Inhibitory Effects on Melanin Production in B16 Melanoma Cells of Fallen Pear (B16F10 Melanoma 세포에서 낙과 배 물 추출물의 멜라닌 생성 저해 효과)

  • This study investigated the water extracts of fallen pear (FPWE) on tyrosinase activity and melanogenesis. In the present study, we examined the effects of FPWE on mushroom tyrosinase activity in vitro, B16F10 melanoma cell tyrosinase activity, melanin contents, and expression of melanogenic enzyme proteins such as tyrosinase. An apparent down-regulatory effect on tyrosinase activity was observed when B16F10 cells were incubated with FPWE. Results of melanin assay using B16F10 cells treated with different concentrations (50, 125, and $250{\mu}g/mL$) of FPWE showed a dose-dependent decrease in melanin content. To determine whether or not FPWE indirectly affects tyrosinase activity, we assessed mushroom tyrosinase activity upon treatment with various concentrations (125, 250, 500, and $1,000{\mu}g/mL$) of FPWE. In addition, we investigated changes in the protein level of tyrosinase by using Western blotting. Tyrosinase and microphthalmia-associated transcription factor expression levels in B16F10 melanoma cells were reduced in a dose-dependent manner by FPWE. These results suggest that FPWE reduced melanin formation by inhibition of tyrosinase activity. Therefore, we suggest that FPWE could be used an effective whitening agent for skin.

Antioxidant and Antipruritic Activities of Ethyl Acetate Fraction from Diospyros lotus Leaves (고욤(Diospyros lotus)잎 유래 Ethyl Acetate 분획물의 항산화 및 항가려움 활성)

  • Diospyros lotus has been cultivated for its edible fruits, which are considered to have medicinal importance. The aim of this study was to evaluate the antioxidant and antipruritic activities of water-soluble, methanol extract, and ethyl acetate (EA) fractions from D. lotus leaves. The EA fraction showed the lowest $IC_{50}$ vale (DPPH: $5.3{\mu}g/mL$, ABTS: $53.8{\mu}g/mL$). Therefore, we further investigated anti-inflammatory and antipruritic effects of the EA fraction. TNF-${\alpha}$ production increased by PMA plus A23187 treatment was significantly inhibited by the EA fraction in a dose-dependent manner. The EA fraction also inhibited histamine release from rat peritoneal mast cells stimulated by compound 48/80, which promotes histamine release. Furthermore, EA fraction had inhibitory effects on scratching behavior induced by compound 48/80 in Balb/c mice. These results suggest that the EA fraction from D. lotus leaves has potential as ameliorative agent against oxidative stress and pruritus-related disease.

The effects of bamboo salt solution on remineralization of artificial caries-like lesions (죽염이 초기인공우식법랑질의 재광화에 미치는 영향)

  • Objectives : The aim of this investigation was to evaluate the remineralization effect of Bamboo salt and NaF+Bamboo salt solutions on bovine enamel formed incipient artificial enamel caries by microcomputed tomography (micro CT). Methods : Experimental solutions were distilled water(negative control), 2% sodium fluoride solution (2% NaF group), 3% bamboo salt solution (3% BS group) and the solution mixed 2% sodium fluoride solution and 3% bamboo salt solution (2% NaF+3% BS group). Specimens were prepared from extracted bovine teeth and divided into 4 groups of 10 specimens each by randomized blocks according to density. Then the specimens surface were divided equally into three parts to observe sound enamel area, incipient enamel carious area and remineralized enamel area. Only one-third of specimen surface was coated with nail varnish and these were exposed to a lactate carbopol buffer system for 72 hours. Then one-half of the demineralized enamel area was coated with nail varnish. The specimens were carried out under pH cycling model for 14 days as follows; samples were immersed in each experimental solution for 2 mins 3 times per day, demineralized for 4 hours and in mixed saliva for the remaining hours. After pH cycling, density was measured using micro CT. Results : All experimental groups showed remineralization effects except for negative control group(p<0.05). The differences of density after experimental solution treatment were statistically significant difference among 4 groups (p<0.01). The density difference values of groups were $0.04{\pm}0.01$ in negative control group, $0.19{\pm}0.01$ in 2% NaF group, $0.14{\pm}0.01$ in 3% BS group, and $0.21{\pm}0.01$ in 2% NaF+3% BS group. Conclusions : The bamboo salt solution showed remineralization effects on incipient artificial enamel caries and the solution mixed sodium fluoride solution and bamboo salt solution showed more remineralization effects than the bamboo salt solution. Thus, it is suggested that bamboo salt can be used as remineralization agent in incipient enamel caries lesion.

