Quality Comparison of Activated Carbon Produced From Oil Palm Fronds by Chemical Activation Using Sodium Carbonate versus Sodium Chloride

  • Using Na2CO3 versus NaCl as chemical activator, we compared the quality of activated carbon produced from oil palm fronds as raw material. These activators were selected for comparison because both are readily available and are environmentally friendly. In the manufacturing, we used Indonesian National Standard (SNI 06-3730-1995) parameters. For the quality comparison, we determined activated-carbon yield, moisture, ash, volatiles, and fixed-carbon contents; and adsorption capacity of iodine. The best characteristics, assessed by morphological surface analysis and Fourier transform infrared (FTIR) spectral analysis, were observed in the carbon activated by Na2CO3 at an activator concentration of 10% and carbonization temperature of 400 ℃. The results were as follows: activated-carbon yield, 84%; water content, 8.80%; ash content, 2.20%; volatiles content, 14.80%; fixed-carbon content, 68.60%; and adsorption capacity of iodine, 888.51 mg/g. Identification using the FTIR spectrophotometer showed the presence of the functional groups O-H, C=O, C=C, C-C, and C-H in the Na2CO3-activated carbon.

Status of Attachment Microalgae Taxa in the Korean Sea and Importance of their Research on Hull Ship Fouling (해양의 부착 미세조류 분류군 현황 및 선체부착 연구의 중요성)

  • Microalgae can attach to the surface of ships and then spread to various areas by means of ship transport. The introduction of invasive species through ships is recognized as a marine problem worldwide. Identification of attached microalgae is necessary to investigate such movement between countries through ships. In the present study, through analytical methods we reviewed research data to identify the taxa of domestic attached microalgae and assess the ecological impacts of such microalgae. A total of 87 genera and 153 species (143 species of diatoms, 10 species of cyanobacteria, and 4 genera of dinoflagellates) were identified as native attached microalgae in Korea, and diatoms accounted for 93% of the total. Most of these attached microalgae were identified through research on natural substrates such as seaweeds and bedrock, and some were also identified through experiments using artificial adherent plates. To date, there is no information on microalgae attached to international ships and introduced into Korea. Molecular genetic analysis and systematic management through on-site sampling of international ships, microscopic analysis, and meta-barcoding are necessary to assess the inflow and spread path of hull-attached marine alien species and evaluate the risk they pose to the domestic ecosystem.

Stability Assessment of Ivory Materials by Cleaning Agents (세척제에 의한 상아류 유물의 안정성 평가 연구)

  • This research was to evaluate conservational stability of cleaning treatment and color recovery by weak acid treatment on ivory. Before this experiment, had an ivory identification such as black light and schreger angle to find whether it is authentic or not. In wet cleaning treatment on ivory, used polar solvents such as distilled water, acetone, ethyl alcohol and acetic acid which usually used for cleaning ivory and bones. After cleaning took a experiment to evaluate conservational stability in surface observation, calculate weight and color difference. Following these results, these polar solvents were stable on the ivory samples. On the other hand, in weak acid treatment, used samples that were exposed by UV-A, UV-B, UV-C for 1 weeks, to restore its color from changed to original color. After treatment, found tiny cracks and exfoliation on the surface. Moreover, the color changed when it is treated by lemon juice, so, it is impossible to adapt.

Development of Microfluidic Chip for Enrichment and DNA Extraction of Bacteria Using Concanavalin A Coated Magnetic Particles (Concanavalin A가 코팅 된 자성 입자를 이용한 미생물 농축 및 유전자 추출 칩 개발)

