Induction of Anticarcinogenic Enzymes by Dichloromethane-soluble Fraction of Physalis alkekengi var. francheti Hort. in Mouse Hepatoma Cells

  • Physalis alkekengi var. francheti Hort. is known as an insecticide and traditional remedy for liver related diseases. Therefore, this study investigated the chemopreventive effects of extracts and several solvent fractions (n-hexane, dichloromethane, n-butanol, water) of Physalis alkekengi var. francheti Hort. First, their cytotoxicity and NQO1 activity were measured using an MTT assay, plus a quinone reductase [NAD(P)H dehydrogenase (quinone); NAD(P)H: (quinone acceptor) oxidoreductase, EC 1.6.99.2]-inducing activity assay was performed using cultured murine hepatoma cells (Hepa1c1c7) and its mutant cells(BpRc1). The reduction of electrophilic quinones by NQO1 is an important detoxification pathway and major mechanism of chemoprevention. When compared with the other solvent soluble fractions with different polarities, the dichloromethane fraction of Physalis alkekengi var. francheti Hort. showed a higher NQO1-inducing activity that was also dose-dependent. Moreover, the dichloromethane fraction of Physalis alkekengi var. francheti Hort. induced ARE-luciferase activities in HepG2-C8 cells that were generated by transfecting the ARE-luciferase gene construct, suggesting the Nrf2-ARE-mediated induction of anti-oxidative enzymes. In conclusion, the dichloromethane-soluble fraction of Physalis alkekengi var. francheti Hort. showed a relatively strong induction of detoxifying enzymes, thereby meriting further study to identify the active components and evaluate their potential as cancer preventive agents.

  • A fraction (CMEx-AH-${\beta}$) of water-soluble polysaccharides, showing selective antitumor activities, was isolated from the mycelia of solid cultured Agaricus blazei Murill by hot-water extraction, ethanol precipitation, and series of chromatography. Chemical characteristics of CMEx-AH-${\beta}$, were as follows: carbohydrate content, 48%; monosaccharide composition, Man:Glu:Gal (2:93:5); molecular weight, $2{\times}10^5$; uronic acid content, 6.2%. Fundamental structure of CMEx-AH-${\beta}$, was deduced as ${\beta}-(1\;{\rightarrow}\;6)$-D-glucan with ${\beta}-(1\;{\rightarrow}\;3)$-D-glucosidic side chains based on results of methylation and $^{13}C$ NMR spectroscopic analyses.

  • Fraction (PMEx-AH-${\beta}$) of water-soluble polysaccharide, showing stimulating activity against macrophages, was isolated from mycelia of solid cultured Phellinus linteus by hot-water extraction, ethanol precipitation, and chromatography. Chemical characteristics of PMEx-AH-${\beta}$ were as follows: carbohydrate content, 71%; monosaccharide composition, Man:Glu:Gal (9:64:27); molecular weight, $1-7{\times}10^4$; uronic acid content, 6.8%. Fundamental structure of PMEx-AH-${\beta}$ is deduced as ${\beta}$-($1{$\rightarrow}6$)-D-glucan with ${\beta}$-($1{\rightarow}3$)-D-glucosidic side chains based on methylation analysis.

  • Extract of water-soluble polysaccharide (CFWx), showing inhibiting activity on ${\alpha}-glucosidase$, was prepared from the fruiting bodies of Cordyceps militaris by hot-water extraction, and ethanol precipitation. Chemical characteristics of CFWx were as follows: carbohydrate content 30% including 16% of uronic acid; 51% protein content; monosaccharide composition, Man:Glu:Gal (30:43:27); molecular weight $3-5{\times}10^4$. CFWx was further purified by ion-exchange, gel-permeation, and affinity chromatography and $CFWx-AH-{\alpha}$ fraction was isolated. Fundamental structure of $CFWx-AH-{\alpha}$ was deduced as ${\alpha}-(1{\to}4$)-D-glucan with ${\alpha}-(1{\to}3$)- and/or ${\alpha}-(1{\to}6$)-D-glycosidic side chains based on methylation analysis.

