Mechanism of aging and prevention (노화의 기전과 예방)

  • Aging is a senescence and defined as a normal physiologic and structural alterations in almost all organ systems with age. As Leonard Hayflick, one of the first gerontologists to propose a theory of biologic aging, indicated that a theory of aging or longevity satisfies the changes of above conditions to be universal, progressive, intrinsic and deleterious. Although a number of theories have been proposed, it is now clear that cell aging (cell senescence) is multifactorial. No single mechanism can account for the many varied manifestations of biological aging. Many theories have been proposed in attempt to understand and explain the process of aging. Aging is effected in individual by genetic factors, diet, social conditions, and the occurrence of age-related diseases as diabetes, hypertension, and arthritis. It involves an endogenous molecular program of cellular senescence as well as continuous exposure throughout life to adverse exogenous influences, leading to progressive infringement on the cell's survivability so called wear and tear. So we could say the basic mechanism of aging depends on the irreversible and universal processes at cellular and molecular level. The immediate cause of these changes is probably an interference in the function of cell's macromolecules-DNA, RNA, and cell proteins-and in the flow of information between these macromolecules. The crucial questions, unanswered at present, concerns what causes these changes in truth. Common theories of aging are able to classify as followings for the easy comprehension. 1. Biological, 1) molecular theories - a. error theory, b. programmed aging theory, c. somatic mutation theory, d. transcription theory, e. run-out-of program theory, 2) cellular theories - a. wear and tear theory, b. cross-link theory, c. clinker theory, d. free radical theory, e. waste product theory, 3) system level theory-a. immunologic/autoimmune theory, 4) others - a. telomere theory, b. rate of living theory, c. stress theory, etc. Prevention of aging is theoretically depending on the cause or theory of aging. However no single theory is available and no definite method of delaying the aging process is possible by this moment. The most popular action is anti-oxidant therapy using vitamin E and C, melatonin and DHEA, etc. Another proposal for the reverse of life-span is TCP-17 and IL-16 administration from the mouse bone marrow B cell line study for the immunoglobulin VDJ rearrangement with RAG-1 and RAG-2. Recently conclusional suggestion for the extending of maximum life-span thought to be the calory restriction.

A Content Analysis of the Psychosocial Maturity of Elderly Korean Residents in Korea and Canada (한국과 캐나다 거주 한국 노인의 심리사회적 성숙경험에 관한 연구)

  • This study examines cultural differneces in psychosocial maturity experiences between collectivistic and individualistic cultures. Personal, relational, and social dimensions were assessed to explore psychosocial maturity experience of elderly Korean residents in Korea and Canada who were psychosocially well developed through in-depth interviews and a content analysis. According to the results, there were diverse similarities and differences in the psychosocial maturity of elderly Korean residents between the two cultures. First, psychosocially mature elderly residents in both cultures showed positive self-concept and self-satisfaction. However, there were differences in their meaning. Second, in terms of interpersonal relations, the most important and satisfying relationship was the spousal relationship for both cultures, and relationships with children were satisfying. However, there were differences in the meaning of relationships between the two cultures as well as gender differences. Third, psychosocially mature elderly residents in both cultures reported psychosocial factors such as mature aging as a social dimension. Elderly residents in Korea regarded mature aging as a relationship oriented phenomenon, whereas those in Canada reported social attitudes toward mature aging. In addition, elderly residents in both cultures reported greed and harm to others as immature aging. Elderly residents in Korea regarded relational discord as immature aging, whereas those in Canada regarded it as demanding adult treatment from future generation. In terms of transcendence, adjustment was reported as living by going with the flow. Elderly residents in Korea reported adjustment based on fatalism, whereas those in Canada showed some religious meaning. Finally, psychosocially mature elderly residents showed a positive view, acceptance and life integration. These results are discussed from the perspective of cultural differences.

  • All of us have no time to predict and prepare for a rapidly-approaching future change in the 21st century, thereby being reality that we feel uneasy about future society. Accordingly, there is necessity for re-consolidating environment and daily culture in preparation for future society, namely, aging society. Given well preparing for super-aging society that will approach in the future, by which this effort becomes what bad luck often brings good luck, our country's role is thought to get probably bigger even in the globally silver industrial market in near future. We need to synthetically recognize a human being's aging psychological and physical conditions through the aging experience exhibition available for publicizing properly this historical phenomenon, and to pay more attention to importance of interaction between a human being and product environment. The experience exhibition is a concept for the exhibition form and the spatial composition, which emphasized interaction, with having escaped from the simply exhibition space. Given trying to communicate information more scientifically with escaping from the conventional exhibition of seeing and listening, it is also important to research Into the basis for exhibition as the experience space based on interaction. After all, a human being's aging is the unavoidably future phenomenon. And, importance of environment, which influences a human being, is very mutual as well. Our country is desired to develop a research on interaction of the exhibition experience space available for broadening recognition of senior-friendly and for properly experiencing this, thereby being an opportunity that the business entity, which plans silver industry, and the academic circles, which research and develop this, do mutually cooperate and can make silver industry grow further.

