with 20 mg/mL of cellobiose. The subsequent confocal microscopy analysis revealed an increase in LC3 puncta, indicating induction of autophagy. In addition, autophagy was further confirmed by western blot analysis, which demonstrated that cellobiose converted LC3-I to LC3- ∏ in a concentration- and time-dependent manners. An analysis of melanin contents after cellobiose treatment at a concentration of 20 mg/mL during 7 days revealed that melanin production was reduced by more than 50%. Additionally, the expression levels of melanogenesis-related proteins TYR and TYRP1 were markedly decreased after cellobiose treatment. Based on these studies, a cosmetic cream formulation containing cellobiose was prepared and the change in formulation was tested for 4 weeks, and it was confirmed that the appearance changed to liquid form at high temperature, but the pH did not change. In conclusion, the present research demonstrated that cellobiose activates autophagy and inhibits melanin production, and showed the potential of this product as a material for whitening cosmetics.

Study on the N-Acetyl-D-glucosamine as the Anti-aging Cosmetic Ingredients (항노화 화장품 원료로서의 N-Acetyl-D-glucosamine에 관한 연구)

  • In this study we applied the NAG obtained by the deacetylation of chitin extracted from the shells of crabs and shrimp as cosmetic ingredient. In order to compare NAG with GLC we identified the influence of cytotoxicity, anti-inflammatory, melanin biosynthesis formation inhibition on skin cell, and we measured the effects of the change of melanin and red spots. The results show that there was not any attentive cytotoxicity on the Raw 264.7 cell and B16F10 cell, and NO formation anti-inflammatory hindrance effect induced from Raw 264.7 by LPS was slight, and NAG suppressed the increase of melanin generation concentration-dependently after we induced the melanin generation with ${\alpha}$-MSH on B16F10 and measured the melanin biosynthesis inhibition. From this result, we identified the applicability of the cosmetics containing NAG as functional cosmetic for enhancing skin-lightening because when cream containing NAG was applied to skin the index of melanin red spots showed statistically meaningful changes.
