Objectives: Despite a rise of an interest in tooth whitening, diverse problems are being caused in case of hydrogen peroxide that is being used as a tooth bleaching agent. Thus, the aim was to examine tooth whitening effectiveness using natural products as a plan for supplementing this. Methods: As a result of having measured a tooth color through using VITA Easyshade V after having developed toothpaste with the application of extracts such as Citrus Peels, Mulberry (Morus alba L.) Root Bark, strawberry, and lemon, and then having used it for 10 weeks, they are as follows. Both upper and lower 6 anteriors mostly got brighter. Results: A statistically significant difference was shown especially in the right canine (p=0.015), in the right central incisor (p=0.007), and in the left central incisor (p<0.001). In consequence of having measured a color change, the tooth got brighter gradually in the higher extract content and in the lengthier use time. In case of canine, it got less bright compared to other teeth. In the outcome of evaluating sensuality, most of the questions were indicated to be high in case of using a whitening toothpaste for 10 weeks. But in what "there is no stickiness in the mouth, the stickiness was more felt in the use up to 5 weeks, but was improved in 10 weeks. Conclusions: The bleaching effectiveness was proved by developing a toothpaste with the application of natural extracts. A short-term effect cannot be seen like a whitening agent of using hydrogen peroxide. But there is a continuous effect in consideration of tooth-brushing more than 3 times a day.
Objectives : The purpose of this study was to research the whitening effects and developing by cosmetics of the extract fromDioscoreae Rhizoma, which is one of the most popular health-promoting herb in herbal medications. Methods : We performed tyrosinase inhibition assay, reverse transcription-polymerase chain reaction (RT-PCR) and western blot for whitening effects. Also we measured MTT assay for cell viability. Results : The results were obtained as follows : For whitening effect, tyrosinase inhibition rate of extract fromDioscoreae Rhizomashowed more than 42.28% at 1,000 ㎍/㎖ concentration. Cell toxicity effect on melanoma cells (B16F10) of extract fromDioscoreae Rhizomashowed 81.97% with toxicity at 50 ㎍/㎖ concentration. So we were measured at a concentrations of 5, 10 and 50 ㎍/㎖ in all experiments involving cell. In addition, whitening related mRNAs including microphthalmia associated transcription factor (MITF), tyrosinase related protein-1 (TRP-1), tyrosinase related protein-2 (TRP-2), tyrosinase were reduced byDioscoreae Rhizoma. We also foundDioscoreae Rhizomatransiently decreased protein kinase A (PKA) which is known to be upstream to the down regulation of MITF and tyrosinase. But phosphorylation of extracellular signal related kinase (pERK) were increased byDioscoreae Rhizoma. These results imply thatDioscoreae Rhizomadecrease melanogenesis via ERK activation and subsequent down regulation of MITF and tyrosinase. Conclusions : Therefore, all these findings suggested the potent usage ofDioscoreae Rhizomaas materials of functional cosmetics by confirming whitening activity related with melanin content.

This study was performed to assess the antioxidant activities and whitening effects of Agrimonia pilosa Ledeb on melanin synthesis. The whitening effects of Agrimonia pilosa Ledeb water extracts were examined by in vitro mushroom tyrosinase assay and B16BL6 melanoma cells. We assessed inhibitory effect of Agrimonia pilosa Ledeb water extract on expression of melanogenic enzyme proteins including tyrosinase, tyrosinase-related protein 1 (TRP-1) and tyrosinase-related protein 2 (TRP-2) in B16BL6 cells. Inhibitory effect of Agrimonia pilosa Ledeb onto free radical generation was determined by measuring DPPH and hydroxyl radical scavenging activitie. Our results indicated that Agrimonia pilosa Ledeb water extract effectively inhibited free radical generation. In DPPH and hydroxy radical scavenging activity, Agrimonia pilosa Ledeb water extract had a potent anti-oxidant activity in a dose-dependent manner. They significantly inhibited tyrosinase activity in vitro and in B16BL6 melanoma cells. Also, Agrimonia pilosa Ledeb suppressed the expression of tyrosinase in B16BL6 melanoma cells. These results show that Agrimonia pilosa Ledeb inhibited melanin production on the melanogenesis. The underlying mechanism of Agrimonia pilosa Ledeb on whitening activity may be due to the inhibition of tyrosinase activity. We suggest that Agrimonia pilosa Ledeb may be useful as new natural active ingredients for antioxidant and whitening cosmetics.

This research was carried out to identify the whitening effect of Banana leaf extract. B16F10 cells were used to measure cell viability, mRNA expression, and tyrosinase activity inhibition assay from B16F10 cell. We also carried out clinical test of the cream product containing banana leaf extract. In this study, we elucidated the effects of banana leaf extract on TRP1 / TRP2 / Tyr mRNA expression and tyrosinase activity inhibition. Quantitative real-time PCR showed that banana leaf extract decreased mRNA level of TRP1, TRP2 and Tyr gene and tyrosinase activity inhibition assay also revealed that banana leaf extract 65% decreased melanin production in B16F10 cell. Banana leaf extract cream can whiten the skin darkness induced by ultraviolet. Therefore, we successfully identified the whitening effect of banana leaf extract, and this finding suggested the banana leaf extract is a considerable potent cosmetic ingredient for skin whitening. Based on this, we anticipated further researches about banana leaf extract for mechanism to develop not only cosmetics but healthcare food or medicines.