A New Attempt to Establish the Extrinsic Aging Hair Model to Evaluate The Response to Aging in Physical Property (모발 노화에 따른 물성변화와 외인성 노화모델의 개발)

  • Human tissue undergoes aging by the oxidant damage via structural change and its physical properties. The skin aging process is well known and many evaluations have been conducted. However, studies on hair aging were relatively few and thus care for aging hair is difficult. This study aims to fabricate an aging hair and identify anti-aging effect with known ingredient in anti-aging. First of all, physical properties of aging hair of age 60s by physiologically intrinsic factors were compared to those of the hair made by various extrinsic factors such as several chemical reactions and iteration numbers of the treatments. The extrinsic aging hair of this study relates to the less amount of lipid and to the hair of perm treated once accordingly, wherein several physical properties, preferably comprise roughness and tensile strength, present a novel concept of the intrinsic aging hair. The penetration of peptide into the aging hair was leading the extrinsic hair towards more structurally directed a younger hair. In addition to the structural change, the penetration of the peptide enhanced texture and tensile strength of the aging hair. These patterns have been also found in addition of propolis. For the first time, these qualitative studies exhibit that indeed our extrinsic aging hair well describes the anti-aging efficacy as a receptor for a cross-linker and the ingredients of human hair.

The Variation of Structure and Physical Properties of XLPE during Thermal Aging Process (가교 폴리에틸렌의 열노화에 따른 구조와 물성의 변화)

  • The variation of chemical structure and physical properties of crosslinked polyethylene (XLPE) during thermal aging process was investigated. The formation of carbonyl functional group resulting from thermal oxidation reaction of XLPE was monitored using X-ray photoelectron spectroscopy and near infrared (NIR) spectroscopy. It was observed that the intensity of carbonyl peak observed at 1715 nm linearly increased with aging time in NIR spectroscopy. The linear relationship between NIR peak absorbance and aging time confirmed that NIR spectroscopy might be used as a proper tool for monitoring the aging process of polymeric materials. Also the formation of crosslinks during the aging process was monitored using thermal mechanical analysis, stress-strain test, and Shore hardness test. The change in the physical properties, such as the increase in the glass transition temperature from 110 to 132$^{\circ}C$, the decrease in the strain from 265 to 110%, as well as the increase in the shore D hardness from 32 to 50, was observed during the aging process.

Effects of Ichungwhan on the Aging Process (이정환(二精丸)이 노화과정에 미치는 영향)

  • Objectives: It is well known that aging and aging-related diseases are linked to the increased level of oxidative stress caused by reactive oxygen species(ROS) and reactive nitrogen species(RNS). Nonprotein-SH decreases during aging, while substances such as ROS, nitric oxide(NO), peroxynitrite($ONOO^-$), myeloperoxidase(MPO), and dityrosine show a significant increase. This study investigated the effect of Ichungwhan on the aging process by examining its effect on the generation of the above-mentioned substances. Methods: Four comparison groups of SD rats were used. They were 6 month-old rats, 24 month-old rats, and 24 month-old rats fed with food containing 0.1% and 0.3% of Ichungwhan extract. The amount of NO, $ONOO^-$, and ROS in the rats' kidneys were examined using a fluorescence microplate reader. The reagents used for this purpose include: dihydrorhodamine 123 (DHR 123), 2',7' -dichlorodihydrofluorescein, diacetate(DCFDA), and 4,5-diaminofluorescein(DAF-2). A spectrophotometer was used to investigate the reactivity of nonprotein-SH and myeioperoxidase(MPO), using reagents such as trichloroacetic acid(TCA) and tetramethylbenzidine(TMB). The amounts of MPO protein and dityrosine were measued by western blot. Results: The observed effects of Ichungwhan on rats were as follows: increase of nonprotein-SH; effective decrease of RNS level by suppression of the generation system of $ONOO^-$ and NO; decrease of ROS level; decrease of the MPO reactivity and the subsequent reduction of amount of MPO protein; retardation of dityrosine formation. It can be hypothesized, therefore, that Ichungwhan affects both the earlier and later phases of the molecular inflammatory process, and retards the aging process. Conclusions: Empirical evidence in this study supports a role for Ichungwhan in generation mechanisms of aging process-linked substances ROS, NO, $ONOO^-$, nonprotein-SH, MPO and dityrosine. Affects contrary to the aging process observed in rats beg further empiricism to investigate potential application of Ichungwhan as a medication for age-related diseases in humans.

