Understanding intestinal health in nursery pigs and the relevant nutritional strategies

  • In the modern pig production, pigs are weaned at early age with immature intestine. Dietary and environmental factors challenge the intestine, specifically the jejunum, causing inflammation and oxidative stress followed by destruction of epithelial barrier and villus structures in the jejunum. Crypt cell proliferation increases to repair damages in the jejunum. Challenges to maintain the intestinal health have been shown to be related to changes in the profile of mucosa-associated microbiota in the jejunum of nursery pigs. All these processes can be quantified as biomarkers to determine status of intestinal health related to growth potential of nursery pigs. Nursery pigs with impaired intestinal health show reduced ability of nutrient digestion and thus reduced growth. A tremendous amount of research effort has been made to determine nutritional strategies to maintain or improve intestinal health and microbiota in nursery pigs. A large number of feed additives have been evaluated for their effectiveness on improving intestinal health and balancing intestinal microbiota in nursery pigs. Selected prebiotics, probiotics, postbiotics, and other bioactive compounds can be used in feeds to handle issues with intestinal health. Selection of these feed additives should aim modulating biomarkers indicating intestinal health. This review aims to define intestinal health and introduce examples of nutritional approaches to handle intestinal health in nursery pigs.

Effects of Acanthopanax senticosus Polysaccharide Supplementation on Growth Performance, Immunity, Blood Parameters and Expression of Pro-inflammatory Cytokines Genes in Challenged Weaned Piglets

  • To investigate the effect of dietary Acanthopanax senticosus polysaccharide (ASPS) on growth performance, immunity, blood parameters and mRNA expression of pro-inflammatory cytokines in immunologically challenged piglets, an experiment employing $2{\times}2$ factorial arrangement concerning dietary ASPS treatment (0 or 800 mg/kg) and immunological challenge (lipopolysaccharide [LPS] or saline injection) was conducted with 64 crossbred piglets (weaned at 28 d of age, average initial body weight of $7.25{\pm}0.21kg$) assigned to two dietary ASPS treatments with 8 replicates of 4 pigs each. Half of the piglets of per dietary treatment were injected with LPS or saline on d 14. Blood samples were obtained at 3 h after immunological injection on d 14 and piglets were slaughtered to obtain spleen samples on d 21. Dietary ASPS did not affect average daily gain (ADG) (p = 0.634), average daily feed intake (ADFI) (p = 0.655), and gain:feed (p = 0.814) prior to LPS challenge. After LPS challenge, for LPS-challenged pigs those fed ASPS had higher ADG and ADFI than the non-supplemented group (p<0.05), and an interaction between $LPS{\times}ASPS$ was observed on the two indices (p<0.05). Dietary ASPS improved lymphocyte proliferation among saline-injected and LPS-injected pigs (p<0.05). Interaction between $LPS{\times}ASPS$ was also revealed on lymphocyte proliferation (p<0.05). Circulatory concentration of IgG was influenced neither by ASPS (p = 0.803) or LPS (p = 0.692), nor their interaction (p = 0.289). Plasma concentration and spleen mRNA expression of interleukin-1beta (IL-$1{\beta}$), interleukin-6 (IL-6), and tumor necrosis factor (TNF)-${\alpha}$ were induced to increase (p<0.05) by LPS challenge, in contrast, these indices were decreased by dietary ASPS (p<0.05), and interactions were found on these cytokines (p<0.05). For LPS-challenged pigs, dietary ASPS also reduced the circulating concentration and spleen mRNA expression of IL-$1{\beta}$, IL-6 as well as TNF-${\alpha}$ (p<0.05). The interaction between $LPS{\times}ASPS$ was also observed on the circulating concentration of insulin-like growth factor-I, ${\alpha}$-acid glycoprotein (${\alpha}$-AGP), nonesterified fatty acid, and glucose (p<0.05). The results of this study demonstrate that dietary ASPS can modulate the release of pro-inflammatory cytokines during immunological challenge, which might enable piglets to achieve better growth performance.

