Seasonal variation of antimicrobial and antioxidative activity in Artemisia princeps var. orientalis

  • Leaves from natural populations of Artemisia princeps var. orientalis in Suncheon of South Korea were examined for antimicrobial and antioxidative activities monthly (April through October). The antimicrobial activity of the ethyl acetate and water fractions of crude methanol extract from the plant collected monthly against three gram-positive, two gram-negative and one lactic acid bacteria were studied. The ethyl acetate and water fractions of crude methanol extract from A. princeps var. orientalis collected in August and September had the highest antimicrobial activity. Minimum inhibitory concentrations (MIC) for each strain by ethyl acetate fraction of methanol extract from A. princeps var. orientalis was different depending on strains and sampling months. MIC for each strain was the highest in the sample of April, followed by October. In specific, MIC for Bacillus cereus was around 0.25mg/disc in the sample of April. The MIC for Staphyllococcus aureus was 0.01mg/disc in the sample of July, August, and September. It was the lowest value among tested strains and samples. And the antimicrobial activity for Lactobacillus plantarum was not found at any concentrations and sampling months. The in vitro antioxidative activities of methanol extract from Artemisia princeps var. orientalis collected monthly were determined by the scavenging of DPPH radical and inhibition of xanthine oxidase activity. The DPPH scavenging activity of the extracts from the plant sampled in May, June and July were higher than any other months. The xanthine oxidase activity of the extract of A. princeps var. orientalis collected in April and May showed the greatest activity, it is different with antimicrobial activity.

Antioxidant Activity of Wa-song (Orostachys japonicus A. Berger) According to Drying Methods (건조 방법에 따른 와송의 항산화 효과)

  • Antioxidant activity of wa-song (Orostachys japonicus A. Berger) was analyzed to clarify the influence of extractive solvent and drying method such as sun, hot-air and freeze drying. The contents of total phenols and flavonoids were significantly higher in 95% ethanol extracts than water extracts. Ability of reducing power and DPPH radical, hydroxyl radical and nitrite scavenging ability were higher in the order of hot-air dried sample> freeze dried sample> sun dried sample and these abilities were also higher in 95% ethanol extracts than water extracts. In conclusion, antioxidant activities of wa-song extracts were in proportion to the contents of total phenols and flavonoids. Also, hot-air drying is the superior method for the enhancement of antioxidant activity of wa-song.

Quality Characteristics of Marinated Chicken Breast as Influenced by the Methods of Mechanical Processing

  • The aim of this study was to investigate the effects of various marination processes on the quality characteristics of chicken breast prepared with chicken feet gelatin and wheat fiber. The chicken feet gelatin was swollen with hydrochloric solution (0.1 N HCl, pH $1.31{\pm}0.02$) and dehydrated by freeze-drying. The composition (w/w) of the marinade was water (10%), soy sauce (12%), phosphate (0.3%), wheat fiber (1.5%), and chicken feet gelatin (1.5%). Three samples of chicken breast were manufactured with Tumbler (only tumbler), Tenderizer (tenderizer and tumbler), and Injector (injector and tumbler). The water content of the Injector sample was significantly higher than those of the Tumbler and Tenderizer samples (p<0.05). During heating, the lightness of all chicken breasts increased and the redness decreased. The tumbling and cooking yield of the Injector sample were significantly higher than those of the Tumbler and Tenderizer samples (p<0.05). The shear force of the Tenderizer sample was significantly lower than that of the Tumbler and Injector samples (p<0.05). No significant differences, except for color, were observed in the sensory analysis of the samples. Thus, the proper selection of mechanical processing is important to improve the quality characteristics of marinated chicken breast, considering the types of final products.

Revision of Ecological Score of Benthic Macroinvertebrates Community in Korea (한국의 저서성 대형무척추동물 생태점수 개선)

  • In 1997, the environmental score (Q) corresponding the tolerance value of Korean benthic macroinvertebrate species and the related biotic score, the ecological score of benthic macroinvertebrates (ESB) were proposed; ESB was similar to Beck's biotic index or Chandler's biotic score. This study was carried out to revise the Q values of individual species and the assessment scheme of ESB based on the taxonomic performance and ecological information accumulated since then. The original ESB was renamed as TESB (total ESB), and AESB (average ESB) was newly proposed. AESB is calculated by dividing the TESB (for a given station) by the number of species present in the sample. In this study, TESB showed a positively skewed distribution, while AESB showed a negatively skewed distribution. The correlation between TESB and the concentration of $BOD_5$ was a little stronger than that of the original ESB. TESB showed a very strong correlation (correlation coefficient r = 0.98) with Margalef's species richness, of which correlation coefficient was higher than that of AESB (r = 0.85). AESB showed a strong correlation (r = -0.79) with the concentration of $BOD_5$, while TESB showed a weaker correlation (r = -0.67). Applying TESB and AESB together in an assessment of the environment may be comprehensive because the physical and chemical states of the environment can be evaluated together. AESB is less dependent on the sample size, while TESB tends to increase as the sample size increases. In the evaluation of the environment using TESB, it is necessary to standardize the methods on monitoring.