  • The real-time enrichment and detection of pathogens are serious issues and rapidly evolving field of research because of the ability of these pathogens to cause infectious diseases. In general, bacterial detection is accomplished by conventional colony counting or by polymerase chain reaction (PCR) after DNA extraction. As colony counting requires considerable time to cultivate, PCR is an attractive method for rapid detection. A small number of pathogens can cause diseases. Hence, a pretreatment process, such as enrichment is essential for detecting bacteria in an actual environment. Thus, in this study, we developed a microfluidic chip capable of performing rapid enrichment of bacteria and the extraction of their genes. A lectin, i.e., Concanavalin A (ConA), which shows binding affinity to the surface of most bacteria, was coated on the surface of magnetic particles to nonspecifically capture bacteria. It was subsequently concentrated through magnetic forces in a microfluidic channel. To lyse the captured bacteria, magnetic particles were irradiated by a wavelength of 532nm. The photo-thermal effect on the particles was sufficient for extracting DNA, which was consequently utilized for the identification of bacteria. Our device will help monitor the existence of bacteria in various environmental situations such as water, air, and soil.

  • Risk assessment considers the situations and characteristics of the exposure environment and host. Various physiological variables of the human body reflects the characteristics of the population that can directly influence risk exposure. Therefore, identification of exposure factors based on the Korean population is required for appropriate risk assessment. It is expected that a handbook about general exposure factors will be used by professionals in many fields as well as the risk assessors of the health department. The process of developing the exposure factors handbook for the Korean population will be introduced in this article, with a specific focus on the general exposure factors including life expectancy, body weight, surface area, inhalation rates, amount of water intake, and soil ingestion targeting the Korean population. The researchers used national databases including the Life Table and the 2005 Time Use Survey from the National Statistical Office. The anthropometric study of size in Korea used the resources provided by the Korean Agency for Technology and Standards. In addition, direct measurement and questionnaire surveys of representative samples were performed to calculate the inhalation rate, drinking water intake, and soil ingestion.

A Review on Nitrate Source Identification using Isotope Analysis (동위원소분석을 이용한 질산염의 오염원 추적에 대한 고찰)

  • Nitrate ($NO_3^-$), a common surface water and groundwater pollutant, poses a serious environmental problem in regions with intensive agricultural activities and dense population. It is thus important to identify the source of nitrate contamination to better manage water quality. Due to the distinct isotope compositions of nitrate among different origins, the dual isotope analysis (${\delta}^{15}N$ and ${\delta}^{18}O$) of nitrate has been widely applied to track contamination sources. This paper provided the underlying backgrounds in the isotope analysis of nitrate, which included typical ranges of ${\delta}^{15}N$ and ${\delta}^{18}O$ from various nitrate sources, isotope fractionation, the analytical methods used to concentrate nitrate from samples, and the potential limitations of the dual isotope analysis along with the resolutions. To enhance the applicability of the dual isotope analysis as well as increase the ability to interpret field data, this paper also introduced several case studies. Furthermore, other environmental tracers including ${\delta}^{11}B$ and $Cl^-/Br^-$ ratios were discussed to accompany the dual isotope analysis for better assignment of contamination sources even when microbial transformation of nitrate and/or mixing between contaminant plumes occur.

Identification of nonregular indication according to change of grain size/surface geometry in nuclear power plant (NPP) reactor vessel (RV)-upper head alloy 690 penetration

  • During the fabrication process of reactor vessel head penetration (RVHP), the grain size of the tube material can be changed by hot or cold work and the inner side of the tube can also be shrunk due to welding outside of the tube. Several nonregular time-of-flight diffraction (TOFD) signals were found because of deformed grains. In this paper, an investigation of nonregular TOFD indications acquired from RVHP tubes using experiments and computer simulation was performed in order to identify and distinguish TOFD signals by coarse grains from those by Primary Water Stress Corrosion Crack (PWSCC). For proper understanding of the nonregular TOFD indications, microstructural analysis of the RVHP tubes and prediction of signals scattered from the grains using Finite Element Method (FEM) simulation were performed. Prediction of ultrasonic signals from the various sizes of side drilled holes to find equivalent flaws, determination of the size of the nonregular TOFD indications from the coarse grains, and experimental investigation of TOFD signals from coarse grain and shrinkage geometry to identify PWSCC signals were performed. From the computer simulation and experimental investigation results, it was possible to obtain the nonregular TOFD indications from the coarse grains in the alloy 690 penetration tube of RVHP; these nonregular indications may be classified as PWSCC. By comparing the computer simulation and experimental results, we were able to confirm a clear difference between the coarse grain signal and the PWSCC signal.