Sexual Dimorphism in Growth of Sucking and Growing Pigs

  • Three studies were conducted to determine the effect of sex on pre- and post-weaning performance of pigs. These studies were conducted in response to observations that female pigs appear to grow faster than male pigs after weaning. In addition, female pigs have been found to grow faster than male pigs when supplied with supplemental milk before weaning. The aims of the present work were to further characterise the ontogeny of sex differences growth of nursing and growing pigs. In the first study, piglets sucking 32 sows were crossfostered to produce litters of 10 boars (n=9), 10 gilts (n=l 1) or 5 boars and 5 gilts (n=12). Liveweight of the sucking pigs was then measured weekly until weaning at 4 weeks of age. In the second study, 80 boars and 80 gilts were weaned at 26 days of age and growth performance measured until 21 days post-weaning. In the third experiment, 40 boars and 40 gilts were weaned into groups of 5 pigs at either 17 or 25 days of age and pigs were weighed until they were approximately 90 kg liveweight. All-boar litters grew more slowly than the all-gilt and mixed litters such that by 14 days of age the all-boar litters were 10% lighter than the all-gilt or mixed litters (39 vs. 43.8 kg, p=0.050). The proportional difference in litter weight appeared to be maintained at 21 days of age (53.9 vs. 59.4 kg, p=0.063) but was diminished by 28 days of age (66.5 vs. 70.8 kg, p=0.28). In the second study, gilts grew more quickly than boars over the first 7 and 21 days post-weaning and as a consequence were 10% heavier than boars at 21 days after weaning (13.7 vs 12.48 kg, p=0.001). In the third study, gilts grew more quickly than boars in the immediate 7 days post-weaning (40 vs. 5 g/day, p=0.014) whereas from 7 until 35 days post-weaning there was no significant difference in growth rate (381 vs. 360 g/day, p=0.19). Gilts also grew more quickly than boars over the 14 days after being moved into the grower (631 vs. 570 g/day, p=0.013) and finisher (749 vs. 688 g/day, p=0.038) sheds. However, these differences were not maintained over the entire period in each shed. These data support the hypotheses that gilts handle the stresses of weaning and other transitions better than boars.

  • Eighteen crossbred pigs were weaned at 4 days of age and fed up to 28 days of age to examine the effect of sulfur amino acid content of three diets upon plasma taurine concentration and hepatic cysteinesulfinate decarboxylase activity. The experimental diets consisted of either whey protein (W) or partialy hydrolyzed soy protein (S) as the source of protein. 0.25% methionine was added to the S diet for the third dietary regimen (SM). Sulfur amino acid content(methionine plus cystine)of the three diets was 1.53%, 1.34% and 1.09% for the W, SM and S diet, respectively. Plasma taurine concentration from the pigs fed the three experimental diets reflected the total sulfur amino acid content of the diet. The S diet resulted in a significantly lower plasma tarrine level than the W and SM diets throughout the experiment. After three weeks, pigs fed the W diet had significantly higher plasma taurine concentration than those fed SM diet. Therfore it appears that taurine requirement of the pig depends on the sulfur amino acid contents of the diets and the conversion o sulfur amino acid to taurine seemed not to be limited by any factor when sulfur amino acid was below 1.53% of the diet. There was no significant difference between three dietary groups in hepatic cysteinesulfinate decarboxylase activity and this suggests that the reduced cysteinesulfinate decarboxylase activity due to high sulfur amino acid in the diet may not occur in the pig liver.

