Antibodies to Heat Shock Protein 70kDa and 90kDa in the Patients with Schizophrenia, and Their Relationship with Clinical Variables

  • Schizophrenia has many clinical expressions and probably different etiologic factors. Infections, autoimmune mechanism and related neurodevelopmental abnormalities have been suggested as possible etiologic factors of schizophrenia. It has been reported that immunoreactivity to heat shock proteins, which play a protective role against environmental stresses in a cell, might be related to the pathogenesis of schizophrenia. Therefore, we examined the immunoreactivity to heat shock protein 70kDa and 90kDa(HSP70 and 90) in 91 patients with schizophrenia and 83 normal controls. Ig G antibodies to HSP70 and 90 of sera were quantitated by ELISA. The optical density(OD) was measured by an automated microplate reader at a wavelength of 490nm. The amounts of antibodies to HSPs were expressed as arbitrary units(AU)/ml related to a standard serum. The limit for elevated antibody titers(anti-HSPs positive or negative) was set at two standard deviations added to the mean of the normal controls. Twenty nine(31.9%) of the 91 patients showed anti-HSP70 positive and 19(20.9%) of those showed anti-HSP90 positive. On the other hand, only 1(1.4%) of the normal controls and 4(4.8%) of those showed anti-HSP70 positive and anti-HSP90 positive, respectively. The titers of anti-HSP70 positive were related with BPRS scores, while those of anti-HSP90 positive were not. There were no relationship between antibody titers and clinical variables including age at onset, duration of illness, family history of schizophrenia or number of admission. The titers of anti-HSP70 positive were significantly associated with anti-HSP90 positive. Our results suggest the presence of abnormal immune reactivity involving HSP70 and HSP90 in a subset of patients with schizophrenia.

Development of Moisture Content Prediction Model for Larix kaempferi Sawdust Using Near Infrared Spectroscopy (근적외선 분광분석법을 이용한 낙엽송 목분의 함수율 예측 모델 개발)

  • The moisture content of sawdust must be measured accurately and controlled appropriately during storage and transportation because biological degradation could be caused by improper moisture. In this study, to measure the moisture contents of Larix kaempferi sawdust, the near-infrared reflectance spectra (Wavelength 1000-2400 nm) of sawdust were used as detection parameter. After acquiring the NIR reflection spectrum of specimens which were humidified at each relative humidity condition ($25^{\circ}C$, RH 30~99%), moisture content prediction model was developed using mathematical preprocessings (e.g. smoothing, standard normal variate) and partial least squares (PLS) analysis with the acquired spectrum data. High reliability of the MC regression model with NIR spectroscopy was verified by cross validation test ($R^2$ = 0.94, RMSEP = 1.544). The results of this study show that NIR spectroscopy could be used as a convenient and accurate method for the nondestructive determination of moisture content of sawdust, which could lead to optimize wood utilization.

Sinigrin content of different parts of Dolsan leaf mustard (돌산갓의 부위별 sinigrin 함량)

  • The aims of this study was to optimize the extraction conditions of sinigrin from Dolsan leaf mustard. Dolsan leaf mustard (Dolsan-eup, Yeosu-si) harvested during at May 2014 was used for sinigrin extraction. After the extraction of sinigrin using 50% $CH_3CN$, 10% $NH_4Cl$, 60% $CH_2OH$, and 70% $CH_3OH$, the sinigrin content was measured by HPLC analysis. The results showed that sinigrin content was highest with 50% $CH_3CN$ solvent extraction and UV detector sensitivity was greater at 228 nm rather than at 242 nm. The sinigrin concentrations of leaf, stem and root with 50% $CH_3CN$ extraction were 345 ppm, 728 ppm, and 539 ppm, respectively. After extraction of the different parts of Dolsan leaf mustard, The standard retention time by HPLC analysis of sinigrin content was 2.054, 2.032, 2.059, and 2.035 min from the root, stalk, and leaf, respectively. On the other hand, HPLC analysis showed that the leaf extracts contained glucoraphanin, one of glucosinolates. The optimum time and extraction solvent for the sinigrin extraction from Dolsan leaf mustard was found to be 24 hr with 50% $CH_3CN$ solvent. In addition, opotimum UV detector k at 228 nm. These results showed that the optimum extraction conditions for Dolsan leaf mustard were 24 hr extraction with 50% $CH_3CN$ solvent. In addition, the optimum wavelength of UV detector was determined to be 228 nm for sinigrin analysis. Therefore, this study could provide a useful information for sinigrin extraction and its systematic analysis during the storage.

