The objective of this study was determined to evaluate ${\alpha}$-amylase, ${\alpha}$-glucosidase, pancreatic lipase inhibition in vitro and DPPH radical scavenging activity of the several Korean resources plants. The ${\alpha}$-amylase inhibitory activity of Salicornia herbacea, Erythronium japonicum (flower) and Phragmites communis (root) in water extract showed relatively high 62.8%, 66.5% and 69.3%, respectively. The ${\alpha}$-amylase inhibitory activity of Citrus junos (pericarp) and Cornus officinalis in methanol extract was found to have an effect with 32.8% in Citrus junos (pericarp) and 60.9% in Cornus officinalis. Corylopsis coreana in both water and methanol extract had the highest ${\alpha}$-glucosidase inhibitory activity of 81.7% and 89.5%, while the extract of Portulaca oleracea, Ficus carica and Citrus junos was not measured ${\alpha}$-glucosidase inhibitory activity at given experiment concentration. Depending on the extraction solvent and the plant species, it was observed that there was a significant difference in ${\alpha}$-glucosidase inhibitory activity. The pancreatic lipase inhibitory activity showed relatively higher in the methanol extract than water extract except pericarp of Citrus junos. The DPPH radical scavenging activity of selected plants was much difference between measured plant species, and showed that the increase was proportional to the concentration. These results suggested that selected plants had the potent biological activity on carbohydrate, lipid Inhibitory activity and antioxidant activity, therefore these plant resources could be a good materials to develop medicinal preparations, nutraceuticals or health functional foods for diabetes or obesity.
In this study, the anticoagulant compounds in Korean red ginseng (KRG) were investigated. KRG powder was extracted using hot methanol, and the methanol extract was fractionated into n-hexane, ethylacetate, n-butanol, and aqueous fractions by solvent partitioning. The remains from the methanol extraction were further extracted with water and then dialyzed to obtain low and high molecular weight fractions. The anticoagulant activities of the seven fractions were evaluated in terms of thrombin time, prothrombin time, and activated partial thromboplastin time. Among these fractions, the ethylacetate fraction showed the most potent anticoagulant activity. The active components in the ethylacetate fraction were identified as the phenolic compounds vanillic, caffeic, ferulic, and p-coumaric acid via TLC and HPLC. These findings suggest that the anticoagulant activities of phenolic compounds contribute to the cardiovascular effects of KRG.

In this study, the antioxidant and anti-skin aging properties of the Korean marine algae Ishige foliacea were investigated. Solvent extracts from I. foliacea were prepared with 70% ethanol, 80% methanol, and water. The extraction yields of various solvent extracts ranged from 9.55% to 35.12%. In terms of antioxidant activity, the ethanol extract showed the highest ABTS (2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt) radical scavenging activity, nitrite oxide scavenging activity, reducing power, and FRAP (ferric reducing antioxidant power). Regarding anti-skin aging activity, evaluation of the skin whitening and anti-wrinkle activities revealed that the methanol, water, and ethanol extracts possessed the highest tyrosinase (IC50=0.98 mg/mL), elastase (IC50=0.15 mg/mL), and collagenase (IC50=0.06 mg/mL) inhibitory activities, respectively. These results suggest that I. foliacea holds potential as an antioxidant and anti-skin aging substance in food and cosmetic materials.

We profiled the health-promoting bioactive components in nine types of soybean seeds with different seed coat colors (yellow, green, brown, and black) and investigated the effects of different extraction solvents (methanol, ethanol, and water) on their antioxidant activities. The carotenoid and anthocyanin compositions varied greatly by seed color, and the phenolic acids, total phenol, and total flavonoid contents differed by genotype. The carotenoid content was relatively higher in soybean seeds with green and black seed coats than in those with a yellow seed coat while lutein was the most plentiful. The anthocyanin content was considerably higher in the soybean seed with the black seed coat. The results of the DPPH assay showed strong antioxidative activities in the methanol- and water-extracts compared to the ethanol-extract, irrespective of the seed coat colors. Moreover, the soybean seeds with the black seed coat exhibited the highest antioxidant activity among the samples, regardless of the extraction solvent used. Eighteen bioactive compounds were subjected to data-mining processes including principal component analysis and hierarchical clustering analysis. Multivariate analyses showed that brown and black seeds were distinct from the yellow and green seeds in terms of the levels of carotenoids and anthocyanins, respectively. These results help our understanding of the compositional differences in the bioactive components among soybean seeds of various colors, providing valuable information for future breeding programs that seek to enhance the levels of compounds with health benefits.