This study was performed to DPPH radical scavenging activity, ABTS radical scavenging activity, tyrosinase inhibition activity and intracellular melanin synthesis inhibition to verify the whitening effect of Fagopyrum tataricum (bitter buckwheat) extract as contrasted with Fagopyrum esculentum (sweet buckwheat) extract. F. tataricum extract in consequence showed higher antioxidant activities, tyrosinase inhibition activity and melanin synthesis inhibition compared with F. esculentum extract. We investigated skin bright value during 8 weeks after induction of pigmentation in human skin from UV irradiation. In result, we obtained statistically a significant skin whitening effect from visual and mechanical evaluation. Accordingly, F. tataricum extract is expected to be high availability as functional cosmetic material for skin whitening.
The aim of this research was to investigate the whitening effects of rutin and rutin metabolites including 3,4-dihydroxyphenyl acetic acid (DHPAA), 3-hydroxyphenyl acetic acid (HPAA), 3,4-dihydroxytolene (DHT) and homovanillic acid (HVA). The potent whitening effect of rutin and rutin metabolites were determined by mushroom tyrosinase inhibition assay and expressed as the half maximal inhibitory concentration ($IC_{50}$) against tyrosinase activity in vitro. The HVA showed the highest inhibitory effect ($IC_{50}=37.10{\mu}M$) of tyrosinase activity, followed by DHPAA ($IC_{50}=45.87{\mu}M$), HPAA ($IC_{50}=50.96{\mu}M$), rutin ($IC_{50}=57.98{\mu}M$), and DHT ($IC_{50}=66.09{\mu}M$), respectively. To evaluate cell cytotoxicity, MTT assay was performed with JB6 P+ mouse epidermal cells and expressed as a relative percentage of untreated control. The results showed that rutin and rutin metabolites had no cytotoxic effects on JB6 P+ cells up to $100{\mu}M$ except for DHT (up to $50{\mu}M$). These results suggests that rutin metabolites may be utilized as a potential tyrosinase inhibitors and the whitening agents for the future.
Juglans mandshurica belongs to the family Juglandaceae is known to contain a wide range of pharmacological activities including anti-cancer, anti-inflammation, astringent, and anti-human immunodeficiency virus-type 1 (HIV-1). Melanogenesis refers to the biosynthesis of melanin pigment in melanocytes. In this study, to investigate the whitening activity of the extracts from Juglans mandshurica, we measured effects on a tyrosinase activity, a melanogenesis, and a tyrosinase synthesis in the B16/BL6 melanoma cells and an antioxidant activity. The extracts significantly scavenged a 1,1-diphenyl-2-picrylhydrazyl (DPPH) and a superoxide anion radicals in a dose-dependent manner with a $SC_{50}$ value of $20\;{\mu}g/mL$ and $25\;{\mu}g/mL$, respectively. Also, the tyrosinase activity and melanogenesis were significantly inhibited by the extracts. Furthermore, the synthesis of tyrosinase protein was significantly decreased by the extracts in enzyme-linked immunosorbent assay. Double blind study on the clinical efficacy of a cream containing 2% of the extracts showed that the extracts have a significant skin whitening effect. Therefore, this study demonstrates that the extracts from Juglans mandshurica may be useful as a potential agent for skin whitening.
To develop a whitening agent, cytotoxicity of the soluble collagen isolated from Todarodes pacificus (CIT) was evaluated. CIT tested for cytotoxicity on human dermal fibroblast (CCD-986sk) was exhibited very low cytotoxicity. Because tyrosinase is the key enzyme for melanin biosynthesis, the use of various tyrosinase inhibitors is a common practice for whitening purpose in cosmetics. Tyrosinase inhibitory activity and melanin production assay were measured to confirm the whitening effect. The inhibitory effect of MMP-1 in UV-irradiated human dermal fibroblast was also performed. CIT showed strong inhibition potency on tyrosinase by 51.5% at 0.2 mg/mL which increased the inhibition by increasing the concentration of CIT, and showed 69.1% inhibition at 1.0 mg/mL. CIT showed strong inhibition effect on melanin production with 82% at 1.0 mg/mL. The CIT also reduced about 76% expression of MMP-1 in UV-irradiated CCD-986sk cell at 1.0 mg/mL. From the preliminary observations, we suggest that the collagen isolated from CIT could be a potential source of natural skin-whitening and anti-aging agents for the photo-damaged skin.

The aim of this study was to investigate whitening effect of combination between arbutine and oil soluble licorice extract. Inhibitory effects of arbutin and oil soluble licorice extract against tyrosinase activity and melanogenesis in B16 melanoma cells were assessed in vitro to determine whitening effect. MTT assay with B16 melanoma cells showed that mixture (arbutin and oil soluble) was not each concentration. Both oil soluble licorice extract and arbutin induced dose-dependent inhibition of mushroom tyrosinase activity. Various concentrations (oil soluble extract : arbutin = 1:1. 1:1.5, 1:2, 1:2.5, 1:5) of mixtures also significantly inhibited tyrosinase activity in B16 melanoma cells by 40-51%. In addition, the mixtures reduced the melanin contents of B16 melanoma cells by more than 50% at each concentration. These results suggest that mixtures of arbutin and oil soluble licorice extract are very effective whitening ingredients.