Effects of Chinae Rhizoma Extracts on the Growth of the Dermatophytes (토복령알코홀추출물의 항진균작용)

  • The majority of drugs used in the treatment of superficial fungal infections has limited values due to its low efficacy or development of resistance. For the purpose of searching efficacious agent on the superficial fungal infections induced by dermatophytes which is regarded as the most malicious one, authors examined whether Chinae Rhizoma Extracts have significant on it. Extracts from Smilax china Linne used for the study are water extract (CRWE), ethanol extract (CREE) and methanol extract (CRME). In in vitro studies, the spores of the dermatophytes were inoculated on Sabouraud's glucose agar media which contained three extracts of Chinae Rhizoma in each concentration of $500\;{\mu}g/ml$, $1,000\;{\mu}g/ml$ and $5,000\;{\mu}g/ml$ respectively, and also $1,000\;{\mu}g/ml$ of salicylic acid and undecylenic acid $1,000\;{\mu}g/ml$ as comparable drugs. The growth of the dermatophytes were observed for 3 weeks. The species of the dermatophytes used in this experiment were Epidermophyton floccosum, Microsporum canis, Microsporum cookei, Microsporum gypseum, Microsporum nanum, Trichophyton mentagrophytes, Trichophyton rubrum, Trichophyton tonsurans and Trichophyton verrucosum distributed from The Institute of Tropical Medicine in Belgium. The results of the studies were as follows: 1. The growth of M. canis, M. nanum, T. mentagrophytes, T. rubrum & T. tonsurans were slightly inhibited in CRWE $1,000\;{\mu}g/ml$ and CRWE $5,000\;{\mu}g/ml$, and only slight inhibition on the growth of E. floccosum, M. canis and M. gypseum were observed in CRWE $5,000\;{\mu}g/ml$. 2. Complete inhibition of T. rubrum, moderate inhibition of M. nanum & T. tonsurans, and slight inhibition of E. floccosunl, M. canis, M. cookei & T. mentagrophytes in growth were observed in concentration of CREE $500\;{\mu}g/ml$. The growth of M. gypseum was slightly inhibited, moderate inhibition on the growth of M. canis, M. cookei & T. mentagrophytes, and complete inhibition of E. floccosum, M. nanum, T. rubrum & T. tonsurans in growth were observeed by CREE $1,000\;{\mu}g/ml$. With $5,000\;{\mu}g/ml$ of CREE, the growth of E. floccosum, M. canis, M. cookei, M. gypseum, T. mentagrophytes, T. rubrum & T. tonsurans were completely inhibited except T. verrucosum being showed slight inhibition. 3. In CRME $500\;{\mu}g/ml$, slight inhibition of T. verrucosum, moderate inhibition of M. gypseum and complete inhibition of E. floccosum, M. canis, M. cookei, T. mentagrophytes, T. rubrum & T. tonsurans in growth were observed. The growth of E. floccosum, M. canis, M. cookei, M. gypseum, M. nanum, T. mentagrophytes, T. rubrum & T. tonsurans were completely inhibited except T. verrucosum being showed moderate inhibition in both CRME $1,000\;{\mu}g/ml$ and CRME $5,000\;{\mu}g/ml$. 4. In $1,000\;{\mu}g/ml$ of undecylenic acid, slight inhibition of T. verrucosum and complete inhibition of E, floccosum, M. canis, M. cookei, M. gypseum, M. nanum, T. mentagrophytes, T. rubrum & T. tonsurans in growth were observed. From the above results, it was found that Chinas Rhizoma Alcoholic Extracts(CREE & CRME) exerted significant antifungal activity, and their effects were probably derived from the pharmacological actions of triterpenoidal saponin and steroidal saponin.

