Antioxidant Activity of Roasted Defatted Perilla Seed

  • The antioxidant activity of roasted defatted perilla (Perilla frutescens) seed was determined by measuring its radical scavenging effect on 1,1-diphenyl-2- picrylhydrazyl (DPPH) radicals, inhibitory activity on total reactive oxygen species generation in kidney homogenates using 2',7'-dichlorodihydro-fluorescein diacetate, and scavenging effect on authentic peroxynitrites. The methanolic extract of roasted defatted perilla seed showed strong scavenging activity in both DPPH and peroxynitrite radicals, and thus fractionated with several solvents. The antioxidant activity potential of the individual fraction was in the order of ethyl acetate>n-butanol>dichloromethane>water>n-hexane fraction. The ethyl acetate soluble fraction exhibiting strong antioxidant activity was further purified by repeated silica gel and Sephadex LH-20 column chromatography. Luteolin was isolated as one of the active principles from the ethyl acetate fraction, together with the inactive chrysoeriol and apigenin.

A brief method for preparation of gintonin-enriched fraction from ginseng

  • Background: Ginseng has been used as a tonic for invigoration of the human body. In a previous report, we identified a novel candidate responsible for the tonic role of ginseng, designated gintonin. Gintonin induces $[Ca^{2+}]_i$ transient in animal cells via lysophosphatidic acid receptor activation. Gintonin-mediated $[Ca^{2+}]_i$ transient is linked to anti-Alzheimer's activity in transgenic Alzheimer's disease animal model. The previous method for gintonin preparation included multiple steps. The aim of this study is to develop a simple method of gintonin fraction with a high yield. Methods: We developed a brief method to obtain gintonin using ethanol and water. We extracted ginseng with fermentation ethanol and fractionated the extract with water to obtain water-soluble and water-insoluble fractions. The water-insoluble precipitate, rather than the water-soluble supernatant, induced a large $[Ca^{2+}]_i$ transient in primary astrocytes. We designated this fraction as gintonin-enriched fraction (GEF). Results: The yield of GEF was approximately 6-fold higher than that obtained in the previous gintonin preparation method. The apparent molecular weight of GEF, determined using sodium dodecyl sulfate-polyacrylamide gel electrophoresis, was equivalent to that obtained in the previous gintonin preparation method. GEF induced $[Ca^{2+}]_i$ transient in cortical astrocytes. The effective dose (ED50) was $0.3{\pm}0.09{\mu}g/mL$. GEF used the same signal transduction pathway as gintonin during $[Ca^{2+}]_i$ transient induction in mouse cortical astrocytes. Conclusion: Because GEF can be prepared through water precipitation of ginseng ethanol extract and is easily reproducible with high yield, it could be commercially utilized for the development of gintoninderived functional health food and natural medicine.

Antimutagenic and Cytotoxic Effects of Ethanol Extract from the Inonotus obliquus (차가버섯 분획물의 항돌연변이 활성 및 암세포 성장억제효과)

  • This study was performed to determine the antimutagenic and cytotoxic effects of ethanol extract, ethylacetate fraction, water fraction and water soluble and insoluble polysaccharides I and II isolated from Inonotus obliquus using Ames test and SRB assay. Each sample itself did not show mutagenic effect. Among samples, the water unsoluble polysaccharides II in the presence of 50 $\mu\textrm{g}$/plate showed the strongest antimutagenic effect with over 90% against MNNG, 4NQO, B(a)P and Trp-P-l. However, ethyl acetate fraction (1 mg/mL) which had 90.8%, 94.3% and 83.5% inhibitory effect against on MCF-7, A549, AGS showed the strong cytotoxic effect compared to other samples.

Desmutagenic Effect of Leonurus sibiricus L. to Aflatoxin B1 in Salmonella Mutation Assay (아플라톡신에 대한 익모초의 돌연변이 억제 효과)

  • By the 505 chromotest which utilized Escherichia bolt PQ 37, Korean medicinal plants had been screened to Investigate the antimutagenic effect to aflatoxin B1(AFBl). Ikmocho(IMC, Leonurus sibiricus L.) was extracted with hot water. The extract was not found to be mutagenic in the Salmonella mutation test with or without metabolic activation, and the extract was showed to possess the antimutagenic properties towards AFB1-induced metation. The mutagenicity of AFB1 was inhibited by methanol soluble fracstion (IMC-MS) in dose-dependent. However, water-soluble fraction exhibited comutagenic activity. The greatest inhibitory effect of IMC-MS on AFB1 mutagenicity occurred when IMC-MS was first incubated, AFB1 followed by a second incubation with the cells and 59 mixture. Also lower inhibition was occurred when S9 mixtures were first incubated, with IMC-MS followed by a second incubation with AFBI. The results of the sequential incubation study support the probability that one mechanism of inhibition could involve the formation of chemical complex between IMC-MS and AFB1 rather than deactivation of S9 enzyme.

Dynamic Runoff of Non-point Sources Pollutants from Agricultural Areas (농촌지역에서 유출시간에 따른 비점오염물질의 유출평가)

  • In this paper, data collected from 22 different rural watersheds during stormflow conditions were analyzed. Those watersheds consisted of forest and cultivated land. EMC data analysis indicates that as agricultural land use increases, EMC values of TSS, COD and TN clearly tends to increase, but TP does not show a significant increase. Pattern of the pollutographs mostly has a similarity in hydrograph shape except nitrogen which inherently shows a variability and complication. The fraction of soluble reactive-P to TP increases as cultivated land use increases while mobile-nitrogen portion was higher in the runoff from forested watersheds than agricultural areas. During stormflow, pollutograph of the nitrogen was determined mainly by change in Particle-TKN as other pollutants but its effect is thought to be masked by decrease of dissolved form of nitrogen due to the dilution.