Effects of sweetener sucralose on diet preference, growth performance and hematological and biochemical parameters of weaned piglets

  • Objective: Two experiments were conducted to investigate the effects of dietary sucralose on diet preference and growth performance of weaned piglets, and a third experiment was a 28-d safety study to examine if high-dose sucralose could affect the health state of weaned piglets. Methods: In experiment one, 48 piglets had free access to a corn-soybean based diet and the same diet supplemented with 150 mg/kg sucralose for 15 d. In experiment two, 180 piglets were blocked into 5 treatments with 6 replications. They were fed basal diets supplemented with 0, 75, 150, 225, and 300 mg/kg sucralose for 28 days. In experiment three, 108 piglets were randomly assigned to 3 treatments and fed diets supplemented with 0, 150 (suitable level), and 1,500 (ten-fold suitable level) mg/kg sucralose for 28 d. Results: The experiment 1 showed that piglets preferred (p<0.05) diets containing sucralose during experimental period. In experiment 2, piglets fed a diet supplemented with 150 mg/kg sucralose had a higher average daily gain (ADG) and average daily feed intake (ADFI) than pigs in the control group and other treatment groups during the experiment period. The concentrations of sucralose over 150 mg/kg may decrease feed intake. However, no difference in feed conversion ratio was observed. In experiment 3, piglets fed diet supplemented with 150 mg/kg sucralose had a higher ADG and ADFI than that of pigs in the control group and 1,500 mg/kg treatment groups during the experiment period. Clinical blood metabolites, organ index and histological morphology were not significantly different between sucralose treatments. Conclusion: Sucralose can promote feed intake and thereby improve growth performance of weaned piglets. Moreover, inclusion of 1,500 mg/kg sucralose was demonstrated to have no observed adverse effects. Supplementing 150 mg/kg sucralose for weaned piglets is recommended in this study.

Phytobiotics and Organic Acids As Potential Alternatives to the Use of Antibiotics in Nursery Pig Diets

  • Two experiments were conducted to determine the effect of phytobiotics and organic acids on growth performance of nursery pigs as an alternative to antibiotics. Phytobiotics refer bioactive compounds from plant materials including essential oils and herbal extracts. In Exp. 1,144 pigs, weaned at 23.4${\pm}$0.3 d age, were allotted to three dietary treatments. Treatment diets were: 1) NC (no antibiotics and no phytobiotics); 2) PC (NC+carbadox, 50 mg/kg); and 3) PB (NC+phytobiotics; 0.1% PEP1000-$1^{(R)}$. Each treatment had six replicates with eight pigs per pen. Pigs were fed the experimental diets for 5 wks in 3 phases (phase 1 for 2 wk; phase 2 for 2 wk; phase 3 for 1 wk). In Exp. 2, 192 pigs, weaned at 19.2${\pm}$0.3 d age, were allotted to three dietary treatments: 1) NC; 2) PC; and 3) PBO (NC+phytobiotics; 0.2% or 0.1% PEP1000-$1^{(R)}$ and organic acids; 0.4% or 0.2% $Biotronic^{(R)}$for the phase 1 and 2, respectively) with eight replicates per treatment and eight pigs per pen. Pigs were fed the assigned diets for 5 wks in 2 phases (phase 1 for 2 wk; phase 2 for 3 wk). Body weights were measured at the beginning of the experiment and at the end of each week in both Exp. 1 and 2. Feed intake was measured at the end of each week in both Exp. 1 and 2. Diarrhea score was measured daily during the entire period for Exp. 1 and during the phase 1 for Exp. 2. In Exp. 1, the PC had a higher (p<0.05) overall ADG than the NC, but the overall ADG of the PB did not differ (p>0.05) from the NC or the PC. In Exp. 2, the overall ADG did not differ (p>0.05) among all the treatments during the entire experimental period. The overall ADFI and the overall gain:feed ratio did not differ (p>0.05) among all the treatments during the entire experimental period in both Exp. 1 and 2. The PC had a higher (p<0.05) overall diarrhea score (harder stools) than the NC and the PB in Exp. 1, and a higher (p<0.05) overall diarrhea score than the NC in Exp. 2. The overall diarrhea score of the PB and the PBO did not differ (p>0.05) from the NC or the PC in Exp. 1 and 2. Results from this study show that the growth of pigs fed the diets with phytobiotics or the combination of phytobiotics and organic acids did not differ from those both with antibiotics and without antibiotics when tested in an environmentally controlled research facility. Further experiments are required to study the growth performance in disease challenged conditions.