Bacterial Fruit Rot of Apricot Caused by Burkholderia cepacia in China

  • An unreported disease of apricot was observed in orchards in Zhejiang province, China. Symptoms started as water soaked lesions on the fruit surface. Later, water-soaked areas developed and spread to the entire fruit, resulting in soft rot of the whole fruit. The causal organism isolated from symptomatic fruits was identified as Burkholderia cepacia based on its biochemical and physiological characteristics and confirmed by the cellular fatty acid composition and Biolog data as well as 16S rRNA gene sequence analysis. The bacterial isolates caused similar symptoms when inoculated onto fruits of apricot. In addition, European plum, Japanese plum, nectarine and kiwifruit were susceptible to the B. cepacia pathogen. However, the B. cepacia pathogen failed to cause any visible symptoms when it was inoculated onto 16 other fruits. This is the first report of a bacterial disease of apricot caused by B. cepacia in China.

Identification and Physiological Characteristics of Microorganism Isolated from Spoiled Sweetened Adzuki Ann (부패된 팥앙금으로부터 분리된 미생물의 동정과 그 균의 생리적 특성)

  • In order to determine the causes of sweetened adzuki ann spoilage, the characteristics of microorganism isolated from spoiled adzuki ann were investigated. The isolated microorganism was gram-positive, roil-shaped and shore-forming bacteria; its surface was smooth and glazed. From the results of the assimilation test of 46 different biochemicals by the Vitec 2 Compact test and comparison of the cellular wall composition of fatty acid by the data bank of Midi sherlock system, the microorganism was identified as Bacillus subtilis, D-value of the B. subtilis spore was 4.85 min at $115^{\circ}C$, 0.69 min at $121^{\circ}C$ and 0.48 min at $125^{\circ}C$; Z-value was 9.71. The Bacillus subtilis growth was not observed below water activity of 0.92 at $45^{\circ}C$. However, bacteria growth increased gradually as water activity increased above 0.93.

Influences of the Sea Surface Wind on Current and Thermal Structures in the Southwestern Part of the East Sea of Korea (동해 남서해역의 해류 및 열구조에 미치는 해상풍의 영향)

  • Temporal variations of the path of the East Korea Warm Current(EKWC) which flows northward along the east coast of Korea were analysed to investigate whether the EKWC directly influences the existence of the so-called Warm Core in the Ulreung basin. From the 13 years(1975-1987) data of the Fisheries Research and Development Agency(FRDA), the $10^{\circ}C$ isotherm at the 100m depth and the depth of $2^{\circ}C$ isotherm and the temperature field at the 200m depth were used for identification of the path and the central position of the Warm Core. Sea surface winds computed from the surface pressure charts gave the monthly-averaged wind stress curl over the East Sea which was used for determination of the Sverdrup transport. And the mass transport stream functions were computed by use of the Sverdrup balance. The variations of the path show that the EKWC does not always have a fixed path and fluctuates with time. And the existence of the Warm Core is independent upon the presence of the EKWC even when the EKWC doesn't flow northward along the east coast of Korea. In view of the mass transport stream functions, the influences of the sea surface winds on the branching of the Tsushima Warm Currents and the presence of the EKWC were investigated. The presence of the EKWC may be hindered by the southward flow driven by the sea surface winds when the Tsushima currents are rather weak. A very weak correlation exists between the north-south component of the Sverdrup transport and the position of the Warm Core. However, a small but significant part of the southward transport across the latitudinal line of $38^{\circ}N$ indicates that cold water from the northern part of the East Sea may be driven and be forced to flow beneath the permanent thermocline in such a way that the thermal structure of the Warm Core and its position might be changed.