Sediment Discharge Based on a Time-Integrated Point Sample (연속점 채취를 이용한 유사량 계산)

  • A procedure for computing total suspended sediment load is presented based on a single point-integrated sample, a power velocity distribution, and Laursen's sediment concentration distribution equation. The procedure was tested with field data from the Rio Grande River. Computed concentrations agreed well with depth-integrated measurements corrected for unmeasured load using nominal values of $\beta$, $\kappa$ and w. Even better agreement was obtained when site-specific data were used to define the x and z exponents of the velocity and concentration distributions. The difference between total suspended load computed using a single measurement and this procedure and conventional computations based on depthintegrated measurements is well within sampling error. There are major advantages in estimating total suspended load using a single time-integrated suspended-sediment point sample. Less field time is required; sampling costs are greatly reduced; and sampling can be more frequent and better timed to measure the changing sediment load. Single-point sampling makes automatic sampling procedures more feasible.

The Development of Continuous Flow Method Through Microwave Oven for the Analysis of Metal Oxides in Water by ICP-AES (ICP-AES에 의한 수중의 금속 산화물 직접분석을 위한 연속흐름 Microwave 용해장치 개발 연구)

  • The method described offers rapid and efficient sample preparation using on-line microwave digestion of metal oxides in water sample with direct elemental detection by ICP-AES. The open tubing digestion system(OTD) and the restraint tubing digestion system(RTD) for flow injection(FI) were designed and tested to find the optimum conditions. Comparison of OTD and RTD indicated that RTD was 3 times faster on the digestion time, and 10 times higher on sample mass. Finally, the results of RTD agree well with those by conventional microwave open vessel in all cases and show good precision; Fe and Cu show good with about 5% of RSD, while Zn and Co more or less than 10% RSD.

  • Throughout the world, physical and chemical analyses of horticultural substrates are carried out in many different ways at the different laboratories. In Europe, standardization in properties and analytical methods of horticultural substrates has been a topic over the last decades. As a result, the CEN methods as European standard methods for the physical and chemical analyses were introduced and the final draft was reported in 1999 by CEN(Committee for European Standardization). Dry matter and moisture content are analyzed after drying samples at $103^{\circ}C$. Laboratory compacted bulk density is analyzed by determining the weight of sample compacted in the test cylinder with constant volume. Dry bulk density, particle density, total pore space, water volume, air volume and volume shrinkage are determined by saturating, draining and drying the sample using double rings and a sand suction table. pH and EC are analyzed by 1:5(sample:distilled water) extraction method on the basis of volume. Organic matter and ash content are determined after drying and combusting the samples. Now, CEN methods are being regarded almost as European standard methods. Further study needs to be carried out for universal applicability of the CEN methods to all the substrates.

Annual Intensities (2016-2017) Analysis of Energy Use and CO2 Emission by End Use based on Measurements of Sample Apartment Units (표본건물 계측에 의한 공동주택 세대에서의 용도별 에너지사용량 및 CO2 배출량 연간 원단위 (2016 - 2017) 분석)

  • In this study, annual site and primary energy use intensities (EUIs) and CO2 emission intensities (CEIs) per area by end use were estimated based on the measurement data from June 2016 to May 2017 of 50 sample apartment units in Seoul. In addition, estimated site EUIs by end use were compared to the U.S. Residential Energy Consumption Survey (RECS) 2009 data. Site EUIs by end use were found to be in the order of heating > electric appliance > domestic hot water > cooking > lighting > cooling > air movement. In the case of primary EUIs and CEIs by end use, electric appliance was found to be the largest. As results of comparison with the RECS 2009 data, it was found that site EUIs were very similar for heating, domestic hot water and electric appliance, etc., but slightly different for cooling. The number of sample apartment units will continue to increase until 2020 (total number of samples 200) and intensities data by end use will be continuously updated through continuous collection of measurement data.

Experimental Evaluation of Particulate-matter Filtration Performance of a Bottom Ash-Silica Sand Mixture (석탄 저회-규사 필터의 입자상물질 여과 성능 실험적 평가)

  • Permeable pavement technology allows the penetration of rainfall into the roadbed, thereby reducing surface runoff and enhancing water quality. The water quality can be improved by adding a filter layer to the permeable pavement. This study analyzes the permeability performance and particulate-matter removal efficiency of a bottom ash-silica sand filter. The performances of five filters with bottom ash and silica sand as the basic materials were evaluated on particulate matter sized 60 ㎛ or smaller. The pure silica sand sample and pure bottom ash sample delivered an average removal efficiency of around 70%. The removal efficiency of the mixed sample was approximately 90%, exceeding the recommended reduction rate (80%) at non-point pollution reduction facilities. In future work, the filter performance should be further verified on permeable pavement.

Qualitative analysis of some kinds of petroleum (thinner, gasoline, kerosene, and diesel oil) by gas chromatography (기체 크로마토그래피를 이용한 몇 가지 석유류(시너, 휘발유, 등유 및 경유)의 정성분석)

  • The evidence containing some kind of petroleum at the fire spot was analyzed by gas chromatography to identify a fire's sources. To extract some petroleum from fire evidence, 10.0 mL of n-hexane was added in this solution, and it was shaken for 30 minutes. To identify a kind of petroleum in fire evidence, the prepared n-hexane solution was injected and analyzed in the gas chromatograph. The chromatogram of sample was different from those of thinner and gasoline that have low boiling point, and shown different peak pattern to heating and boiler oils. But it was similar to the chromatogram of diesel oil. After small amount of diesel oil was added to the sample, the area of characteristic peaks was increased more than those of raw sample. From the results, the kind of petroleum in the fire evidence was diesel oil.