이유자돈에 있어 대두유, 건조팜유분말과 Monoglyceride의 첨가가 성장 및 혈청 지질변화에 미치는 영향 (The Effect of Soybean Oil, Dried Palm Oil Powder and Monoglyceride Supplementation on Growth Performance and Serum Lipid Changes in Weaned Pigs)

  • 본 실험은 이유자돈에 있어 대두유, 건조팜유분말과 monoglyceride의 첨가가 성장 및 혈청 내 콜레스테롤 변화에 미치는 영향을 조사하기 위하여 실시하였다. 3원 교잡종[(Y×L)×D] 자돈 125두(개시시 체중 6.00±0.79kg)를 공시하여 21일간 사양시험을 실시하였다. 시험설계는 기초사료 내 일반대두유 5%를 함유한 처리구(SOY), 건조팜유분말 5.5%를 함유한 처리구(PALM), 일반대두유 2.5%와 0%, 12.5%, 25%의 monoglyceride를 각각 첨가한 건조팜유분말 2.7%를 함유한 처리구(SOPM0, SOPM12.5, SOPM25)의 5개 처리를 두었다. 0-21일간의 총 사양시험기간동안, 일당사료섭취량은 monoglyceride를 함유한 SOPM 구와 PALM 구가 SOY 구, monoglyceride를 함유하지 않은 SOPM 구와 비교하여 증가하였다(P<0.05). 일당증체량 및 사료효율에 있어서는 처리구간 차이를 보이지 않았다. 지방소화율에 있어서는 SOY 구가 가장 높았으며(P<0.05), monoglyceride를 함유한 SOPM 구는 PALM 구와 비교하여 유의적으로 소화율이 증가하였다(P<0.05). SOPM12.5 구와 SOPM25 구는 SOY 구와 비교하여 통계적인 차이를 보이지 않았다(P>0.05). 그러나, DM, N 및 DE의 소화율에 있어서는 처리구간 차이를 보이지 않았다(P>0.05). 등지방 두께는 처리구간 유의적인 차이를 나타내지 않았다(P>0.05). 혈청 내 콜레스테롤 농도에 있어서는 SOY 구는 다른 처리구와 비교하여 혈청 내 total cholesterol, HDL- cholesterol과 total lipid 함량에 있어서는 유의적으로 감소하였으며(P<0.05), free fatty acid 함량에 있어서는 증가하였다(P<0.05). 또한, triglyceride 함량에 있어서는 PALM 구가 SOPM 구와 비교하여 증가하였다(P=0.06). 결론적으로, 이유자돈에 있어 대두유의 급여는 건조팜유분말의 단일 급여보다 높은 지방 소화율을 나타내었으며, 혈청 콜레스테롤 및 triglyceride 함량에 있어서도 감소하는 것으로 나타났다. 또한, monoglyceride가 함유된 건조팜유분말은 건조팜유분말만을 급여한 처리구와 비교하여 이유자돈의 지방 소화율을 개선하는 효과를 가지는 것으로 사료된다.(색인: 건조팜유분말, 모노글리세라이드, 성장율, 혈청 지질, 이유자돈)

Effect of Partial Substitution of Dietary Spray-dried Porcine Plasma or Fishmeal with Soybean and Shrimp Protein Hydrolysate on Growth Performance, Nutrient Digestibility and Serum Biochemical Parameters of Weanling Piglets