감과 대추의 연화중 가용성 단백질의 변화 (Changes in Soluble Proteins during Softening of Persimmon and Jujube Fruits)

  • 과실의 성숙 연화동안에 단백질의 변화에 대해 연구하고자 완숙 연화된 감과 대추에서 용해성에 따라 추출한 수용성 단백질, 염가용성 단백질 및 세포벽 단백질의 함량 변화와 각 단백질의 gel filtration chromatogram의 pattern 변화를 조사하였다. 두 과실의 수용성 단백질은 완숙감과 대추에서 각각 98.2, 117.1mg/100g-fr.wt.이었으며, 연화된 감과 대추에서는 각각 112.6, 183.4mg/100g-fr.wt.로 과실의 연화에 따라 증가하였다. 염가용성 단백질도 감의 경우 완숙감에서 51.7mg, 연시에서 133.2mg/100g-fr.wt.로 증가하였고, 대추의 경우에도 완숙·연화 대추 각각 101.8, 139.5mg/100g-fr.wt.로 현저한 증가를 보였다. 이 에 반해 세포벽 단백질 함량은 두 과실 모두 감소하였다. Gel filtration에서 연시의 수용성 단백질 chromatogram은 완숙감에 나타난 1개의 peak 이외에 다소 분자량이 작은 2개의 peak가 함께 존재 했으며 , 대추의 수용성 단백질은 완숙 대추와 연화 대추 모두 3개의 peak로 존재했다 염가용성 단백질 및 세포벽 단백질의 get filtration에서는 완숙 연화 과실에서 거의 유사한 경향의 chromatogram을 나타내었다. 또한 연좌된 감과 대추의 세포벽 단백질에서 단백질 함량이 감소하는 fraction은 수용성 및 염가웅성 단백질 분획에서 단백질 함량이 증가하는 fraction과 거의 일치하였다.

화살나무 및 느릅나무 추출물이 면역계세포의 활성에 미치는 영향 (Effect of Euonymus alatus and Ulmus clavidiana var japonica on the immune system)

  • We have previously shown that crude water extract of Euonymus alatus (EA) had strong prophylactic effect against chemically induced-and tumor cell implanted-cancer, and that the mechanisms responsible for its antitumor effects were due to nonspecific enhancement of the NK cell activities and the cell mediated immunity. However, it was unknown that any components of crude extract did work so, since it consisted of several components. In this paper, we fractionated the crude watar EA-extract into several fraction such as hexane-, ethylether-, ethyl acetate-, n-butanol- and water soluble-fraction, and screened the immune regulating activities of each fraction by the evaluation of lymphokine production and activated lymphocyte proliferation. As a result of the component fraction of EA-extract, it was found that n-butanol fraction was a potent immunostimulator, and the remained water soluble fraction also contained some stimulator, But, other fraction did not showed any remarkable effect. It is therefore suggested that EA-glycosides in n-butanol fraction may be new one of the potent biological response modifiers. The present study was also undertaken in an efforts to investigate the effects of elm-bark(EB, Ulmus clavidiana var japonica), which has been used for curing ulcer and inflammation as a folk medicine without any kind of experimental evidence to support this, on the cellular- and humoral-immune responses, lymphocyte function and NK cell activities in mice. Regardless of time and duration of EB-treatment, Arthus reaction and antibody response to SRBC were not modified by EB, but delayed hypersensitivity to SRBC was significantly enhanced only when EB was treated prior to SRBC-sensitization. EB slightly inhibited the proliferation responses of splenocytes to PHA-stimulation, but it significantly augmented the responses of these cells to S aureus Cowan 1 and Con A-activation, and these effects were manifested only when EB was added at culture initiation. EB did not influence Ig secretion of spleen cells but it significantly augmented the Con A-induced 1L 2 and MIF production of splenocytes. NK cell activities of splenocytes were markedly riled when effector cells were pretreated with EB and this augmentation was dine to the increase of binding affinity of effector cells to target cells and the target cell lytic activities of effector cells. These results led to the conclusion that EB triggers increase of cellular immune responses, such as delayed hypersensitivitiy, lymphokine production and NK cell activities. Also these results suggested that EB contains potent immune stimulants, which may provide the rational basis for their therapeutic use as one of the new biological response modifiers.

교외지역 수용성유기탄소 내 식생연소 및 2차 유기탄소에 의한 기여량 연구 (Contribution of Biomass Burning and Secondary Organic Carbon to Water Soluble Organic Carbon at a Suburban Site)

  • The $PM_{2.5}$ samples were collected for every 6th day during one year at a suburban site in the Namwonsi, Jeollanamdo, Republic of Korea. Samples were analyzed for elemental carbon (EC), organic carbon (OC), and water-soluble organic carbon (WSOC), and levoglucosan. Although the water-soluble fraction of fine particulate OC consistently showed over a year, levoglucosan fraction of WSOC varied considerably from month to month. In this study, non-biomass-burning WSOC ($WSOC_{NBB}$) and biomass-burning $WSOC_{BB}$ were calculated with measurements of organic source tracer, levoglucosan, to better understand the temporal distribution and sources of WSOC. Two methods of predicting the secondary organic carbon from the biomass-burning $WSOC_{BB}$ Method and the EC-tracer Method were compared. Poor correlations between SOC estimated between two methods suggested that the use of the EC tracer method to estimate SOC may be significantly flawed. Direct measurements of levoglucosan and WSOC can provide a reasonable estimate of secondary organic carbon concentrations.