  • Currently, reticulocyte experimental calculation technology used in clinical laboratories are divided two types: manual and automated. Manual reticulocyte counting using a microscopy lacks accuracy due particularly to its low reproducibility, affecting the accuracy of manual reticulocyte count. Moreover, Automatic blood corpuscle analyzer flow cytometry is difficult to be used in underdeveloped countries and small scale laboratories due to relatively high cost. Therefore, this study tried to find a new method to complement these drawbacks. The aim of this study was to compare the stained reticulocytes count by spectrophotometer and also to analyze the statistics of spectrophotometer and flow cytometer. The same 8 EDTA samples were repeated 36 times to compare the agreement between spectrophotometer and flow cytometer. This study measured the specimen diluted 600 times at 700~780 nm by 10 differences. Wavelength between 710 to 730 by absorbance showed a positive correlation between standard data and test data (r=0.967, p<0.01), presenting a correlation between variables. Statistical analyses of regression for test and standard parametric data, the optimal dilution factor was 600 times. Therefore, this study tried to technical utilizes such as contributing economical for the reticulocyte absorbance apply from the auto spectrophotometer, a monitoring system for the reticulocyte relation anemia, etc. Therefore, more extensive studies, including an auto chemical analyzer application, will be needed.

Evaluation of Beef Freshness Using Visible-near Infrared Reflectance Spectra (가시광선-근적외선 반사스펙트럼을 이용한 쇠고기의 신선도 평가)

  • The objective of this study was to develop models to predict freshness factors (total viable counts (TVC), pH, volatile basic nitrogen (VBN), trimethylamine (TMA), and thiobarbituric acid (TBA) values) and the storage period in beef using a visible and near-infrared (NIR) spectroscopic technique. A total of 216 beef spectra were collected during the storage period from 0 to 14 d at a $10^{\circ}C$ storage. A spectrophotometer was used to measure reflectance spectra from beef samples, and beef freshness spectra were divided into a calibration set and a validation set. Multi-linear regression (MLR) models using the stepwise method were developed to predict the factors. The MLR results showed that beef freshness had a good correlation between the predicted and measured factors using the selected wavelength. The correlation of determination ($r^2$), standard error of prediction (SEP), and ratio of standard deviation to SEP (RPD) of the prediction set for TVC was 0.74, 0.64, and 2.75 Log CFU/$cm^2$, respectively. The $r^2$, SEP, and RPD values for pH were 0.43, 0.10, and 1.10; those for VBN were 0.73, 1.45, and 2.00 mg%; those for TMA were 0.70, 0.19, and 2.58 mg%; those for TBA values were 0.73, 0.13, and 2.77 mg MA/kg; and those for storage period were 0.77, 1.94, and 2.53 d, respectively. The results indicate that visible and NIR spectroscopy can predict beef freshness during storage.

Prediction of Chemical Composition and Fermentation Parameters in Forage Sorghum and Sudangrass Silage using Near Infrared Spectroscopy

  • This study was conducted to assess the potential of using NIRS to accurately determine the chemical composition and fermentation parameters in fresh coarse sorghum and sudangrass silage. Near Infrared Spectroscopy (NIRS) has been increasingly used as a rapid and accurate method to analyze the quality of cereals and dried animal forage. However, silage analysis by NIRS has a limitation in analyzing dried and ground samples in farm-scale applications because the fermentative products are lost during the drying process. Fresh coarse silage samples were scanned at 1 nm intervals over the wavelength range of 680~2500 nm, and the optical data were obtained as log 1/Reflectance (log 1/R). The spectral data were regressed, using partial least squares (PLS) multivariate analysis in conjunction with first and second order derivatization, with a scatter correction procedure (standard normal variate and detrend (SNV&D)) to reduce the effect of extraneous noise. The optimum calibrations were selected on the basis of minimizing the standard error of cross validation (SECV). The results of this study showed that NIRS predicted the chemical constituents with a high degree of accuracy (i.e. the correlation coefficient of cross validation ($R^2{_{cv}}$) ranged from 0.86~0.96), except for crude ash which had an $R^2{_{cv}}$ of 0.68. Comparison of the mathematical treatments for raw spectra showed that the second-order derivatization procedure produced the best result for all the treatments, except for neutral detergent fiber (NDF). The best mathematical treatment for moisture, acid detergent fiber (ADF), crude protein (CP) and pH was 2,16,16 respectively while the best mathematical treatment for crude ash, lactic acid and total acid was 2,8,8 respectively. The calibrations of fermentation products produced poorer calibrations (RPD < 2.5) with acetic and butyric acid. The pH, lactic acid and total acids were predicted with considerable accuracy at $R^2{_{cv}}$ 0.72~0.77. This study indicated that NIRS calibrations based on fresh coarse sorghum and sudangrass silage spectra have the capability of assessing the forage quality control

Utilization of Near Infrared Spectroscopy for Analysis of Proximate Composition and Starch in Alaska Pollack Surimi (명태 수리미의 일반성분 및 전분의 분석을 위한 근적외선 분광분석법의 이용)