  • The present experiment was conducted to study the effects of partial replacement of spray-dried porcine protein (SDPP) or fish meal with soybean and shrimp protein hydrolysate (SSPH) on growth performance, nutrient digestibility and serum biochemical parameters in weaned pigs. Two hundred and forty 21${\pm}$2 d old pigs ((Pietrain${\times}$Duroc)${\times}$(Landrace${\times}$Large Yorkshire)) with initial weight of 6.9${\pm}$0.5 kg were randomly allocated to five dietary treatments with six replicates per treatment and eight piglets per replicate. The control diet (T1) contained 2% SDPP and 6% fishmeal, and SDPP for experimental diets T2 and T3 was replaced with 1% and 2% SSPH, respectively, on an iso-nitrogenous basis. The fishmeal for experimental diets T4 and T5 was replaced with 1% and 2% SSPH, respectively, also on an iso-nitrogenous basis. The experimental period was 21 days. The results showed that weaned piglets fed the diets containing 1% and 2% SSPH as a replacement for SDPP had similar average daily gain (ADG), average daily feed intake (ADFI), feed/gain (F/G), diarrhea rate and serum biochemical indices e.g. blood urea nitrogen (BUN), total serum protein (TP), albumin to globulin ratio (A/G), globulin (GLO), serum glucose (GLU), and immunoglobulin G (IgG) to those fed the control diet during 0-10 d and 0-21 d of the experiment. The substitution of 1% SSPH on an iso-nitrogenous basis for fish meal appeared to be beneficial for ADG (p = 0.59) and ADFI (p = 0.23) of piglets during the overall period. The digestibility of calcium was higher (p<0.01) in pigs fed diets containing SSPH than on the control diet. Addition of 1% SSPH on an iso-nitrogenous basis for fish meal could increase the digestibilities of dry matter and energy of the diet. Dietary replacement of fish meal with 1% and 2% SSPH had no effect on the concentrations of BUN, TP, A/G, GLO, GLU, and IgG. In conclusion, dietary SDPP or fish meal could partially replace SSPH without any adverse effect on growth performance, nutrient digestibility and serum biochemical parameters in weaned piglets.

난용성 식이섬유의 첨가가 이유자돈의 생산성 및 영양소 소화율에 미치는 영향 (Effects of Insoluble Dietary Fiber Supplementation on Performance and Nutrient Digestibility of Weanling Pigs)

  • 사료에 대한 난용성 식이섬유(Vitacel®)의 첨가가 이유자돈의 생산성 및 영양소 소화율에 미치는 영향을 알아보기 위해 평균 체중 6.49 kg(SD=0.52) 및 평균 일령 23일(SD=2.1)인 교잡종(Landrace$\time$Yorkshire) 거세돈 96두를 4처리 12반복으로 배치하여 40일간 사양시험을 실시하였다. 시험은 4처리를 두었으며, 시험처리는 1) 대조구, 2) 대조구+0.3% 난용성 식이섬유(IDF, Insoluble Dietary Fiber) 첨가구, 3) 대조구+0.6% IDF 첨가구 및 4) 대조구+0.9% IDF 첨가구였다. 사료 영양소 소화율 측정을 위한 분 채취는 diet 2를 급여 후 8-9일 및 18~19일째에 각 돈방 별로 실시하였으며, 소화율을 측정하기 위한 표식제로는 Celite-545(Fluka)를 1 % 사용하였다. 시험 전 기간(0~40일간) 동안, 일당증체량은 IDF 0.3% 첨가구가 기초사료 급여군, IDF 0.6% 첨가군 및 IDF 0.9% 첨가군에 비해 높았으며(P<0.05), IDF 0.6% 첨가군은 기초사료 급여군에 비해 일당증체량이 높았으나(P<0.05), IDF 0.9 % 첨가구와 기초사료 급여군 간에는 유의차가 인정되지 않았다(P>0.05). 일당섭취량은 IDF 0.3% 첨가구가 모든 처리구에 비해 유의적으로 높았으며(P<0.05), 기초사료 급여구, IDF 0.6% 첨가구 및 IDF 0.9% 첨가구 사이에서는 통계적인 유의차가 인정되지 않았다(P>0.05). 유기물, 조단백질, 조섬유 및 에너지 소화율은 IDF 0.3% 첨가구에서 기초사료 급여구 보다 유의적으로 높았다(p<0.05), 그러나 IDF 0.6% 첨가구와 IDF 0.9% 첨가구간에는 양양소 소화율에서 유의적인 차이가 관찰되지 않았으며, IDF 0.6% 이상의 첨가구와 기초사료 급여구 간에도 ADF(산성세제섬유소) 소화율을 제외한 다른 영양소 소화율에는 유의적인 차이를 보이지 않았다 (P>0.05). 본 실험에서 이유자돈 사료에 IDF의 첨가는 첨가수준에 따른 quadratic 생산성 개선 효과를 보여주었으며, 이러한 결과는 IDF 첨가에 따른 영양소 소화율 개선 효과로부터 기인된다고 판단해 볼 수 있다.