Aging of Immune System (면역 반응체계의 노화)

  • Immune system provides defense integrity of body against external invaders. In order to accomplish the important defending role immune system is composed of many different components which are regenerated continuously during lifespan. The key components are professional killing cells such as macrophage, neutrophil, natural killer cell, and cytotoxic T cell and professional blocking molecule, antibody, which is produced by plasma cell, the terminal differentiated B cell. Immune response is orchestrated harmoniously by all these components mediated through antigen presenting cells such as dendritic cells. Immune responses can be divided into two ways: innate immune response and adaptive immune response depending on induction mechanism. Aging is a broad spectrum of physiological changes. Likewise other physiological changes, the immune components and responses are wane as aging is progressing. Immune responses become decline and dysregulating, which is called immunosenescense. Immune components of both innate and adaptive immune response are affected as aging progresses leading to increased vulnerability to infectious diseases. Numbers of immune cells and amounts of soluble immune factors were decreased in aged animal models and human and also functional and structural alterations in immune system were reduced and declined. Cellular intrinsic changes were discovered as well. Recent researches focusing on aging have been enormously growing. Many advanced tools were developed to bisect aging process in multi-directions including immune system area. This review will provide a broad overview of aging-associated changes of key components of immunity.

Physicochemical attributes, oxidative stability, and microbial profile of boneless sirloin and bone-in T-bone steaks from Hanwoo steer with reference to dry-aging

  • We investigated the comparative physicochemical attributes, oxidative stability, and microbial characteristics of 28 days dry-aged meat in between boneless sirloin (gluteus medius) and bone-in T-bone steaks (infraspinatus) muscles from Korean Native Hanwoo Steer (KNHS). Results reveal that regardless of the muscles, dry-aging increased protein content and water-holding capacity (WHC) (p < 0.05). Meat from infraspinatus-aged muscle led to darker meat with higher pH values than un-aged meat (p < 0.05). However, fat content, CIE a*, and CIE b* remained unchanged in both muscles at aging. At aged meat, thiobarbituric acid reactive substances (TBARS) values from bone-in infraspinatus muscle was 2.5-fold higher than boneless gluteus medius muscle (p < 0.05). Dry-aging led to an increase in the contents of total unsaturated fatty acids (UFAs), monounsaturated fatty acids (MUFAs), and UFA/saturated fatty acids (SFA) in both muscles (p < 0.05). Furthermore, gluteus medius aged muscle concentrated with olic acid (C18:1) compared to infraspinatus aged muscle. Irrespective of the muscles, dry-aging enhanced the total free amino acids (FAAs) as well as tasty, and bitter amino acid contents whereas decreased the tasty/bitter amino acids (p < 0.05). Aromatic amino acid, tryptophan that converted to serotonin was 2-fold higher in boneless gluteus medius muscle than bone-in infraspinatus muscle at pre and post aging processes (p < 0.05). Aged Infraspinatus muscle increased total bacteria (p < 0.05) while no salmonella spp. was detected in both muscles. Taken together, our study confirms that 28 days dry-aging profiling the quality characteristics of boneless sirloin (gluteus medius) and bone-in T-bone steaks (infraspinatus) distinctly while gluteus medius aged steak performs better owing to oxidative stability and functional compounds than infraspinatus aged steak.

The Effects of Perceived Health and Ageism Experience on Successful Aging (주관적 건강 및 노인차별경험이 성공적 노화에 미치는 영향)

  • In this study, we investigated the effects of perceived health and ageism experience of the elderly on successful aging, which were respectively presumed as predictors of biomedical model and psychosocial model of the successful aging. As important research results, firstly, negligence among ageism experience constructs appeared higher than other discriminatory experiences. Secondly, male showed significantly higher discriminatory experience than female in the area of 3 constructs such as property, negligence and employment ageism. Also there were some different discriminative experiences in the area of perceived health and ageism in terms of age, wealth and education. Finally, the results of hierarchical multiple regression hired to find out factors influencing successful aging showed that the factors such as sex, wealth, perceived health and property discrimination of ageism affected successful ageing. Based on the analytic results, we verified that perceived health, i.e., predictor of biomedical model had higher effect on successful aging than ageism experience, i.e., predictor of psychosocial model of successful aging. Thus, it was suggested that health promotion policy should be considered with priority and also combating ageism as well for successful aging.