  • Near infrared spectroscopy (NIR), is recently applied to analysis of food, and it gives non-destructive and highly reproducible results. The objectives of this study were to assay protein, lipid and starch content in Alaska pollack surimi and to investigate application in seafood. NIR spectra showed the absorbance signal at 1,510 nm, 2,050 nm, 2,170 nm and 2,180 nm for protein, and signal were increased with protein content. Standard error of equation (SEE) was 0,296 and standard error of prediction (SEP) was 0.327. In analysis of lipid in Alaska pollack surimi by NIR, near infrared spectra of lyophilized Alaska pollack surimi were scanned and the signals of absorbance from C-H functional groups in lipid were identified at 1,730 nm, 1,740 nm and 2,300 nm and these signals were risen as Increasing lipid contents of Alaska pollack surimi as samely protein. SEE and SEP were 0.319, 0,353, respectively. In starch analysis using NIR the signals of starch distinctly changed at wavelength of 1,450 nm and 1,950 nm. Collected values of SEE and SEP were 0.304 and 0.318, respectively and the range of errors was $0.0186\~0.6470$ in starch contents.

  • TACAN(TACtical Air Navigation) was created to support military aircraft's short range navigation (200~300 mile). TACAN must fulfill a condition of MIL-STD-291C, the U.S. Military Standards, which addresses the sum of 15Hz and 135Hz should be within 55%, following the factor of modulations for both to be $21{\pm}9%$ each. Within the existing TACAN antenna, modulation factor for 15Hz and 135Hz are created differently depending on its diameter, wavelength, angle of gradient, internal modulation method or using frequency code. It brings up a problem where applications needed to be stopped and repaired when modulating signal exceeds the standard of MIL-STD-291C since the existing TACAN antenna does not have coordination function. Hence, plan and produce a modulating signal generator using FPGA, and check the changes in the modulation factor for 15HZ and 135Hz, depending on the values that have been set in each criteria. Moreover, allow the modulating signal generator to be automatically adjusted based on the monitoring signal emitted by antenna, and place alarm sound just in case if it exceeds the standard.

  • Wi-Fi is a rapidly spreading communications network with Smart phone's publication, the technology has become Ubiquitous-based core network which is connected to personal computers, laptops, and tablet PC. Wi-Fi can send currently a variety of data standard due to developed wireless LAN communications network. One of Wi-Fi standard protocols, which is IEEE 802.11n, use 2.4GHz and 5GHz band. 2.4GHz band is used for 802.11b/g protocol because wavelength is long, diffraction and receiving distance is enough to connect other device. 5GHz band has more available channels to use than 2.4GHz band, so there is no frequency interference of other wireless device such as Bluetooth, RFID. Moreover, there is low interference between channels due to small users in each bandwidth level. In the thesis, we are going to analyze 802.11a/b/g protocol which has used since the beginning of Wi-Fi protocol and 802.11n protocol which is used lately. Furthermore, we look into development and direction for standardization of the next generation wireless LANs which are 802.11ac and 802.11ad. In addition, we will consider for the security, vulnerabilities and its countermeasure in Wireless LAN.

Comparisons between Micro-Kjeldahl and Near Infrared Reflectance Spectroscopy for Protein Content Analysis of Malting Barley Grain (근적외분광분석법과 Micro-Kjeldahl 법 간의 맥주보리 종실의 단백질함량 분석 비교)

  • Near Infrared Reflectance Spectroscopy(NIRS) has been used as a tool for the rapid, accurate, protein assay of malting barley. NIRS used in this study was filter type instruments, Neotec 102. The objective of this study was to obtain the best calibration equation, for the rapid, ease and accurate protein content analysis of malting barley using NIRS system. The optimum wavelength for protein content analysis used NIRS were 2095nm, 2095/1941nm, 2095/1941/2282nm, 2905/1941/2282/2086nm, respectively. Mean protein content with this calibration equation in NIRS analysis was 10.59%, while 10.60% in Micro-Kjeldahl one. The range of protein content in Micro-Kjeldahl was 8.66~12.66% and that in NIRS was 8.80~12.35%. When 18 other varieties produced in 1992 were analysed with 2095nm, 2095/1941nm, 2095/1941/2282nm, 2095/1941/2282/2086nm equation, standard deviation of difference (SDD)and standard error of performence(SEP) and $R^2$ values were 0.47, 0.43, 0.95, respectively. Both the mean protein content by Micro-Kjeldahl and by NIRS was 10.25%. With this equation, analysied 31 varities produced in 1993, SDD and SEP and r values were 0.69, 0.67, 0.91, respectively, and that bias value was 0.65. In this analysis, mean protein content by Micro-Kjeldahl was 10.17% and by NIRS was 10.81%. The range of protein content in Micro-Kjeldahl was 7.58~14.29%, What that in NIRS was 8.63~13.93%. After adjusted bias in the best calibration equation, mean protein content of Micro-Kjeldahl was 10.17% and that of NIRS was 10.09%, without variance of SDD, SEP and r values.