Production and Characterization of a Recombinant Antibody Neutralizing Botulinum Neurotoxin A (보툴리눔 신경독소 A를 중화하는 재조합 항체의 제조와 특성 분석)

  • Botulinum neurotoxin (BoNT/A) is a neurotoxin that selectively attacks the peripheral cholinergic nerve endings. It is produced by Gram -positive, endospore-forming strict anaerobic bacteria, Clostridium botulinum. Since BoNT/A could be a biothreat agent, as well as a contaminator of food and water supplies, the development of sensitive assays for toxin detection and potent antitoxin for the treatment of intoxication is necessary. In this study, for the purpose of producing monoclonal antibodies (mAbs) that are capable of neutralizing Botulinum neurotoxin type A (BoNT/A), scFv (single-chain variable domain fragment) libraries from the rabbit antisera against BoNT/A was fused to a human IgG. The resulting recombinant scFvIgG antibody protein was expressed in stable cell lines and was purified using a protein A affinity chromatography. The efficacy of scFvIgG mAb was confirmed by ELISA and was evaluated for the neutralization of BoNT/A in vivo. Such an in vivo toxin neutralization assay was performed using mice. Although scFvIgG antibody proteins (10 ug) failed to fully protect the mice challenged with BoNT/A (100,000 $LD_{50}$), it significantly prolonged the survival time. These results suggest that scFvIgG mAb may be capable of neutralizing BoNT/A single-chain variable domain fragment.

On the dimensional stabilization of woods with treatment of Polyethylene Glycol-400 (폴리에치렌 글리콜-400에 의한 목재(木材)의 칫수안정화(安定化))

  • How to stabilize wood against shrinking and swelling in variable atmospheric moisture conditions is important to the wood-using industry and a challenge to research. Polyethylene glycol stabilize wood by bulking the fiber. PEG also serve as a chemical seasoning agent, suppress decay in high concentrations, and have slight effect on physical properties, gluing or finishing. The study designed to determine the effect of PEG-400 on the dimensional stabilization of local hardwoods for wood carvings that could supply a greatly expanding tourist trade and making curved furniture parts, lamp stands and other decorative objects, and possible gunstock. The species examined were 6 species, Seo-Namoo (Carpinus laxiflora), Cheungcheung-Namoo (Cornus controversa), Gorosae-Namoo (Acer mono), Karae-Namoo (Juglans mandshurica), Jolcham-Namoo (Quercusserrata) and Sanbud-Namoo (Prunus sargentii), used as block of 5cm thick radially to the grain, 7cm wide tangentially, and 70cm long parallel to the wood grain. All these test piecies were conditioned above the fiber saturation point before impregnation. The stabilization effects were determined for PEG-400 treated woods in a 50 percent solution for 20 days. The following conclusions were obtained. PEG retentions increased with treating time. It was more effective to treat at 60$^{\circ}C$ than at room temperature. In degree of PEG-400 impregnation on species, Cheungcheung-Namoo havinglow specific gravity had the highest retentions, 68.77% but the lowest, 56.33% was shown in Jolcham-Namoo with high specific gravity. Specific gravity of treated wood increased considerably with effectiveness of polymer loading. The increases in specific gravity were 5.36 to 13.16 percent. The highest was Jolcham-Namoo, the lowest Karae-Namoo. On the dimensional stability, a 40 percent of effectiveness of polymer loading was just as effective as 60 percent in reduction in water absorptivity (RWA), antishrinkage efficiency (ASE) and antiswelling efficiency (AE), and from over 60 percent they increased more rapidly. Also species response varied considerably. ASE was 30.12 to 69.97 percent tangentially and 27.86 to 56.37 percent radially, AE 34.06 to 73.76 percent tangentially and 30.11 to 70.12 percent radially, and RWA 42.31 to 65.32 percent. No differences in volume swelling among the 6 species were observed. Its values were ranged from 14.98 to 19.55 percent and also increased with PEG retentions. On the mechanical properties, the strengths very much decreased with PEG-400 loadings as shown in Figure 12; that were 11.41 to 22.90 percent in compression, 21.61 to 34.35 percent in bending and 22.83 to 36.83 percent in tensile strength. PEG retention in cell wall was less than 1 percent and the most of PEG were immersed in cell lumen. Except for Korae-Namoo, effectivenesses of polymer loading were as much high as 61.58 to 75.02 percent. This is believed to be due to the effect of PEG-400 on excellant dimensional stability of treated woods.