Effects of Lactobacilli on the Performance, Diarrhea Incidence, VFA Concentration and Gastrointestinal Microbial Flora of Weaning Pigs

  • Two experiments were conducted to evaluate the effects of a complex Lactobacilli preparation on performance, resistance to E. coli infection and gut microbial flora of weaning pigs. In exp. 1, twelve pigs (7.65$\pm$1.10 kg BW), weaned at 28 d, were randomly allotted into 2 groups and placed in individual metabolic cages. During the first 7 d, one group of pigs was provided ad libitum access to water containing $10^5$ colony forming units (CFU) Lactobacilli per ml and the control group was provided tap water. The Lactobacilli preparation included Lactobacillus gasseri, L. reuteri, L. acidophilus and L. fermentum, which were isolated from the gastrointestinal (GI) tract mucosa of weaning pigs. On d 8, 20 ml of $10^8$ CFU/ml E. coli solution (serovars K99, K88 and 987P at the ratio of 1:1:1) was orally administered to each pig. Diarrhea scores and diarrhea incidence were recorded from d 7 to 14. On d 14, pigs were euthanized and digesta and mucosa from the stomach, duodenum, jejunum, ileum, cecum and colon were sampled using aseptic technique to determine microflora by culturing bacteria in selective medium. The results showed that Lactobacilli treatment significantly decreased E. coli and aerobe counts (p<0.01) but increased Lactobacilli and anaerobe counts (p<0.01) in digesta and mucosa of most sections of the GI tract. A 66 and 69.1% decrease in diarrhea index and diarrhea incidence, respectively, was observed in the Lactobacilli treated group. In exp. 2, Thirty-six crossbred Duroc$\times$Landrace$\times$Yorkshire piglets, weaned at 28$\pm$2 days, were selected and randomly allocated into 2 groups. There were 18 piglets in each group, 3 piglets in one pen and 6 replicates in each treatment with 3 pens of barrow and 3 pens of female piglet in each treatment. Piglets had ad libitum access to feed and water. The initial body weight of piglet was 7.65$\pm$1.09 kg. Dietary treatments included a non-medicated basal diet with Lactobacilli ($10^5$ CFU/g diet) or carbadox (60 mg/kg) as control. On d 21, six pigs per group (one pig per pen) were euthanized. Ileal digesta was collected to determine apparent amino acid digestibility. Microflora content was determined similarly to exp.1. The results showed that Lactobacilli treatment significantly improved average daily feed intake (ADFI) of pigs compared to carbadox (p<0.05) during the first 2 wks after weaning and average daily gain (ADG) and ADFI increased significantly (p<0.05) from d 8 to 14. Nitrogen and total phosphorus digestibility also increased (p<0.05). Bacterial counts were similar to exp. 1. The results indicate that the complex Lactobacilli preparation improved performance for 2 wks after weaning, enhanced resistance to E. coli infection, and improved microbial balance in the GI tract.