Radical Scavenging and Antioxidant Activities of Fermented Laminaria japonica Extracts

  • Radical scavenging and antioxidant activities of Laminaria japonica and fermented its extracts were evaluated. Freeze-dried L. japonica was fermented by Aspergillus oryzae and extracted with distilled water. The extract solution was mixed with ethanol and centrifuged. The supernatant was ethanol soluble fraction, non-polysaccharide fraction (ESF), and residue was ethanol insoluble precipitation, polysaccharide fraction (EIP). ESF was subjected to sequential fractionation with dichloromethane, ethyl acetate, butanol and water. To determine the radical scavenging and antioxidant activities of these, DPPH radical, hydroxyl radical, superoxide anion radical scavenging activities and linoleic acid oxidation were tested. Among the extracts, ESF of fermented L. japonica showed the highest radical scavenging activity. The ESF showed DPPH radical scavenging activity of 64.33% at concentration of $50{\mu}g/mL$. It was higher than 57.70% of vit. C. Ethyl acetate and butanol fraction had high value of radical scavenging and antioxidant activities, especially butanol fraction of fermented L. japonica was 79.48 % of hydroxyl radical scavenging activity at concentration of $50{\mu}g/mL$. The fermented L japonica had radical scavenging and antioxidant activities higher than L. japonica. These results suggest that fermented L japonica is healthy food having radical scavenging and antioxidant activities.

Extraction and Bioassay of Allelochemicals in Jerusalem Artichoke

  • Helianthus tuberosus has been known to inhibit the growth of weeds and other plants sharing its habitat. This study was conducted to identify the allelochemicals of Helianthus tuberosus which were extracted with water and solvents. Aqueous extracts of leaf, stem, root, tuber and tuber peel of Helianthus tuberosus except tuber did not show significant differences in phytotoxicity to alfalfa seedlings. It was considered that Helianthus tuberosus contained fewer or less potential water-soluble substances that were toxic to alfalfa. Methanol extract of leaves of Helianthus tuberosus was sequentially partitioned in increasing polarity with n-hexane, ethylacetate and n-butanol. Each extract had a yield of 148, 12, 15.7 and 9.5g, respectively. Inhibitory effects on germination of alfalfa seeds treated with four fractions were not significantly different. But the significant reductions on hypocotyl length were observed for all the solvent extracts. Among the four fractions, the ethylacetate fraction showed the most significant inhibition effect on bioassay with alfalfa. Further separation of the active ethylacetate fraction by open column chromatography led to the 25 subfractions. In bioassay of each sub-fraction with alfalfa seeds, sub-fraction No. 13 showed the most inhibitory effect on seedling growth. $^1$H NMR and gas chromatography-mass spectrometry analysis revealed that sub-fraction No. 13 was the mixture of straight-chain saturated fatty acids.

감마선 조사된 장류 물추출 분획의 유전독성학적 안전성 평가 (Genotoxicological Safety on Water-Soluble Fraction of Gamma Irradiated Korean Soybean Fermentation Foods)

  • 우리나라 전통 장류인 간장, 된장, 고추장, 청국장의 위생화를 위한 방사선 조사기술의 이용 가능성을 검토할 목적으로 방사선 조사를 실시한 후 유전독성학적 안전성 실험의 일환으로 Ames test를 실시하였다. 감마선 조사 및 비조사된 간장, 된장, 고추장, 청국장 물추출 분획의 Salmonella typhimurium TA98, TA100, TA1535 및 TA1537에 대한 복귀변이 집락수를 조사한 결과, 대사활성계 도입 및 부재시 모두, 모든 시험균주에서 시험적용 농도인 10,000 $\mu\textrm{g}$/plate까지의 농도에서 복귀변이 집락수의 농도 의존적인 증가 혹은 감소를 보이지 않아 20 kGy의 고선량 감마선 조사된 간장, 된장, 고추장, 청국장은 돌연변이원성이 없는 것을 확인할 수 있었다.