In this study, seven species of seaweeds (Sargassum horneri, Grateloupia filicina, Kjellmaniella crassifolia, Porphyra tenera, Ecklonia stolonifera, Scytosiphon lomentaria, Agarum cribrosum) from Gangwon coast were collected for antioxidative assays. The methanol soluble and insoluble (water soluble) fraction which were prepared from 75% methanol extraction were assayed by DPPH method and deoxyribose assay. The antioxidative activity of methanol soluble fraction of Agarum, Scytosiphon and Sargassum reached to 26.60%, 24.28% and 23.40%, respectively. The antioxidative activities of methanol insoluble fractions of the Kjellmaniella and Porphyra were assayed to 29.70% and 21.01% which were higher than methanol soluble fractions with the values of 9.26% and 8.66%, respectively. The results from DPPH assay and deoxyribose assay showed the methanol soluble fractions of Agarum and Sargassum have strong antioxidative activity. However the methanol insoluble (water soluble) fractions of seaweeds did not show any meaningful antioxidant activity excepted Kjellmaniella and Porphyra.
Grayanotoxin-contaminated honey exhibits toxicity. In this study, a reliable and sensitive liquid-chromatography-tandem-mass-spectrometric method (LC-MS/MS) was developed and validated for the quantitation of grayanotoxin I and grayanotoxin III in honey. The grayanotoxins were extracted from honey via solid phase extraction and separated on a biphenyl column with a mobile phase consisting of 0.5 % acetic acid in water and methanol. Mass spectrometric detection was performed in the multiple-reaction monitoring mode with positive electrospray ionization. The calibration curve covered the range 0.25 to 100 ㎍/g. The intra- and inter-day deviations were less than 10.6 %, and the accuracy was between 94.3 and 114.0 %. The validated method was successfully applied to the determination of grayanotoxins in mad honey from Nepal. The concentrations of grayanotoxin I and grayanotoxin III in 33 out of 60 mad honey samples were 0.75 - 64.86 ㎍/g and 0.25 - 63.99 ㎍/g, respectively. The method established herein would help in preventing and confirming grayanotoxin poisoning.
Berberine is one of alkaloids extracted from Phellodendri Cortex or Coptidis Rhizoma and has extensively used as an antibacterial and antidiarrheal drug. In order to increase the bioavailability of berberine preparation, berberine pamoate was synthesized and investigated on its usage in vitro and in vivo. Berberine was more rapidly extracted from herbal plants by hot water extraction method than soxhlet extraction method. Berberine pamoate was easily synthesized from berberine hydrochloride and potassium pamoate solution and identified using the infrared spectrum. Quantitative analysis of berberine was possible in methanol solution by fluorometric determination. The dissolution rate of berberine pamoate was more decreased than that of berberine hydrochloride in simulated gastric fluid and simulated intestinal fluid. The remaining proportion of berberine pamoate in the small intestine of rat was maintained at high concentration for a long time as compared with that of berberine hydrochloride.

This study was designed to investigate the effects of antioxidant and antibacterial activities of water, ethanol and methanol extracts from root, stem and flower stalk of Hagocho (Prunella vulgaris). The solvent extract yields from root were higher than those from stem and flower stalk, and water extract showed the highest extraction yield against ethanol and methanol extracts. The contents of total phenolic and flavonoid in ethanol extract were significantly higher in stem extract compared with those of root and flower stalk. In the case of water and methanol extracts, however, the contents were the highest in flower stalk. The electron donating ability and reducing power in all test groups were significantly increased in a dose-dependent manner, and antioxidant activities were the highest in methanol extract. In extracts from different parts of Hagocho, the antioxidant activity was the highest in flower stalk followed by stem and root. ABTS radical scavenging ability of water and methanol extracts was above 50% at $100\;{\mu}g/mL$ concentration. Antibacterial activities did not show significant differences depending on parts of Hagocho. However, antibacterial activity of ethanol extract was higher than those of other extracts.

Liquid Chromatographic behavior of Ni(II) in Isonitrosoethylacetoacetate Imine(IEAA-NR), Ni(IEAA-NH)(IEAA-NR)(R = H, CH_3, C-2H_5, n-C_4H_9, C_6H_5-CH_2) chelates were investigated by reversed-phase HPLC on Micropak MCH-5 column using methanol/water as mobile phase. The optimum conditions for the separation of Ni(IEAA-NH)(IEAA-NR) chelates were examined with respect to the effect of the flow rate, sample solvent, mobile phase strength and column temperature. It was fo$und that metal chelates were properly eluted in an acceptable range of capacity factor value(0{\le}logk'{\le}1). The dependence of the logarithm of capacity factor (k') on the volume fraction of water in the binary mobile phase as well as on the liquid-liquid extraction distribution ratio (Dc) in methanol-water/n-alkane extraction system showed the good linearties, and the dependence of the logarithm of capacity factor (k') on the column temperature and on the enthalpy exhibited the good linearties, and the compensation temperature ({\beta}) from the slope was 773.47{\circ}K. It was suggested that the retention of metal chelates was largely affected by the hydrophobic effect.
Acanthoside-D, contained in Acanthopanax chilsanensis is known as a ginseng-like substance. This work was focused to set up analytical and preparative conditions for Acanthoside-D purification. The ethanol extract from the powder of the trunk of Acanthopanax chilsanensis was partitioned with hexane. A $\mu$-Bondapak $C_{18}$ (3.9$\times$300 mm, $10\mu\textrm{m}$) column was used to separate Acanthoside-D from the trunk of Acanthopanax chilsanensis. From the experimental results, the mobile phase used for isolating Acanthoside-D from the extract was water/acetonitrile/methanol=80/14/6 %(v/v). The flow rate of mobile phase was 1.0 mL/min, and UV wavelength was fixed at 210 nm. Finally on a semi-preparative column (3.9$\times$ 300 mm, $15\mu\textrm{m}$, Lichrospher 100RP-18) with the same mobile phase composition, the allowable maximum injection volume increased to 250 $\mu\ell$