High Throughput-compatible Screening of Anti-oxidative Substances by Insect Extract Library (약용곤충추출물 라이브러리를 이용한 항산화 활성의 초고속 검색)

  • Oxidant stress is well-known for a pivotal parameter related to neuro-inflammatory diseases including Alzheimer's disease, Parkinson's disease, and ALS (Lou Gehrig's disease). In order to effectively screen for anti-inflammatory agents, we first established the infrastructure of high throughput screening for anti-oxidant agents from medicinal insect library extracted with water, methanol, ethanol, and dimethyl sulfoxide. By the screening system, we found that Tenodera angustipennis Saussure, Pyrocoela rupa Olivier and Papilio maackii Mntris had strong anti-oxidant activity. Moreover, Tenodera angustipennis Saussure and Tenodera aridifolia (Stoll) exhibited protection effects of cellular damage by treatment of an oxidant hydrogen peroxide. Together, the results suggest that some selected hits could be a potential agent against neuro-inflammation, although the in vivo studies should be clearly tested.

Inhibitory Effect of Ecklonia cava Extracts against Lipase Activity and Stability Effect of Temperature and pH on Their Activity (감태 추출물이 지니는 Lipase 저해활성의 열 및 pH 안정성)

  • This study was performed to investigate the inhibitory activity of Ecklonia cava (EC) against lipase and the stability of this activity under various heat and pH conditions. As a result, EC ethanol extract showed lipase inhibitory activity of 59, 34 and 19% at concentrations of 5, 2.5 and 1 mg/mL, whereas the water extract showed low inhibitory activity at all concentrations compared to that of the ethanol extracts. In a heat and pH stability test, the inhibitory activity of the EC ethanol extract increased with heat treatment at $121^{\circ}C$ for 15 min compared with the control and was stable in the pH range of 2~10. Therefore, the EC ethanol extract could be useful as a natural anti-obesity agent.

Effects of Aging Temperature and Time on the Conversion of Garlic (Allium sativum L.) Components (온도 및 숙성기간이 마늘의 화학적 성분변화에 미치는 영향)

  • Some thermally processed foods have higher biological activities due to their various chemical changes during heat treatment. Especially, 5-hydroxymethylfurfural (HMF) is derived from dehydration of sugars and has been identified in processed garlic. The biological function of HMF have revealed as antisickling agent and thyrosinase inhibitor. This study was carried out to examine the formation of HMF and free sugars from the aged garlics when it is treated at 60 and $75^{\circ}C$ and different incubation periods from 7 to 35 days. HMF and free sugars from the hot-water extracts of aged garlics were analyzed with GC/MS, LC/MS, and HPLC. The amount of HMF was higher than at $75^{\circ}C$ and increasing incubation period. Among free sugars, the only fructose except glucose and sucrose was formed and converted to HMF at high temperature and long incubation period. However, fructose formed in low temperature during making of aged garlic was rarely converted to HMF. This result indicates that formation of HMF can be dependent on the temperature and incubation period for making aged garlic.

Anticarcinogenic Effect of S-allylcysteine (SAC) (S-allylcysteine의 항암효과)

  • S-allylcysteine (SAC) is an aged garlic derived water soluble organosulfur compound and has been suggested to have anticarcinogenic activity against diverse types of cancer cells. This review summarizes the cellular signaling pathways and molecular mechanisms whereby SAC exerts its effects on cellular proliferation, apoptosis, cell cycle progression and metastasis based on the results from both in vitro and in vivo studies. SAC activates proapoptotic proteins including Bax and caspase-3, but suppresses antiapoptotic Bcl-2 family proteins to bring about cancer cell death through mitochondria-mediated intrinsic pathway. SAC also inhibits cellular proliferation by inducing cell cycle arrest in which SAC reduces expression and activation of NF-κB, cyclins, Cdks, PCNA and c-Jun, but elevates expression of cell cycle inhibitor proteins p16 and p21 through suppression of both PI3K/Akt/mTOR and MAPK/ERK signaling pathways. And, SAC inhibits invasion and metastasis of cancer cells by inducing suppression of both angiogenesis and epithelial-mesenchymal transition (EMT) through decreased cyclooxygenase (COX)-2 expression and increased E-cadherin expression which were then caused by suppression of inhibitory transcription factors Id-1 and SLUG from SAC-mediated inactivation of both MAPK/ERK and PI3K/Akt/mTOR/NF-κB signaling pathways. Furthermore, SAC prevents toxic compound-induced carcinogenesis by inducing antioxidant enzymes such as glutathione-s-transferase (GST). Thus, SAC can be considered as a potential chemotherapeutic agent for the prevention and treatment of cancer.