Antioxidative Activity of Methanolic Extracts from Seaweeds (해조류 메탄올 추출물의 항산화효과)

  • In this study, seven species of seaweeds (Sargassum horneri, Grateloupia filicina, Kjellmaniella crassifolia, Porphyra tenera, Ecklonia stolonifera, Scytosiphon lomentaria, Agarum cribrosum) from Gangwon coast were collected for antioxidative assays. The methanol soluble and insoluble (water soluble) fraction which were prepared from 75% methanol extraction were assayed by DPPH method and deoxyribose assay. The antioxidative activity of methanol soluble fraction of Agarum, Scytosiphon and Sargassum reached to 26.60%, 24.28% and 23.40%, respectively. The antioxidative activities of methanol insoluble fractions of the Kjellmaniella and Porphyra were assayed to 29.70% and 21.01% which were higher than methanol soluble fractions with the values of 9.26% and 8.66%, respectively. The results from DPPH assay and deoxyribose assay showed the methanol soluble fractions of Agarum and Sargassum have strong antioxidative activity. However the methanol insoluble (water soluble) fractions of seaweeds did not show any meaningful antioxidant activity excepted Kjellmaniella and Porphyra.

  • Addition of one percent cucumber, as a Kimchi component, showed the stimulation of lactic acid fermentation, by reducing the lagphase, and resulted the early matured Kimchi. Alcohol-soluble fraction from cucumber serum accerelated the growth of Lactobacillus plantarum, p-1, p-2 newly isolated strains from cucumber pickle, but had no effect on the growth rate of Streptococus faecalis. The growth promoting activity was inactivated by dialysis against water and heat treatment at $120^{\circ}C$ for 10 minutes. Softness of cucumber tissues enables to extract easily out a substance, which is thought one of the important factors for the stimulation of me growth of Lactic acid bacteria.

Isolation of Flavonol Glycoside Related to Antioxidant Activity from Hippophae rhamnoides Leaves (비타민나무 잎으로부터 항산화활성 관련 Flavonol Glycoside 분리)

  • The leaves of Hippophae rhamnoides were extracted with methanol and then further fractioned with hexane, ethyl acetate, butanol and water in order to investigate biological activity. Two flavonol glycosides were isolated and identified from ethyl acetate soluble fraction, which showed the strongest antioxidant activity ($RC_{50}$ = 4.33 ${\mu}g/ml$). Isolated two compounds have shown strong free radical scavenging activity. Especially, quercetin 3-O-glucoside ($RC_{50}$ = 2.60 ${\mu}g/ml$) was more active than ${\alpha}$-tocopherol ($RC_{50}$ = 4.67 ${\mu}g/ml$). Total phenol and flavonoid contents were the highest to 4.17 and 1.14 mg/ml in the ethyl acetate soluble fraction. In addition, ethyl acetate soluble fraction was shown to the strongest ${\alpha}$-glucosidase inhibitory activity ($IC_{50}$ = 137.88 ${\mu}g/ml$) among the fractions. The results suggest that leaves of H. rhamnoides could be a potential source of natural antioxidant.

Antioxidant Activity of the Extractives from the Needles of Picea abies Karsten (독일가문비(Picea abies Karsten) 잎 추출성분의 항산화 활성)

  • The dried needles (1.5 kg) of Picea abies Karsten were ground, extracted with acetone-$H_2O$ (7:3, v/v), concentrated, and fractionated with a series of n-hexane, methylene chloride, ethyl acetate and water on a separatory funnel. Each fraction was freeze dried, then a portion of ethyl acetate soluble powder was chromatographed on a Sephadex LH-20 column using a series of aqueous methanol and ethanol-n-hexane mixture as eluents. The isolated compounds were identified by cellulose TLC, $^1H$-, $^{13}C$-NMR, FAB and EI-MS. (+)-catechin (compound I), (-)-epicatechin (compound II), kaempferol-3-O-${\beta}$-D-glucopyranoside (compound III), 4-hydroxyacetophenone (compound IV) were isolated from the ethyl acetate soluble fraction and (+)-catechin (compound I), protocatechuic acid (compound V) were isolated from the $H_2O$ soluble fraction of P. abies needle. The antioxidative activities of each fraction and the isolated compounds were tested by DPPH radical scavenging method, and EtOAc soluble fraction, (+)-catechin and (-)-epicatechin showed similar values to ${\alpha}$-tocopherol and BHT as controls.

Diagnosis of Wastewater Treatment Processes through the Wastewater COD Fractionation and Process Simulation I : Wastewater COD Fractionation (유입하수 유기물 분류 및 공정모사를 통한 하수처리공정 진단 I : 유입하수 유기물 분류)

  • The simulation programs used for diagnosis and design of activated sludge process require organic fractions in municipal wastewater as the input variables. However, methods for characterizing organic fractions are still under development, and are not standardized. In this study, total COD of municipal wastewater was experimentally subdivided into readily and slowly biodegradable COD as well as soluble and particulate inert COD. The COD fractionation of the three municipal wastewater for one year shows linear relationship between each COD fraction and TCOD concentration with around 100% COD balance. This result means that the COD fraction do not vary very much with time, although the actual influent concentrations vary significantly with time and day. Therefore, the experimentally subdivided COD fractions can be utilized as wastewater specific parameters for the simulation of activated sludge processes.
